Description

Technical Field

[0001] The present application relates to the field of biological control technology, and specifically to a lipopeptide composition and its use. Background Art

[0002] The emergence and outbreak of agricultural pests and diseases will not only affect the yield of crops, but also reduce the quality and market value of crops, further hindering the sustainable development of the agricultural sector. Pests may show characteristics such as periodicity, continuity, and uncertainty due to differences in natural factors such as region, climate, and insect species. If management and response are not timely, it is easy to cause large-scale crop damage. How to achieve effective prevention and control of crop pests is a key issue that needs to be urgently addressed in agricultural production. At the same time, pathogenic microorganisms in the soil are also an important cause of crop diseases. Factors such as excessive development of soil resources and unscientific and rational use of fertilizers and pesticides have caused soil to become the main transmission medium for a variety of crop pathogens. Among them, fungal microorganisms represented by mold have caused more than 70% of crop diseases. Under conditions that are conducive to the spread of rice blast fungi, rice blast can cause a reduction of up to 50% in rice yield, seriously restricting rice production worldwide. In addition to major diseases of cereal crops such as rice blast, wheat head blight, and corn ear rot, solanaceous wilt caused by Fusarium oxysporum and root rot of solanaceous and melons caused by Fusarium solani have also become a major obstacle to high-quality and efficient production of crops. Taking potatoes, which are widely planted around the world, as an example, potato wilt caused by Fusarium fungi can cause nearly 30-50% yield loss and quality decline, and has now become one of the most serious diseases in the world's crop industry.

[0003] The prevention and control of crop diseases and insect pests actively responds to the policy of "prevention first, integrated prevention and control". At present, it mainly includes three categories: chemical control, biological control and mechanical control. Mechanical control is to interfere with the growth and development of harmful organisms by physical factors or mechanical action. The actual operation is relatively complicated and is mostly used for prevention or emergency remediation. The relatively common chemical control mainly works through pesticides. Long-term use or improper use can easily cause problems such as pesticide residues, environmental pollution, and drug resistance, which in turn threaten human health. Compared with traditional pesticides, biological control has received widespread attention in the field of crop disease and insect pest control due to its environmental friendliness and sustainability. Among them, the rational use of microorganisms plays an important role in achieving plant disease control. For example, Pseudomonas, Bacillus, Trichoderma, etc. can effectively prevent soil-borne diseases of crops caused by pathogens, and Bacillus thuringiensis, Beauveria bassiana, etc. can be used as low-toxic insecticides for pest control. However, most microorganisms that can be used for biological control have complex metabolite spectra, the types of effective compounds are not clear, and the control effect is often unstable due to restrictions on host species, pathogen types, environmental conditions and other factors. Therefore, how to effectively control plant diseases and insect pests based on abundant microbial resources and related metabolites has become a difficult and hot topic in agricultural research.

[0004] Some studies have shown that the secondary metabolites of biocontrol microorganisms are the key to their control. Among them, lipopeptides are a class of low molecular weight biosurfactants that contain both hydrophilic amino acids and hydrophobic fatty acid chains, mainly produced by fermentation of microorganisms such as Bacillus, Pseudomonas, and Streptomyces. Lipopeptide compounds have diverse structures and functions. Due to different carbon chain lengths and amino acid compositions, they are divided into different families and have different functions. In addition to application fields such as petrochemicals, food industry, and environmental protection, lipopeptides have shown great application potential in the fields of medicine and agriculture due to their broad-spectrum antibacterial activity. Studies have shown that lipopeptide compounds have excellent effects in antibacterial and insecticidal effects, and are suitable for development as new environmentally friendly agents for the control of crop diseases and pests. However, a single type of lipopeptide has certain limitations in the application process, such as limited effect on inhibiting certain specific pathogens or being easily affected by environmental factors, resulting in unsatisfactory effects. Therefore, how to compound single lipopeptide compounds to achieve broad-spectrum control of plant diseases and insect pests is one of the important directions for high-quality development in the field of biological control. [0118] Example 2 Evaluation of the Indoor Antibacterial Effect of Lipopeptide Compositions on Common Plant Pathogenic Bacteria

[0119] Activation of pathogenic bacteria. Different pathogenic bacteria required for the experiment were inoculated on LB medium for activation and placed in a 37°C incubator for culture until the colonies grew to 2 / 3 of the medium.

