[0001] CROSS-REFERENCE TO RELATED APPLICATIONS

[0002] This application claims priority under 35 U.S.C. §119(e) to U.S. Provisional Application No. 62 / 858,050, filed on June 6, 2019, which is incorporated herein by reference in its entirety for all purposes.

[0006] not applicable Background of the Invention

[0008] Adenosine is a purine nucleoside compound that contains a complex of adenine and a ribose molecule (furanose). Adenosine is naturally present in mammals and plays an important role in several biochemical processes, including energy transfer (such as adenosine triphosphate and adenosine monophosphate) and signal transduction (such as cyclic adenosine monophosphate). Adenosine also plays a role in processes related to vasodilation (including cardiac vasodilation) and acts as a neuromodulator (for example, it is believed to be involved in promoting sleep). In addition to participating in these biochemical processes, adenosine is also used as a therapeutic antiarrhythmic agent to treat, for example, supraventricular tachycardia. As further discussed herein, tumors evade host responses by suppressing immune function and promoting tolerance, and adenosine has been shown to play an important role in mediating tumor escape from the immune system. Adenosine is expressed on various immune cell subsets and endothelial cells through the A 2A R and A 2B Adenosine signaling by R has been established to have an important role in protecting tissues during inflammatory responses. Thus, in certain instances, adenosine protects tumors from immune destruction (e.g., see Fishman, P et al., (2009) Handb Exp Pharmacol 193:399-441).

[0009] Adenosine receptors are a class of purinergic G protein-coupled receptors that use adenosine as an endogenous ligand. The four types of adenosine receptors in humans are called A1, A 2A , A 2B Modulation of A1 has been suggested to manage and treat, for example, neurological disorders, asthma, and heart and kidney failure; A 2A Antagonists are used to manage and treat, for example, Parkinson's disease; modulation of A 2BUseful in the management and treatment of, for example, chronic lung disease, including asthma; and modulation of A3 has been suggested for the management and treatment of, for example, asthma and chronic obstructive pulmonary disease, glaucoma, cancer, and stroke.

[0010] Historically, modulators of adenosine receptors have been non-selective. However, recent work has identified modulators that specifically target adenosine A 2A Receptor (A 2A R) or adenosine A 2B Receptor (A 2B R) useful adenosine modulators. This work includes compounds described in WO2018 / 136700, which include an aminopyrimidine moiety.

[0011] Despite these advances, the synthetic methods described for preparing the compounds of WO2018 / 136700 include chemical transformations with relatively low yields, which hinder large-scale production and impede the commercial availability of the compounds.

[0012] Therefore, there is a need in the art to provide improved methods for preparing aminopyrimidine compounds. The present disclosure satisfies this need and also provides related advantages. Summary of the invention

[0179] Example

[0180] Example 1: Trifluoroethanol-assisted condensation of β-keto esters to provide hydroxypyrimidines (and chloropyrimidines).

[0181]

[0182] Step 1: Add 12M HCl to the (水溶液) 3-bromo-2-methylaniline (18.6 g, 100 mmol) was added dropwise to a mixture of 4-nitropropene (26 mL) and water (26 mL) to form a fine white suspension. The mixture was cooled to 0 ° C, and a solution of sodium nitrite (7.31 g, 106 mmol) in water (15.1 mL) was added dropwise. The mixture was stirred at 0 ° C for 30 minutes. Sodium bicarbonate (17.8 g, 212 mmol) was added to the homogeneous mixture at 0 ° C to avoid excessive gas evolution. The aqueous phase of the brown suspension was found to have a pH of about 7. This suspension was maintained at 0 ° C.

[0183] In a separate flask, copper cyanide (9.85 g, 110 mmol), potassium cyanide (13.0 g, 200 mmol) and water (31 mL) were heated to 60 ° C to form a homogeneous solution. At 60 ° C, the above suspension was added dropwise to this stirred solution to avoid excess gas evolution. After addition, the mixture was stirred at 100 ° C for 30 minutes. The mixture was cooled, MTBE (200 mL) was added, the mixture was stirred, and filtered to remove any solids, washed with MTBE. The organic phase was dried over Na2SO4 and concentrated. The resulting crude product was purified by vacuum distillation to obtain the desired product (13.6 g, 69%) as a light orange solid.

