HETERODIMERIC PROTEINS AGAINST IL-12 RECEPTORFIELD OF THE INVENTION

[0001] This application generally relates to heterodimeric protein. More specifically, the application relates to a truncated IL-12 cytokine against IL-12 receptor and uses thereof, a masked IL-12 cytokines against IL-12 receptor and uses thereof and a nanobody binding to IL-12 cytokine and uses thereof.BACKGROUND OF THE INVENTION

[0002] Interleukin-12 (IL-12) is a pro-inflammatory cytokine that plays an important role in innate and adaptive immunity. IL-12 is a heterodimeric cytokine composing an alpha subunit, IL-12p35, and a beta subunit, IL-12p40. The IL-12 receptor is composed of IL-12Rβ1, which mainly interacts with IL-12p40 subunit, and IL-12Rβ2, which mainly interacts with IL-12p35 subunit.

[0003] IL-12 promotes Th1 polarization of CD4+ T cells and potentiates cytotoxicity of NK and CD8+ T cells. IL-12 induces master transcription factor Tbet to drive IFNγ-dependent anti-tumor immunity. Systemic administration of IL-12 has shown overwhelming efficacy in animal models of tumor. Therefore, IL-12 protein has been studied as a promising anti-cancer therapeutic. However, despite high expectations, early clinical studies did not yield satisfactory results [1] . Severe toxicity was observed in clinical trials with recombinant IL-12 administration.

[0004] Jung et al. constructed heterodimeric Fc-fused IL-12 which exhibited longer plasma half-life and enhanced anti-tumor immunity [2] . However, there is no evidence showing that the IL-12-Fc fusion protein is less toxic.

[0005] One strategy to improve the therapeutic window of IL-12 is to deliver it to tumor tissue via fusing IL-12 with tumor-targeting antibodies [3] . However, there is still a very large portion in the periphery tissues due to the high binding affinity of IL-12 to its receptors.

[0006] Another strategy is to mask IL-12 with its receptor or blocking antibodies [4] . The masking moiety is usually fused to IL-12 through a cleavable linker which is sensitive to proteases that are highly expressed in tumor tissue. The masked IL-12 could remain unreactive in peripheral tissues but become effective in tumor microenvironment. Nevertheless, their anti-tumor efficacy is dependent on the expression profile of tumor-specific proteases, which is generally heterogeneous in patients.

[0007] Glassman et al. constructed potency reduced IL-12 via introducing amino acid mutations to IL-12p40 subunit which interacts with IL-12Rβ1 [5] . It showed decreased toxicity in mouse models. However, introducing amino acid mutations may increase the risk of immunogenicity.

[0008] Thus, there is a need in the art for an improved therapeutic approach for using IL-12 to treat tumors.SUMMARY OF THE INVENTION

[0009] Cytokines can provide powerful therapy, but are accompanied by undesired effects that are difficult to control clinically and which have limited the clinical use of cytokines. IL-12 has been shown to have potent anti-tumor activity, however, its clinical application is limited by severe systemic toxicity. This disclosure relates to new forms of IL-12 cytokines that can be used in patients with reduced or eliminated undesired effects.

[0010] The present disclosure is directed to a method to decrease the toxicity of IL-12 via reducing binding affinity of IL-12p40 subunit with IL-12Rβ1. In some aspect, the method comprises deleting amino acids interacting with IL-12Rβ1 to decrease the binding affinity of IL-12 to its receptors. In some aspect, the method comprises truncating part of N-terminal, C-terminal or middle amino acids of IL-12p40 subunit to decrease the binding affinity of IL-12 to its receptors. In some aspect, the method comprises masking the p40 subunit of IL-12 by a masking moiety to decrease the binding affinity of IL-12 to its receptors. In some aspect, the method comprises masking the p40 subunit of IL-12 by a nanobody to decrease the binding affinity of IL-12 to its receptors. By using a masking moiety or truncated IL-12p40 subunit, the systemic side effects of an administered IL-12 cytokine or functional fragment thereof can be reduced by interfering with the binding capability of the IL-12 cytokine or functional fragment thereof to its cognate receptor.

[0013] The present invention provides novel truncated IL12-Fc fusion proteins with decreased toxicity, longer half-life and lower immunogenicity.

[0332] The six-well plate was pre-coated with 100 ng / ml OKT3 overnight at 4℃. PBMCs were freshly isolated from healthy donors and adjusted to 1×106 cells / mL in RPMI-1640 complete medium supplemented with 50 U / mL recombinant human IL-2. The PBMCs were then added into the OKT3-coated plate and stimulated at 37℃, 5%CO2 for 3 days. After incubation, the PBMCs were collected by centrifuging at 300g for 5 min at 25℃ and adjusted to 1×106 cells / mL in RPMI-1640 complete medium supplemented with 50 U / mL recombinant human IL-2. The PBMCS were then added into a 96-well plate at 100,000 cells / well and stimulated with a dilution series of IL-12 proteins (10-fold serially diluted from 2000nM to 1×10-6nM) . The plates were kept in an incubator set to 37℃ and 5%CO2 for 5 days. Afterward, the supernatants were harvested for IFN-γ measurement by ELISA.

[0333] Human IFN-γ release was measured by ELISA using matched antibody pairs. Recombinant human IFN-γ was used as standards. The two-fold serial concentrations from 8 ng / mL to 0.0078 ng / mL of IFN-γ were used for standard curve. The plates were pre-coated with capture antibody specific for human IFN-γ at a concentration of 0.75 μg / mL. After blocking with 1×PBS / 2%BSA for 2 hours, 50 μL of standards or samples were added into each well and incubated for 2 hours at ambient temperature. Following removal of the unbound substances by washes, the biotin-conjugated IFN-γ detecting antibody was diluted to 0.5 μg / mL in 1×PBS / 2%BSA, added into the wells (50 μL / well) and incubated for 1 hour at ambient temperature. The plates were washed 3 times after incubation. Streptavidin-HRP (1: 20000) was then added into the plates and incubated for 0.5 hour at ambient temperature. After washing 6 times, the color was developed by dispensing 50 μL of TMB substrate, and then stopped by 50 μL of 2M HCl. Absorbance was read at 450 and 540 nm using a Microplate spectrophotometer (SpectraMax M5e) , followed by four-parameter non-linear regression analysis using GraphPad Prism 5 software. The concentration of IFN-γ in supernatant was quantitated according to the standard curve.
