[0019] Synthesis of N-(2-cyanophenyl)-N-methylacrylamide:

[0020]

[0021] According to a previous preparation report (Org. Chem. Front., 2024, 11, 4436-4441), a 100 mL round-bottom flask equipped with a magnetic stir bar was charged with 2-aminobenzonitrile (1.18 g, 10 mmol, 1.0 equiv.), anhydrous triethylamine (1.52 g, 15 mmol, 1.5 equiv.), and DCM (40 mL), then cooled to 0°C in an ice-water bath. Methacryloyl chloride (1.14 g, 11 mmol, 1.1 equiv.) was slowly added to the solution at 0°C, followed by stirring at room temperature for 12 hours. The solution was concentrated under reduced pressure, and the mixture was purified by flash column chromatography on silica gel (1:20 to 1:5 EtOAc:PE) to afford N-(2-cyanophenyl)methacrylamide (1.67 g, 9 mmol, 90%) as a white solid.

[0022] To a solution of N-(2-cyanophenyl)methacrylamide (1.67 g, 9 mmol) in THF (30.0 mL) at 0°C was added a magnetic stir bar and 60% NaH (540 mg, 13.5 mmol, 1.5 equiv.) and stirred for 30 minutes. Methyl iodide (1.92 g, 13.5 mmol) was then added and the reaction was warmed to room temperature. After stirring for 8 hours, the reaction was quenched with water and the aqueous layer was extracted with EtOAc (10 mL x 3). The combined organic layers were washed with brine, dried over Na2SO4, and concentrated in vacuo. The residue was purified by flash column chromatography on silica gel (1:10 to 1:3 EtOAc:PE) to afford 1aN-(2-cyanophenyl)-N-methylacrylamide (1.40 g, 7 mmol, 78%) as a yellow solid. Most substrate preparations followed the reaction pathway of 1a.

[0023]

[0024] According to a previous preparation report (Org. Chem. Front., 2023, 10, 3995), 2-bromo-6-fluorobenzonitrile A (20.0 mmol, 1.0 equiv.), MeNH2·HCl (30.0 mmol, 1.5 equiv.), and NaOH (30.0 mmol, 1.5 equiv.) were added to a 100 mL pressure bottle equipped with a magnetic stir bar. The mixture was stirred in CH3CN (40 mL) at 80°C under air for 12 h. After completion of the reaction, the mixture was cooled to room temperature, extracted with ethyl acetate (100 × 3 mL), and washed with saturated brine (100 mL). The organic layer was dried over anhydrous sodium sulfate, filtered, and concentrated in vacuo to obtain intermediate B.

[0025] To a 100 mL pressure bottle equipped with a magnetic stir bar, substituted phenylboronic acid (15 mmol, 1.5 equiv.), PdCl2(PPh3)2 (0.3 mmol, 0.02 equiv.), and Cs2CO3 (42.0 mmol, 2.8 equiv.) were added, along with MeCN (150 mL) and H2O (7.5 mL), respectively. The reaction mixture was stirred at 80°C under a nitrogen atmosphere for 12 h. After completion, the mixture was cooled to room temperature and extracted with ethyl acetate (100 x 3 mL). The combined organic layers were washed with saturated brine (100 mL), dried over anhydrous Na2SO4, filtered, and concentrated in vacuo to afford crude product C, which was not further purified.

[0026] Compound C (10 mmol) and triethylamine (20 mmol, 2.0 equiv.) obtained above were dissolved in DCM (24 mL) and the corresponding methacryloyl chloride (1.5 equiv.) was added dropwise at 0°C. The reaction mixture was stirred at 0°C overnight. After completion of the reaction, the resulting solution was extracted with DCM (30 × 3 mL). The combined organic layers were washed with saturated brine (30 mL), dried over anhydrous Na2SO4, filtered, and concentrated in vacuo. Purification by column chromatography on silica gel using petroleum ether / ethyl acetate (20:1 to 10:1) as the eluent afforded the desired substrate 1. BRIEF DESCRIPTION OF THE DRAWINGS

[0047] To a solution of N-(2-cyanophenyl)methacrylamide (1.67 g, 9 mmol) in THF (30.0 mL) at 0°C was added a magnetic stir bar and 60% NaH (540 mg, 13.5 mmol, 1.5 equiv) and stirred for 30 minutes.
