Abstract

The present disclosure provides a TL1A antibody or antigen-binding fragments thereof. The present disclosure provides an antibody against TNF-like ligand 1a (TL1A) or antigen-binding fragments thereof, wherein the antigen-binding protein or active fragment thereof comprises at least one light chain variable region and at least one heavy chain variable region. TL1A ANTIBODY AND USE THEREOFFIELD OF THE INVENTION

[0001] The present disclosure belongs to the field of biotechnology, specifically relates to TL1A antibodies or antigen-binding fragments thereof and use thereof.BACKGROUND OF THE INVENTION

[0002] TNF-like ligand 1a (TL1A) , one of the members of the tumor necrosis factor superfamily, is a type II membrane protein that is expressed by antigen-presenting cells (including dendritic cells, B cells, and macrophages) , CD4+ and CD8+ T cells, and endothelial cells and can be expressed on the cell surface or secreted as soluble cytokines.

[0003] TL1A mediates signaling pathways that promote the secretion of pro-inflammatory cytokines or promote apoptosis by binding to DR3. Depending on the cellular environment, the ligation of TL1A to DR3 can trigger one of two signaling pathways: activation of the transcription factor NF-kB or activation of caspase and apoptosis. TL1A has been involved as a potential therapeutic target for a variety of diseases and conditions. Currently, there is no medicament for this target on the domestic or foreign market.

[0004] Therefore, there is an urgent need for therapeutic agents and methods targeting TL1A.SUMMARY OF THE INVENTION

[0005] In order to address at least one of the above issues, the present disclosure provides an antibody or antigen-binding fragment thereof that specifically recognizes and / or binds to TNF-like ligand 1a, and use thereof. The antibodies or antigen-binding fragments thereof provided by the present disclosure have good affinity and biological activity and have broad application prospects in the treatment or prevention of TL1A-related diseases.

[0140] Example 7. Transient expression, purification and identification of IgG4 humanized antibody

[0141] ExpiCHO-STM cells with a density of 2×106 cells / ml were configured with culture medium, placed in erlenmeyer flasks, and cultured at 100 rpm, 37℃ and 5%CO2 for 20-25 h, and only when the cell density reached about 5×106 cells / ml, and the cells were in the logarithmic growth phase with a viability of more than 95%, they could be used for transfection experiments. The recombinant plasmid expressing the humanized antibody was added to Trans Buffer A (Medium Bank) and gently mixed and added with PEI to mix gently. The diluted PEI solution was added to the corresponding diluted plasmid, mixed gently and incubated at room temperature for 5 min. The mixture was added into an erlenmeyer flask containing ExpiCHO-STM cells and cultured in a shaker at 100 rpm, 37℃, and 5%CO2. After 24 h post-transfection, the transfected cells were cultured in a shaker at 100 rpm, 32℃, and 5%CO2. After 10-14 d post-transfection, the cell cultures were collected by centrifugation at 3000 rpm for 15 min. The supernatant obtained was subjected to affinity chromatography using a HiTrap MabSelect SuRe prepacked column. The column was rinsed with pure water and equilibrated with PB buffer, then the supernatant was loaded onto the chromatography column for binding. After loading, the column was rinsed with PB buffer to the baseline, and the protein was eluted with 0.1 M citrate eluent. After neutralizing with 1 M Tris-HCl, the eluted protein was concentrated appropriately and loaded onto the Superdex200 well equilibrated with PBS for further purification. The collected protein was concentrated to a certain concentration, and subjected to SDS-PAGE electrophoresis for identification, and the results are shown in Figure 1A and Figure 1B, and subjected to SEC-HPLC, and the results are shown in Table 6. The IgG4 humanized antibodies, corresponding parental hybridoma antibodies and anti-TL1A positive control antibodies were simultaneously tested for affinity and biological activity, and the results are shown in Table 6. The biological activity and affinity of 2G11-1 and 41E10-1 humanized antibodies were enhanced, and the biological activity was higher than that of the positive control antibodies.

