Abstract
Description
Technical Field
[0001] The invention belongs to the technical field of organic synthesis, and specifically relates to two chiral spirodihydrophene structures and an asymmetric synthesis method thereof. Background Art
[0002] Asymmetric catalytic synthesis is a frontier field of organic synthesis, playing a vital role in drug development, preparation of chiral chemicals and synthesis of chiral materials. Among them, the design and development of new chiral catalysts and chiral ligands are the core of asymmetric catalytic research. The introduction of spirocyclic structures has greatly enriched the diversity of chiral catalysts and chiral ligands, and has been successfully applied to asymmetric transformation reactions such as asymmetric catalytic hydrogenation, asymmetric carbon-carbon bond formation or carbon-heterobond formation, and synthesized many valuable chiral products. However, the inherent rigidity of the spirocyclic structure also limits the scope of application of the corresponding chiral catalyst, and the development of structurally diverse spirocyclic structures is the key to solving this problem. Spirodihydrophenanthene is an important new type of spirocyclic structure. The literature (J.Am.Chem.Soc.2021,143,12445) reports an asymmetric synthesis method (E) of chiral spirodihydrophenanthene structures, and the chiral catalysts and chiral ligands derived from them show excellent reactivity and chiral induction ability. However, the development of new spirodihydrophenanthene structures is still very limited.
[0003]
[0004] The present invention develops two novel spirodihydroquinone structures containing different aromatic substituents at the 3,3, positions, and develops an asymmetric synthesis method and an asymmetric catalytic synthesis application thereof. Summary of the invention
[0005] Purpose of the Invention
[0006] The invention provides two chiral spirodihydrophene structures and asymmetric synthesis methods and applications thereof.
[0007] Technical Solution
[0008] In order to achieve the above technical objectives, the present invention adopts the following technical solutions:
[0009] Two chiral spirodihydroquinone structures and derivative compounds thereof, characterized in that they are the following compounds A, enantiomers or racemates, and compounds B, enantiomers or racemates
[0010]
[0011] Wherein Ar is phenyl, 4-methylphenyl, 4-fluorophenyl, 4-bromophenyl, 3-methylphenyl, 3-bromophenyl, 3-chlorophenyl, 3.5-di-tert-butylphenyl
[0012] A method for asymmetric synthesis of two chiral spirodihydrophene structures and derivative compounds thereof, characterized in that:
[0013]
[0014] The following steps are involved:
[0015] Step (1): Acetone undergoes a condensation reaction with different aromatic substituted aldehydes under alkaline conditions to generate I;
[0016] Step (2): Under the catalysis of a chiral rhodium catalyst, compound I and arylboron reagent II undergo an asymmetric addition reaction to generate intermediate III, which is separated and purified; when boron trifluoride etherate and potassium phosphate are added, intermediate III undergoes an intramolecular Friedel-Crafts reaction to prepare compound IV; boron trifluoride etherate and trifluoroacetic acid are added to generate compound V; a mixture of compound IV and V is obtained by adding only boron trifluoride etherate;
[0017] in:
[0018] In step (1), the reaction solvent is selected from an aqueous solution of an inorganic base, which is one or more of potassium carbonate, cesium carbonate, tripotassium phosphate, lithium hydroxide, sodium hydroxide and potassium hydroxide, preferably potassium hydroxide; preferably, the amount of the inorganic base added is 2 equivalents of different aromatic substituted aldehydes; wherein the molar ratio of different aromatic substituted aldehydes to acetone is 2 to 5:1, preferably 2:1; the reaction temperature is 0 to 50°C, preferably 15 to 25°C; and the reaction time is 6 to 12 hours, preferably 8 to 12 hours.
[0019] Further, compound I has the following structure:
[0020]
[0021] In step (2):
[0022] The chiral rhodium catalyst has a general structural formula of [RhCl(L)x]2 (x=1 / 2), wherein the chiral ligand L is selected from a chiral diene ligand, a chiral heteroatom ligand, a chiral heteroatom olefin ligand, etc.;
[0023] Further, the chiral ligand is selected from the following structures:
[0024]
[0025]
[0026] Furthermore, the chiral ligand is L: Ar = 2-naphthy.
