[0078] 1. Effects of Chinese herbal compresses on lipopolysaccharide-induced acute lung injury in rats
[0079] Experimental methods:
[0080] 1. Animal handling and grouping:
[0081] Healthy male Wistar rats, weighing 280-300g, were purchased from Beijing Weitong Lihua Experimental Animal Technology Co., Ltd. They were kept for 3 days before the experiment to adapt to the environment, and were given rat feed and free drinking water. The rats were randomly divided into 11 groups, namely the normal group, the model group, the embodiment 1-3 groups and the comparative example 1-6 groups, 5 rats in each group, a total of 55 rats. The abdomen of all rats was depilated before the experiment. Except for the normal group, the other groups were all used for model rat experiments.
[0082] 2. Establishment of rat model of acute lung injury (ALI) induced by lipopolysaccharide (LPS):
[0083] After the rats were anesthetized by intraperitoneal injection of sodium pentobarbital (50 mg / kg), the rats in the embodiment group and the comparative group were instilled with 5 mg / kg LPS poisoning in the trachea by a one-time tracheal puncture method. Method: The rats were fixed in a head-high position with an inclination angle of 60°, and the trachea was punctured at the cricoid cartilage with a No. 9 injection needle, and the LPS solution diluted with sodium chloride injection (LPS was purchased from Sigma-Aldrich, USA) was slowly instilled, with an instillation volume of 2 ml / kg and an instillation time of ≥15 s to prevent suffocation; the rats in the model group and the normal group of the positive control group were instilled with an equal volume of sodium chloride injection in the same way.
[0084] 3. Drug administration in nursing experiments:
[0085] The acute lung injury (ALI) model was successfully established 4 hours after administration. The rats in the normal group and the model group were injected with 10 ml / kg of normal saline of the same volume; the rats in the Example 1-3 group and the Comparative Example 1-6 group were respectively smeared with the corresponding ointment on the depilated part of the abdomen, and the ointment was applied at a dose of 150 mg / kg, and the drug was administered once / d for 7 consecutive days; among them, on the basis of applying the ointment, the oral prescription was administered by gavage in Example 2, and the oral prescription dose was 10 ml / kg, once / day, for 7 consecutive days. The specific medication is shown in Table 1; after the last gavage or application, fasting for 8 hours, and alveolar lavage fluid and bilateral lung tissue were collected for subsequent experiments.
[0086] 4. Bronchoalveolar lavage fluid (BALF) collection:
[0087] After the last administration, the rats were fasted for 8 hours, anesthetized, and the trachea was exposed by midline incision of the neck. After blunt dissection, a 1 ml syringe connected to a hose of an indwelling needle was inserted into the trachea, and air leakage was minimized. The right lung was lavaged 4 times with 1 ml of pre-cooled phosphate buffered saline (PBS) injected with a syringe. During the lavage process, the chest cavity was gently massaged to obtain about 3.2 ml of bronchoalveolar lavage fluid, which was centrifuged at 4 ° C, 3000 r / min for 15 min, and the supernatant was obtained and stored at -80 ° C.
[0088] 5. Lung tissue collection: After the rats were killed, they were fixed in supine position on a sterile operating table. The neck, chest, and abdomen were disinfected with ethanol. The thorax was opened aseptically and the complete lungs were removed. The left lung tissue was stored at -80°C.
[0089] Detection Methods
[0090] 1. Determination of the content of (TNF-α, IL-6, IL-1β) in bronchoalveolar lavage fluid BALF:
[0091] The contents of IL-6, TNF-α, and IL-1β in BALF were detected by enzyme-linked immunosorbent assay. The contents of IL-6, TNF-α, and IL-1β in BALF were detected strictly according to the instructions of the kit, and the results are shown in Table 2. Among them, IL-6 and IL-1β ELISA detection kits were purchased from Shanghai Biyuntian Biotechnology Co., Ltd.; TNF-α ELISA detection kits were purchased from Wuhan Boster Biotechnology Co., Ltd.