[0143] Example 8. Relative binding activity of IgG4 humanized antibodies

[0144] ELISA was used to detect binding ability of 2G11-1-3, 41E10-1-3 and anti-TL1A positive control antibody PC (PRA023) to human TL1A. The human TL1A was diluted to 3 μg / ml, and at 100 μL / well, coated onto the wells at 4℃ overnight. On the following day, the plates were washed and blocked with 5%skimmed milk for 2h. Then the plate was washed and the antibody samples with different concentration gradients were added and incubated at 37℃ for 1h. The plates were washed, 100 μL of HRP-labeled goat anti-human secondary antibody was added to each plate, and the plates were incubated at 37℃ for 30 min. TMB substrate solution was then added for color development at 37℃ for 5 minutes, followed by the termination of the reaction with the addition of 50 μL of H2SO4, and the OD450 values were subsequently read using a TECAN multifunctional microplate reader, and the data were also plotted using Prism 5 (GraphPad) software and the results are shown in Figure 2A and Figure 2B. 2G11-1-3, 41E10-1-3 humanized antibodies have good binding activity to hTL1A.

[0145] Example 9. Evaluation of the in vitro activity of IgG4 humanized antibody

[0146] Firstly, fresh anticoagulant blood was diluted 1: 1 with PBS, and the diluted blood samples were slowly added to the top of lymphocyte separation medium Ficoll-Paque, centrifuged to collect the PBMCs. The PBMCs were washed twice with sterile PBS, and then resuspended in 1640 medium (Gibco) containing three cytokines, IL12, IL15, and IL18, and cultured in an incubator for 48h. Human TL1A was diluted and the antibody to be tested was diluted in different concentration gradients and added into 96-well plates at the same time with PBMC cells cultured for 48h, and incubated in 37℃, 5%CO2 incubator for 48 hours. The levels were then quantified using the Solarbio Human INF-γ kit and plotted using Prism 5 (GraphPad) software, and the results are shown in Figure 3. The data showed that both 2G11-1-3 and 41E10-1-3 humanized antibodies had good in vitro activity.

[0147] Example 10. Non-specific detection of IgG4 humanized antibodies with TL1A homologs (TNF family factor)

[0148] Non-specificity of 2G11-1-3 and 41E10-1-3 with different TNF family factors (TL1A homologs) was detected by ELISA, respectively. Human TL1A (TNFSF15) , TNFSF10, TNFSF6, TNFSF14, TNFα, and TNFβ (SinoBiological) were all diluted to 3 μg / ml, and at 100 μL / well, coated onto the wells at 4℃ overnight. On the following day, the plates were washed and blocked with 5%skimmed milk for 2h. Then the plate was washed and the antibody samples with different concentration gradients were added and incubated at 37℃ for 1h. The plates were washed, 100 μL of HRP-labeled goat anti-human secondary antibody was added to each plate, and the plates were incubated at 37℃for 30 min. TMB substrate solution was then added for color development at 37℃ for 5 minutes, followed by the termination of the reaction with the addition of 50 μL of H2SO4, and the OD450 values were subsequently read using a TECAN multifunctional microplate reader, and the data were also plotted using Prism 5 (GraphPad) software and the results are shown in Figure 4A and Figure 4B. Although TNFSF6 (FAS ligand) , TNFSF10 (TRAIL) , TNFSF14 (LIGHT) , TNF-β, and TNF-α had 36%, 35%, 31%, 30%, and 29%homology with TL1A, respectively, 2G11-1-3 or 41E10-1-3 IgG4 humanized antibodies did not non-specifically bind to these TL1A homologs, suggesting that the humanized antibodies had better specific binding to TL1A.

