Description

TECHNICAL FIELD [1] The present disclosure relates to the field of gel preparation technology, in particular to aWestern Blot gel preparation device. BACKGROUND ART [2] Western Blot is also called a protein imprinting method, which is commonly used in molecular biology, biochemistry and immunogenetics. The basic principle is that cells or biological tissue samples are colored with specific antibodies after being processed by gel electrophoresis. The expression of a particular protein in the sample is determined by analyzing the location of the staining and the depth of staining. [3] In practical application, the parameters of the SDS-PAGE gel to be prepared should be determined firstly according to the molecular weight range of the mixed protein sample and the final molecular weight of the target protein to be detected. After preparing the SDS-PAGE gel, the protein mixed sample is separated by the gel electrophoresis, and then the protein in the gel is transferred to a chemically inert macromolecule membrane by protein electroimprinting, then the antibody (primary antibody) of the target protein to be detected is immunologically combined with the target protein on the membrane, and then the labeled (such as HRP) secondary antibody is combined with the primary antibody. The presence and abundance of the target protein are determined by detecting the marker of the secondary antibody. [4] The preparation process of the conventional SDS-PAGE gel generally uses 8 reagents: deionized water, acrylamide (Acr), methylenebisacrylamide (Bis), Tris base, HC1, Ammonium persulfate (AP), and TEMED. Four reagents are prepared in advance: 30% acrylamide solution (mass ratio of acrylamide: methylene bisacrylamide is 29:1), pH 8.8 separation gel buffer, pH 6.8 concentrated gel buffer and 10% (m / v) Ammonium persulfate solution. TEMED is generally a commercial reagent. [5] The preparation of the conventional SDS-PAGE gel includes three stages: I) according to the molecular weight range of the target protein to be separated, the corresponding SDS-PAGE system is selected, which can prepare such as 10%, 12%, 15% and other different concentrations of separation gel; 2) the corresponding concentration of separation gel is prepared according to the selected system, after adding TEMED, the solution is quickly mixed, and the separation gel solution is injected along the side of the gel preparation plate to a distance of 1.5 cm from the upper edge of the second glass plate. And then the separation gel is covered with isopropanol (or deionized water), a clear straight line will appear at the interface between the separation gel and the isopropanol layer after about 30-60 minutes, which indicates that the separation gel has fully polymerized, and at this point, the gel preparation plates are inclined and residual liquid are absorbed by using filter paper; 3) the corresponding concentration of concentrated gel is prepared according to the selected system, after adding TEMED, the solution is quickly mixed, and the concentrated gel solution is injected along the side of the gel preparation plate to the top of the short glass plate, and the sample comb is inserted as soon as possible. After about 15-30 minutes, the gel preparation is completed and the electrophoresis may be performed. [6] Since there are many samples running in practical operation, many SDS-PAGE gels usually need to be prepared at the same time, and are generally operated by pure manual operation. However, two problems are brought about: firstly, due to a narrow dispensing plate opening, a small injection amount and a slow operation, the injection time is too long, so that the gel solution is frozen before injection. Especially, when the concentrated gel is injected, it is not uncommon for the concentrated gel to solidify before inserting a sample comb; secondly, a lot of gel is injected manually, and the problem of inconsistent height of the separation gel often exists. Especially, when using a gel preparation mould with a long time, gel leakage may not be avoided. Separation gels with different heights will cause the target protein to stay at different positions, which affects subsequent strip analysis. Therefore, there is an urgent need for an efficient, simple and time-saving gluing device, which can meet the requirement of simultaneously preparing multiple pieces of gels. [7] The patent publication NO. CN 111545138 A provides an automated gel preparation system, which is too complex and inconvenient to operate. The patent with publication No. CN 217910206 U is optimized on the basis of the patent publication NO. CN 11545138 A, but is also very cumbersome, and the whole gel preparation system is too large, occupies space and is not easy to clean. The patent with publication No. CN 211785304U mainly aims to improve the leakage phenomenon, and is only limited to preparing one piece of SDS-PAGE gel. The patent publication NO. CN 103013982 A only provides a new formulation scheme for an SDS-PAGE gel, without involving a gel preparation device. DETAILED DESCRIPTION OF THE EMBODIMENTS

[37] The technical solutions in the embodiments of the present disclosure will be clearly and completely describes below with reference to the drawings in the embodiments of the present disclosure. It is apparent that the described embodiments are only a part rather than all of the embodiments of the present disclosure. All other embodiments obtained by those skilled in the art based on the embodiments of the present disclosure without creative efforts shall belong to the scope of protection of the present disclosure.

[38] The embodiments aim to provide a highly efficient and simple, time-saving and labor-saving Western Blot gel preparation device, which not only can meet needs of preparing multiple pieces of gels at the same time, but also can ensure easy operation and easy cleaning without occupying space.