[0092] 2. Detection of SOD, MPO and MDA content in lung tissue
[0093] The lung tissue of the left lung was homogenized with cold physiological saline at a ratio of 1:9 (mass to volume ratio), and the supernatant was collected by centrifugation at 8000 rpm for 5 min. The protein content in the supernatant was quantified by the BCA method. According to the instructions, the MPO, SOD and MDA content in the supernatant were detected by the corresponding ELISA kit; the protein content was then used to quantify the corresponding indicators in the lung tissue. The results are shown in the figure. Figure 1 , Figure 2 and Figure 3 Among them, MPO, SOD, and MDA detection kits were purchased from Nanjing Jiancheng Bioengineering Institute.

[0096]
[0097] Table 2 Contents of TNF-α, IL-6 and IL-1β in bronchoalveolar lavage fluid of rats in each group
[0098]
[0099] The experimental results in Table 2 show that compared with the normal group, the content of TNF-α, IL-6, and IL-1β inflammatory mediators in the bronchoalveolar lavage fluid of the model group rats increased. After treatment with the Chinese medicine compress prepared by the present invention, the content of inflammatory mediators TNF-α, IL-6, and IL-1β in the rats of the embodiment group decreased significantly. The content of inflammatory mediators TNF-α, IL-6, and IL-1β in the comparative example decreased but not significantly. After the intervention of the Chinese medicine compress and the treatment of the oral prescription in Example 2, the content of inflammatory mediators TNF-α, IL-6, and IL-1β in the bronchoalveolar lavage fluid of the rats decreased significantly, and the treatment effect was better than that of other groups. It is suggested that the use of the Chinese medicine compress prepared by the present invention during treatment can improve the efficacy, alleviate clinical symptoms, reduce the level of inflammatory mediators, and improve neurological function, and on this basis, the effect can be increased by coordinating the oral prescription treatment.
[0100] Depend on Figure 1 , Figure 2 , Figure 3 It can be seen that compared with the normal group, the MPO activity and MDA content of the lung tissue of the model group were significantly increased, and the SOD activity was significantly decreased; compared with the model group, the embodiment group and the comparative example group reduced the MPO activity and MDA content of the lung tissue of ALI rats, and increased the SOD activity; compared with comparative examples 1-6, the MPO activity and MDA content of the lung tissue of the embodiment group were more significantly reduced, and the SOD activity was more significantly increased. The effect of the embodiment group was significantly higher than that of the comparative example group, and the effect of the embodiment group 2, which used both Chinese medicine compresses and oral medications, was slightly better than that of embodiments 1 and 3.
[0101] Inflammatory response is one of the important characteristics of ALI. This study showed that Chinese herbal compress can reduce the MPO activity and MDA content in the lung tissue of ALI rats, and increase the activity of SOD, suggesting that Chinese herbal compress has the effect of clearing away heat and detoxifying, and is suitable for reducing LPS-induced oxidative stress.
[0102] 2. Experiment on the treatment of deep burns with Chinese herbal compresses
[0103] (1) Experimental methods:
[0104] 1. Burn treatment:
[0105] 70 C57BL / 6J mice were purchased from Liaoning Changsheng Biotechnology Co., Ltd., half male and half female, 6-8 weeks old. The mice were allowed to fully adapt to the environment for 1 week before the experiment, and they had free access to feed and water. After anesthesia, the mice were fixed on the surgical board in a prone position, the back skin was fully exposed, and the limbs were fixed. The back skin of the mice was repeatedly wiped with 1% iodine cotton balls, and the hair on the back of the mice was gently shaved with a stainless steel double-sided blade clamped with a needle holder. After skin preparation, the back skin was wiped with 1% iodine again 2-3 times. A brass probe with a diameter of 8mm was used to induce full-thickness contact burns on the back skin of the mice. The temperature of the brass probe was set to 300℃ and contacted the skin for 2-3 seconds. After injury, the mice were intraperitoneally injected with 0.9% sodium chloride for fluid resuscitation. The recovery of the mice was closely observed within 2 hours after injury, and the vital signs of the mice were closely observed within 24-48 hours. After burns, the mice were housed separately according to their respective experimental groups, with free access to food and water.
