[0001] The present invention relates to the technical field of biological medicine, and particularly relates to a circular RNA vaccine for protecting a subject from a disease-associated protein, and application thereof.BACKGROUND OF THE INVENTION

[0002] Research on tumor immunotherapy have made tremendous progress in the past decades. Cancer vaccines, which are designed to induce anti-tumor responses by expression tumor antigens in vivo, are also an attractive immunotherapeutic option. A factor which is essential for the efficacy of cancer vaccine is optimal induction of specific T cell responses. Due to the central T-cell tolerance, clinical trials have been largely unsuccessful. Most showed only low rate of clinical responses which was correlated to the moderate T cell immunity. Therefore, the medical need is still high to develop vaccines with robust and specific T cell responses. Here we present a method for inducing both CD4+ and CD8+ T cells immune response through elaborate circular RNA vaccines.

[0003] In the present invention, we have designed several vaccines with different elements to facilitate MHC I and MHC II presentation. Lysosome-associated membrane protein (LAMP1) is a lysosome membrane glycoprotein which facilitates lysosomal acidification for optimal hydrolase activity. This protein shuttles between lysosomes, endosomes, and the plasma membrane. We fused several functional domains of murine or human LAMP1 / Dendritic-cell LAMP to reside antigens in MHC class II processing compartment. MHC class II associated invariant chain (Ii) , a type II membrane protein, serves as both a targeting subunit and guardian of the peptide-binding site, targeting signals in the cytoplasmic domain of the Ii chain can drive Ii-bound complex into the endosomal-lysosomal pathway. Whereas LAMP proteins are predominantly located in acidified late endosomes and lysosomes, antigens fused with Ii chain are targeted to early endosomes. The helper epitopes may also be combined with the antigen coding sequence to break immunological tolerance. The helper epitopes P2P16 are derived from tetaus toxoid, which is known to provide CD4+ mediated T cell response. We show that linking these functional domains to antigens allows robust T cells response, resulting in distinct CD4 / 8+ T cell specificities.SUMMARY OF THE INVENTION

[0188] The protein circular RNA encoded consists of several functional domains: a signal peptide leading at the N-terminal, tumor associated antigen to trigger tumor cell specific immune response, helper peptide derived from tetanus toxoid in order to enhance resulting immune response, and amino acids sequence to increase MHC presentation. All these functional domains are joined by protein linkers. Nucleotides encoding this protein are codon optimized with internal algorithms and the translation is initiated through a coxsackievirus B3 (CVB3) internal ribosome entry sites (IRESs) . The nucleotide sequences of IRES and coding region were synthesized by Azenta Life Sciences (Suzhou, China) .

[0190] EXAMPLE 2: Manufacturing of Circular RNA.

[0191] The plasmid was linearized by digestion with BspQ I restriction endonuclease (Vazyme, DD4302-PC-02) , the reaction was incubated at 50 ℃ for 3 hours and the linear DNA template was purified using DNA Clean &Concentrator-25 Kit (ZYMO RESEARCH, D4034) . Following the linearization step, an aliquot of the linearized DNA template was added to an IVT reaction to get the precursor RNA. In a traditional IVT reaction system, reaction components were assembled with 400 ng linearized DNA template, 100 U T7 RNA Polymerase (Vazyme, DD4101-PC-03) , a NTP solution mix including 100 mM GTP (Vazyme, DD4108-PA-02) , 600 mM GMP solution, 100 mM ATP (Vazyme, DD4106-PA-02) , 100 mM CTP (Vazyme, DD4107-PA-02) , 100 mM UTP (Vazyme, DD4105-PA-02) , 10×reaction buffer (Vazyme, DD4101R) , 10 U Rnase Inhibitor (Vazyme, DD4102-PA-01) and 0.008 U Pyrophosphatase (Vazyme, DD4103-PC-01) , the IVT reaction was incubated at 37 ℃ for 2 hours. Then the linear products were exposed to Dnase I (Vazyme, EN401) digestion to remove the template DNA, and the RNA transcripts were column purified by RNA Cleanup Kit (NEB, T2040L) . To get the circular RNA, we then used T4 Rnl2 (Kactus, TRL-BE103-C1) to circularize the RNA transcripts, an enzyme ligation reaction was assembled, 10 U T4 RNA Ligase II, 10×reaction buffer and 10 μg linear RNA were added to a 40 μL reaction mixture and the reaction vessels were incubated at 25 ℃ for 2 hours. At last Rnase R exonuclease (Novoprotein, GMP-E224-M001) was added to degrade the linear RNA and enrich the circular product, 1 μg column purified RNA product, 1 U Rnase R was assembled to the 6 μl reaction system, and the reactions were incubated at 37 ℃ for 30 minutes. The circular RNA products were then purified using  RNA Cleanup Kit (NEB, T2040L) .

