[0002] This application claims the benefit of U.S. Non-Provisional Application No. 18 / 136,419, filed on April 19, 2023, which is incorporated herein by reference in its entirety. Technical Field
[0003] The present invention relates generally to the field of devices for cryopreservation of biological specimens. More specifically, the present invention relates to devices capable of remote identification via RFID technology. Background Art
[0004] In life sciences, cryopreservation is performed for the purpose of stopping the biological activity of living cells for a prolonged period of time. Vitrification is one of the techniques used for cryopreservation.
[0005] Vitrification involves converting a solution containing a biological sample (i.e., oocyte or embryo) into a glassy amorphous solid without any crystalline structure, and then cooling extremely quickly. One of the main challenges of this method is to prevent the intracellular liquid in the oocyte or embryo from forming ice crystals. Therefore, the first step is to use a fluid containing a cryoprotectant called "vitrification medium" to dehydrate one or more cells as much as possible. The biological sample is then rapidly frozen by being immersed in a cryogenic fluid such as liquid nitrogen (LN2). Due to the appropriate combination of freezing speed and cryoprotectant concentration, the water in the cell will reach a solid, harmless, glassy (vitrification) state, rather than an orderly, destructive crystalline ice state. Vitrification can be described as a rapid increase in fluid viscosity, which captures water molecules along a random orientation. However, the vitrification medium may contain a relatively high level of cryoprotectant, which is toxic to cells except in a vitrified state. Therefore, the time that cells are exposed to the vitrification medium during dehydration and warming must be carefully controlled to avoid cell damage, and therefore it is expected that freezing will be performed as quickly as possible.
[0006] The CRYOLOCK cryopreservation device was developed as a versatile, simple and efficient vitrification device intended for preserving, freezing and storing oocytes or embryos in liquid nitrogen. It is a cryopreservation device that allows samples to be rapidly cooled and stored without direct contact with sterile liquid nitrogen (LN2). The CRYOLOCK cryopreservation device is described in U.S. Patent Application Publication No. US2016 / 0174545, published on June 23, 2016, which is incorporated herein by reference in its entirety.
[0007] The CRYOLOCK cryopreservation device uses a combination of semi-flexible plastics and functional design, including specific shapes and tight gaps, to hermetically seal samples within the cryopreservation device at room temperature and maintain that hermetic seal as the cryopreservation device and samples cool to liquid nitrogen temperatures.
[0008] Due to the expense of storing samples in liquid nitrogen, multiple samples are stored in a given cryostat. Since the entire cryostat is stored in the cryostat, the size of each cryostat and the packaging characteristics of such cryostat affect the amount of space required and, therefore, the cost of storage. Beyond simple space issues, quality management and auditing are required to ensure that the identification and storage conditions of each sample are tracked over time. If a cryostat needs to be removed from a cryostat during a quality audit, temperature fluctuations may affect the long-term viability of the sample.
[0009] There is a need to be able to identify each sample / device in a cryopreservation tank without removing the cryopreservation device from the cryopreservation tank. There is a need to maintain or reduce the size of cryopreservation devices that are self-identifying, particularly at the level of individual biological specimens. There is a need to maintain the ability to identify each sample / device throughout its lifecycle of collection, storage, retrieval, and use.
