[0110] Results Figure 1-7 As shown, compared with gradient 1, under the optimized elution procedure of gradient 7, the number of chromatographic peaks increased from 100 to 111, the separation degree between the chromatographic peak where S peak (peonidin) is located and the surrounding peaks increased from 3.03 (gradient 1) to 3.39, the separation degree between peak 9 and the surrounding peaks under gradient 1 was only 1.35, which was increased to 2.27 under gradient 7, the separation degree between peak 1 (crystal orchid glycoside) and the surrounding peaks increased from 2.05 (gradient 1) to 3.16, and the number of plates increased from 13645 to 16823. Under gradient 1, the separation degree between peak 6 and peak 7 was only 1.35 (less than 1.5), which was increased to 2.27. Under gradient 1, peak 11 (ferulic acid) was not visible, and the separation degree between peak 12 and peak 13 (liquiritin) was only 1.21 (less than 1.5), which was increased to 2.03 under the optimized gradient 7. Therefore, it was determined that the chromatographic conditions of gradient 7 were selected for fingerprint determination.
[0111] 3. Selection of detection wavelength
[0112] The test solution prepared in item 1 of this example was tested by HPLC, and different absorption wavelengths were investigated; the absorption wavelengths were 200nm, 240nm, 280nm, and 300nm respectively. A Shimadzu Inert Sustain C18 chromatographic column (column length 250mm, inner diameter 4.6mm, particle size 5μm) was selected, acetonitrile was used as mobile phase A; 0.1% phosphoric acid solution was used as mobile phase B, and gradient elution was performed according to the provisions in Table 7; the flow rate was 0.8ml per minute; the column temperature was 35°C; the results are shown in Table 7. Figure 8 .
[0113] By comparing the fingerprint spectra of different wavelengths of 200nm, 240nm, 280nm and 300nm, it can be seen that the baseline wavelength is larger at 200nm, some chromatographic peak information is missing at 280nm and 300nm, and the chromatographic peak information is rich at 240nm, and the response of each chromatographic peak is balanced. Therefore, the detection wavelength of the fingerprint spectrum of Tiaogan Decoction is determined to be 240nm.
[0114] 4. Investigation of flow rate
[0115] This section investigates the flow rate of the mobile phase, and the test solution prepared in Item 1 of this Example is tested by HPLC. The effects of flow rates of 0.7 ml per minute, 0.8 ml per minute, and 0.9 ml per minute on the fingerprint of Tiaogan Decoction were investigated respectively. A Shimadzu Inert Sustain C18 chromatographic column (column length 250 mm, inner diameter 4.6 mm, particle size 5 μm) was selected, acetonitrile was used as mobile phase A; 0.1% phosphoric acid solution was used as mobile phase B, and gradient elution was performed according to the provisions in Table 7, the column temperature was 35°C, and the detection wavelength was 240 nm; the results are shown in Fig. 9 .
[0116] The results showed that when the flow rate was 0.7 ml / min, 21 chromatographic peaks appeared, of which peak 12 was merged with peak 13, and peaks 5, 11 and 19 were merged with the original surrounding peaks, resulting in a large difference in peak area; when the flow rate was 0.9 ml / min, 22 chromatographic peaks appeared, and the peak shapes of some peaks were poor, of which peak 3 was merged with the surrounding peaks, and peak 14 was split; while at 0.8 ml / min, there were no such problems. Therefore, in order to ensure the good reproducibility of the method, the recommended optimal flow rate was 0.8 ml / min.
[0117] 5. Investigation of column temperature
[0118] The test solution prepared in item 1 of this example was tested by HPLC. The column temperature was investigated, and the effects of column temperatures of 30°C, 35°C, and 40°C on the fingerprint of Tiaogan Decoction were investigated. A Shimadzu Inert Sustain C18 chromatographic column (column length 250 mm, inner diameter 4.6 mm, particle size 5 μm) was selected, acetonitrile was used as mobile phase A; 0.1% phosphoric acid solution was used as mobile phase B, and gradient elution was performed according to the provisions in Table 7, the flow rate was 0.8 ml / min, and the detection wavelength was 240 nm. The results are shown in Table 7. Fig.10 .
[0119] The results showed that different chromatographic column temperatures had no significant effect on the fingerprint of Tiaogan Decoction, indicating that this method can adapt to certain changes in column temperature. However, considering the chromatographic peak shape, peak separation, baseline and other factors, the preferred chromatographic column temperature for the fingerprint study of Tiaogan Decoction in this experiment was 35°C.
