[0107] The compositions described herein can be made by conventional techniques generally known to those skilled in the art. For example, the main water is added to a container equipped with a central turbine agitator, and the prebiotic (e.g., inulin) is premixed with 4-5% water; the gum is premixed with glycerin. The premix is then added to the main batch and heated to about 60°C. After the mixture has cooled to about 40°C or less, caprylyl / caprylyl glucoside and decyl glucoside are added. Then, the oil and antioxidant are slowly added, followed by the aroma. Then, the organic acid is added and titrated to reach the desired pH of about 4.
[0108] Example 2
[0109] Foam testing or characterization can be performed using a variety of methods. For example, foam structure and characterization can be performed using a Dynamic Foam Analyzer Model DFA 100 available from Kruss USA of Matthews, North Carolina.
[0110] Foam distribution can also be measured using a mechanical cylinder vibration foam method, such as the Gaum Foam Method.
[0111] The foam test measurement is used to evaluate the foam distribution of the exemplary body wash and hand soap of the present invention and the comparative examples described herein. The measurement is performed using a mechanical graduated cylinder vibration foam method and a Gaum foam height machine purchased from Gaum, Inc. of Robbinsville, New Jersey. Flash foam, maximum foam and drain time are measured.
[0112] Procedure: Weigh 75g of the test body wash or hand soap in a beaker. Weigh 425g of deionized water in a 1000mL glass beaker and place a 1" magnetic stir bar in it. Place the beaker on a stir / hot plate and begin mixing until the body wash or hand soap is completely dispersed. Mix at a speed that produces very little foam. Heat the solution to 41-42°C. Once the solution reaches 41-42°C, the solution is ready. Make enough to use in 3 graduated cylinder tests. Keep the sample at a constant temperature of 41-42°C when not being used for testing. Heat the DI water to 45°C. Fill a 500mL graduated cylinder with 45°C DI water. Allow the DI water to sit in the graduated cylinder for a few minutes. Drain the DI water from the graduated cylinder before pouring the body wash solution and testing. When the body wash test solution temperature is reached, drain the 500mL glass graduated cylinder. When the body wash test solution reaches 41-42°C, pour 100g of the body wash test solution into the 500mL graduated cylinder. Tilt the graduated cylinder at an angle and pour it into the graduated cylinder at a slow rate so that almost no foam is generated when pouring. Put in a Teflon rod and seal it with a rubber stopper. Connect a 500mL graduated cylinder to a rotator assembly (Gaum foam machine) to keep the graduated cylinder in a vertical plane while rotating. Rotate the graduated cylinder at a constant speed (30RPM) for the following cycle:
[0113] a. Flash foam measurement
[0114] 8 cycles: After 8 cycles the foam volume is read. For each product the average and standard deviation of 3 samples are taken.
[0115] b. Maximum foam and drainage time measurement
[0116] 12 cycles: Start timing immediately after the cycle is completed. Read the maximum foam volume after 12 cycles of maximum foam and stop timing once the liquid portion in the measuring cylinder reaches the discharge time of 100 mL.
[0117] For each product, the average and standard deviation of 3 samples were taken. Drain time is defined as the time measured from the completion of 12 cycles to the discharge of 100 mL of liquid. Drain time is a measure of the wetness and stability of the foam.

[0133] Tables 12 to 18 describe the foam evaluation test results of exemplary body wash and hand soap formulations including various alkyl polyglucoside surfactant systems (e.g., octanoyl / decanoyl glucoside alone, decyl glucoside alone, and combinations of octanoyl / decanoyl glucoside and decyl glucoside in different ratios (ratios of 3.5:1 to 1:3.6). Higher flash foam (mL) and maximum foam height (mL) indicate greater foam volume. The longer the drain time, the more stable the foam. Both the flash foam test and the drain time test provide an overall foam distribution. Varying the combined ratio of octanoyl / decanoyl glucoside and decyl glucoside from 3.5:1 to 1:3.6 provides the best ratio for foam distribution.
[0134] Example 3: Probiotics Testing
[0135] The prebiotic effects of the pure ingredients inulin and carrageenan and the combination of inulin and carrageenan were tested in vitro. Growth inhibition assays were performed to test whether the pure prebiotic ingredients inhibited or promoted the growth of skin bacteria. Pure colonies of Escherichia coli, Corynebacterium fasciatum, Staphylococcus aureus and Staphylococcus epidermidis were grown overnight in tryptic soy broth (TSB) medium. The turbidity of the bacterial culture solution was adjusted to 0.1 OD (optical density) at 610 nm using a UV-VIS spectrometer Lambda 45 purchased from Perkin-Elmer. The pure ingredients were diluted into a certain concentration of tryptic soy broth (TSB) medium. 2 ml of bacterial culture was incubated with 2 ml of 2% or 5% inulin or 0.3% carrageenan at 37°C for 24 hours. Bacteria incubated only with the medium were considered as controls. After incubation, the turbidity of each tube was read at 610 nm using a UV-VIS spectrometer. Each treatment was repeated. The mean OD number of the prebiotic was subtracted from the control number. If the value is negative, the prebiotic inhibits bacterial growth, and if the value is positive, the prebiotic promotes bacterial growth. The results show that 0.3% carrageenan, 2% and 5% inulin, and the combination of carrageenan and inulin selectively inhibited the growth of undesirable bacteria (Escherichia coli, Corynebacterium fasciatum, and Staphylococcus aureus) in vitro, but promoted the growth of desirable bacteria (Staphylococcus epidermidis) (see, e.g. Figure 1 ).
