[0003] The present disclosure relates to RNA interference (RNAi) agents for inhibiting INHBE, such as double-stranded RNAi agents, pharmaceutical compositions comprising the INHBE RNAi agents, and methods of using same.BACKGROUND

[0004] Body fat distribution is a critical risk factor for cardiovascular and metabolic diseases including type 2 diabetes (T2D) , coronary heart disease (CHD) , circulating lipids, and blood pressure, while there is a lack of therapeutic options to modify improper fat storage. By genome-wide association studies (GWAS) , INHBE has been successfully identified as a genetic variant correlated with fat distribution and reduced risk of diabetes and coronary disease. INHBE encodes inhibin βE subunit, a member of the transforming growth factor-β (TGF-β) , which dimerizes to form activin E, a hepatokine whose signaling has not been fully-characterized. INHBE expression was found to increase in human livers from subjects with insulin resistance. And its elevated expression was also observed in high fat diet (HFD) induced mouse obesity model. On the other hand, INHBE suppression reduced body weight by loss of fat mass rather than lean mass.

[0005] Due to the lack of treatments for modifying excessive fat stores, there’s a need for development of new therapies to selectively and efficiently modulate metabolic syndrome associated genes like INHBE, e.g., INHBE RNAi agents, which is potential to bring durable and safe management of body weight for subjects with such a need.SUMMARY

[0006] The present disclosure provides an iRNA composition that causes cleavage of the RNA transcript of the INHBE gene mediated by an RNA-induced silencing complex (RISC) . The INHBE can be located within a cell, such as a cell of a subject, such as a human subject.

[0007] The present disclosure relates to a double-stranded ribonucleic acid (dsRNA) agent that inhibits the expression of INHBE, wherein the dsRNA agent comprises a sense strand and an antisense strand, the sense strand being as follows: 5’-Q1-X-Q2-3’, wherein Q1 and Q2 are 0, 1 or 2 nucleotide sequences selected from A, U, T, C, and G nucleotide motifs; X is a sequence containing positions 1 to n1 of the sense strand sequence as shown in Table 1-1, Table 1-3, Table 4-1, Table 5-1, Table 6-1, Table 7-1, Table 8-1 or Table 11-1, and n1 is position 18, 19, 20, or 21; the antisense strand being as follows: 5’-Q3-Y-Q4-3’, wherein Q3 and Q4 are 0, 1 or 2 nucleotide sequences selected from A, U, T, C, and G nucleotide motifs; Y is a sequence containing positions n2 to n3 of the antisense strand sequence as shown in Table 1-1, Table 1-3, Table 4-1, Table 5-1, Table 6-1, Table 7-1, Table 8-1 or Table 11-1, n2 is position 1 or 2, and n3 is position 18, 19, 20, 21, 22, or 23.