[0149] Example 11. Cross-reactivity of IgG4 humanized antibody with other species of TL1A

[0150] The cross-reactivity of IgG4 molecules with other species of TL1A was determined using the BLI technique. The 2G11-1-3 and 41E10-1-3 IgG4 humanized antibodies were first diluted to 15 nM and absorbed onto the ProA probe at 400 rpm  / min. Different species of TL1A were serially diluted from 100nM by doubling dilution to 5 concentration gradients. The program was set up so that the diluted TL1A bound to probes with attached 2G11-1-3 and 41E10-1-3 humanized antibodies, respectively for a fixed period of time. After binding was complete, the probes with bound complex were then transferred to analyte-free Q buffer to dissociate the bound analytes. KD values were obtained by fitting the data using the built-in software of the Gator instrument. The results are shown in Table 7.2G11-1-3 IgG4 humanized antibody did not bind to mouse TL1A and bound strongly to monkey and rat TL1A; 41E10-1-3 bound weakly to mouse TL1A and strongly to monkey and rat TL1A.

[0152] Example 12. Expression, purification and identification of IgG1 humanized antibody

[0153] The heavy chain constant region and Fc region of the IgG4 humanized molecule obtained in Example 6 were replaced with the heavy chain constant region and Fc region of IgG1. The modified 2G11-1-3 and 41E10-1-3 IgG1 humanized antibodies (with the heavy chain amino acid sequences SEQ ID NO: 134 and 135, respectively) were obtained, expressed, purified (see Example 7 for steps) and identified. Purity, isoelectric point, and affinity were assayed, respectively, and the results are shown in Table 8. The relative binding activity is shown in Figure 5, and the biological activity is shown in Figures 6A to 6C. The data showed that the biological activity and affinity of the IgG1 humanized antibody did not change significantly.

[0155] Example 13. Thermal Stability Analysis of IgG1 Humanized Antibody

[0156] The thermal stability of IgG1 humanized antibodies was analyzed by the Institute of Process Engineering, Chinese Academy of Sciences, by means of a MicrolCalTM VP-DSC system (Malvern, UK) , based on the principle of differential scanning calorimetry (DSC) . The samples and buffers were degassed by a vacuum pump for 5 min and then assayed. The instrument parameters were set as follows: heating rate: 60℃ / h; temperature range: 25-95℃; feedback mode: none. Sample spectra were corrected by subtracting buffer spectrum, and data analysis was performed by the MicrolCal Origin 7.0 (Origin-Lab Corp., MA) software provided with the system. The results are shown in Table 9 and Figure 7. Tm1 is the melting temperature at which the interface of the two CH2 domains melts, and Tm2 is the melting temperature at which the interface of the heavy and light chains melts. The data show that the Tm1 > 65℃ for 2G11-1-3 IgG1 and 41E10-1-3 IgG1 humanized antibodies, and Tm2 corresponding to the Fab domain > 80℃, and the antibodies are relatively stable.

[0159] Bio-Layer Interferometry (BLI) was used to determine Fc Function of IgG1 Humanized Antibodies.

[0160] (1) Affinity assay of IgG1 humanized antibodies with FcγR. The hFcγRI (NP_000557.1) , hFcγRIIa (131H) (P12318-1) , hFcγRIIa (131R) (P12318-1) , hFcγRIIb (NP_001002274.1) , hFcγRIIIa (158F) (P08637-1) , hFcγRIIIa (158V) (P08637-1) , hFcγRIIIa (158V) (P08637-1) ) fusion proteins were labeled with a biotin labeling kit. These labeled proteins were then diluted to a concentration of 60 nM and loaded onto the SA probe at 400 rpm / min, and then the diluted IgG1 humanized antibodies with different gradients were conjugated to the SA probe attached with FcγR. The probes bound with antigen-antibody complexes were placed into the analyte-free Q buffer to dissociate the bound analytes, and the affinity values were obtained by fitting with the software provided with the instrument and the results are shown in Table 10. The results showed that the IgG1 humanized antibody only bound weakly to FcγRI and not to other FcγR, indicating that the ADCC function of the humanized antibody was greatly reduced.

