[0074] Experimental Example 1. Evaluation of the ability to inhibit nitric oxide (NO) production

[0075] To evaluate the inhibition of nitric oxide production, experiments were performed using the GRIESS method. Raw 264.7 cells pre-cultured in growth medium (DMEM supplemented with 10% FBS) were seeded at 1–2 × 10 in 24-well tissue culture plates. 5 cells / ml and cultured for one day in a 5% CO2 incubator. The medium was removed, and starvation was maintained for 12 hours with serum-free medium, and then the samples were pretreated for 30 minutes at various concentrations, and then lipopolysaccharide (LPS) was added at a concentration of 500 ng / ml and cultured for 18 hours. After culture, the supernatant was transferred to a 96-well plate, and the GRIESS reactant was added and reacted at room temperature for 15 minutes. The absorbance at 540 nm was measured using an ELISA reader, and the inhibition rate of NO production was calculated. According to the following mathematical equation 1, the inhibition rate of NO production compared to the negative control group (LPS 500 ng / ml treatment group) was calculated as follows. The results are shown in Table 4.

[0080]

[0081] Experimental Example 2. Evaluation of inhibition of melanin production and tyrosinase activity.

[0082] MNT-1 melanoma cells were seeded at 1–2 x 10 in 6-well plates. 5 The cells were dispensed at a concentration of 10 cells / ㎖ and cultured for 24 hours. After treating the sample and culturing for 72 hours, the cells were harvested. The cells were centrifuged at 13,000 rpm for 1 minute, the supernatant was removed, and 300 μl of 0.5% Triton X-100 solution was added to the pellet to lyse the cells. This was centrifuged again at 13,000 rpm for 3 minutes, and the pellet and supernatant were collected separately. The cell pellet was added with 100 μl of 0.5 N NaOH and incubated overnight to dissolve melanin, and the absorbance at 450 nm was measured using an ELISA reader.

[0083] To measure tyrosinase activity, 100 μl of the cell lysis supernatant recovered above was taken, 1 mM L-DOPA solution was added thereto, and the reaction was performed at 37°C for 1 hour. The reaction solution was measured for absorbance at 450 nm using an ELISA reader. The melanin production inhibition rate and tyrosinase activity were calculated according to the following mathematical equations 2 and 3. In this experimental example 2, the absorbance of the negative control group was set at 100%. The results are shown in Table 5 below.