[0010] The addition of RFID tags in each cryogenic storage device has brought several challenges. First, the part of the device that keeps the biological sample must be sterilized, and it is usually sterilized using gamma rays, and gamma rays can damage the ability of the RFID tag to function. In addition, the addition of RFID tags must be suitable for the workflow during vitrification, which means that before collecting the biological sample, the RFID tag needs to be firmly engaged with a part of the cryogenic storage device, because the time between collecting the sample and being immersed in liquid nitrogen is necessarily shorter. The RFID tag needs to be firmly maintained by the cryogenic storage device under ambient conditions and liquid nitrogen temperature, and the cryogenic storage device cannot be damaged when such a large temperature fluctuation. Any size increase in the cryogenic storage device that can keep the RFID tag needs to be minimized. Ideally, the RFID tag can be fixed to the cryogenic storage device without the need for an adhesive that may be affected by low temperatures. Summary of the invention

[0107] Example
[0108] Comparative Example 1, Comparative Example 2, and Example 3 were performed using a slender rod of a cryopreservation device injection molded from polystyrene. Examples 1 and 2 were performed using a prototype of the slender rod of the cryopreservation device. The prototype was 3D printed by Digital ABS Plus from Statasys in Eden Prairie, Minnesota. The slender rod of each device / prototype was made of a single plastic integral piece. The slender rod in each example was designed with an RFID chamber to accommodate a transponder RFID microchip tag (RFID tag), which is cylindrical, has a diameter of 2 mm and a length of 12 mm, and can be obtained from Shenzhen Manruta Technology Co. Ltd. in Guangdong Province, China. The sample collection tip of each slender rod used for the comparative example and example can be sealed with a CRYOLOCK cap (Biotech, Inc., Alpharetta, GA).
[0109] Comparative Example 1
[0110] Design and injection molding Figure 1a to Figure 1d Cryogenic storage device shown. Figure 1a , Figure 1b , Figure 1c and Figure 1d , the cryopreservation device 10 includes an elongated stem 12 and a cap 14. The elongated stem includes an RFID section 16, a circumferential groove 18a, an elongated body 20, a truncated cone-shaped boss 22 extending from a first end of the elongated body 20, and a sample collection tip 24 extending from a narrow end of the boss 22. The cap 14 includes a circumferential groove 18b and an elongated hollow chamber 25 defined along a long axis of the cap 14 and sized to accommodate the sample collection tip 24 and the truncated cone-shaped boss 22. The circumferential grooves 18a, 18b disposed near the end of the cap 14 and the distal end of the elongated stem 12 are used to grip the cryopreservation device 10 with tweezers (not shown), thereby making it easier to manipulate the cryopreservation device 10 in a changing temperature environment. The RFID section 16 includes an RFID transition region 26, an RFID chamber 28, and an RFID termination region 30. The RFID termination region 30 is located farther from the elongated body 20 along the longitudinal axis than the RFID transition region 26, and the RFID cavity 28 is located between the RFID termination region 30 and the RFID transition region 26. The RFID cavity 28 is sized to hold an RFID tag (not shown). As such, the RFID cavity 28 is cylindrical in shape, and the opening 32 is circular. Figure 1c and Figure 1d 1 are side and cross-sectional views of the cryopreservation device 10 shown in FIG. 1 with the cap 14 removed so that the entire elongated rod 12 is shown.
[0111] The RFID tag was manually inserted into the RFID chamber 28 through the opening 32 under ambient conditions. The RFID tag was securely held in the RFID chamber 28 and the chip could not be removed from the RFID chamber 28 by shaking the elongated rod 12. The elongated rod 12 was immersed in liquid nitrogen for approximately 2 minutes. The elongated rod 12 was inspected when removed from the liquid nitrogen. The RFID chamber 28 had several cracks visible to the naked eye.
[0112] Example 1
[0113] Design as Figure 2a to Figure 2d The elongated rod 12 for a cryopreservation device is shown and a prototype is produced by 3D printing. Figure 2a , Figure 2b , Figure 2c and Figure 2d , the elongated rod 12 includes an RFID section 16, an elongated body 20, a truncated cone-shaped boss 22 extending from a first end of the elongated body, and a sample collection tip 24 extending from a narrow end of the truncated cone-shaped boss 22. The RFID section 16 includes a tapered RFID transition region 26, an RFID chamber 28, and an RFID terminal region 30. The RFID terminal region 30 is located farther away from the elongated body 20 along the longitudinal axis than the RFID transition region 26, and the RFID chamber 28 is located between the RFID terminal region 30 and the tapered RFID transition region 26. Because the RFID chamber 28 has a larger cross-section than the elongated body 20, the RFID transition region 26 is tapered or gradually becomes thinner. The size of the RFID chamber 28 is designed to hold an RFID tag (not shown).
