[0004] The invention related to orally disintegrating nicotine tablets according to the claims. Also, the invention relates to an oral nicotine tablet according to the claims.

[0005] BACKGROUND

[0006] Nicotine-releasing tablets applied for the purpose of providing a release of nicotine in a user’s mouth over a certain period of time is well-known. Much effort has in prior art been put into emulating the nicotine release and oral perception of a cigarette when it is smoked by a user, which means that release profiles from nicotine tablets have been thoroughly investigated in prior art.

[0011] The invention relates to an orally disintegrating nicotine tablet for fast nicotine craving relief, the tablet comprising nicotine, disintegrant, and a filler, wherein the filler comprises no more than 10% by weight of sugar alcohol. An advantage of the invention may be that the minimized amount of sweeteners, such as sugar alcohol, facilitates a decreased salivation for users of the tablets, i.e. a decreased perceived salivation. While salivation on one hand may help to disintegrate the tablet, it has been found that users may not prefer a very pronounced salivation associated with a large content of sugar alcohol, and that minimizing the sugar alcohol content while the tablet remains an orally disintegrating nicotine tablet is advantageous. Also, without being bound by theory, it is contemplated that less saliva generation facilitates buccal absorption due to increased nicotine residence time and limits swallowing of nicotine.

[0012] Furthermore, the present inventors unexpectedly found that nicotine may still be taste masked without the use of sweeteners, such as sugar alcohol, for example by using flavor and / or other taste sensates.

[0013] A further advantage of the invention may be that it has a reduced calorie content due to the reduced content or even absence of sugar alcohol and other sweeteners.

[0014] A further advantage of the invention may be that it facilitates fast nicotine craving relief while maintaining nicotine burning at an acceptable level.

In embodiments of the invention, the orally disintegrating nicotine tablet may thus disintegrate within 4 minutes upon oral administration, such as within 3 minutes upon oral administration, preferable when administered between the gum and the lip. In embodiments of the invention, the orally disintegrating nicotine tablet may disintegrate within 60 seconds in vitro, preferably when measuring vitro disintegration time in accordance with European Pharmacopoeia 9.0, section 2.9.1, Disintegration of tablets and capsules.

[0017] According to an embodiment of the invention, the in vivo disintegration time is measured by at least 6 trained assessors, the trained assessors abstaining from eating and drinking at least 30 minutes before initiation of any test, the tablet is weighted and placed in the mouth, on the tongue, where the in vivo disintegration time is registered as the time point where the tablet disintegration is substantially complete. During the test, the assessors refrained from chewing and did not substantially move the tablet from its initial position. The trained assessors are selected based on the following criteria. Then, nicotine burning at fixed time points 30 and 180 seconds was calibrated across the test panel. In the test, each assessor evaluated the burning sensation in the throat on a scale from 0 to 15, where 15 is the most intense burning. Each assessor evaluated all samples twice. The evaluations were noted for the time points indicated. Average values of all assessors were calculated.

[0354] It was therefore concluded that the inventive tablets with the claimed little to no amount of sugar alcohol or bulk sweetener are effective in delivering nicotine while unexpectedly not increasing nicotine burning. This result was obtained even though the sweetening effect resulting from large amount of sweetener in conventional tablets is conventionally considered to contribute to taste masking and thereby often considered to be important or indispensable to reduce nicotine burning to acceptable levels.

Claims

CLAIMS1. An orally disintegrating nicotine tablet for fast nicotine craving relief, the tablet comprising nicotine, disintegrant, and a filler, wherein the filler comprises no more than 10% by weight of sugar alcohol.

2. The orally disintegrating nicotine tablet according to claim 1, wherein the tablet comprises a filler in an amount of at least 40% by weight of the tablet, such as at least 50% by weight of the tablet, such as at least 60% by weight of the tablet, such as at least 70% by weight of the tablet, such as at least 80% by weight of the tablet.

3. The orally disintegrating nicotine tablet according to claim 1 or 2, wherein the filler comprises a fiber.

4. The orally disintegrating nicotine tablet according to any of claims 1-3, wherein the filler comprises a fiber in an amount of at least 50% by weight of the filler, such as at least 60% by weight of the filler, such as at least 70% by weight of the filler, such as at least 80% by weight of the filler, such as at least 90% by weight of the filler.

5. The orally disintegrating nicotine tablet according to any of claims 1-4, wherein the filler comprises a water-soluble fiber.

6. The orally disintegrating nicotine tablet according to any of claims 1-5, wherein the filler comprises a water-soluble fiber selected from the group consisting of indigestible maltodextrin, polydextrose, inulin, and any combination thereof.

