[0004] BACKGROUND
[0005] The graying of hair is one of the earliest and most noticeable signs of aging in humans. The phenomenon transcends cultural, geographic, and ethnic boundaries, holding deep social significance across the globe. As a result, there has been increasing interest in developing methods to reverse or slow down the process. Hair pigmentation, which gives rise to various hair colors such as black, brown, blonde, and red, is determined by the production of melanin, a pigment synthesized by melanocytes. These melanocytes are situated in the hair follicles and are responsible for producing two primary types of melanin-eumelanin and pheomelanin. The balance and ratio of these melanins, influenced by factors such as pH and cysteine levels within melanosomes, determine the resulting hair color.
[0006] Hair graying happens when melanocytes, the cells that produce melanin in hair follicles, gradually decrease. As people age, melanogenesis, the process of melanin production, slows down. Thus, leads to less melanin in the hair, causing it to turn gray or white. One major cause is oxidative stress, where reactive oxygen species (ROS) damage melanocytes. Psychological stress also speeds up graying by disrupting hormones, which affects melanocyte function. Malnutrition, particularly a lack of essential vitamins and minerals, further hampers melanin production.

Thus, results in fewer melanocytes and less melanin in the hair shaft, leading to gray or white hair. [0009] In an aspect of the present disclosure, a composition for anti-graying of hair is disclosed. The composition comprising: Aloe barbadensis Leaf Extract in a range of 0.05 to 0.20% by volume of the composition, Camellia sinensis leaf extract in a range of 0.05 to 0.30% by volume of the composition, Rosmarinus officinalis Leaf Oil in a range of 0.02 to 0.1% by volume of the composition, Hamamelis virginiana extract in a range of 0.5 to 2.00% by volume of the composition, hyaluronic acid in a range of 0.01 to 0.1% by volume of the composition, panthenol in a range of 0.2 to 2.00% by volume of the composition, saccharide isomerate in a range of 0.1 to 0.5% by volume of the composition, alpha glucosyl hesperidin in a range of 0.1 to 0.5% by volume of the composition, dimethyl isosorbide in a range of 0.5 to 2.00% by volume of the composition, Neopicrorhiza scrophulariiflora root extract in a range of 2.0 to 4.0% by volume of the composition, mandarin peel pastes in a range of 4.0 to 7.0% by volume of the composition, N-acetyl L-tyrosine in a range of 3.0 to 6.0% by volume of the composition, and one or more excipients.

[0080] The test composition was dissolved in dimethyl sulfoxide (DMSO) to prepare a stock concentration of 50% v / v. For cell treatment, a working solution of 5% v / v was prepared.

[0082] The WM 266-4 cells (2 x 105) were seeded on 60 mm cell culture plates containing DMEM medium supplemented with 10% FBS and incubated at 37°C in a 5% CO2 atmosphere. The cells were further incubated for 48 hours, after which they were trypsinized, and the resulting cell suspension was pelleted by centrifugation at 10,000 rpm for 10 minutes. The cell pellet was then resuspended in 500 pl of RIP A buffer, followed by sonication for 10 minutes to prepare the lysate. The protein concentration of the cell lysate was subsequently estimated using the Bradford method.

[0084] Standard Preparation- A stock solution of melanin was prepared at a concentration of 5 mg / mL in DMSO. The melanin stock solution was then diluted with assay buffer to obtain a 500 pg / mL melanin standard solution (Ml). A series of 1:2 serial dilutions was subsequently performed to generate the remaining melanin standards (M2 to M7). Sample and Standard Assay- Each sample well was loaded with 250 micrograms of protein lysate, and the volume was adjusted to 50 microliters using assay buffer. Subsequently, 50 microliters of signal enhancer were added to each well, and the plate was incubated at room temperature for 30 to 60 minutes.