[0136] The prebiotic effect of the prototype shower gel formula incorporating prebiotics was also tested in vitro. A conventional competitive shower gel was included as a control. 2% inulin AGA was incorporated into the shower gel formula. The shower gel was diluted to a 1% solution in TSB medium. Pure colonies of Corynebacterium striata and Staphylococcus epidermidis were grown overnight in TSB medium. The turbidity of the bacterial culture solution was adjusted to 0.1OD (optical density) at 610nm using a UV-VIS spectrometer. Then, 2ml of the bacterial culture solution was incubated with 2ml1% shower gel solution at 37°C for 3 hours. After incubation for 3 hours, the mixed solution was diluted 10 times in TSB. 100ul of bacterial solution was spread on a tryptic soy agar plate and incubated overnight at 37°C. The bacterial colonies on the plate were counted the next day. In Minitab software, the statistical significance of the number of bacterial colonies between treatments was analyzed by ANOVA at a 95% confidence level. The results showed that the prototype shower gel containing 2% inulin exhibited a prebiotic effect compared to conventional shower gel. It significantly increased the growth of desirable bacteria (Staphylococcus epidermidis) and reduced undesirable bacteria (odor-causing bacteria) compared to conventional shower gel (Comparative Example II). Figure 2 ).
[0137] Example 4: Barrier function test
[0138] External company Genoskin uses The model tested the ex vivo skin benefits of a prototype body wash containing prebiotics. Prior to model production, skin biopsies collected from human donors during surgery were tape stripped using 50 consecutive strips (MonaDerm, DS100) to remove the stratum corneum. The model was maintained under cell culture conditions (37°C, 5% CO2, and maximum humidity) using 1 mL of standard The culture medium was cultured for 3 or 5 days. 10 μL of shower gel was topically applied to the skin surface so that the compound covered the entire surface in a uniform manner. The shower gel sample was kept in contact for 60 seconds and then rinsed with phosphate buffered saline (PBS). This procedure was repeated 3 times, and the model was finally rinsed 3 times. The treatment was renewed every day. On the 3rd and 5th days, the model was divided into 2 batches, the skin tissue was processed for fluorescent yellow analysis, and the rest was processed for histological analysis. 50 μL of 1mM fluorescent yellow was topically applied to the surface of the skin model. Then, the model was incubated for 4 hours under cell culture conditions (at 37°C, in a CO2 incubator with a water-saturated atmosphere). Then, a cotton swab was used to remove the fluorescent yellow, and the skin biopsy was fixed in 10% buffered formalin (e.g., HT501128 purchased from Sigma Aldrich) for 48 hours and processed for paraffin embedding. Fluorescent yellow dye penetration indicates skin barrier, and the less dye penetration, the stronger the skin barrier. Hematoxylin and eosin staining was performed on 5 μm thick paraffin-embedded skin cross sections. For each sample, 3 transmitted light photographs were obtained using a Leica DMi1 microscope at 40X magnification. Histological staining demonstrated the integrity of the skin structure.
[0139] In vitro studies have shown that conventional shower gels have a strong negative effect on skin integrity and vitality. On day 5, the skin structure of the conventional shower gel treatment group was damaged, while the prototype shower gel containing inulin maintained the normal structure of the skin and the stratum corneum was regenerated after treatment (see e.g. Figure 3 ).
[0140] Lewis Yellow dye penetration assays showed that skin tissue treated with the prototype shower gel containing prebiotics had less dye penetration into the epidermis, suggesting that the tissue treated with the prototype prebiotic shower gel had a stronger skin barrier than tissue treated with a conventional shower gel (Comparative Example II) and even untreated tissue (see e.g. Figure 4 ).
[0141] Example 5: Analysis of Alkyl Polyglucoside
[0142] The analysis of the carbon chain distribution of different alkyl polyglucosides (APGs) in the raw material and the finished product was used to determine the optimal ratio for the foaming curve. The carbon chain distribution of the alkyl polyglucosides (APG) raw material was determined by an Agilent 1200 series HPLC equipped with an Agilent 1290 InfinityII evaporative light scattering detector (ELSD) and an Agilent 1260 Infinity II HPLC equipped with a single quadrupole mass spectrometer. The carbon chain homologues were separated using a reverse phase mobile phase gradient of 0.1% formic acid in water and 0.1% formic acid in methanol using a Zorbax SB-C18 column (Agilent part number 863953-902) and an Agilent Eclipse Plus phenyl-hexyl column (Agilent part number 959941-912). The ELSD parameters were 35°C evaporator temperature / 35°C nebulizer temperature / 1.6L / min nitrogen flow. The concentration of the raw material sample in a diluent containing 10% water and 90% methanol was 2.0mg / mL. Carbon chain homologues are identified by relative retention times and single ion monitoring on a single quadrupole mass spectrometer. The expected elution order is characterized by the carbon number in the alkyl homologues: increased chain length leads to longer retention times (Kühn, Characterization of Mixtures of Alkyl Polyglycosides (Plantacare) by Liquid Chromatography-Electrospray Ionization Quadrupole Time-of-Flight Mass Spectrometry, 2004, pp. 2347-2353).
[0143] The results were calculated by normalized area percentage and they showed that the decyloctyl glucoside raw material (Plantaren 810 UP) contained C8 and C10 homologues, while the decyloctyllauryl glucoside raw material (Plantaren 818 UP, Plantaren 2000N UP) contained C8, C10, C12 and C14 homologues.