[0106] 2. Post-burn care:
[0107] After the mice woke up from anesthesia, they were able to move freely and eat normally, indicating that the modeling was successful. The mice with successful burn model construction were randomly divided into: model group, positive control group, Example 1-3 group, Comparative Example 1-8 group, 5 mice in each group, a total of 13 groups, and 5 mice were randomly selected as the normal group. The temperature of the brass probe was set to 37°C in the normal group, and the rest were treated with the same method as the burn model construction; after the scald, the wound was locally disinfected with iodine tincture, and the positive control group was smeared with sulfadiazine silver cream, which was directly applied to the wound, with a thickness of about 1.5mm; the mice in Example 1-3 group and Comparative Example 1-8 group were respectively smeared with corresponding ointments, which were directly applied to the wound, with a thickness of about 1.5mm, and the wound was disinfected once every 2 days, and the corresponding drugs were replaced, and the treatment was 4 weeks in total. The wounds of mice in the normal group and the model group were disinfected once every 2 days, and no medicine was applied. Among them, Example 2 was based on the application of ointment, and the oral prescription was administered, and the dosage of the oral prescription was 10ml / kg, once a day.
[0108] 3. Collection of materials:
[0109] The measurement method mainly uses standard transparent squares to measure and scan into the computer, and then uses ImageJ image analysis software to calculate the wound area and the wound healing area ratio. Starting from the wound, the wound area of the back of the mouse was evaluated on the 0th, 7th, 14th, 21st, and 28th day of intervention. The camera was fixed with a tripod, the distance between the camera and the mouse was fixed each time, and the appropriate light source was selected to ensure that the multiple dependent variables were the same as much as possible. After taking pictures, the digitized wound photos were used by ImageJ software (NIH, Bethesda, MD) to measure the wound area at different time points to evaluate the wound healing rate.
[0110] (2) Detection indicators and methods
[0111] 1. Wound healing rate calculation method: Healing rate (%) = (SD0-SD 天数 ) / SD0×100%, where SD0 represents the wound area before treatment, SD 天数 Indicates the wound area after a certain number of days of treatment; the results are shown in Table 3.
[0112] Table 3 Comparison of wound healing rate (%) of mice at different time
[0113]
[0114] As shown in Table 3, compared with Comparative Examples 1-8, it can be seen that the wound healing ratio of Example 1 is larger and the speed is faster, which further proves the particularity of the fermentation strain of Salvia miltiorrhiza. Only the concentrated solution of Salvia miltiorrhiza fermented by the combined fermentation of Lactobacillus plantarum ATCC 8014 and Bacillus subtilis ATCC 6633 has good heat-clearing and detoxicating and burn-treating effects. Compared with Comparative Examples 5 and 6, it is obvious that Example 1 has good effects in terms of speed and area of treatment. The obtained experimental results can prove that the concentrated solution of Salvia miltiorrhiza fermented, Licorice extract and other Chinese medicine ingredients produce synergistic effects together, and the effects of heat-clearing and detoxicating and burn-treating are remarkable. Compared with Comparative Examples 7 and 8, the healing effect of Example 1 is significantly increased, indicating the importance of sesame stalk forging and storage treatment, and also embodies that the tung oil decoction liquid makes the effective ingredients in Chinese medicine fully boiled out, and then its heat-clearing and detoxicating and burn-treating effects increase.
[0115] The above is only a preferred specific implementation of the present application, but the protection scope of the present application is not limited thereto. Any changes or substitutions that can be easily conceived by any technician familiar with the technical field within the technical scope disclosed in the present application should be covered within the protection scope of the present application.