[0120] Plate confrontation antibacterial experiment of different pathogenic bacteria. The plate confrontation method was used to detect the inhibitory effect of the lipopeptide composition 1-64 on the growth of pathogenic bacteria. Under aseptic operation conditions, the pre-melted sterilized culture medium was quantitatively added to a sterile conical flask, and the lipopeptide composition 1-64 in Example 1 was quantitatively aspirated and added to the conical flask respectively. After fully shaking, an equal amount of plate culture medium containing the lipopeptide composition was prepared (specifically, 24 mL of culture medium was taken, 1% of the lipopeptide composition was added and fully shaken, and 3 culture dishes with a diameter of 6 cm were taken and poured with an equal amount of about 8 mL of culture medium to prepare a plate culture medium containing the corresponding lipopeptide composition). The experiment set up a clean water treatment group as the control group, and each group was treated with no less than 3 replicates.

[0121] After the center of the plate was determined by the cross method, a sterile puncher with a diameter of 5 mm was used to punch holes at the edge of the activated pathogenic bacterial colony and take the bacterial cake. The side with mycelium on the bacterial cake was transferred to the center of the above LB plate with a sterile toothpick facing down and placed in a 37°C incubator for culture. After the culture was completed, according to the growth of pathogens in the blank control culture dish, the diameter, antibacterial bandwidth and other indicators of each pathogen in the different lipopeptide composition treatment groups were observed and measured by the cross method, and the antibacterial rate was calculated by the following formula. The results verified the inhibitory effect of different lipopeptide compositions on the growth of pathogens.

[0122] Bacterial inhibition rate (%) = [(pathogen diameter of control group - diameter of bacterial cake) - (pathogen diameter of experimental group - diameter of bacterial cake)] / (pathogen diameter of control group - diameter of bacterial cake) × 100%. The experimental results are shown in Table 2.

Place a medium-sized, fresh rapeseed leaf in a culture dish containing a 9-cm diameter filter paper, select 20 3rd-instar larvae of the test insect diamondback moth with basically the same growth condition and without any treatment and place them on the leaf, use a micro-syringe to extract 1 μL of the prepared lipopeptide composition solution, and drip it on the pronotum of the test insect. Similarly, place a medium-sized, fresh watermelon leaf in a culture dish containing a 9-cm diameter filter paper, select 20 3rd-instar nymphs of the test insect whitefly with basically the same growth condition and without any treatment and place them on the leaf, use a micro-syringe to extract 1 μL of the prepared lipopeptide composition solution, and drip it on the pronotum of the test insect. After the injection treatment, they were placed in a greenhouse for breeding, and the blank control was the clean water treatment group. Each treatment group was repeated 3 times. After 48 hours, the number of insect deaths in each treatment group was recorded, and the mortality rate and corrected mortality rate were calculated. The relevant calculation formula is as follows:

[0137] Mortality rate (%) = (number of test insect deaths / total number of test insects) × 100; Corrected mortality rate (%) = [(mortality rate of test insects in the treatment group - mortality rate of test insects in the control group) / (100 - mortality rate of test insects in the control group)] × 100. The experimental results are shown in Table 4.

[0138] Table 4

[0139]

[0140]

[0141]

[0142]

[0143] After different lipopeptide compounds were compounded, the growth inhibition effect on both insects was improved. The mortality rate after treatment with the two lipopeptide compositions 12-49 and the three lipopeptide compositions 50-64 was higher than that after treatment with the single lipopeptide composition 1-11 under the same conditions.

