FIELD OF THE INVENTION The present invention relates to antibacterial compounds, more particularly to 5- substituted 1H-pyrazolo[4,3-d]pyrimidines which act as Staphylococcus species (spp) inhibitors specifically Staphylococcus aureus. BACKGROUND OF THE INVENTION Staphylococcus aureus is a human pathogen that can cause a range of illnesses, from minor skin infections to life-threatening diseases, such as pneumonia, meningitis, endocarditis, toxic shock syndrome (TSS), bacteremia, and sepsis. It is one of the most common causes of nosocomial infections and is often the cause of postsurgical wound infections. Methicillin-resistant S. aureus (MRSA) strains and Vancomycin Intermediate Resistant Staphylococcus aureus (VISA), which have become resistant to most antibiotics, are most often found associated with institutions such as hospitals, but they are also becoming increasingly prevalent in community-acquired infections The danger of MRSA and VISA infections result from not only the emergence of multidrug resistance but also the occurrence of strong biofilm-forming bacteria. Linezolid is one of the powerful antibiotics and during the course of time, Linezolid started getting resistant resulting different Linezolid resistant microbes especially Staphylococcus spp. Thus, there is a continuing need to develop newer and more potent antibiotics. Particularly, there is a need to combat drug-resistant bacterial infections, which present a significant threat to public health globally. More specifically, there is an urgent requirement for antibiotics capable of overcoming the challenges posed by Linezolid-resistant Staphylococcus spp. and other multidrug-resistant pathogens. DETAILED DESCRIPTION OF THE INVENTION Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by those of ordinary skill in the art to which the invention belongs. Although any methods and materials similar or equivalent to those described herein can be used in the practice or testing of the present invention, preferred methods and materials are described. For the purposes of the present invention, the following terms are defined below. The articles "a" and "an" are used herein to refer to one or to more than one (i.e. to at least one) of the grammatical object of the article. By way of example, "an element" means one element or more than one element. The term "alkyl" refers to a straight or branched chain saturated aliphatic hydrocarbon that may be substituted or unsubstituted. In certain embodiments, the alkyl is C1-C6 alkyl. As used herein, the term "cycloalkyl" used herein, either alone or in combination with other radicals, denotes mono, bicyclic, or polycyclic saturated, partially saturated hydrocarbon ring system of about 3 to 12 carbon atoms which may be substituted or unsubstituted. Exemplary "cycloalkyl" groups include but are not limited to cyclopopyl, cyclobutyl, cyclopentyl, cyclohexyl and cycloheptyl, perhydronapthyl, adamantyl, noradamantyl and spirobicyclic groups such as spiro (4,4)non-2-yl. The term "halogen" or “halo” refers to fluorine, chlorine, bromine, or iodine. As used herein, the term "heteroaryl" refers to monocyclic aromatic ring systems or fused bicyclic aromatic ring systems comprising two or more aromatic rings, preferably two to three ring systems. The present disclosure also provides methods for formulating the disclosed compounds for pharmaceutical administration. The compositions and methods of the present invention may be utilized to treat a subject in need thereof. In certain embodiments, the subject is a mammal such as a human or a non-human mammal. When administered to an animal, such as a human, the composition or the compound is preferably administered as a pharmaceutical composition comprising, for example, a compound of the invention and a pharmaceutically acceptable carrier. Pharmaceutically acceptable carriers are well known in the art and include, for example, aqueous solutions such as water or physiologically buffered saline or other solvents or vehicles such as glycols, glycerol, and oils such as olive oil or injectable organic esters. Formulations of the pharmaceutical compositions for administration to the mouth may be presented as a mouthwash or an oral spray or an oral ointment. Alternatively, or additionally, compositions can be formulated for delivery via a catheter, stent, wire, or other intraluminal device. Delivery via such devices may be especially useful for delivery to the bladder, urethra, ureter, rectum, or intestine. Formulations which are suitable for vaginal administration also include pessaries, tampons, creams, gels, pastes, foams, or spray formulations containing such carriers as are known in the art to be appropriate. Dosage forms for the topical or transdermal administration include powders, sprays, ointments, pastes, creams, lotions, gels, solutions, patches, and inhalants. The active compound may be mixed under sterile conditions with a pharmaceutically acceptable carrier and with any preservatives, buffers or propellants that may be required. The ointments, pastes, creams, and gels may contain, in addition to an active compound, excipients, such as animal and vegetable fats, oils, waxes, paraffins, starch, tragacanth, cellulose derivatives, polyethylene glycols, silicones, bentonites, silicic acid, talc and zinc oxide or mixtures thereof. Sprays can additionally contain customary propellants, such as chlorofluorohydrocarbons and volatile unsubstituted hydrocarbons, such as butane and propane. Transdermal patches have the added advantage