[39] In order to make the above-mentioned objectives, features, and advantages of the present disclosure clearer and more comprehensible, the following further describes the present disclosure in detail with reference to the drawings and specific embodiments.

[40] As shown in Figs. 1-14, the present embodiment provides a Western Blot gel preparation device for Western Blot protein electrophoresis, including a gel preparation base 1, an injection plug-in component 2 and multiple gel preparation plates 3. The gel preparation base 1 is provided with a first liquid adding tank 4. The multiple gel preparation plates 3 are located in the gel preparation base 1. The first liquid adding tank 4 is in communication with an inner space of each of the gel preparation plates 3. The injection plug-in component 2 is provided on the gel preparation plates 3. The injection plug-in component 2 includes a liquid adding structure and multiple sample combs 25. The liquid adding structure is provided with a second liquid adding tank 15. The second liquid adding tank 15 is in communication with the inner space of each of the gel preparation plates 3. The sample combs 25 are detachably connected to the liquid adding structure, and each of the sample combs 25 is insertable into the inner space of a corresponding one of the gel preparation plates 3.

[41] Specifically, in the present embodiment, the gel preparation base 1 further includes a base 6 and transverse pinch doors 5. The transverse pinch door 5 is hinged at a mounting opening 12 of the lower end of the base 6 through a notch protrusion. One end of the base 6 is provided with the first liquid adding tank 4, and an outer side of a bottom of the first liquid adding tank 4 is arc-shaped, which is convenient for gel liquid to flow into the base 6 while saving the amount of the gel liquid. Multiple bottom gel strips 9 are provided in the base 6. Gel plate grooves 8 are formed between adjacent bottom gel strips 9 and between the outermost bottom gel strip 9 and the transverse pinch doors 5. A bottom of the gel preparation plate 3 is arranged in a corresponding one of the gel plate grooves 8. A bottom of the first liquid adding tank 4 is in communication with the gel plate grooves 8 and the inner space of the gel preparation plate 3. After the gel preparation plates 3 are inserted into the gel plate grooves 8 of the gel preparation base 1, side surfaces of the gel preparation plates 3 may be closely fitted with the bottom gel strips 9, so that the bottom gel strips 9 separates the gel preparation plates 3 one by one, and the gel liquid is limited at bottom ends of the gel preparation plates 3 by closely fitting the bottom gel strips 9 and the side surfaces of the gel preparation plates 3. The transverse pinch doors 5 are closed, so as to enable a lower portion of each gel preparation plate 3 to contact with the bottom gel strip 9 closely via clamping opening protrusions, so that the gel liquid may only enter the gel preparation plate 3 from a lower side of the gel preparation plate 3 and may not leak out of a gap between the gel preparation plate 3 and the bottom gel strip 9.

[42] In the present embodiment, two ends of the base 6 are provided with respective fixing plates. One fixing plate is directly connected with a bottom plate of the base 6, a gap is left between a bottom of the other fixing plate and the bottom plate, and the gap is lower than the bottom gel strip 9, so that the base 6 is in communication with the first liquid adding tank 4 to form a connector, and the heights of separation gels in all the gel preparation plates 3 are consistent during injection. A distance between the two fixing plates is consistent with the length of the gel preparation plates 3. The inner walls of the two fixing plates are provided with respective multiple fixing notches 7. The distance between every two adjacent fixing notches 7 is consistent with the thickness of the gel preparation plate 3. The gel preparation plate 3 is slidably connected with the fixing notch 7.

[43] In present embodiment, the injection plug-in component 2 further includes multiple liquid outlets 22. The multiple liquid outlets 22 are located at a bottom of the injection plug-in component 2. The multiple liquid outlets 22 are in communication with respective second liquid adding tanks 15. A lower end of an interior of the second liquid adding tank 15 is in a slope shape, and the liquid outlets 22 facing towards an inner side of the second liquid adding tank 15 increase flow of the gel liquid while saving the amount of the gel liquid. Each of the liquid outlets 22 is in one-to-one correspondence with and in communication with the inner space of a corresponding one of the gel preparation plates 3.

[44] In present embodiment, the injection plug-in component 2 further includes a bracket. Each of the liquid adding structures further includes a transverse plate 16 extending inward from an upper end of the second liquid adding tank 15. The length of the transverse plate 16 is approximately 1 / 4-1 / 5 of the length of the comb tooth of the sample comb 25. The transverse plate 16 is located at the inner side of the second liquid adding tank 15. The transverse plate 16 is detachably connected with the bracket. The bracket is detachably connected with each of the sample combs 25. The inner side of each second liquid adding tank 15 is provided with multiple vertical slots 17. The transverse rod 19 of the bracket is slidably connected with the vertical slot 17.