[0184] Step 2: In a two-liter two-necked flask, aryl bromide (101.9 g, 520 mmol, 1.0 eq) was dissolved in THF (520 mL) under N2 atmosphere, and the mixture was cooled in an ice-water bath. iPrMgCl·LiCl (400 mL, 1.3 M in THF, 520 mmol, 1.0 eq) was added via cannula. When the addition was complete, the ice bath was removed. After 4 hours, the flask was cooled in an ice-water bath and dry ice (about 230 g, 5.2 mol, 10 eq) was added portionwise to prevent overheating or excessive bubbling (Note: CO2 gas can be bubbled through the solution instead of solid dry ice). When the bubbling of the addition was complete, the mixture was diluted with MTBE (500 mL) and 2M HCl (250 mL). The layers were separated, and the aqueous layer was washed with additional MTBE (500 mL). The organic layer was extracted with 10% NaOH (190 mL x 2), and the combined aqueous layers were cooled in an ice-water bath and acidified with concentrated HCl until a white precipitate formed. The precipitate was isolated by filtration and washed with water, then dried in a vacuum oven at 80°C overnight to give benzoic acid (64.1 g, 76% yield) as a white solid.

[0185] Step 3: benzoic acid (50g, 311mmol, 1.0 equivalent) is suspended in CH2Cl2, and oxalyl chloride (40mL, 466mmol, 1.5 equivalent) is added, followed by DMF (about 30 drops). Gas emission is observed immediately, and the reaction flask is opened to the atmosphere under the positive pressure of N2. As determined by LCMS and visual inspection (starting material is completely dissolved), after the initial acid is completely consumed, the reaction mixture is concentrated. Excess oxalyl chloride is removed by azeotropic distillation with toluene to obtain the corresponding acyl chloride in a light brown-brown solid.

[0186] In a separate two-necked flask equipped with an overhead stirrer, ethyl malonate potassium salt (66.1 g, 388 mmol, 1.25 eq.), triethylamine (108 mL, 777 mmol, 2.5 eq.) and MeCN (777 mL) were cooled in a salt / ice-brine bath. Solid MgCl2 (74 g, 777 mmol, 2.5 eq.) was added and the resulting suspension was stirred vigorously at about -10 °C. After 1 hour, the solid acyl chloride was added at a rate that ensured dissolution in the concentrated suspension. The suspension quickly became homogeneous and the stirring rate was reduced to avoid splashing. The ice bath was removed. As determined by TLC analysis, after complete consumption of the starting material, the reaction mixture was cooled in an ice-water bath, 2M HCl (971 mL, 1.9 mol, 6.25 eq.) was added, and the ice bath was removed. After 30 minutes, the layers were separated and the aqueous layer was extracted with MTBE. The combined organic layers were washed with saturated NaHCO 3 and brine, dried over sodium sulfate, filtered and concentrated to give the keto-ester as a light brown-brown solid (67 g, 93% yield).

[0187] Step 4: A round-bottom flask was charged with 42.0 g (181.8 mmol) of β-keto-ester, 32.7 g (181.8 mmol) of guanidine carbonate and 227 mL of trifluoroethanol. The suspension was heated to reflux under N2 for 16 h.

[0188] Workup: The reaction was cooled to room temperature and the solvent was evaporated under reduced pressure to give a viscus red oil. The oil was redissolved in 250 mL of H2O and the aqueous solution was extracted with dichloromethane (2 x 250 mL). The reaction mixture was then eluted with 1.0 M HCl (水溶液) The aqueous phase was acidified to pH about 2-3. The precipitated product was collected by filtration, washed thoroughly with H2O and dried in a vacuum oven at 70°C. Yield 30.81 g (75%), purity >99%.

[0189] Step 5: A round-bottom flask was charged with 50.0 g (221.2 mmol) of the pyrimidinone from step 4 and 100.8 g (442.2 mmol) of benzyltriethylammonium chloride. The mixture was suspended in 442.2 mL of anhydrous acetonitrile and 31.0 mL (331.8 mmol) of POCl3 was added. The suspension thus obtained was then heated to reflux under N2 for 4 h.

[0190] Workup: The reaction was cooled to room temperature and about 200 g of crushed ice was added. The mixture was then stirred for 30 min, followed by dropwise addition of ice-cold 15% NH4OH aqueous solution to about pH 10-11. (Note: Slow addition of cold NH4OH is recommended to avoid sudden exotherm due to quenching of excess POCl3). The suspension was then stirred at room temperature for another 1.5 h. The precipitated product was collected by filtration, washed thoroughly with H2O and dried in a vacuum oven at 70°C. Yield 48.2 g (89%), purity>99%.