of providing controlled delivery of a compound of the present invention to the body. Such dosage forms can be made by dissolving or dispersing the active compound in the proper medium. Absorption enhancers can also be used to increase the flux of the compound across the skin. The rate of such flux can be controlled by either providing a rate controlling membrane or dispersing the compound in a polymer matrix or gel. Ophthalmic formulations, eye ointments, powders, solutions, and the like, are also contemplated as being within the scope of this invention. If desired, liquid ophthalmic formulations have properties similar to that of lacrimal fluids, aqueous humor or vitreous humor or are compatable with such fluids. A preferred route of administration is local administration (e.g., topical administration, such as eye drops or administration via an implant). The phrases "parenteral administration" and "administered parenterally" as used herein means modes of administration other than enteral and topical administration, usually by injection and includes, without limitation, intravenous, intramuscular, intraarterial, intrathecal, intracapsular, intraorbital, intracardiac, intradermal, intraperitoneal, transtracheal, subcutaneous, subcuticular, intraarticular, subcapsular, subarachnoid, intraspinal and intrasternal injection and infusion. Pharmaceutical compositions suitable for parenteral administration comprise one or more active compounds in combination with one or more pharmaceutically acceptable sterile isotonic aqueous or nonaqueous solutions, dispersions, suspensions or emulsions or sterile powders which may be reconstituted into sterile injectable solutions or dispersions just prior to use, which may contain antioxidants, buffers, bacteriostats, solutes which render the formulation isotonic with the blood of the intended recipient or suspending or thickening agents. Examples of suitable aqueous and nonaqueous carriers that may be employed in the pharmaceutical compositions of the invention include water, ethanol, polyols (such as glycerol, propylene glycol, polyethylene glycol and the like) and suitable mixtures thereof, vegetable oils, such as olive oil and injectable organic esters, such as ethyl oleate. Proper fluidity can be maintained, for example, by the use of coating materials, such as lecithin, by the maintenance of the required particle size in the case of dispersions and by the use of surfactants. These compositions may also contain adjuvants such as preservatives, wetting agents, emulsifying agents, and dispersing agents. Prevention of the action of microorganisms may be ensured by the inclusion of various antibacterial and antifungal agents, for example, paraben, chlorobutanol, phenol sorbic acid and the like. It may also be desirable to include isotonic agents, such as sugars, sodium chloride and the like into the compositions. In addition, prolonged absorption of the injectable pharmaceutical form may be brought about by the inclusion of agents that delay absorption such as aluminium monostearate and gelatin. In some cases, in order to prolong the effect of a drug, it is desirable to slow the absorption of the drug from subcutaneous or intramuscular injection. This may be accomplished by the use of a liquid suspension of crystalline or amorphous material having poor water solubility. Various slow-release polymeric devices have been developed and tested in vivo in recent years for the controlled delivery of drugs, including proteinaceous biopharmaceuticals. A variety of biocompatible polymers (including hydrogels), including both biodegradable and non-degradable polymers, can be used to form an implant for the sustained release of a compound at a particular target site. Actual dosage levels of the active ingredients in the pharmaceutical compositions may be varied so as to obtain an amount of the active ingredient that is effective to achieve the desired therapeutic response for a particular patient, composition, and mode of administration, without being toxic to the patient. The selected dosage level will depend upon a variety of factors including the activity of the particular compound or combination of compounds employed or the ester, salt or amide thereof, the route of administration, the time of administration, the rate of excretion of the particular compound(s) being employed, the duration of the treatment, other drugs, compounds and / or materials used in combination with the particular compound(s) employed, the age, sex, weight, condition, general health and prior medical history of the patient being treated and like factors well known in the medical arts. A physician or veterinarian having ordinary skill in the art can readily determine and prescribe the therapeutically effective amount of the pharmaceutical composition required. For example, the physician or veterinarian could start doses of the pharmaceutical composition or compound at levels lower than that required in order to achieve the desired therapeutic effect and gradually increase the dosage until the desired effect is achieved. By "therapeutically effective amount" is meant the concentration of a compound that is sufficient to elicit the desired therapeutic effect. It is generally understood that the effective amount of the compound will vary according to the weight, sex, age, and medical history of the subject. Other factors which influence the effective amount may include, but are not limited to, the severity of the patient's condition, the disorder being treated, the