[45] In present embodiment, the gel preparation plate 3 includes a first glass plate 13 and a second glass plate 14. The first glass plate 13 is arranged on one side of the second glass plate 14. A lower end of the first glass plate 13 is flushed with a lower end of the second glass plate 14. An upper end of the second glass plate 14 is lower than an upper end of the first glass plate 13. Two ends of the first glass substrate 13 have respective protrusions extending towards a direction of the second glass plate 14. The height of the protrusion is equal to a thickness of the SDS-PAGE gel that needs to be prepared. A surface of the protrusion is subjected to a glassification treatment, and the glassification surface of the protrusion is attached to the second glass plate 14 to prevent the gel liquid from leaking. The first glass plate 13, the second glass plate 14 and the protrusions form the inner space of the gel preparation plate 3.

[46] In present embodiment, a first base 10 and a second base 11 are disposed in each of the gel plate grooves 8. The first base 10 is in contact with a bottom of the first glass plate 13. The second base 11 is in contact with a bottom of the second glass plate 14. The length of each of the first base 10 and the second base 11 is equal to the width of the glassification surface. The height of each of the first base 10 and the second base 11 is equal to the height of a gap between a bottom of the other fixing plate and the bottom plate. An inner side of an upper portion of each protrusion is provided with a slope 23. The liquid outlet 22 of the injection plug-in component 2 has an angular surface. The angular surface of the liquid outlet 22 is attached to the slope 23 of the protrusion, which ensures that the concentrated gel liquid may flow into an upper end of the gel preparation plate 3 via the raised slope 23. If the angular surface of the liquid outlet 22 protrudes too far, it may be blocked by an edge of the front end of the sample comb 25. After the gel preparation plates 3 are inserted into the gel plate grooves 8 of the gel preparation base 1, the gel preparation plates 3 does not completely abut against the base 6, but are elevated by a height of 2-3 mm from the bottom plate of the base 6 through the first base 10 and the second base 11, so that the gel liquid in the first gel liquid adding tank 4 may drain into the gel preparation plates 3 from the gap between the bottom of the other fixing plate and the bottom plate.

[47] In present embodiment, the bracket includes a connector 20 and multiple transverse rods 19. Each of the transverse rods 19 is connected with the connector 20. Handles 21 are provided on the connector 20. Two ends of each of the transverse rods 19 are connected with the respective vertical slots 17 in a sliding manner. When the transverse rods 19 slide to the bottom ends of the vertical slots 17, each of the sample combs 25 is fully inserted into the concentrated gel in the gel preparation plate 3. A magnetic element 18 is provided on an upper side of each of the transverse rods 19. The width of the magnetic element 18 is equal to the width of the transverse rod 19. A first iron sheet 24 is provided on a position, corresponding to the magnetic element 18, of the transverse plate 16. A connection between the bracket and the liquid adding structure is achieved by means of the attraction of the magnetic element 18 and the first iron sheet 24. A second iron sheet is provided at a position, corresponding to the magnetic element 18, on an upper surface of each of the sample combs 25. A connection between the bracket and the sample comb 25 is achieved by the attraction between the magnetic element 18 and the second iron sheet. In present embodiment, the transverse rod 19, the connector 20 and the handle 21 are made of plastic. The sample combs 25 may be inserted into the respective gel preparation plates 3 at the same time, which saves time and labour and ensures the uniformity of the prepared gels, and greatly simplifies the process and time for the gel preparation. During the process of conventional preparation of concentrated gel, it is necessary to insert the sample combs 25 as soon as possible after injection of the gel liquid. Therefore, when preparing multiple pieces of SDS-PAGE gels with conventional devices, the operator is often in a hurry to prevent the gel liquid from solidifying. In the present embodiment, since integrated injection plug-in component 2 is used, the sample combs 25 may be inserted into all the gel preparation plates 3 at the same time by only pressing gently after injection, thereby greatly simplifying operation steps, and saving operation time.

[48] As shown in Fig. 10, in an empty state, the sample combs 25 are hung on the gel preparation plates 3 through the transverse rods 19 and the transverse plates 16. When a downward external force is applied by the handles 21, as shown in Fig. 11, the transverse rods 19 are detached from the magnetic force attraction between the transverse plates 16 and the transverse rods 19, and drive the sample combs 25 to be inserted into the gel preparation plates 3.

[49] Taking simultaneous preparation of six pieces of SDS-PAGE gels as an example, the height of the gel preparation base 1 is 6.35 cm, the width of the first liquid adding tank 4 is 0.8-1.0 cm, the gap between the bottom of the other fixing plate and the base plate is 2-3 mm, the height of the bottom gel strip 9 is 8 mm, and the width of the gel plate groove 8 is 4.51 mm (taking a gel preparation plate with a thickness of 1.0 mm as an example, the width of the gel plate groove 8 may be adjusted appropriately according to different gel preparation plates). The width of the fixing notch 7 is 4.51 mm, the height of the second liquid adding tank 15 is 10.5 mm, and the width of the transverse plate 16 is about 16-20 mm.