Claims

1.A double-stranded ribonucleic acid (dsRNA) agent for inhibiting the expression of INHBE, wherein the dsRNA agent comprises a sense strand and an antisense strand, wherein the antisense strand comprises a region complementary to the transcript of INHBE, and the complementary region comprises at least 15 contiguous nucleotides differing by no more than 3 nucleotides from the nucleotide sequences of any one of the antisense strands listed in Table 1-1.2.A double-stranded ribonucleic acid (dsRNA) agent for inhibiting the expression of INHBE, wherein the dsRNA agent comprises a sense strand and an antisense strand,wherein the sense strand has the structure as shown in formula (i) :5’-Q1-X-Q2-3’, formula (i)wherein Q1 and Q2 are 0, 1 or 2 nucleotides selected from A, U, T, C, and G nucleotide motifs; X is a sequence comprising positions 1 to n1 of any sense strand sequence as shown in Table 1-1, and n1 is position 18 or 19;the antisense strand has the structure as shown in formula (ii) :5’-Q3-Y-Q4-3’, formula (ii)wherein Q3 and Q4 are 0, 1 or 2 nucleotides selected from A, U, T, C, and G nucleotide motifs; Y is a sequence containing positions n2 to n3 of any antisense strand sequence as shown in Table 1-1, n2 is position 1 or 2, and n3 is position 18, 19, 20 or 21.3.The dsRNA agent according to claim 1 or 2, wherein the dsRNA agent comprises at least one modified nucleotide.4.The dsRNA agent according to claim 1 or 2, wherein all or substantially all nucleotides on the antisense strand are modified nucleotides, and preferably all nucleotides on the sense strand and antisense strand are modified nucleotides.5.The dsRNA agent according to claim 1 or 2, which comprises a sense strand and an antisense strand, wherein(1) the sense strand comprises a nucleotide sequence X, the antisense strand comprises a nucleotide sequence Y, the nucleotide sequence X and the nucleotide sequence Y are at least partially reverse-complementary to form a double-stranded region; and,(2) each nucleotide in the sense strand and the antisense strand is independently a fluorinated modified nucleotide or a non-fluorinated modified nucleotide; and,(3) the fluorinated modified nucleotide is located in the nucleotide sequence X and the nucleotide sequence Y;and,(4) in the 5′to 3′direction, sequence X has the general formula (iii) :5'(N) NsNsNsNsNsNsNZNZNZNsNsNsNsNsNsNsNsNsNs-3'in the 5′to 3′direction, sequence Y has the general formula (iv) :5'(N) NsNFNiNiNiNiNiNiNiNiNiNdNiNFNiNFNsNsNsNsNs-3'Wherein:NF each independently represents a 2′-fluorine-modified nucleotide;Nd each independently represents a 2′-deoxy-modified nucleotide;Ns each independently represents a 2′-O-methyl (2′-OMe) -modified ribonucleotide;Ni independently represents a 2’-O-methyl (2’-OMe) modified ribonucleotide or 2’-deoxy (DNA) -modified nucleotide;NZ each independently represents either a 2′-fluorine-modified ribonucleotide or a 2′-deoxy (DNA) -modified nucleotide and;(5) the dsRNA targets and inhibits INHBE.6.The dsRNA agent according to claim 3 or 4, wherein the at least one modified nucleotide includes: 2'-O-methyl nucleotide, 2'-fluoro nucleotide, 2'-deoxynucleotide, locked nucleic acid (LNA) , unlocked nucleic acid (UNA) , glycol nucleic acid (GNA) , 2'-F-arabinose nucleotide, 2'-methoxyethyl nucleotide, abasic nucleotides, ribitol, reverse nucleotides, reverse abasic nucleotides, reverse 2'-OMe nucleotide, reverse 2'-deoxynucleotide, 2'-amino modified nucleotide, 2'-alkyl modified nucleotide, morpholino nucleotide and 3'-OMe nucleotide, nucleotides containing a 5'-phosphorothioate group, or terminal nucleotides linked to cholesterol derivatives or dodecanoate bisdecylamide groups, 2'-amino modified nucleotides, phosphoramidates, or unnatural bases containing nucleotides.7.The dsRNA agent according to any one of claims 1 to 6, wherein in at least one phosphate ester group in the phosphor-sugar backbone of at least one strand of the sense strand and antisense strand, said phosphate ester group has a modifying group.8.The dsRNA agent according to claim 7, wherein the modified phosphate ester group is a phosphorothioate group formed by substitution of at least one oxygen atom in the phosphodiester bond with a sulfur atom.9.The dsRNA agent according to claim 7 or 8, wherein the phosphorothioate linkage is present in at least one location selected from the group consisting of:Between nucleotides 1 and 2 at the 5′-end of the sense strand;Between nucleotides 2 and 3 at the 5′-end of the sense strand;Between nucleotides 1 and 2 at the 3′-end of the sense strand;Between nucleotides 2 and 3 at the 3′-end of the sense strand;Between nucleotides 1 and 2 at the 5′-end of the antisense strand;Between nucleotides 2 and 3 at the 5′-end of the antisense strand;Between nucleotides 1 and 2 at the 3′-end of the antisense strand; andBetween nucleotides 2 and 3 at the 3′-end of the antisense strand.10.The dsRNA agent according to any one of claims 1-9, wherein the 5′-terminal nucleotide o 25.The oligonucleotide conjugate according to any one of claim 16-24, wherein said conjugates are QLAD-003220, QLAD-004458, QLAD-003151, QLAD-003132, QLAD-003117, QLAD-003135, QLAD-003126, QLAD-004465, QLAD-004466, QLAD-004467, QLAD-004468 or QLAD-004469.26.A pharmaceutical composition comprising the dsRNA agent according to any one of the preceding claims for inhibiting the expression of the gene encoding INHBE; preferably, in which the expression of INHBE was reduced by at least 50%in animals.27.The pharmaceutical composition according to claim 26, wherein the pharmaceutical composition further comprises a pharmaceutically acceptable carrier, a buffer solution, a non-buffer solution, or an additional therapeutic agent.28.The pharmaceutical composition according to claim 26 or 27, wherein the pharmaceutical composition is packaged in a kit, a container, a wrapper, a dispenser, a prefilled syringe or a vial.29.The pharmaceutical composition according to any one of claims 26-28, wherein the pharmaceutical composition is formulated for subcutaneous administration or is formulated for intravenous (IV) administration.30.A cell comprising the dsRNA agent according to any one of the preceding claims.31.The cell according to claim 30, wherein, the cell is a mammalian cell, optionally a human cell.32.A method for inhibiting the expression of INHBE gene, comprising contacting the cell with the dsRNA agent according to any one of claims 1-15, or the conjugate according to claim 16-25, or the pharmaceutical composition according to any one of claims 26-29, thereby inhibiting the expression of the INHBE gene in the cell.33.A method for treating or preventing a subject suffering from a condition that would benefit from reduced expression of INHBE, comprising administering to the subject a therapeutically or prophylactically effective amount of the dsRNA agent according to any one of claims 1-15, or the conjugate according to claim 16-25, or the pharmaceutical composition according to any one of claims 26-29, thereby treating or preventing the subject suffering from a condition that would benefit from reduced expression of INHBE.34.The method according to claim 32 or 33, wherein(i) the cell is in the body of a subject, and preferably, the subject is a human; and / or(ii) the subject suffers from INHBE -related condition; and / or(iii) the cell is in the body of a subject and the dsRNA agent is administered subcutaneously to the subject; and / or(iv) the cell is in the body of a subject and the dsRNA is administered intravenously (IV) to the subject; and / or(v) contacting the cell with the dsRNA agent, thereby the expression of INHBE is inhibited by at least 50%.35.The method according to claim 33 or 34, wherein the condition is an INHBE -associated disease, including but not limited to metabolic disorders, type 2 diabetes, obesity, hypertriglyceridemia, hypercholesterolemia, lipodystrophy, fatty liver disease, cardiovascular disease, and high blood pressure.