[0144] Example 5 Evaluation of the contact killing effect of the lipopeptide composition on southern root-knot nematodes

[0145] The second-instar larvae of southern root-knot nematodes were used as the test object, and the immersion method was used for determination, with 3 replicates for each treatment group. Take a 24-well cell culture plate, drop 10 μL of nematode suspension into each well, add 500 μL of composition 1-64, repeat 3 times, put it in a 28°C incubator, add an equal amount of clean water to the blank treatment, observe the second-instar larvae of root-knot nematodes under a microscope after 24 hours and 96 hours, judge the number of deaths and hatched eggs according to the morphology of the nematodes, and calculate the corrected mortality rate.

[0146] The calculation formula is as follows:

[0147] Mortality rate (%) = (number of 2nd instar larvae dead / total number of nematodes tested) × 100; Corrected mortality rate (%) = [(2nd instar larvae mortality rate in treatment group - 2nd instar larvae mortality rate in control group) / (100 - 2nd instar larvae mortality rate in control group)] × 100. The experimental results are shown in Table 5.

[0148] Table 5

[0149]

[0150]

[0151]

[0152]

[0153] After the different lipopeptide compounds were compounded, the growth inhibition effect on nematodes was improved. The mortality rate of the second-instar larvae after treatment with the two lipopeptide compositions 12-49 and the three lipopeptide compositions 50-64 was higher than that of the single lipopeptide composition 1-11 treatment group under the same conditions. At 96 hours, after treatment with the corresponding lipopeptide composition, the mortality rate of the second-instar larvae of the root-knot nematode was able to reach 100% (see Figure 2 ).

[0154] Example 6 Indoor antibacterial activity of lipopeptide compositions in different proportions against common plant pathogens

[0155] Based on the results of Example 2, based on the typical two-lipopeptide compound compositions 14, 20, 21, 26, 32, 38, 39, the antibacterial activity of common plant pathogens was explored according to the compounding ratio. The antibacterial rate calculation method is the same as that of Example 2. The EC of each agent was calculated by the software. 50 values, and the theoretical EC of the composition 50 Value, toxicity ratio (SR) = theoretical EC 50 Value / Determination EC 50 If the toxicity ratio SR>1.5, it is a synergistic effect, SR<0.5 is an antagonistic effect, and 0.5≤SR≤1.5 is an additive effect. The experimental results are shown in Table 6.

[0156] Table 6

[0157]

[0158]

[0159] The experimental results show that when the compounding ratio of the two lipopeptides is controlled between 50:1 and 1:50, there is an additive or synergistic effect. The application of the composition can greatly improve the antibacterial effect and save time and effort. When the compounding ratio is maintained between 1:0.1 and 1:10, the composition has a synergistic effect, and the antibacterial activity against common plant pathogens can be further improved through compounding.

[0160] Example 7 Evaluation of lipopeptide compositions for the control of common plant diseases in the field

[0161] 1. Effects of seed soaking with lipopeptide composition on plant seedlings

[0162] Seed treatment. For plants such as highland barley (Gramineae), peanut (Leguminosae), rapeseed (Cruciferae), potato (Solanaceae), cucumber (Cucurbitaceae), spinach (Leguminosae), lettuce (Asteraceae), and cotton (Malvaceae), select full seeds of uniform size and soak them with compositions 2, 3, 5, 8, 9, 10, 21, 22, 26, 27, 38, 40, 53, and 57 at a ratio of seeds: treatment solution = 1:1.5 for 24 hours. The water treatment was used as a blank control, and each treatment was repeated 3 times.

[0163] After the seeds were sown and the seedlings emerged, the growth of each plant seedling was investigated and recorded every 5 days, including the number of dead seedlings and the total number of plants, until no more seedlings died, and the death rate was calculated. The experimental results are shown in Table 7.

