AI Technical Summary
Existing skin care compositions are difficult to effectively deliver hydrophilic vitamins such as ascorbic acid to the living areas of the skin, especially due to their hydrophilicity, which leads to poor penetration through the skin barrier layer, resulting in short or ineffective efficacy.
A combination of low molecular weight hyaluronic acid (average molecular weight less than 75 kDa, eg about 50 kDa) and hydrophilic vitamins is used to enhance its absorption and penetration depth in the skin.
It significantly improves the absorption and penetration depth of ascorbic acid in the skin, extends the efficacy, and avoids degradation or membrane formation problems caused by excessive molecular weight of hyaluronic acid.

Abstract
Description
[0001] Cross-references to related patent applications
[0002] This application claims priority to U.S. Application No. 63 / 348,630, entitled SKIN CARE COMPOSITIONS AND METHODS OF USE, filed on June 3, 2022, the contents of which are incorporated herein by reference in their entirety for all purposes. Technical Field
[0003] The present disclosure relates to skin care compositions comprising one or more hydrophilic vitamins and low molecular weight hyaluronic acid or alkali metal hyaluronate (< avg 75 kDa) (e.g., avg 35 kDA to avg 60 kDA) (e.g., about avg 50 kDA) and methods of using and making these compositions. Background Art
[0004] Effective topical delivery of hydrophilic ingredients, including hydrophilic vitamins such as ascorbic acid (vitamin C), to viable regions of the skin has long been a challenging task. For example, pure ascorbic acid is a very hydrophilic active, which can limit its transport through the lipid channels that separate the keratinocytes of the outermost stratum corneum and form the primary barrier to active delivery. Although some hydrophilic vitamins can be "derivatized" to increase their hydrophobicity, thereby becoming more permeable alternatives, these derivatives lack the proven efficacy of topical applied hydrophilic vitamins such as ascorbic acid.
[0005] Although skin care compositions are often used to deliver active ingredients, the effects may be short-lived or ineffective because the active agent may not penetrate the layers of the skin. This challenge may be even more significant with the various hydrophilic ingredients used as surface active ingredients in many skin care compositions. Therefore, there is a need to enhance the surface delivery of hydrophilic ingredients such as ascorbic acid so that they can be effectively delivered across the protective barrier layer of the skin to the active areas of the skin. Summary of the invention
[0006] It has been unexpectedly discovered that hyaluronic acid ("HA") has a specific molecular weight correlation that can enhance and increase the skin penetration depth of hydrophilic vitamins (e.g., ascorbic acid). At a defined lower molecular weight (<average 75 kDa) (e.g., average 35 kDa to average 60 kDA) (e.g., about average 50 kDA), hyaluronic acid can have the effect of unexpectedly increasing the absorption and penetration of surface hydrophilic vitamins in the skin.
[0007] Hyaluronic acid (also known as hyaluronan or hyaluronate) is an anionic non-sulfated glycosaminoglycan (GAG) widely distributed in the connective tissue of vertebrates and is the most abundant high molecular weight glycosaminoglycan in the extracellular matrix of soft periodontal tissue.
[0008] In one aspect, the present disclosure provides a skin care composition (Composition 1.0) comprising:

[0157] Example 1 - Topical delivery of a combination of ascorbic acid and hyaluronic acid
[0158] In order to observe the absorption enhancement characteristics of hyaluronic acid of different weights, samples of pig ear skin were first "pretreated" with an aqueous solution of 1% appropriate hyaluronic acid. The control sample was treated with water only. After this pretreatment, the sample was then treated with a market-based skin care composition containing 20% by weight of ascorbic acid and placed on a Franz cell (Logan Instruments, DHC-6T) for a diffusion time of 24 hours. After treatment, excess product was removed from the skin surface and the stratum corneum was removed from the skin by tape stripping 20 times. The remaining skin was composed of the living epidermis and dermis, which were then homogenized and quantitatively analyzed for ascorbic acid content by an Agilent 1200 series high performance liquid chromatography (HPLC) with a UV-visible detector. For each experiment, the "enhancement factor" of the living skin area was calculated. The "enhancement factor" is calculated as follows:
[0159] Enhancement factor = 100*(((ascorbic acid permeated in sample pretreated with HA) / ((ascorbic acid permeated in sample pretreated with water only)
[0160] Ascorbic acid permeated into the treated control sample))-1)
[0161] This calculation facilitates comparisons between the many different experiments performed, the results of which are as follows:
[0162] Table 1.
[0163]
[0164] Table 1 shows the enhancement factors for various samples, each incorporating different molecular weights of HA or chemically modified HA.