[0161] (2) Affinity assay of IgG1 humanized antibody with FcRn (Acrobiosystems) at different pH buffer systems. The hRcRn was labeled with biotin labeling kit, diluted to a concentration of 25 nM, loaded onto the SA probe, and then reacted with IgG1 humanized antibodies of different gradients under pH 6.0 and pH 7.4 buffer systems, respectively, and then dissociated under the buffer of the corresponding pH, and the affinity values were obtained by fitting with the software provided with instrument, and the results are shown in Table 10. The results showed that the humanized antibody bound to FcRn only at pH 6.0 and not at pH 7.4.

[0162] (3) Affinity assay of IgG1 humanized antibody with C1q (Quidel) . The IgG1 humanized antibody and the positive control antibody was labeled simultaneously with a biotin labeling kit, diluted to a concentration of 10 μg / ml, loaded onto the SA probe, and then conjugated to diluted C1q of different gradients. The probes bound with antigen-C1q complexes were placed into the analyte-free Q buffer to dissociate the bound analytes, and the affinity values were obtained by fitting with the software provided with the instrument and the results are shown in Table 10. The results showed that the humanized antibody did not bind to C1q, suggesting that mutations in the corresponding binding site on Fc reduced the antibody CDC function.

[0165] The IgG1 humanized antibody and Rituximab were added to a 96-well plate at 200 μg / ml as the first well concentration, following serial 5-fold dilutions across 8 gradients, respectively. The HEK293 / human TL1A target cells were resuspended with medium and inoculated into 96-well plates at a density of 2×104 cells per well. The plates were gently mixed and incubated for half an hour at room temperature. The PBMC cells were washed with medium and resuspended, then at 2×l05 cells / well plated into the corresponding 96-well plates and incubated in the incubator for a total of 6 hours. The 96-well plate was centrifuged at 250g for 10 minutes, and 100μL of the supernatant was aspirated and transferred into a new 96-well transparent flat-bottom plate. Each well was then filled with 100μL of working solution and incubated in the dark at room temperature for 30 minutes. The absorbance at dual wavelengths of 490nm and 620nm were detected on the multifunctional microplate reader to calculate the cell killing rate, and the data were plotted using Prism 5 (GraphPad) software. The results are shown in Figure 8. Rituximab showed an ADCC effect, and neither of the two IgG1 humanized antibodies, 41E10-1-3 and 2G11-1-3, had the ADCC effect.

[0166] Example 16. CDC Assay of IgG1 Humanized Antibody

[0167] The IgG1 humanized antibody and Rituximab were added to a 96-well plate at 1000 μg / ml as the first-well concentration, following serial 5-fold dilutions across 8 gradients, respectively. The treated HEK293 / human TL1A target cells were resuspended with DPBS and inoculated into 96-well plates at 5×104 cells / well. The cell culture plate was incubated at room temperature for 30 min, and then incubated for 16 h in a 37℃, 5%CO2 incubator after adding 10%guinea pig serum (10 μL) . Then 100 μL of Cell Titer-Glo reagent was added to each well, gently mixed, and the chemiluminescence signals were detected on a multifunctional microplate reader, and the data were plotted using Prism 5 (GraphPad) software, and the results are shown in Figure 9. Rituximab showed a CDC effect, and neither of the two IgG1 humanized antibodies, 41E10-1-3 and 2G11-1-3, had the CDC effect.

[0168] Example 17. Stability Assay of IgG1 Humanized Antibody

[0169] Stability tests were performed on two IgG1 humanized antibodies, 2G11-1-3 and 41E10-1-3, each set at two concentrations of 30 mg / ml and 2 mg / ml. The high concentration samples were subjected to accelerated stability testing for 0 week, 1 week, 2 weeks, and 4 weeks at 40℃. The low concentration samples were divided into two groups; one group was subjected to repeated freeze-thaw cycle treatments of 3, 5, and 10 times, while the other group was subjected to accelerated treatments for 0 week, 1 week, 2 weeks, and 4 weeks at 40℃. The prepared samples were assayed for protein content, purity, isoelectric point, charge heterogeneity, reduced and non-reduced CE-SDS, biological activity and relative binding activity to evaluate the stability of IgG1 humanized antibody. The results are shown in Tables 11, 12 and 13. Taken together all the data, it was shown that both IgG1 humanized antibodies, 2G11-1-3 and 41E10-1-3, are relatively stable under freeze-thaw conditions.

