[0005] Microfluidic technology facilitates the precise manipulation and control of fluid flows ranging from microliters to femtoliters within channels of micrometer dimensions. This technology leverages the distinct behavior of fluids at the microscale, which arises from variations in physical properties compared to their behavior at larger scales. Such capabilities make microfluidics an invaluable tool across multiple disciplines, including engineering, physics, chemistry, biochemistry, nanotechnology, synthetic biology, and biotechnology.
[0006] In the domain of droplet-based microfluidics, this technology employs immiscible fluids to generate an emulsion, thereby forming discrete reaction vessels, called droplets. In lipid vesicle construction, transitioning lipid vesicles from an oil / water to a water / water system requires a surfactant that can create a metastable system, robust enough for stabilization of w / o droplets but also capable of being destabilized for vesicle release. For cell encapsulation, additional nutrients must be included for the survival and functionality of the encapsulated cells. This creates a high osmotic pressure, which increases the fusion forces of the droplets. In droplet digital molecular biology, droplets have to maintain stability during chemical reactions and analyses, to ensure accurate results. [0480] To determine the interfacial tension between the selected formulation and water, the inventors employed the pendant drop method. Initially, the formulation was diluted to a total concentration of 2.5 mM using Novec 7500 Engineered Fluid and loaded into a syringe equipped with a flat-tipped needle. The surrounding phase comprised deionized water, which was carefully introduced into a cuvette to serve as the continuous phase.

The mammalian cells are cultivated in appropriate cell culture medium in a CO2 incubator at 37°C. Upon reaching 70-80% confluency, cells are detached using trypsin-EDTA solution. The trypsin is neutralized with fresh medium, and cells are transferred to a centrifuge tube. After centrifugation at 300 x g for 5 minutes, the supernatant is discarded. The cell pellet is resuspended, and cells are counted followed by adjusting the cell concentration 1 x 10A5 cells / mL in cell culture media with 10% FBS. 13. The formulation according to any one of claims 1 - 12, wherein the at least one amphiphilic polymer has a purity of at least 95% as determined by NMR spectroscopy.
