Description

[0001] This application is a divisional application of an application with an application date of September 30, 2021, application number 202180067119.9, and invention name “Composition for inhibiting dental plaque formation”. Technical Field

[0002] The present disclosure relates to a composition for inhibiting the formation of dental plaque (tooth plaque), etc. It should be noted that the contents of all the documents described in this specification are incorporated into this specification by reference. Background Art

[0003] Dental plaque (tooth plaque) is a biofilm of microorganisms in the oral cavity, which is believed to be the cause of dental caries and periodontal disease. Therefore, it is important to control dental plaque, especially to inhibit the formation of dental plaque.

[0004] Generally speaking, dental plaque is formed as follows: First, a thin film of protein derived from saliva or physiological gingival sulcus fluid called "pellicle" is formed on the tooth surface, and facultative anaerobes such as streptococci (early adherent bacteria) adhere to the tooth surface through this pellicle. Mediating bacteria such as Fusobacterium that co-aggregate with various oral bacteria adhere to the early adherent bacteria, and late adherent bacteria such as Porphyromonas gingivalis and Treponema denticola, which are anaerobic bacteria, adhere and aggregate through the mediating bacteria, and dental plaque matures. It is known that late adherent bacteria in particular can cause periodontal disease and are directly or indirectly related to periodontal tissue damage.

[0005] Prior art literature

[0006] Non-patent literature

[0007] Non-patent document 1: J Dent Res 90(11):1271-1278, 2011 Summary of the invention

[0008] Problem that the invention aims to solve

[0009] In view of the above, in preventing periodontal disease, it is important to inhibit the formation of dental plaque on tooth surfaces, especially to inhibit the adhesion and aggregation of advanced adherent bacteria and the maturation of dental plaque.

[0010] Solutions for solving problems

[0011] The present inventors have focused on and conducted research on Fusobacterium, a mediating bacterium, because it is believed that if the action of Fusobacterium, a mediating bacterium that can coaggregate with various bacteria, can be inhibited, the adhesion and aggregation of late-adherent bacteria on the tooth surface can be inhibited, and even the maturation of dental plaque formation can be inhibited.

[0012] Therefore, research has been conducted on methods for sterilizing Fusobacterium, but despite attempts to study various bactericides commonly used in oral compositions, no bactericide has been found that can efficiently sterilize Fusobacterium.

[0013] As an example of a bactericide commonly used in oral compositions, cetylpyridinium chloride is no exception, and no efficient bactericidal effect is observed against Fusobacterium. However, after further research, the present inventors found that since cetylpyridinium chloride is a C16 alkylpyridinium chloride, there is a possibility that the length of the alkyl group has an effect on the bactericidal effect against Fusobacterium, and further improvements have been made.

[0126] Bacteria 1: Fusobacterium nucleatum subsp. nucleatum ATCC23726

[0127] Bacteria 2: Fusobacterium nucleatum subsp. nucleatum ATCC25586

[0128] The test bacteria were inoculated into 10 ml of GAM broth medium (Nissui Pharmaceutical Co., Ltd.), and cultured anaerobically at 37° C. for 2 days. This culture solution was used as the test bacterial solution.

[0129] 200 μl of the test bacterial solution was mixed with 200 μl of the fungicide solution of different concentrations. After mixing for 30 seconds, 100 μl of the mixture was collected and 900 μl of the drug-inactivated PBS prepared by adding soybean lecithin and Tween 80 to phosphate buffered saline (PBS) at a final concentration of 0.07% and 0.5%, respectively, was added thereto (the mixture was diluted 10 times) to inactivate the fungicide. In addition, the mixture was diluted stepwise with the drug-inactivated PBS until the mixture was diluted to 10 1 ~10 7 times (stage mixture dilution).

[0130] 100 μl of each of the prepared step mixed solution dilutions was spread on CDC sheep blood agar medium for anaerobic bacteria (Becton Dickinson Japan Co., Ltd.), anaerobically cultured at 37°C for 3 days, and the number of viable bacteria was counted.

[0131] The results are shown in Figure 1 , Figure 2 and Figure 3It should be noted that these graphs are graphs with Log (viable count) CFU / ml as the vertical axis, and the detection limit of this value is 2 (a value less than 2 indicates sterilization to less than 100 CFU). In addition, the concentrations of CPC, DPC, OPC, BKC12, BKC14, and BKC16 in these graphs and the following graphs represent the concentrations of the components in the prepared disinfectant solution.

[0132] It can be seen that CPC and OPC did not show bactericidal effect on Fusobacterium, while DPC showed bactericidal effect on Fusobacterium even at a lower concentration. In addition, it can be seen that although CPC alone did not show bactericidal effect on Fusobacterium, when combined with DPC, the bactericidal effect of DPC on Fusobacterium was enhanced.

[0133] Here, the results of using OPC, DPC or CPC separately are divided into Figure 1 and Figure 2 Extracted from the Figure 4a and Figure 4b .

[0134] In addition, it can be seen that BKC16 did not show a bactericidal effect on Fusobacterium, while BKC14 or BKC12 showed a bactericidal effect on Fusobacterium even at a lower concentration. In addition, it can be seen that although CPC alone did not show a bactericidal effect on Fusobacterium, when combined with BKC14 or BKC12, the bactericidal effect of BKC14 or BKC12 on Fusobacterium was enhanced.

[0135] Study on the bactericidal effect of a composition containing quaternary ammonium salt and CPC on Fusobacterium

[0136] In addition to the quaternary ammonium salt and CPC, a fragrance, a preservative, a solubilizer (polyoxyethylene hydrogenated castor oil), etc. were mixed in water and a solvent (glycerin) to prepare a liquid composition containing the quaternary ammonium salt and CPC at various concentrations.

[0137] The same operation as above was carried out using this liquid composition to examine the bactericidal effect. The results are shown in Figure 5 .

[0138] In addition, the liquid composition was subjected to the same operation as above to examine the bactericidal effect on more Fusobacterium subspecies. That is, the following bacteria 3, bacteria 4, bacteria 5, bacteria 6 and bacteria 7 were used instead of bacteria 1 and 2 to examine the bactericidal effect. The results are shown in Figure 6 .

[0139] Bacteria 3: Fusobacterium necrophorum subsp. necrophorum ATCC25286

[0140] Bacteria 4: Fusobacterium nucleatum subsp. polymorphum ATCC10953

[0141] Bacteria 5: Fusobacterium nucleatum subsp. Fusiforme ATCC51190

[0142] Bacteria 6: Fusobacterium nucleatum subsp.vincentii ATCC49256

[0143] Bacteria 7: Fusobacterium nucleatum subsp.animalis ATCC51191

[0144] It could be confirmed that the studied compositions containing DPC and CPC sterilized Fusobacterium to below the detection limit.

[0145] In addition, similarly to the above, the bactericidal effects of compositions containing alkylpyridinium chlorides at various concentrations alone or in combination against Fusobacterium were also examined again. The results are shown in Figure 7a and Figure 7b .

[0146] The following are examples of compounding ratios. It should be noted that the values of each component in the following examples of compounding ratios are expressed in mass %.