[0027] Step (2) The reaction solvent is selected from one or more of tetrahydrofuran, ethanol, toluene and water, preferably a mixed solvent of ethanol and water; preferably, 0.5 to 2 mL of the reaction solvent is added to 0.1 mmol of compound I, more preferably 1 to 1.5 mL;
[0028] The molar ratio of compound I, aryl boron reagent II and chiral rhodium catalyst is 1:1-5:0.005-0.05, the reaction temperature for converting compound I into aryl boron reagent II is 40-100°C, and the reaction time is 10-16h.
[0029] When compound III is converted to compound IV in step (2), 5-10 molar equivalents of boron trifluoride etherate and 1-2 molar equivalents of potassium phosphate are added.
[0030] When compound III is converted to compound V in step (2), the added boron trifluoride etherate is 5-10 molar equivalents and the added trifluoroacetic acid is 4-5 molar equivalents.
[0031] Beneficial Effects
[0032] 1. The present invention generates compounds IV and V with completely new structures, which have not been reported in the literature and are completely new compounds. The inventors further found that according to the prior art (J.Am.Chem.Soc.2021,143,12445), the presence of an aromatic substituent at the 3,3, position, i.e., the presence of steric hindrance, makes it difficult to form the compounds described in the present invention according to the method;
[0033] Drawing on the article published by the applicant (https: / / doi.org / 10.1002 / anie.201812266) and the patent application CN 109400445A, which discloses an asymmetric synthesis method of 3,3'-diaryl-substituted chiral spirocyclic diphenol compounds, the inventor creatively uses the same chiral rhodium catalyst for the present invention. It should be noted that the difference between the two is that the present invention contains a naphthalene structure, while the patent discloses a phenol structure, that is, the present invention is a spirodihydrophenanthene structure, while the patent discloses a spirocyclic diphenol structure, which belong to different types of compounds. Further, in Example 2, a chiral rhodium catalyst is used to produce intermediate III; further, the inventor surprisingly found that when adding boron trifluoride etherate and potassium phosphate, intermediate III undergoes an intramolecular Friedel-Crafts reaction to prepare compound IV; adding boron trifluoride etherate and trifluoroacetic acid to generate compound V; adding only boron trifluoride etherate or adding boron trifluoride etherate and other bases to obtain a mixture of compounds IV and V, that is, the inventor discovered a method for directional generation of IV or V.
[0034] 2. The present invention relates to compounds of a completely new structure, and for the first time realizes the enantioselective synthesis of two chiral spirodihydroquinone derivatives through an asymmetric synthesis method catalyzed by chiral rhodium. This method has the advantages of high yield and good enantioselectivity (ee value>99%), and has high industrial production value. BRIEF DESCRIPTION OF THE DRAWINGS

[0085] Example 5
[0086]
[0087] Under the protection of high-purity nitrogen, compound Ia (23.4 mg, 0.1 mmol), 7-hydroxynaphthaleneboric acid (56.4 mg, 0.3 mmol), [RhCl(L)]2 (1.9 mg, 2 mol%), potassium hydroxide aqueous solution (0.1 mL, 0.05 M, 5 mol%) and anhydrous ethanol (1 mL) were added to a Schlenk tube in sequence. After the addition was completed, the mixture was reacted at 60 °C for 12 h under the protection of nitrogen. Cool to room temperature, add water (5mL) and ethyl acetate (5mL) to the Schlenk tube for extraction, take out the organic layer, extract the water layer with ethyl acetate (5mL) once more, evaporate the solvent to obtain a crude product, and purify it by column chromatography (PE:EA=4:1, v / v) to obtain intermediate III-a. Add III-a (52.2mg, 0.1mmol) and trifluoroacetic acid (31μL, 0.4mmol) to Schlenk, add anhydrous toluene (1.0mL), slowly drop boron trifluoride ether solution (126μL, 1mmol) to the reaction bottle, move to 60℃, and react for 12h. Add water (2mL) to quench, extract with ethyl acetate (3mL×3), and remove the solvent under reduced pressure to obtain crude IV-a. Column chromatography gave 42.3mg of white solid (PE:EA=10:1, yield 95%, dr value>20:1, ee value>99%.