7. The orally disintegrating nicotine tablet according to any of claims 1-6, wherein the filler comprises a water-soluble fiber selected from the group consisting of polydextrose, inulin, and any combination thereof.

8. The orally disintegrating nicotine tablet according to any of claims 1-7, wherein the filler comprises an indigestible fiber.

9. The orally disintegrating nicotine tablet according to any of claims 1-8, wherein the filler comprises a water-insoluble fiber.

10. The orally disintegrating nicotine tablet according to claim 9, wherein the waterinsoluble fiber comprises one or more selected from the group consisting of wheat fibers, pea fibers, rice fiber, maize fibers, oat fibers, tomato fibers, barley fibers, rye fibers, sugar beet fibers, buckwheat fibers, potato fibers, apple fibers, cocoa fibers, bran fibers, bamboo fibers, powdered cellulose, microcrystalline cellulose, and any combination thereof.

11. The orally disintegrating nicotine tablet according to claim 9 or 10, wherein the water insoluble fiber comprises microcrystalline cellulose.

12. The orally disintegrating nicotine tablet according to any of claims 1-11, wherein the filler comprises protein, such as pea protein.

13. The orally disintegrating nicotine tablet according to any of claims 1-12, wherein the filler comprises no more than 5% by weight of sugar alcohol, such as no more than 2% by weight of sugar alcohol, such as no more than 1% by weight of sugar alcohol, such as free of sugar alcohol.

14. The orally disintegrating nicotine tablet according to any of claims 1-13, wherein the filler is free of sugar alcohol.

15. The orally disintegrating nicotine tablet according to any of claims 1-14, wherein the sugar alcohol comprises sugar alcohol selected from the group consisting of xylitol, maltitol, mannitol, erythritol, isomalt, sorbitol, lactitol and any combination thereof.

16. The orally disintegrating nicotine tablet according to any of claims 1-15, wherein the filler comprises no more than 10% by weight of bulk sweetener, such as no more no more than 5% by weight of bulk sweetener, such as no more than 2% by weight of bulk sweetener, such as no more than 1% by weight of bulk sweetener, such as free of bulk sweetener.

17. The orally disintegrating nicotine tablet according to any of claims 1-16, wherein the filler comprises no more than 10% by weight of sugar, such as no more no more than 5% by weight of sugar, such as no more than 2% by weight of sugar, such as no more than 1% by weight of sugar, such as free of sugar.

18. The orally disintegrating nicotine tablet according to any of claims 1-17, wherein the tablet comprises said disintegrant in an amount of at least 0.5% by weight of the tablet, such as at least 1% by weight of the tablet, such as at least 2% by weight of the tablet, such as at least 3% by weight of the tablet, such as at least 5% by weight of the tablet.

19. The orally disintegrating nicotine tablet according to any of claims 1-18, wherein said disintegrant is selected from the list consisting of starch, pregelatinated starch, modified cellulose, ion-exchange resin, calcium silicate, crosslinked cellulose, crosslinked polyvinyl pyrrolidone, crosslinked starch, crosslinked alginate, and combinations thereof.

20. The orally disintegrating nicotine tablet according to any of claims 1-19, wherein said disintegrant comprises pregelatinated starch.

21. The orally disintegrating nicotine tablet according to any of claims 1-20, wherein said disintegrant consists of pregelatinated starch.

22. The orally disintegrating nicotine tablet according to any of claims 1-21, wherein the disintegrant comprises a superdisintegrant.

23. The orally disintegrating nicotine tablet according to any of claims 1-22, wherein the tablet comprises superdisintegrant in an amount of at least 0.5% by weight of the tablet, such as at least 1% by weight of the tablet, such as at least 2% by weight of the tablet, such as at least 3% by weight of the tablet, such as at least 5% by weight of the tablet.

24. The orally disintegrating nicotine tablet according to claim 22 or 23, wherein the superdisintegrant is selected from the group consisting of crosslinked cellulose, crosslinked polyvinyl pyrrolidone, crosslinked starch, crosslinked alginate, and any combinations thereof.

25. The orally disintegrating nicotine tablet according to any of claims 22-24, wherein the disintegrant is a super disintegrant selected from the group consisting of crosslinked cellulose, crosslinked polyvinyl pyrrolidone (PVP), crosslinked starch, and any combinations thereof.

26. The orally disintegrating nicotine tablet according to any of claims 22-25, wherein the disintegrant is a super disintegrant selected from the group consisting of crosslinked polyvinyl pyrrolidone, croscarmellose, sodium starch glycolate, and any combinations thereof.

27. The orally disintegrating nicotine tablet according to any of claims 1-26, wherein the tablet is a compressed tablet.