[0120] 6. Determination of optimal chromatographic conditions
[0121] Based on the above experiments, the chromatographic conditions were determined as follows: octadecylsilane bonded silica gel as filler (InertSustain C18, column length 250 mm, inner diameter 4.6 mm, particle size 5 μm); acetonitrile as mobile phase A, 0.1% phosphoric acid as mobile phase B; gradient elution according to the provisions in Table 7; flow rate 0.8 ml per minute; column temperature 35°C; detection wavelength 240 nm. The theoretical plate number calculated based on the paeoniflorin peak should not be less than 10,000.
[0122] Example 2 Investigation of the preparation method of the test solution
[0123] 1. Investigation of extraction methods
[0124] Different extraction methods will affect the extraction efficiency of each chromatographic peak. Therefore, the extraction efficiency of each chromatographic peak of ultrasonic extraction and heating reflux extraction is compared, and the appropriate extraction method is determined in combination with the peak area of each characteristic peak; take about 0.5g of Tiaogantang granules, grind them into powder, accurately weigh them, put them in a stoppered conical flask, accurately add 25ml of 50% methanol aqueous solution by volume, plug it, and use ultrasonic treatment (power 250W, frequency 40kHz) for 30 minutes or reflux extraction for 30 minutes, respectively, take it out, cool, shake well, filter, and take the filtrate. Use high performance liquid chromatography to detect under the optimal chromatographic conditions of Example 1, see Fig.11 shown.
[0125] The results showed that 22 characteristic peaks appeared when ultrasonic or reflux extraction was used. There was no difference in the ratio of peak area to sampling volume. Considering the convenience of sample preparation, ultrasonic extraction was the preferred extraction method for the sample.
[0126] 2. Investigation of extraction solvent
[0127] Different proportions of solvents will affect the extraction efficiency of each chromatographic peak. Therefore, the extraction efficiency of each chromatographic peak extracted by 50% methanol, 80% methanol and methanol is compared, and the appropriate extraction solvent is determined in combination with the peak area of each characteristic peak. Specifically, about 0.5g of Tiaogantang granules is taken, ground, accurately weighed, placed in a stoppered conical flask, accurately added with 50% or 80% methanol aqueous solution or 25ml of methanol by volume percentage, plugged, and ultrasonically treated (power 250W, frequency 40kHz) for 30 minutes respectively, taken out, cooled, shaken, filtered, and the filtrate is obtained. Detection by high performance liquid chromatography under the optimal chromatographic conditions of Example 1, see Fig.12 shown.
[0128] Analysis of the results showed that 50% methanol, 80% methanol and methanol all had 22 chromatographic peaks. Comparing the peak area / sampling volume results of each characteristic peak, the peak area was higher when the solvent was 80% methanol, so the preferred solvent for preparing the test sample was 80% methanol.
[0129] 3. Determine the preparation method of the test solution
[0130] Take about 0.5 g of the test sample, weigh it accurately, put it in a stoppered conical flask, accurately add 25 ml of 80% methanol, stopper it tightly, and treat it with ultrasound (power 250 W, frequency 40 kHz) for 30 minutes. Take it out, cool it, shake it well, filter it, and take the filtrate to obtain the product.
[0131] Example 3 Peak attribution and selection of references
[0132] (1) Construction of reference fingerprint
[0133] Fifteen batches of samples of Tiaogan Decoction were taken respectively, and the sample solutions prepared by the preparation method of the sample solution determined in Example 2 were measured under the optimal chromatographic conditions of Example 1 to construct multiple batches of fingerprints. The fingerprint detection AIA data was imported into the "Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System" calculation software, and a total of 22 peaks were calibrated to form a common pattern diagram, and a control spectrum was established. Fig.18 Peak 10 was selected, and the paeoniflorin peak was used as the reference peak, which was marked as Peak S.

The average retention time of peak No. 21 was 97.081 min, and the RSD was 0.01%. The average retention time of peak No. 22 was 97.531 min, and the RSD was 0.01%.
[0135] Taking the paeoniflorin peak as the reference peak, marked as Peak S, the relative retention time of each characteristic peak and Peak S is within the range of ±10% of the specified value; the specified values of Peak 1 to Peak 30 are: 0.19, 0.30, 0.61, 0.62, 0.67, 0.69, 0.71, 0.79, 0.87, 1.00, 1.32, 1.40, 1.44, 1.47, 1.56, 1.69, 1.78, 1.96, 2.07, 2.24, 2.42 and 2.43, respectively.
[0136] According to the similarity evaluation system of traditional Chinese medicine chromatographic fingerprints, the similarity was calculated based on the common peaks. The similarities between the test sample fingerprint and the control fingerprint were both higher than 0.900. The differences between the fingerprints of each batch of particles were small based on the similarity evaluation.