[0114] The RFID tag is manually inserted into the RFID chamber 28 through the opening 32, which is sized to allow the RFID tag to pass through the opening 32 and into the RFID chamber 28. A locking mechanism 34 defines the opening 32. Figure 2a and Figure 2d The locking mechanism 34 shown ensures that the RFID tag (not shown) remains inside the RFID chamber 28 throughout the handling and storage of the elongated rod 12. The RFID tag is inserted at an angle that allows a portion of the RFID tag to enter the portion of the RFID chamber 28 beyond the RFID opening 32 and adjacent to the RFID termination area 30. Then as the RFID tag is pushed into the RFID chamber 28, once the RFID tag is fully inserted into the RFID chamber 28, the flaps 36a and 36b bend downward and rest on top of the RFID tag. Figure 2cAs shown, openings 37a and 37b are longitudinally spaced along the outer edge of the RFID chamber 28, and opening 32 is placed on opposite sides of the elongated rod 12 perpendicular to the longitudinal direction. The RFID tag is firmly held in the RFID chamber 28, and the chip cannot be removed from the RFID chamber 28 by shaking the elongated rod 12. The elongated rod 12 was immersed in liquid nitrogen. The RFID segment 16 was inserted into the liquid nitrogen for about 2 minutes and then returned to ambient conditions, and the RFID segment 16 showed no visible signs of cracking. The RFID segment 16 was observed at 10X magnification, and still no signs of cracking were observed.
[0115] Example 2
[0116] Design as Figure 3a The slender rod 12 for the cryopreservation device shown in FIG. 3 d is provided, and a prototype is 3D printed. The slender rod 12 includes a groove 18a, an RFID section 16, an elongated body 20, a truncated cone-shaped boss 22 extending from the first end of the elongated body 20, and a sample collection tip 24 extending from the narrow end of the truncated cone-shaped boss 22. The RFID section 16 includes a tapered RFID transition zone 26, an RFID chamber 28, and an RFID terminal zone 30. The RFID terminal zone 30 is positioned farther away from the elongated body 20 along the longitudinal axis than the RFID transition zone 26, and the RFID chamber 28 is located between the RFID terminal zone 30 and the RFID transition zone 26. The size of the RFID chamber 28 is designed to hold an RFID tag 38. The tapered RFID transition zone 26 is designed to transition from a smaller cross section of the elongated body 20 to a larger cross section of the RFID section 16, while allowing the resting side of the elongated body 22 and the RFID section 16 to be straight for easy handling.
[0117] The RFID tag is manually inserted into the RFID chamber 28 through the opening 32 under ambient conditions. The RFID tag is inserted at an angle that allows a portion of the RFID tag to enter a portion of the RFID chamber 28 that is beyond the RFID opening 32 and adjacent to the RFID termination area 30. The locking mechanism 34 includes a circular tab 39 that bends downward when the RFID tag 38 is loaded into the RFID chamber 28 and is located above the RFID tag 38 after the RFID tag 38 is fully inserted into the RFID chamber 28. The locking mechanism 34 holds the RFID tag 38 within the RFID chamber 28. The RFID tag is securely held in the RFID chamber 28 and cannot be removed from the RFID chamber 28 by shaking the elongated rod 12. The elongated rod 12 is immersed in liquid nitrogen. When the elongated rod 12 is immersed in liquid nitrogen for approximately 2 minutes and then returned to ambient conditions, the RFID section 16 shows no visible signs of cracking. The RFID segment 16 was viewed at 10X magnification, and again no signs of cracking were observed.