[0173] Example 18. Preventive and Therapeutic Effects of IgG1 Humanized Antibody on a Rat Model of DSS-Induced Ulcerative Colitis

[0174] (1) Animal selection and grouping, modeling, and administration protocols. Twenty-four male SD rats (6-7 weeks old) were randomly divided into four groups according to body weight: G1 was a healthy control group, G2 was a model control group, G3 was a low-dose group of TL1A antibody (41E10-1-3 IgG1) (5 mg / kg, twice a week, s. c. ) , and G4 was a high-dose group of TL1A antibody (41E10-1-3 IgG1) (50 mg / kg, twice a week, s. c. ) , 6 animals in each group. Animals were acclimatized for 3 days and then randomly grouped according to their body weights. Animals in the modeling groups began drinking water supplemented with dextrose sodium sulfate (DSS) at a concentration of 5% (wt / vol) on day 1 of the test (D1) for 7 days (defining days 1-8 as the DSS period) , to induce the colitis model. The rats were euthanized on D8. The corresponding drugs or D-PBS were injected subcutaneously before and after modeling on D1, D4, and D7 for a total of 3 times.

[0175] (2) Detection indicators

[0176] a. General clinical observations. During the test period, general clinical observations were conducted twice daily on all surviving animals in each of the above groups, monitoring their health status and mortality. Routine examinations encompassed the appearance and physical signs, mental state, behavioral activities, food and water intake, fecal property, weight changes, and any other abnormalities of the test subjects. Special attention was paid to conditions such as weight loss, lethargy, disheveled fur, reduced activity levels, diarrhea, perianal filthy, fecal shape, and fecal occult blood.

[0177] b. Body weight. Body weights were determined for each of the above groups of animals: one weighing before grouping, one daily weighing during the test period, one weighing before euthanasia, and one weighing of any of unexpectedly dead animals.

[0178] c. Fecal property and degree of hematochezia. The fecal property and degree of hematochezia were scored once daily from the day of modeling (D1) for the surviving animals in each of the above groups. The rules are as follows: fecal property: 0 = normal; 1 = Soft but formed feces; 2 = Soft unformed feces; 3 = Loose feces; 4 = watery feces; degree of hematochezia: 0 = normal; 1=positive for occult blood + to ++; 2=positive for occult blood +++ to ++++; 3=blood streaks visible in feces; 4=blood in the feces visible to the naked eye.

[0179] d. Colon length and weight measurement. Before dissection, the rats were required to fast for at least 4 h, euthanized, and the abdominal cavity was rapidly opened to remove colorectal tissues. The tissue was measured for length, weighed, scored for colorectal mucosa, and photographed. During the test, near-death animals and animals found dead were subjected to gross anatomy.

[0180] e. Splenic index analysis. After euthanasia of the animal, the abdominal cavity was quickly dissected, the spleen was removed and weighed for wet weight using an analytical balance after removing the surrounding adherent fat and connective tissue to calculate the organ coefficient.

[0182] (3) Statistical analysis. Measurements data are described using mean and standard deviation (Mean±SD) . Graphs were plotted and statistically analyzed using GraphPad Prism 9 software in the following process: the data were subjected to the Homogeneity of Variance test; the data with homogeneity of variance (P>0.05) were subjected to the One-way ANOVA, and the data that were tested to be different (P≤0.05) were further analyzed using LSD multiple comparison analysis; the data without he Homogeneity of Variance (P≤0.05) were subjected to Kruskal-Wallis nonparametric test, and the data that were tested to be different (P≤0.05) were subjected to pairwise comparisons using Mann-Whitney two-by-two comparative analysis, and P≤0.05 was considered to be significantly different. The intergroup differences in the assays between the healthy control group and the model control group, and between the model control group and each of the administering groups were mainly examined.