28. The orally disintegrating nicotine tablet according to any of claims 1-27, wherein the tablet is composed of a plurality of compressed particles.

29. The orally disintegrating nicotine tablet according to any of claims 1-28, wherein the tablet is compressed using a compression force of 1 to 35 kN, such as 2 to 30 kN, such as 2 - 20 kN30. The orally disintegrating nicotine tablet according to any of claims 1-29, wherein the nicotine is selected from the group consisting of a nicotine salt, nicotine free base, a nicotine-ion exchange resin combination, nicotine bound to a fatty acid such as oleic acid, a nicotine inclusion complex or nicotine in any non-covalent binding, nicotine bound to zeolites, nicotine bound to cellulose, nicotine bound to starch microspheres, and any combination thereof.

31. The orally disintegrating nicotine tablet according to any of claims 1-30, wherein the nicotine comprises nicotine free base.

32. The orally disintegrating nicotine tablet according to any of claims 1-31, wherein the nicotine is provided in association with a fatty acid such as oleic acid.

33. The orally disintegrating nicotine tablet according to any of claims 1-32, wherein the nicotine comprises nicotine salt.

34. The orally disintegrating nicotine tablet according to claim 33, wherein the nicotine salt is selected from the group consisting of nicotine ascorbate, nicotine aspartate, nicotine benzoate, nicotine monotartrate, nicotine bitartrate, nicotine chloride, nicotine citrate, nicotine fumarate, nicotine lactate, nicotine mucate, nicotine laurate, nicotine levulinate, nicotine malate, nicotine perchlorate, nicotine pyruvate, nicotine salicylate, nicotine sorbate, nicotine succinate, nicotine sulfate, and any combination thereof.

35. The orally disintegrating nicotine tablet according to any of claims 1-34, wherein the nicotine comprises nicotine bitartrate.

36. The orally disintegrating nicotine tablet according to any of claims 1-35, wherein the tablet comprises nicotine in an amount of at least 0.2 mg, such as at least 0.5 mg, such as at least 1.0 mg.

37. The orally disintegrating nicotine tablet according to any of claims 1-36, wherein the tablet comprises nicotine in an amount of at least 0.2% by weight of the tablet, such as at least 0.3% by weight of the tablet, such as at least 0.5% by weight of the tablet.

38. The orally disintegrating nicotine tablet according to any of claims 1-37, wherein the tablet comprises a pH regulating agent.

39. The orally disintegrating nicotine tablet according to claim 38, wherein the pH regulating agent is an alkaline pH regulating agent, such as an alkaline buffering agent.

40. The orally disintegrating nicotine tablet according to claim 38 or 39, wherein the pH regulating agent comprises pH regulating agent selected from the group consisting of sodium carbonate, sodium bicarbonate, potassium carbonate, potassium bicarbonate, trometamol, amino acids, di-alkali hydrogen phosphate, tri-alkali phosphate, or any combination thereof.

41. The orally disintegrating nicotine tablet according to any of claims 38-40, wherein the pH regulating agent is selected from the group consisting of sodium carbonate, sodium bicarbonate, potassium carbonate, potassium bicarbonate, or any combination thereof.

42. The orally disintegrating nicotine tablet according to any of claims 38-41, wherein the pH regulating agent comprises or consists of sodium carbonate.

43. The orally disintegrating nicotine tablet according to any of claims 38-42, wherein the tablet comprises the pH regulating agent in an amount of at least 0.2% by weight of the tablet, such as at least 0.5% by weight of the tablet, such as at least 1% by weight of the tablet, such as at least 2% by weight of the tablet.

The orally disintegrating nicotine tablet according to any of claims 1-43, wherein the tablet comprises the pH regulating agent in an amount of at most 2% by weight of the tablet, such as at most 1.5% by weight of the tablet, such as at most 1% by weight of the tablet, such as at most 0.5% by weight of the tablet, such as at most 0.2% by weight of the tablet, such as at most 0.1% % by weight of the tablet.

45. The orally disintegrating nicotine tablet according to any of claims 1-44, wherein the tablet is free of effervescent agents.

The orally disintegrating nicotine tablet according to any of claims 1-45, wherein the tablet has a weight of no more than 600 mg, such as no more than 500 mg, such as no more than 400 mg, such as no more than 300 mg, such as no more than 200 mg, such as no more than 150 mg, such as no more than 120 mg, such as no more than 100 mg, such as no more than 50 mg.

47. The orally disintegrating nicotine tablet according to any of claims 1-46, wherein the tablet is a multilayer tablet comprising a first layer and a second layer.

