Racemic mixture 14a-P2 (71 mg, 0.2 mmol) of (2R,3S,4S,5R)-3-(3,4-difluoro-2-methoxyphenyl)-4,5-dimethyl-5-(trifluoromethyl)tetrahydrofuran-2-carboxylic acid was added, along with anhydrous N,N-dimethylformamide (0.0025 mL) and anhydrous dichloromethane (1 mL). The mixture was cooled to 0-5°C under argon protection, and a solution of oxaloyl chloride (51 mg, 0.4 mmol) in anhydrous dichloromethane (0.2 mL) was slowly added dropwise, producing numerous bubbles. The reaction proceeded at room temperature for 2 hours, followed by direct post-processing and concentration. The crude product was then added to tert-butanol (1 mL), 2-amino-5-fluorobenzamide (31 mg, 0.2 mmol), and potassium tert-butoxide (49 mg, 0.4 mmol). The mixture was heated to 95°C under argon protection and reacted overnight. LC-MS showed that the starting material had almost completely reacted. Acetonitrile (about 1 mL), a small amount of water and 1 drop of acetic acid were added to the reaction solution, and enantiomer 59 (13.21 mg, yield: 14.5%) was prepared directly by high performance liquid chromatography in acidic conditions as a white solid.