stability of the compound and, if desired, another type of therapeutic agent being administered with the compound of the invention. In general, a suitable daily dose of an active compound used in the compositions and methods of the invention will be that amount of the compound that is the lowest dose effective to produce a therapeutic effect. Such an effective dose will generally depend upon the factors described above. If desired, the effective daily dose of the active compound may be administered as one, two, three, four, five, six or more sub-doses administered separately at appropriate intervals throughout the day, optionally, in unit dosage forms. In yet another embodiment of the present invention, the active compound may be administered two or three times daily. In preferred embodiments, the active compound will be administered once daily. It is understood that in any of the above schemes, any reactive group in the substrate molecule may be protected according to any conventional procedure known in the prior art. Suitable protecting groups comprise N-Boc, N-Cbz, N-Fmoc, alkyl, benzophenoneimine for protection of amino group, acetal protection for aldehyde, ketal protection for ketone. The invention also encompasses prodrugs of compounds of the invention, which on administration undergo chemical conversion by metabolic processes before becoming active pharmacological substances. After completion of reaction (monitored by TLC), the reaction mixture was poured into a beaker and 5mM solution of Sodium bicarbonate was added slowly into the mixture with continuous stirring until the generation of fumes was stopped and then ice-cold water (200 ml) was added into the mixture. After stirring for another 15 minutes the reaction mixture was extracted with EtOAc (3 x 50). The organic layers were collected, combined, dried over anhydrous Na2SO4, filtered and concentrated under vacuum. EXMAPLE 15: INHIBITORY EFFECT OF COMPOUNDS ON DIFFERENT STRAINS OF STAPHYLOCOCCUS AUREUS: Compounds were tested to evaluate their inhibitory effect on different strains of staphylococcus aureus. An experiment was conducted to assess the minimum inhibitory concentration (MIC) of the compound of Formula (I). Assay for antimicrobial susceptibility testing by broth micro-dilution method (Limbago 2019; CLSI 2020, herein incorporated by reference) • The stock solutions of different antimicrobial agents were prepared with a concentration of 2 mg / mL in DMSO. The aliquots were prepared and stored at - 20°C for further use. • Before starting the experiment, the stock aliquots were thawed at the controlled lab conditions and used for the preparation of the working stocks by diluting further in the test medium to 2X, the highest concentration required in the test. For instance, if the highest desired concentration was 64 µg / ml, the stock solution was diluted and adjusted to 128 µg / ml. • Labelled the 96-well U bottom polypropylene plate (Tarson, 980040, Korea) and dispensed 100 µL of the test medium (Cation-adjusted Muller Hinton Broth) into all wells except the 2nd column. • Added 200 µL of the test drugs (2X) in the each well of the column 2 and hence, eight antimicrobial agents were dispensed into the eight wells of the column followed by 9-point serial dilution (1:1) with gentle mixing from column 2 to column 11 using a multichannel micropipette (Eppendorf, Germany). • In the column11, 100 µL volume was taken out and discarded to adjust the volume uniformity across the plate. • Overnight grown fresh bacterial culture was used to prepare the inoculum, adjusted to 0.5 McFarland (1.5x10^8 CFU / ml) using a densitometer (DEN1-B). The culture inoculum was further diluted to 1:100 ratio in the test medium and resulting in the bacterial suspension of 1.5x10^6 CFU / ml.100 µL of this suspension was dispensed and inoculated into each well of the 96-well plate excluding the column1. • The first column only with the medium and the column 12 with the bacterial inoculum were considered as the negative and positive control for this experiment respectively. • The plate was kept for overnight incubation in ambient air at 37°C and after 12-14 hours incubation, the inhibition of microbial growth was visually assessed using the MIC mirror and the results were interpreted as per the CLSI guidelines. Compound 4a is sensitive against MRSA (Methicillin Resistant Staphylococcus aureus), VISA (Vancomycin Intermediate Resistant Staphylococcus aureus) and / or clinical isolates of Linezolide resistant Staphylococcus spp. MIC of compound 4a against different strains of Staphylococcus aureus is evaluated as described below. It is a highly potent molecule against MRSA, VISA, and Linezolid resistant clinical isolates of Staph. spp strains and has the potential to be a very effective antibiotic. Claims
What is claimed is:
1. A compound of formula (I):or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof; wherein: each R1and R2is independently selected from hydrogen, halogen, hydroxyl, optionally substituted alkyl, optionally substituted alkoxy, optionally substituted cycloalkyl, optionally substituted aryl, optionally substituted heterocyclyl, and optionally substituted heteroaryl; R3is selected from haloalkyl, O-(haloalkyl), and S-(haloalkyl); ring A is an aryl or heteroaryl; wherein aryl and heteroaryl are optionally further substituted with one or more groups selected from halo, haloalkyl, haloalkoxy, alkyl, and alkoxy; and Het represents an optionally substituted heterocyclyl or heteroaryl connected through a C-C bond.