48. The orally disintegrating nicotine tablet according to any of claims 1-47, wherein the tablet is a one-layer tablet.

49. The orally disintegrating nicotine tablet according to any of claims 1-48, wherein the tablet comprises flavor.

50. The orally disintegrating nicotine tablet according to any of claims 1-49, wherein the tablet is free of flavor.

51. The orally disintegrating nicotine tablet according to any of claims 1-50, wherein the tablet comprises high intensity sweetener.

52. The orally disintegrating nicotine tablet according to any of claims 1-51, wherein the tablet comprises less than 0.4% by weight of high intensity sweetener, such as no more than 0.2% by weight of high intensity sweetener, such as no more than 0.1% by weight of high intensity sweetener, such as free of high intensity sweetener.

53. The orally disintegrating nicotine tablet according to any of claims 1-52, wherein the tablet further comprises dissolution modifiers in an amount of at most 2 % by weight of the tablet, such as at most 1% by weight of the tablet, such as at most 0.5% by weight of the tablet, such as at most 0.25% by weight of the tablet, such as at most 0.1% by weight of the tablet.

54. The orally disintegrating nicotine tablet according to any of claims 1-53, wherein the tablet is free of dissolution modifiers.

55. The orally disintegrating nicotine tablet according to any of claims 1-54, wherein the tablet is free of mucoadhesives.

56. The orally disintegrating nicotine tablet according to any of claims 1-55, wherein the tablet comprises a lubricant, such as sodium stearyl fumarate (SSF) or magnesium stearate.

57. An oral nicotine tablet comprising the orally disintegrating nicotine tablet according to any of claims 1-56 as a first module and further comprising a second module, wherein the second module is different in composition compared to the first module.

58. The oral nicotine tablet according to claim 57, wherein the second module is a lozenge module.

59. The oral nicotine tablet according to claim 57 or 58, wherein the second module comprises a filler.

60. The oral nicotine tablet according to any of claims 57-59, wherein the second module comprises a filler in an amount of at least 40% by weight of the second module, such as at least 50% by weight of the second module, such as at least 60% by weight of the second module, such as at least 70% by weight of the second module, such as at least 80% by weight of the second module.

61. The oral nicotine tablet according to any of claims 57-60, wherein the filler of the second module comprises at least one sugar alcohol.

62. The oral nicotine tablet according to any of claims 57-61, wherein the filler of the second module comprises the at least one sugar alcohol in an amount of no more than 10% by weight of the filler, such as no more than 5% by weight of the filler, such as no more than 2% by weight of the filler, such as no more than 1% by weight of the filler, such as the second module being free of sugar alcohol.

63. The oral nicotine tablet according to any of claims 57-62, wherein the at least one sugar alcohol of the second module comprises sugar alcohol selected from the group consisting of xylitol, maltitol, mannitol, erythritol, isomalt, sorbitol, lactitol and any combination thereof.

The oral nicotine tablet according to any of claims 57-63, wherein the filler of the second module comprises no more than 10% by weight of bulk sweetener, such as no more than 5% by weight of the bulk sweetener, such as no more than 2% by weight of the bulk sweetener, such as no more than 1% by weight of the bulk sweetener, such as the second module being free of bulk sweetener.

65. The oral nicotine tablet according to any of claims 57-64, wherein the filler of the second module comprises a fiber.

The oral nicotine tablet according to any of claims 57-65, wherein the filler of the second module comprises a fiber in an amount of at least 50% by weight of the filler, such as at least 60% by weight of the filler, such as at least 70% by weight of the filler, such as at least 80% by weight of the filler, such as at least 90% by weight of the filler.

67. The oral nicotine tablet according to any of claims 57-66, wherein the filler of the second module comprises a water-soluble fiber.

68. The oral nicotine tablet according to any of claims 57-67, wherein the filler of the second module comprises a water-soluble fiber selected from the group consisting of indigestible maltodextrin, polydextrose, inulin, and any combination thereof.

73. The oral nicotine tablet according to any of claims 57-72, wherein the water insoluble fiber of the second module comprises microcrystalline cellulose.

74. The oral nicotine tablet according to any of claims 57-73, wherein the second module comprises a dissolution modifier.

75. The oral nicotine tablet according to any of claims 57-74, wherein the second module comprises dissolution modifier in an amount of at least 1% by weight of the second module, such as at least 2% by weight of the second module, such as at least 3% by weight of the second module.

76. The oral nicotine tablet according to any of claims 57-75, wherein the second module comprises nicotine.

77. The oral nicotine tablet according to any of claims 57-76, wherein the second module comprises 1-10% by weight of flavor, such as 2-8% by weight of flavor, such as 3-6% by weight of flavor.

78. The oral nicotine tablet according to any of claims 57-77, wherein the second module comprises less than 0.4% by weight of flavor.