[0165] Without being bound by theory, those skilled in the art may initially expect that there is such a linear relationship: samples treated with lower molecular weight HA will show greater enhancement of surface ascorbic acid delivery. This expectation may be due to the potential of lower molecular weight HA to increase dermal absorption. However, as shown in Table 1, the relationship is not linear. Surprisingly, when using sample C (about 50kDa hyaluronic acid), the enhancement of surface ascorbic acid delivery is the highest. Unexpectedly, both sample A and sample B, although respectively incorporated with hyaluronic acid having an average molecular weight of 3.6kDa and 20kDa, both show reduced ascorbic acid enhancement, which is significantly lower than sample C. Without being bound by theory, although variants with lower molecular weight are considered to be more easily absorbed into the skin, these variants with lower molecular weight may also be more susceptible to degradation, which may limit their effectiveness. Alternatively, hyaluronic acid with lower molecular weight may be less effective in water absorption. Therefore, in order to enhance or effectively deliver ascorbic acid to hyaluronic acid, the molecular weight must be low enough to be easily absorbed into the skin, but it cannot be too low to easily degrade and limit its water absorption capacity.
[0166] It is noteworthy that two samples treated with higher molecular weight HA, sample D and sample E, both showed reduced ability to enhance the surface delivery of ascorbic acid. Regarding sample E, the enhancement effect is actually negative. This should be interpreted as meaning that, compared with the control sample pretreated with water alone, the sample pretreated with hyaluronic acid having an average molecular weight of 1,000kDa to 1,400kDa has a reduced surface ascorbic acid delivery level. Visually, the skin sample pretreated with hyaluronic acid having an average molecular weight of 1,000kDa to 1,400kDa seems to form a film on its surface. Without being bound by theory, this observation may be because the molecular weight of the HA used in sample E is too large, so that it cannot be absorbed into the skin, and may only remain on the skin surface to form a film. Similarly, without being bound by theory, this film formation effect can explain why sample E shows a negative value in terms of the surface delivery of ascorbic acid.
[0167] In order to further explore the possible mode of action of how HA enhances the surface delivery of ascorbic acid, sample F was treated with acetylated hyaluronic acid with an average molecular weight of 20kDa. Although the low molecular weight of acetylated hyaluronic acid indicates that it should be easily absorbed into the skin, it is not bound by theory, and it is possible that the polar hydroxyl groups terminated with acetyl moieties may significantly reduce the overall water binding capacity. As shown in Table 1, similar to hyaluronic acid (e.g., sample E) with much larger molecular weight, acetylated hyaluronic acid (average molecular weight of 20kDa) also hinders the surface delivery of pure ascorbic acid, although the molecular weight of acetylated hyaluronic acid is about 1 / 50 of the molecular weight used in sample E. Similarly, without being bound by theory, this may indicate that the strong water binding capacity of (non-acetylated) HA is crucial in its ability to enhance the surface delivery of ascorbic acid. In addition, it is obvious from the molecular weight range of hyaluronic acid in Table 1 that the delivery of ascorbic acid is not linear with the relationship of the molecular weight of hyaluronic acid. Again, without being bound by theory, this unexpected relationship may be due to the fact that hyaluronic acid, with an average molecular weight of 50 kDa, has a low enough molecular weight to be readily absorbed into the skin, but not so low that its water binding properties are greatly reduced.
[0168] The composition "market-based skin care composition" contains the ingredients listed below: Cyclopentasiloxane, Ascorbic Acid, Ethylhexyl Hydroxystearate, Polysilicone-11, Tocopherol, Isosorbide Dioctanoate, Polymethylsilsesquioxane, Hexylresorcinol, Silybum Marianum Fruit Extract, Hordeum Distichon / Barley Extract, Citrus Aurantium Dulcis / Orange Oil, Silica, Phellodendron Amurense Bark Extract, Santalum Album / Sandalwood Wood Extract.
[0169] Example 2 - Topical delivery of ascorbyl glucoside in combination with hyaluronic acid
[0170] Ascorbyl glucoside is a derivative of ascorbic acid. However, it is believed that ascorbyl glucoside also has several properties that one skilled in the art might believe would hinder its surface delivery into the skin. For example, compared to ascorbic acid, ascorbyl glucoside is not only larger (MW of ascorbyl glucoside is 338 g / mol, while MW of ascorbic acid is 176 g / mol) but also more polar (LogP of ascorbyl glucoside is -2.5, while LogP of ascorbic acid is -1.85), and therefore more hydrophilic.