[0164] Table 7

[0165]

[0166]

[0167] It was determined that compared with the blank control (11.72-15.67%), the seedling death rate after the seed was treated with the above composition was reduced, and the seedling emergence was better than that of the control group. Among them, the average seedling death rate of the plants in the single lipopeptide composition 2, 3, 5, 8, 9, 10 treatment groups was 10.27-14.51%, and the average seedling death rate of the plants in the lipopeptide compound composition 21, 22, 26, 27, 38, 40, 53, 57 treatment groups was 6.26-12.89%. For the composition 38 treatment group, the seedling death rate was reduced by 25.77-40.45% compared with the control group under different plant growth conditions. Although the single lipopeptide composition treatment can also significantly reduce the seedling death rate, the effect of the lipopeptide compound composition treatment group is significantly better than the single lipopeptide composition treatment group under the same period, and the plant seedling death situation is significantly improved.

[0168] 2. Effects of spraying lipopeptide combination on plant diseases during growth period

[0169] The above composition was taken and sprayed on plants (highland barley, peanuts, potatoes, cucumbers, and cotton) 60 days after the seeds of the above plants emerged, and the spraying was continued for 3 times with an interval of 10 days. The growth of the plants was randomly sampled and investigated 5 days after the last application of the pesticide, and 5 points were randomly sampled in each area to check the incidence of leaf spots, grade them, and calculate the disease index and relative control effect.

[0170] The grading standard is as follows: 0: normal leaves, no lesions; 1: a small number of lesions on the leaves (1-2); 2: a moderate number of lesions on the leaves (3-5); 3: a large number of lesions on the leaves (6-8); 4: more than 9 lesions on the leaves, the area of lesions accounts for less than 50% of the total leaf area; 5: the area of lesions accounts for more than 50% of the total leaf area. Among them, the disease index = 100 × ∑ (number of diseased plants at each level × level) / (total number of plants × highest level representative value), relative control effect (%) = (control group disease index - treatment group disease index) / control group disease index × 100. The experimental results are shown in Table 8.

[0171] Table 8

[0172]

[0173]

[0174] It was determined that continuous spraying of the lipopeptide combination can alleviate the disease situation of each plant. The average control effect of the treatment groups of composition 2, 3, 5, 8, 9, and 10 was 23.82-36.08%, and the average control effect of the treatment groups of lipopeptide compound composition 21, 22, 26, 27, 38, 40, 53, and 57 was 32.86-50.15%, respectively, which were significantly higher than the treatment groups of the single lipopeptide combination under the same conditions. The treatment of the lipopeptide combination can promote the growth of the plants while effectively preventing diseases, and the height of the plants has been significantly improved (see Figure 3 , Figure 4 ).

[0175] 3. Effect of lipopeptide composition irrigation treatment on leaf spot disease during plant growth period

[0176] Field test. Plant crops such as chestnut trees (Fagaceae), walnut trees (Juglandaceae), peach trees (Rosaceae), grapevines (Vitaceae), kiwifruit trees (Actinidiaceae), citrus trees (Rutaceae), and banana trees (Musaceae) were transplanted in the fields in different regions. Clean water treatment was used as a blank control, and the above-mentioned lipopeptide composition was irrigated. Each plant was irrigated 1500mL, once a month, and continued to be cultivated for 9 months after continuous irrigating for 3 months. The water and fertilizer management during the planting process was uniformly handled by the plantations in each region. Random sampling surveys were conducted at 5 points in each area to record the growth of the trees, check the incidence of lesions on the leaves of the trees, and classify them to calculate the disease index and relative control effect. The calculation method of the disease index and relative control effect is the same as that of plant growth period diseases. The experimental results are shown in Table 9.

[0177] Table 9

[0178]

[0179]

[0180] As for the plant crops involved in the experiment, the irrigation treatment of the lipopeptide combination can effectively slow down the incidence of plant crops and achieve the prevention and control of related diseases. The prevention and control effect varies depending on the type of plant crops. Among them, the prevention and control effect of the lipopeptide compound combination treatment group is significantly higher than that of the single lipopeptide composition 2, 3, 5, 8, 9, 10 treatment groups under the same conditions.