2. The compound as claimed in claim 1 is a compound of formula (Ia):or a pharmaceutically somer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof; wherein: R1, R2,R3,and ring A are same as defined in claim 1; R is independently selected from the group consisting of hydrogen, halogen, hydroxyl, alkyl, alkoxy, cycloalkyl, aryl, heterocyclyl and substituted heteroaryl; X is hydrogen, halo, alkyl, alkoxy, aryl, or -NHSO2Ph; and Y is hydrogen, halo, alkyl, alkoxy, aryl, or -NHSO2Ph.
3. The compound as claimed in claim 1 or 2 is a compound of formula (Ib):or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof; wherein R1, R2,and R3, R, X, and Y are same as defined in claim 2.
4. The compound as claimed in any one of claims 1 to 3, wherein R1 and R2 are alkyl.
5. The compound as claimed in claim 4, wherein R1and R2are C1-C6alkyl.
6. The compound as claimed in any one of claims 1 to 5, wherein R1 and R2 are independently methyl, ethyl, or propyl.
7. The compound as claimed in claim 6, wherein R1is propyl and R2is methyl.
8. The compound as claimed in any one of claims 1 to 7, wherein R3is haloalkyl or - O-haloalkyl.
9. The compound as claimed in claim 8, wherein R3 is -CF3 or -OCF3.
10. The compound as claimed in any one of claims 1 to 9, wherein R is hydrogen.
11. The compound as claimed in any one of claims 1 to 10, wherein X is hydrogen and Y is hydrogen, halo, alkyl, or alkoxy.
12. The compound as claimed in any one of claims 1 to 10, wherein X is hydrogen and Y is fluoro, chloro, bromo, methyl, or methoxy.
13. A pharmaceutical composition comprising a compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, optionally admixed with a pharmaceutically acceptable carrier or excipient or diluent.
15. A method of inhibiting Staphylococcus species comprising administering to a subject in a need thereof a therapeutically effective amount of a compound as claimed in any one of claims 1-13 or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, a prodrug, a polymorph, N-oxide, or bioisostere thereof, or a pharmaceutical composition as claimed in claim 14.
16. A method of preventing or treating a condition, disease, disorder caused by Staphylococcus species in a subject, comprising administering to the subject a therapeutically effective amount of a compound as claimed in any one of claims 1- 13 or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or a pharmaceutical composition as claimed in claim 14.
17. The method as claimed in claim 15 or 16, wherein Staphylococcus species is Staphylococcus aureus (S. aureus).
18. The method as claimed in claim 17, wherein S. aureus is multidrug-resistant Staphylococcus aureus (MRSA).
19. A compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or a pharmaceutical composition as claimed claims 14, for use as a medicament or a therapeutic agent.
20. A compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or a pharmaceutical composition as claimed in claim 14, for use in preventing or treating a condition, disease, or disorder caused by Staphylococcus species or other Gram-positive bacteria.
21. A compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or a pharmaceutical composition as claimed in claim 14, for use in inhibiting Staphylococcus species.
22. The compound as claimed in claim 20 or 21, wherein Staphylococcus species is S. aureus.
23. The compound as claimed in claim 22, wherein S. aureus is MRSA.
24. Use of a compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, orbioisostere thereof, or of a pharmaceutical composition as claimed in claim 14, for preventing or treating a condition, disease, or disorder caused by caused by Staphylococcus species.
25. Use of a compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or of a pharmaceutical composition as claimed in claim 14, in the inhibition of Staphylococcus species.
26. Use of a compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or of a pharmaceutical composition as claimed in claim 14, in the manufacture of a medicament.

Use of a compound as claimed in any one of claims 1-13, or a pharmaceutically acceptable salt, stereoisomer, hydrate, solvate, prodrug, polymorph, N-oxide, or bioisostere thereof, or of a pharmaceutical composition as claimed in claim 14, in the manufacture of a medicament for inhibiting Staphylococcus species or for preventing or treating a condition, disease, or disorder caused by Staphylococcus species.
28. The use as claimed in any claim 24, 25, or 27, wherein Staphylococcus species is S. aureus.
29. The use as claimed in claim 28, wherein S. aureus is MRSA.