[0171] The topical delivery of ascorbyl glucoside was performed using the same experimental procedure as described in Example 1. Briefly, 20 wt% of ascorbyl glucoside was loaded into the base formulation of the "market-based skin care composition" described in Example 1, such that the ascorbyl glucoside-containing skin care composition contained a total of 20 wt% of ascorbyl glucoside relative to the total weight of the ascorbyl glucoside-containing skin care composition. However, it should be noted that the active ingredients were removed from the market-based skin care composition, such that the ascorbyl glucoside-containing skin care composition did not contain ascorbic acid, tocopherol, hexylresorcinol, or milk thistle fruit extract.
[0172] Pig ear skin explants were treated with either pure water (control) or a 1 wt % aqueous solution of hyaluronic acid ("HA") having an average molecular weight of 50 kDa, wherein the solution comprised a 1 wt % aqueous solution of hyaluronic acid and the balance was water. Subsequently, the skin pieces were treated with a skin care composition containing ascorbyl glucoside and then placed on a Franz diffusion cell for approximately 24 hours. After this period of time, the top twenty layers of the skin were removed using adhesive tape. All of these tape layers and the remainder of the skin were analyzed for ascorbyl glucoside content using high performance liquid chromatography (HPLC).
[0173] The results of the various tests of Sample G and Sample H can be seen in Table 2 andFigure 1 and Figure 2 In both cases, the trends were very similar. The area showing the highest amount of ascorbyl glucoside was the "rest of skin," also referred to as the "rest of skin." The "rest of skin" refers to the area of skin outside the stratum corneum that is considered to represent roughly the living area of the skin and a representative of the surface delivery of enhanced hydrophilic vitamins such as ascorbyl glucoside. In both experiments, the levels of ascorbyl glucoside in the skin samples treated with a 1 wt % hyaluronic acid solution (about 50 kDa) were significantly higher (p<0.10) relative to the skin samples treated with water alone. The absolute levels of ascorbyl glucoside in these areas are detailed in Table 2, where the unit levels in the "rest of skin" are expressed in micrograms.
[0174] Table 2
[0175]
[0176] *p value: about 0.055
[0177] **p value: about 0.061
[0178] The overall observable trend is very similar to that seen with 20 wt % pure ascorbic acid in anhydrous matrix used in Example 1, where skin samples were pre-treated with a 1 wt % solution of hyaluronic acid having an average molecular weight of 50 kDa.
[0179] In summary, the results in Table 2 indicate that a 1 wt% solution of hyaluronic acid having an average molecular weight of 50 kDa can significantly enhance the surface delivery of ascorbyl glucoside in a composition containing 20 wt% ascorbyl glucoside.
[0180] Example 3 - Topical delivery of a combination of carnosine and hyaluronic acid
[0181] Topical delivery of carnosine was performed using the same experimental procedures as described in Examples 1 and 2. Briefly, a carnosine composition was prepared by incorporating carnosine into the market-based skin care composition described in Example 1 in an amount such that the carnosine composition contained 5 wt% carnosine relative to the total weight of the carnosine composition. Active ingredients were removed from the market-based skin care composition such that the carnosine skin care composition did not contain ascorbic acid, tocopherol, hexylresorcinol, or milk thistle fruit extract.
[0182] Pig ear skin explants (Sample I) were prepared, which were treated with an aqueous solution of 1% by weight of hyaluronic acid with an average molecular weight of 50 kDa and the balance of water and a carnosine composition. A control (Sample J) was prepared by treating the pig ear skin explants with water and a carnosine composition. Specifically, the pig ear skin explants received an aqueous solution of 1% by weight of hyaluronic acid with an average molecular weight of 50 kDa and the balance of water or water, and then treated with the carnosine composition.
[0183] After the carnosine composition was applied to the pig ear skin explants, the pig ear skin explants were placed in a Franz diffusion cell for about 24 hours. After this period of time, the top twenty layers of the skin were removed using adhesive tape. All of these tape layers and the remainder of the skin were analyzed for carnosine content using high performance liquid chromatography (HPLC).
[0184] The results of two tests evaluating the combination of a carnosine composition and an aqueous solution of hyaluronic acid having an average molecular weight of 50 kDa compared to a carnosine composition and water (control) can be seen in Table 3 (below) and Figure 3 and Figure 4 middle.
[0185] Table 3
[0186]
[0187] The area showing the highest amount of carnosine was the "rest of skin." In both experiments, carnosine levels were significantly higher in skin samples treated with a 1 wt% solution of hyaluronic acid (approximately 50 kDa) relative to skin samples treated with water alone.