[50] In practical application, the Western Blot gel preparation device may be used to prepare 6, 8 or 10 pieces of SDS-PAGE gels at the same time. If it is not necessary to prepare so many gels at one time, plastic plate models may also be used to replace the gel preparation plates 3 and be inserted into the gel plate grooves 8 to prevent the gel liquid from leaking from the bottom. It is worth mentioning that the gel plate grooves 8 at the bottom of the Western Blot gel preparation device may be slightly adjusted according to the thickness of the SDS-PAGE gel to be prepared, which may be 1 mm, and can also be 0.75 mm or 1.5 mm. The sample combs 25, which are attached to the transverse rods 19 of the injection plug-in component 2 above, may be 10-hole combs, 12-hole combs or 15-hole combs according to experimental requirements.

[51] When used in Western Blot experiment, firstly, all the gel preparation plates 3 are inserted into the gel plate grooves 8 of the gel production base 1, and the transverse pinch doors 5 are closed to ensure that there are no gaps exist, except for the communication between the bottoms of the gel preparation plates 3 and the bottom of the first liquid adding tank 4 of the gel production base I. A concentration of SDS-PAGE separation gels to be prepared is selected according to experimental requirements, and separation gels of corresponding concentrations are prepared according to the selected preparation system, and some more pieces of separation gels are prepared (in the present embodiment, about one more piece of gel is approximately prepared, i. e„ 6+1 pieces of gels are prepared). After being mixed, the separation gel liquid is injected along a side wall of the first liquid adding tank 4 of the gel production base 1 to the required height, and the injection of the gel liquid is stopped. At this time, the separation gel liquid enters into the gel preparation plates 3 through the lower communicating gaps, and the heights of the separation gels in all the gel plates 3 are consistent with the height of the remained separation gel liquid in the first liquid adding tank 4. The injection plug-in component 2 is supported on upper ends of the gel preparation plates 3 (in this case, the sample combs 25 are not connected), isopropyl alcohol (or deionized water) is injected into the second liquid adding tank 15 on both sides thereof, flows into the gel preparation plates 3 through the liquid outlets 22, and covers the separation gels for 30-60 minutes. When it is observed that the separation gels in the first liquid adding tank 4 of the gel preparation base 1 solidifies, the SDS-PAGE separation gels are prepared.

[52] The injection plug-in component 2 is removed from the upper ends of the gel preparation plates 3, the gel preparation base 1 and the gel preparation plates 3 are inclined integrally, residual liquid is adsorbed by using filter paper and then put back into place. The injection plug-in component 2 is naturally air-dried and then connected with the sample combs 25, and are again placed at the upper ends of the gel preparation plates 3. Concentrated gels with a corresponding concentration are prepared according to the selected system, and are quickly blended uniformly after addition of TEMED, and are injected through the second liquid adding tanks 15 at left and right sides of the injection plug-in component 2, and when it is observed that the concentrated gels are injected from the liquid outlets 22 to a top of the second glass plate 14, the injection of the concentrated gels are stopped. The handles 21 above the injection plug-in component 2 are slightly pressed downwards, so that the bracket (the transverse rods 19, the connector 20 and the handles 21) is detached from the magnetic attraction between the transverse plates 16 and the bracket, and drive the sample combs 25 to be inserted into the gel preparation plates 3. After standing for 15-30 min, all of multiple pieces of homogeneous SDS-PAGE gels are prepared, and may be extracted and subjected to electrophoresis.

[53] The present embodiment provides an efficient and simple, time-saving and labour-saving gel preparation device, which solves the problem of separately injecting a lower layer separation gel and an upper layer concentrated gel by means of a simple and small gel preparation base 1 and the injection plug-in component 2. The first liquid adding tank 4 is connected with the gel preparation base 1 to form a structure of communicating vessel, so that the heights of the separation gels in all the gel preparation plates 3 are consistent during injection, so that the requirement of uniform gel preparation is completely satisfied. By designing such as bracket (the transverse rods 19, the connector 20 and the handles 21) and the magnetic elements 18, the second iron sheets which are integrated on the injection insert 2, the sample combs 25 are inserted into all the gel preparation plates 3 at the same time, and the gel preparation process is further simplified. In conclusion, the gel preparation device for the Western Blot protein electrophoresis provided by the present embodiment not only ensures fast and efficient multi-block gel preparation, but also ensures the homogeneity of the prepared gels, and is very suitable for popularization and application.
