[0001] The present disclosure belongs to the field of medicinal chemistry. Specifically, the present disclosure relates to a Pt (IV) chemotherapeutic prodrug and its controlled release for treating tumors.Background of the invention

[0002] Cancer is one of the most serious contemporary diseases that threaten human life and health. Surgery, radiotherapy and chemotherapy are collectively referred to as the three major methods for treating tumors.

[0003] Radiation therapy is a local treatment method that uses radiation to treat tumors. The curative effect of radiotherapy depends on radiosensitivity, and different tissues and organs and various tumor tissues have different responses of changes after being irradiated. For example, fibrosarcoma, osteosarcoma, and melanoma are radiation-insensitive (resistant) tumors. Radiation therapy is still limited by challenges inherent to radiation, including toxicity to surrounding healthy tissues and limited efficacy against radioresistant tumors. Radiotherapy is also constrained by tumor heterogeneity, encompassing factors such as variability in individual patient and tissue tolerance to radiation, differences in tumor distribution, and cumulative radiation doses across multiple sessions. These limitations collectively undermine the effectiveness and safety of radiotherapy in specific clinical contexts, ultimately leading to recurrence and metastasis in a significant proportion of patients.

[0004] Chemotherapy uses chemical drugs to kill cancer cells for treatment purposes. Divalent platinum-based drugs have high-efficiency and broad-spectrum anticancer activity, and have become clinically important first-line chemotherapy drugs. Divalent platinum-based drugs are widely used in the treatment of common malignant tumors such as lung cancer, bladder cancer, ovarian cancer, cervical cancer, esophageal cancer, gastric cancer, colorectal cancer and head and neck tumors. The first generation of platinum-based anticancer drugs is represented by Cisplatin, the second generation of platinum-based anticancer drugs is represented by Carboplatin and Nedaplatin. and the third generation of platinum-based anticancer drugs is represented by Oxaliplatin and Lobaplatin. The side effects of divalent platinum-based drugs, such as nephrotoxicity, gastrointestinal toxicity, hematological system toxicity, nervous system toxicity and ototoxicity, limit their application, and the drug resistance of tumors also limits their therapeutic effect. In order to expand platinum-based drugs, studies on tetravalent platinum drugs have also been carried out. Tetravalent platinum complexes function as prodrugs of cytotoxic divalent platinum-based species and generally exhibit greater chemical stability in plasma compared to their divalent platinum counterparts. These compounds are designed to be selectively activated within the reductive microenvironment characteristic of tumor tissues, thereby enabling localized release of the active agent and reducing systemic toxicity. In addition, the octahedral coordination geometry of tetravalent platinum complexes allows for axial ligand modifications, offering a flexible platform for optimizing pharmacokinetic behavior, improving tumor selectivity, and incorporating synergistic therapeutic functionalities. Despite these advantages, the bioreductive activation of tetravalent platinum complexes depends on endogenous reducing agents within tumors, a process that can be compromised by tumor heterogeneity, resulting in inefficient drug release and suboptimal therapeutic outcomes. Furthermore, undesired reduction in non-tumor tissues may lead to premature activation and off-target toxicity.

[0006] Accordingly, there remains an urgent need to develop advanced tetravalent platinum prodrug systems with enhanced chemical and biological stability in non-tumor environments and improved tumor-specific activation efficiency, in order to fully realize their clinical potential and therapeutic efficacy.Summary of the invention

[0007] After in-depth research and creative work, the present inventors found that Pt (IV) complexes can be used as prodrugs, and the prodrugs release divalent platinum drugs through irradiation to realize the combination therapy of tumors. If radiotherapy is efficiently combined with Pt (IV) prodrugs-where radiation activation triggers the localized release of active chemotherapeutic Pt(II) species and simultaneously enhances radiosensitization, the therapeutic efficacy can be significantly improved.. This selective activation provides dual benefits: enhancing the localized chemotherapy effects of radiotherapy region while sparing non-target tissues from systemic toxicity. Pt (IV) complexes, by harnessing the spatiotemporal precision of radiotherapy, have the potential to enhance the therapeutic index of Pt (II) -based chemotherapy and to overcome both chemoresistance and radioresistance in certain tumor types.

[0415] The plasma stability was performed to detect the stability of Ab-Pt (IV) incubated with human plasma. Briefly, the plasma, oxaliplatin-spiking solutions, non-albumin-binding platinum (AcPt) -spiking solutions, and Ab-Pt (IV) -spiking solutions were pre-warmed at 37℃ for 5 min. Then, the pre-warmed plasma and Ab-Pt (IV) -spiking solutions were added into the wells designated for all the time points (0, 60, 120, 240, 480, 1440, 2880 min) . At different time points, 1000 μL of MEOH: ACN (1: 1) containing internal standard (IS) was added to the wells of corresponding plates, respectively, to stop the reaction. After quenching, the plates were shaken for 5 min, centrifugated at 600 rpm for 10 min, and analyzed by LC / MS / MS.

[0436] Six-week-old female Nu / Nu nude mice were ordered from Shanghai Model Organisms (Shanghai, China) and kept under specific pathogen-free condition with free access to standard food and water. Approximately 2×106 HCT116 cells suspended in 100 μL of PBS were implanted subcutaneously into the right shoulder of Nu / Nu mice. Tumor volumes were calculated with the formula: the volume = 1 / 2×length×width2. When the volume of tumor reached ~100 mm 3 (about 10 days after inoculation) , tumor-bearing mice were then randomly divided, intravenous Compound was followed by X-ray irradiation twice a week for two consecutive weeks. If the volume of tumors exceeds 1500 mm3 , mice are sacrificed.