[0184] a. Animal survival and diarrhea incidence. By the endpoint of the test on D8, a total of three animals in the G2 model group had died, and no animals in the remaining groups had died. By the endpoint of the test on D8, the diarrhea rates of animals in the healthy control group (G1) , the model control group (G2) , 41E10-1-3 low dose group (G3) , and 41E10-1-3 high dose group (G4) were 0%, 100%, 50%, and 100%, respectively. The rates of diarrhea for animals in each of groups at D7 were 0%, 100%, 33.33%, 33.33%, respectively. The rates of diarrhea in animals in each of groups at D6 were 0%, 83.33%, 33.33%, 50%, respectively. Table 14 shows animal survival and diarrhea. According to the results in Table 14, prophylactic administration of 41E10-1-3 at 5 mg / kg injected twice weekly reduced mortality and diarrhea in rats with DSS-induced colitis.

[0186] b. Body Weight. The changes in body weight and the trend of body weight difference of the animals in the four test groups are detailed in Figures 10 and 11. Figure 10 shows the effect of 41E10-1-3 on body weight in a SD rat model with DSS-induced colitis. As can be seen from Figure 10, there was a fluctuating trend of increasing body weight in healthy control (G1) animals during the test period. The body weight of animals in the modeling groups (G2-G4) increased slowly from the 3rd day of DSS drinking (D4) , and was significantly lower than that of the G1 group at D7 and D8 (P<0.05) , and the difference in body weights was highly significant lower than that of the G1 group (P<0.001) ; among them, the body weights of the 41E10-1-3 high dosage group (G4) declined more slowly, and the degree of body weight loss was lower than that of the model control group (G2) , and the body weight of the 41E10-1-3 low dose group (G4) decreased more slowly at D7 and D8, and the degree of body weight loss was lower than that in the model control group (G2) . According to the results of Figures 10 and 11, 41E10-1-3 can improve the body weight loss in rats with the DSS-induced ulcerative colitis.

[0187] c. Scores of fecal property and degree of hematochezia. The statistical results of the scores of fecal property and degree of hematochezia in the four groups of animals are shown in Figures 12 and 13. Figure 12 shows the effect of 41E10-1-3 on fecal property scores in a SD rat model with DSS-induced colitis. Figure 13 shows the effect of 41E10-1-3 on fecal occult blood scores in a DSS-induced colitis model in SD rats. It can be seen from the results shown in Figures 12 and 13 that, compared to the healthy control group (G1) , the model control group (G2) and the administering groups (G3, G4) exhibited clinical symptoms associated with colitis, such as soft feces and positive fecal occult blood, starting from the second day of drinking DSS water (D3) . The clinical scores continuously increased, and by D4, the fecal property and fecal scores were significantly higher than those of the healthy control group (G1) . The fecal property score of animals in the 41E10-1-3 low-dose group (G3) from D5 to D8 was milder than that of the model control group (G2) , and the fecal occult blood condition of animals in the 41E10-1-3 low-dose group (G3) from D6 to D8 was milder than that of the model control group (G2) . According to the results in Figures 12 and 13, the 41E10-1-3 low-dose group can ameliorate diarrhea and fecal occult blood in rats with the DSS-induced ulcerative colitis.

[0188] d. Colon length and weight. In this test, the colon tissue of each group of animals was assessed at the test endpoint (D8) . The colon length is shown in Figure 14, and the colon weight is shown in Figure 15. Figure 15 shows the effect of 41E10-1-3 on colon weight in a SD rat model with DSS-induced colitis. Figure 14 shows the effect of 41E10-1-3 on colon length in a SD rat model with DSS-induced colitis. As can be seen from Figures 14 and 15, the colon length and weight of the animals in the model control group (G2) were significantly shortened compared to the healthy control group (G1) ; the colon length and weight of the animals in the 41E10-1-3 low-dose group (G3) were somewhat increased compared to the model control group (G2) . According to the results in Figure 14 and Figure 15, 41E10-1-3 low dose can ameliorate colon shortening in rats with DSS-induced ulcerative colitis and had a preventive effect on colon shortening in rats with DSS-induced colitis.