[0192] EXAMPLE 3: Induction of Antigen-Specific Immune Responses

[0193] C57BL / 6 mice were repetitively immunized intramuscularly with lipid nanoparticles formulated circular RNA vaccines at Day0, Day3 and Day8.20 μg circRNA-LNP were injected into the tibialis anterior muscle. At Day14, mice were sacrificed and spleen were collected for enyzme-linked immune spot (ELISPOT) and flow cytometry analysis. For single cell suspension preparation, spleens were homogenized by a syringe plunger at a 70 μm cell strainer ( 352350) in RPMI-1640 medium (HyClone, SH30027.01) . Red blood cells were removed by applying ACK lysis buffer (GibcoTM, A1049201) to splenocyte for 4 min at room temperature. Cells were washed with DPBS (GibcoTM, 14040133) twice and resuspended at 1×107 cells / mL. for ELISPOT, 3×105 splenocyte were seeded in 96-plates coated with IFN-γ capture antibody (MABTECH, 3321-4AST) . NY-ESO-1 peptide pool (Miltenyi Biotec, 130-095-381) was added then to a concentration of 0.25 μg / mL and incubated 40 h for in vitro stimulation. IFN-γ secretion was detected by biotinylated anti-IFN-γ antibody. Spots were developed following the manufacturer’s instructions. The final spots were counted using an AID SLISPOT reader. (Figure 2)

[0194] EXAMPLE 4: Induction of Antigen-Specific CD8+ and CD4+ T-Cell Responses

[0195] For antigen specific CD4+ and CD8+ T cells response detection, 3 million splenocytes were stimulated with NY-ESO-1 peptide pool (0.25 μg / mL, Miltenyi Biotec, 130-095-381) and anti-CD28 monoclonal antibody (1 μg / mL, BioLegend, 102116) in a U-bottom 96-well plate. 4 hours later, BD GolgiStop monensin (1: 1000 dilution, BD Bioscience, 554715) was added to block protein transport for 12 hours. Cells were washed with PBS and followed by anti-CD16 / 32 Fc-receptor blockade (BD Pharmingen, 553142) before staining. Cells were stained with PE anti-mouse CD3 Antibody (BioLegend, 100205) , FITC Rat Anti-Mouse CD8a (BD Pharmingen, 553030) , APC Rat Anti-Mouse CD4 (BD Pharmingen, 553051) , and Fixable Viability Stain 510 (BD Pharmingen, 564406) for surface staining at 4 ℃ for 30 minutes. Then cells were washed with stain buffer (BD Pharmingen, 554656) , fixed and permeabilized using the Cytofix / Cytoperm kit (BD Biosciences, 554714) , and stained intracellularly for another 30 minutes in stain buffer with antibodies including PerCP-CyTM5.5 Rat Anti-Mouse IFN-γ (BD Pharmingen, 560660) and PE-CyTM7 Rat Anti-Mouse IL-4(BD Pharmingen, 560699) . (Figure 3) .
