AI Technical Summary

Technical Problem

Current treatments for malignant tumors with high microsatellite instability (MSI-H) are limited, and there is a need for specific targeted inhibitors to overcome immune tolerance and improve treatment outcomes in cancers such as colorectal, endometrial, and ovarian cancers.

Method used

Development of spiro-triazolopyrimidine analogues that act as inhibitors of the Werner Syndrome RecQ DNA helicase (WRN), which are designed to selectively target and inhibit WRN in MSI-H tumor cells, thereby inducing DNA cleavage, apoptosis, and cell cycle arrest.

Benefits of technology

The spiro-triazolopyrimidine analogues effectively inhibit WRN, leading to selective killing of MSI-H tumor cells and potentially enhancing the efficacy of existing treatments for MSI-H cancers.

Description

SPIRO-TRIAZOLOPYRIMIDINE ANALOGUES AS WRN INHIBITORS AND USES THEREOFFIELD

[0001] This application relates to spiro-triazolopyrimidine analogues as inhibitors of Werner Syndrome RecQ DNA helicase (WRN) , their preparation and uses thereof.BACKGROUND

[0002] DNA mismatch repair deficiency (dMMR) is a common cause of malignant tumor development. dMMR can be found in about 10-30%of colorectal cancer, endometrial cancer, ovarian cancer and gastric cancer. dMMR can lead to frequent insertion or deletion of the microsatellite (Microsatellite, MS) DNA recombination list during the recovery process, and the recovery errors of MS can not be corrected. The resulting accumulation of errors can change the sequence length or base pairs of MS, which is called Microsatellite instability (MSI) . Currently, some progress has been made in the treatment of malignant tumors with high frequency of microsatellite instability (MSI-high, MSI-H) . For example, pembrolizumab (anti-PD-1 monoclonal antibody) , which can significantly prolong progression-free survival rate in metastatic colorectal cancer with MSI-H and dMMR (MSI-H-dMMR) , has been approved as first-line therapy in these cancers. However, the clinical application of these immunotherapies is still limited, and immune tolerance can occur. Therefore, there is an urgent need to develop specific targeted inhibitors in colorectal cancer, endometrial cancer, ovarian cancer and other MSI-H indications.

[0003] WRN is a member of the RecQ helicase family. It can bind and hydrolyze ATP. With the energy supply of ATP, ATPase can unwind DNA double strands and G-quadruplexes and other complex DNA secondary structures. Studies have found that depletion or inhibition of WRN can selectively kill colorectal tumors and ovarian cancer with MSI-H characteristics (Chan, E.M et. al, WRN helicase is a synthetic lethal target in microsatellite unstable cancers, Nature 568, 551-556 (2019) ; Lieb, S et. al, Werner syndrome helicase is a selective vulnerability of microsatellite instability-high tumor cells. eLife 8, e43333 (2019) . The anti-tumor mechanism via WRN inactivation / absence or inhibition may be achieved by interfering with the unwinding process of the secondary structure of DNA formed by the extensively expanded dinucleotide TA repeat sequence in MSI-H tumor cells, thereby inducing double-stranded DNA cleavage in the  cells and promoting apoptosis and cell cycle arrest (van Wietmarschen, N. et. al. Repeat expansions confer WRN dependence in microsatellite-unstable cancers. Nature 586, 292-298, 2020) . Therefore, WRN can be a synthetic lethal target for a class of malignant tumors with MSI-H characteristics, including, for example, colorectal cancer, endometrial cancer, gastric cancer, and ovarian cancer. Specific WRN inhibitors can be useful in the treatment of tumors with dMMR and MSI-H.SUMMARY

[0392] II. Results

[0393] The above in vitro assays were used to measure the activity of the compounds disclosed herein to inhibit the ATPase hydrolase of WRN and BLM in vitro, and the IC50 is shown in Table 1.

[0394] Table 1: IC50 (nM) inhibition on WRN ATPase and BLM ATPase

[0395] It can be seen from the data in Table 1 that the compounds disclosed herein have substantially selective inhibitory activity on WRN ATPase: their inhibition on BLM enzyme activity is weak. The compounds disclosed herein can be considered as selective WRN inhibitor.

[0397] MSI-H Tumor Cell Proliferation Inhibition Assay

[0398] I. Assay

[0399] Cell Titer Glo method is used to measure the anti-proliferation activity of the compound on tumor cell lines, and its growth half inhibitory concentration (IC50, μM) is determined under the experimental conditions.

[0400] Culture of SW48 and SK-CO-1 cells (Cell Titer Glo method) -Colon cancer cell lines SW48 (Cat#CCL-231) and SK-CO-1 (Cat#HTB-39) are purchased from the American Type Culture Collection (ATCC) . SW48 cells are cultured in RPMI-1640medium containing 20% fetal bovine serum (Gibico) , and SK-CO-1 cells are cultured in EMDM medium containing 10%fetal bovine serum (Gibico) . These two types of cells are cultured in a cell incubator with a constant temperature of 37 degrees and 5%CO2. SW48 cancer cells have MSI-H characteristics while SK-CO-1 cancer cells have MSS characteristics.

[0401] Measurement of IC50 post 96 hour treatment

[0402] Day 0: after the cells are digested into single cells by 0.05%trypsin (Gibico) , they were seeded at a concentration of 1000 / well (SW48 cell) and 750 / well (SK-CO-1) on 384 well culture plate with white opaque bottom (Corning Cat # 3764) . After inoculation, the cells are cultured overnight in a cell incubator at 37℃containing 5%CO2.

[0403] Day 1: after the compound is formulated into a 10mM stock solution with DMSO, 9 working concentrations are serially diluted (the dilution factor was 3) . The compound solution is then added to the culture medium at a ratio of 1: 1000. DMSO solution is used as negative control. Each sample is assayed in duplicate.

[0404] Day 5: after 96 hours, the CellTiter-Glo kit (Cat#G7573) from Promega is used to measure the ATP level in the cells to evaluate the proliferation activity of the cells. 10μL CellTiter Glo detection reagent is added to each well and the wells are incubated at room  temperature for 10minutes. The luciferase signal is read by using the Envision multifunctional detector from PerkinElmer.

[0405] Data analysis: the level of cell proliferation activity is significantly correlated with the intracellular ATP level measured by the above-mentioned CellTiter Glo method, so this luciferase signal reading can be used to represent the intracellular proliferation activity. Graphpad Prism9software is used for data analysis, IC50 is calculated using the following formula:

[0406] Y=Bottom+ (Top-Bottom)  /  (1+10^ ( (LogIC50-X) *HillSlope) , obtained by fitting the curve

[0407] with the four-parameter method, where Y values are raw luciferin readings per well minus average of media-only no-cell control wells.

[0408] II. Results

[0409] The above SW48 and SK-CO-1 proliferation inhibition assay measured the activity of the compounds disclosed herein, and the IC50 inhibition data is shown in Table 2.

[0410] Table 2: IC50 (nM) inhibition on the proliferation of SW48 and SK-CO-1 cells

[0411] It can be seen from the data in Table 2 that the compounds disclosed herein have substantially selective inhibitory activity on cell SW48, and their inhibition on cell SK-CO-1 is weak. The compounds can selectively kill tumor cells with MSI-H characteristics.