[0118] Comparative Example 2
[0119] Design and injection mold the elongated rod 12. Figure 4a , Figure 4b , Figure 4c 4d, the elongated rod includes an RFID segment 16, a circumferential groove 18a, an elongated body 20, a truncated cone-shaped boss 22 extending from a first end of the elongated body 20, and a sample collection tip 24 extending from a narrow end of the truncated cone-shaped boss 22. The circumferential groove 18a is located between the end of the elongated body 20 away from the sample collection tip 24 and the RFID segment 16. The RFID segment 16 includes an RFID transition zone 26, an RFID chamber 28, and an RFID terminal zone 30. The RFID terminal zone 30 is positioned farther away from the elongated body 20 along the longitudinal axis than the RFID transition zone 26, and the RFID chamber 28 is located between the RFID terminal zone 30 and the RFID transition zone 26. The size of the RFID chamber 28 is designed to hold an RFID tag (not shown). As such, the RFID chamber 28 is cylindrical in shape, and the opening 32 is circular. In Figure 4a One of the two longitudinal slits 40a is shown in FIG. Figure 4b and Figure 4c Two longitudinal slits 40a, 40b are shown in FIG. 2. In this comparative example, the longitudinal slits are of equal size and each slit begins at the RFID opening 32 and removes a piece of the wall of the RFID chamber 28 that is approximately 90% of the length. Figure 4b yes Figure 4a A cross-sectional view of the elongated rod 12 is shown, and Figure 4c yes Figure 4a A detailed cross-sectional view of the RFID section 16 of the elongated shaft 12 is shown. The locking mechanism includes an "elbow" or ledge 42a, 42b on at least a portion of the wall of the RFID chamber 28 nearest the RFID opening 32.
[0120] Under ambient conditions, an RFID tag is inserted into the RFID chamber 28 through the opening 32. The RFID tag cannot be removed from the RFID chamber 28 by shaking the elongated rod 12. However, because the RFID tag can be removed from the RFID chamber 28 by simply moving a block of the wall of the RFID chamber 28 in an outward direction, the RFID tag is not securely held in the RFID chamber 28. The elongated rod 12 with the RFID tag in the RFID chamber 28 is immersed in liquid nitrogen. The RFID segment 16 is inserted into the liquid nitrogen for approximately 2 minutes and then returned to ambient conditions, and the RFID segment 16 shows no visible signs of rupture. The RFID segment 16 is observed at 10X magnification, and still no signs of rupture are observed. This design only addresses one of the two necessary criteria, namely that the RFID segment 16 does not rupture, but the RFID tag is not securely held in place during processing in the cryopreservation device.
[0121] Example 3
[0122] Design and injection mold the elongated rod 12. Figure 5a , Figure 5b and Figure 5c , the elongated rod includes an RFID segment 16, a circumferential groove 18a, an elongated body 20, a truncated cone-shaped boss 22 extending from a first end of the elongated body 20, and a sample collection tip 24 extending from a narrow end of the truncated cone-shaped boss 22. Figure 4b , Figure 4cTwo longitudinal slits 40a, 40b are shown in FIG. In this example, the longitudinal slits are of different sizes, and the longitudinal slit 40a removes a piece of about 50% of the length of the wall of the RFID chamber 28, while the longitudinal slit 40b removes a piece of about 90% of the length of the wall of the RFID chamber 28. Figure 4b yes Figure 4a A cross-sectional view of the elongated rod 12 is shown, and Figure 4c yes Figure 4a A detailed cross-sectional view of the RFID section 16 of the elongated shaft 12 is shown. The locking mechanism includes a "knock" or land 42a, 42b on at least a portion of the wall of the RFID chamber 28 nearest the RFID opening 32.
[0123] Under ambient conditions, an RFID tag was inserted into the RFID chamber 28 through the opening 32. The RFID tag was firmly held in the RFID chamber 28, and the chip could not be removed from the RFID chamber 28 by shaking the elongated rod 12. Unlike in Comparative Example 2, the RFID tag could not be removed from the RFID chamber 28 by moving the wall of the RFID chamber 28 outward.

Figure 6c4 is a cross-sectional view illustrating an RFID tag 38 positioned within the RFID chamber 28 and secured by an RFID plug 44. In this embodiment, the RFID chamber 28 is slightly oversized relative to the RFID tag 38 to allow for heat shrinkage when the cryopreservation device is submerged in liquid nitrogen. The RFID plug 44 is used to Figure 2a to Figure 2d and Figure 3a to Figure 3c By blocking the RFID opening 32, the RFID plug 44 retains the RFID tag 38 in the RFID chamber 28 throughout the handling and storage of the cryopreservation device.