The composition as claimed in claim 2, wherein the humectants are selected from a group comprising pentylene glycol in a range of 5.0 to 10.0% by volume of the composition, propanediol in a range of 1.00 to 3.00% by volume of the composition, and propylene glycol in a range of 1.0 to 3.0% by volume of the composition.
4. The composition as claimed in claim 2, wherein the moisturising agents are selected from a group comprising ethylhexylglycerin in a range of 0.2 to 0.6% by volume of the composition.
5. The composition as claimed in claim 2, wherein the scalp hydrator is selected from a group comprising: gluconolactone in a range of 0.30 to 0.70% by volume of the composition.
6. The composition as claimed in claim 2, wherein the anti greying agent is biotin in a range of 0.01 to 0.1% by volume of the composition.
7. The composition as claimed in claim 2, wherein the thickening and stabilising agent is selected from a group comprising acrylates / C 10-30 alkyl acrylate crosspolymer in a range of 0.1 to 0.5% by volume of the composition.
8. The composition as claimed in claim 2, wherein the chelating agent is selected from a group comprising tetrasodium ethylenediamine tetraacetic acid (EDTA) in a range of 0.01 to 0.1% by volume of the composition.
9. The composition as claimed in claim 2, wherein the conditioning agent is selected from a group comprising polyalkylene glycol mono butyl ether in a range of 2.0 to 5.0% by volume of the composition.

The composition as claimed in claim 2, wherein the preservatives are selected from a group comprising phenoxyethanol in a range of 0.2 to 0.6% by volume of thecomposition, sodium benzoate in a range of 0.4 to 0.80% by volume of the composition, and potassium sorbate in a range of 0.4 to 0.80% by volume of the composition.

A method of preparing the composition for anti-graying hair serum, the method comprising: a. adding demineralized (DM) water to a mixing vessel, followed by gradual addition of aery lates / cl 0-30 alkyl acrylate crosspolymer in a range of 0.1 to 0.5% by volume of the composition, tetrasodium ethylenediamine tetraacetic acid (EDTA) in a range of 0.01 to 0.1% by volume of the composition, Aloe Barbadensis Leaf Extract in a range of 0.05 to 0.20% by volume of the composition, dimethyl isosorbide in a range of 0.5 to 2.00% by volume of the composition, and propanediol in a range of 1.00 to 3.00% by volume of the composition; b. preparing one or more premixes in separate vessels, and adding sequentially in the mixing vessel; and c. adding propylene glycol in a range of 1.00-3.00 %, saccharide isomerate in a range of 0.1-0.5 %, Hamamelis virginiana (witch hazel) in a range of 0.5- 2.00 %, polyalkylene glycol mono butyl ether in a range of 2.00-5.00%, Rosmarinus officinalis Leaf Oil in a range of 0.02-0.1 %, and ethylhexylglycerin and phenoxyethanol in a range of 0.2-0.6 % to form the composition.
12. The method as claimed in claim 11, wherein: a. premix A comprises sodium benzoate in a range of 0.4 to 0.80% by volume of the composition and potassium sorbate in a range of 0.4 to 0.80% by volume of the composition, b. premix B comprises alpha glucosyl hesperidin in a range of 0.1 to 0.5% by volume of the composition, c. premix C comprises biotin in a range of 0.01 to 0.1% by volume of the composition and panthenol in a range of 0.2 to 2.00% by volume of the composition,d. premix D comprises Camellia sinensis leaf extract in a range of 0.05 to 0.30% by volume of the composition, e. premix E comprises hyaluronic acid in a range of 0.01 to 0.1% by volume of the composition, and f. premix F comprises Neopicrorhiza scrophulariiflora root extract in a range of 2.0 to 4.0% by volume of the composition, mandarin peel paste in a range of 4.0 to 7.0% by volume of the composition, and n-acetyl 1-tyrosine in a range of 3.0 to 6.0% by volume of the composition.
13. The method as claimed in claim 11, wherein the composition is prepared in a topical form selected from a group comprising: serum, lotion, cream, gel, spray, ointment, emulsion, or oil.
14. A method of treating a subject in need of hair anti-graying treatment, the method comprising: administering a composition to scalp of the subject, wherein the composition reduces the appearance of gray or white hair by stimulating melanocyte activity, enhancing melanin production, and promoting scalp health and hair pigmentation.
15. The method as claimed in claim 14, wherein the composition is administered in a dosage of 0.5 to 1.0 milliliters per application, once or twice daily.
16. Use of a composition for reducing gray or white hair, restoring natural hair pigmentation, stimulating melanogenesis, and improving scalp condition and overall hair appearance in a subject in need thereof.
