Description

Technical Field

[0001] The invention belongs to the technical field of lignin degradation, and in particular relates to a lignin microbial degradation amplification reactor. Background Art

[0002] Lignin (Latin, English, German: Lignin) is a class of complex organic polymers that form important structural materials in the supporting tissues of vascular plants and some algae. Lignins are particularly important in the formation of cell walls, especially in wood and bark, because they impart rigidity and resistance to decay. Chemically, lignins are cross-linked phenolic polymers. The xylem of plants (a structure responsible for transporting water and minerals) contains a large amount of lignin, which allows the xylem to maintain its high rigidity and support the weight of the entire plant.

[0003] Lignin can be degraded by microorganisms, and white rot fungi are common microorganisms used to degrade lignin. The suitable degradation temperature of white rot fungi is 20 degrees to 30 degrees. White rot fungi are aerobic microorganisms, and the degradation process of lignin by white rot fungi does not require light.

[0004] The degradation rate of lignin by white rot fungi is slow and the time is long. The high degradation efficiency achieved in degradation experiments in small containers under laboratory environment and conditions is very difficult to maintain the same degradation efficiency in the same environment and conditions in large industrial reactors. Generally, special large reaction vessels are required for efficient degradation design and experiments. That is, the transition from small experimental containers to large industrial containers requires a lot of work.

[0005] In addition, although the environment and conditions for degradation using white rot fungi have been greatly improved, the degradation cycle still needs to be measured in weeks, which is still relatively long and the efficiency is relatively low. At present, most people use fermentation degradation tanks to achieve degradation, that is, a large number of degradation tanks are required during the cycle of large-scale lignin degradation, resulting in higher degradation costs.

[0006] The present invention designs a lignin microbial degradation amplification reactor to solve the above problems. Summary of the invention

[0007] Based on this, it is necessary to provide a lignin microbial degradation amplification reactor to address the problems existing in the current lignin degradation reactor. The fermentation component in the present invention utilizes 75 small-capacity reaction bottles evenly divided into three layers to form an overall large-capacity fermentation reaction container. While maintaining the laboratory research environment and conditions of the small container, the transformation of small-capacity container fermentation to large-capacity container fermentation is effectively completed, so that the overall large-capacity reactor can effectively ferment lignin under the same environment and conditions as the laboratory research. There is no need to carry out a large amount of additional experimental work on the transformation of laboratory small-container fermentation to large-container fermentation specifically for the required fermentation environment and conditions of the large-capacity reactor. The suitable and favorable fermentation environment and conditions that can be quickly achieved in the laboratory are quickly applied to the actual large-container fermentation and degradation, effectively improving the efficiency of the lignin fermentation and degradation experiment. The double-layer reaction bottle formed by the plastic film in the fermentation component of the present invention can continuously deliver oxygen-rich clean air to the interlayer between its inner wall and outer wall through the air delivery pipe at the bottom thereof, and the oxygen-rich clean air entering the interlayer enters the fermentation reaction mixture in the reaction bottle through the first air vents densely distributed on its inner wall to provide sufficient required oxygen for the fermentation reaction in the reaction bottle, so that the fermentation mixture in the reaction bottle is fully fermented, and the fermentation reaction efficiency and fermentation quality of the fermentation mixture in the reaction bottle are effectively improved. The fermentation components of the present invention can be combined in groups of three after completing the filling of the fermentation mixture of all the reaction bottles, so that the reaction bottles in the three combined fermentation components are distributed up and down, thereby effectively saving the space occupied by the fermentation components after all the reaction bottles are filled with the fermentation mixture and pushed into the warehouse to wait for the fermentation process, so that more fermentation components can be placed in the warehouse and lignin can be effectively fermented and degraded in large quantities, effectively improving the efficiency of the lignin fermentation reaction. In the present invention, the film sleeves provided on the outside of the fermentation components formed in groups of three can slow down the loss of oxygen-rich clean air released from the reaction bottles, so that all the reaction bottles in the three fermentation components are always in an oxygen-rich clean environment during the fermentation reaction of the fermentation mixture, preventing the fermentation mixture in the open reaction bottles from being contaminated, improving the fermentation and degradation reaction efficiency of the fermentation mixture in the reaction bottles, and improving the product quality after the fermentation and degradation reaction. The reaction bottles in the fermentation components of the present invention are formed of plastic films, which effectively reduces the cost of a large number of large-capacity reaction vessels required for the large-capacity fermentation and degradation reaction of lignin. [0040] The fermentation component 100 in the present invention utilizes 75 small-capacity reaction bottles 111 evenly divided into three layers to form an overall large-capacity fermentation reaction container. While maintaining the laboratory research environment and conditions of the small container, the conversion of small-capacity container fermentation to large-capacity container fermentation is effectively completed, so that the overall large-capacity reactor can effectively ferment lignin under the same environment and conditions as the laboratory research. There is no need to carry out a large amount of additional experimental work on the conversion of laboratory small-container fermentation to large-container fermentation specifically for the required fermentation environment and conditions of the large-capacity reactor. The suitable and favorable fermentation environment and conditions that can be quickly achieved in the laboratory are quickly applied to the actual large-container fermentation and degradation, effectively improving the efficiency of the lignin fermentation and degradation experiment. The double-layer reaction bottle 111 formed by a plastic film in the fermentation component 100 of the present invention can continuously transport oxygen-rich clean air to the interlayer between its inner wall 114 and the outer wall 112 through the air supply pipe 415113 at the bottom thereof. The oxygen-rich clean air entering the interlayer enters the fermentation reaction mixture in the reaction bottle 111 through the first air vents 115 densely distributed on the inner wall 114 thereof, thereby providing sufficient oxygen required for the fermentation reaction in the reaction bottle 111, so that the fermentation mixture in the reaction bottle 111 is fully fermented, thereby effectively improving the fermentation reaction efficiency and fermentation quality of the fermentation mixture in the reaction bottle 111. The fermentation components 100 of the present invention can be combined in groups of three after completing the filling of the fermentation mixture of all the reaction bottles 111, so that the reaction bottles 111 in the three fermentation components 100 formed are distributed up and down, thereby effectively saving the space occupied by the fermentation components 100 in which all the reaction bottles 111 are pushed into the warehouse to wait for the fermentation process after completing the filling of the fermentation mixture, so that more fermentation components 100 can be placed in the warehouse and the lignin can be effectively fermented and degraded in large quantities, effectively improving the efficiency of the lignin fermentation reaction. In the present invention, the film sleeve 118 provided on the outside of the fermentation components 100 combined in groups of three can slow down the loss of oxygen-rich clean air released from the reaction bottles 111, so that all the reaction bottles 111 in the three fermentation components 100 are always in an oxygen-rich clean environment during the fermentation reaction of the fermentation mixture therein, preventing the fermentation mixture in the open reaction bottles 111 from being contaminated, improving the fermentation and degradation reaction efficiency of the fermentation mixture in the reaction bottles 111, and improving the product quality after the fermentation and degradation reaction. The reaction bottles 111 in the fermentation assembly 100 of the present invention are formed of plastic film, which effectively reduces the cost of a large number of large-capacity reaction vessels required for large-capacity fermentation and degradation reactions of lignin. The first adding assembly 300, the second adding assembly 400 and the third adding assembly 500 in the feeding assembly 200 of the present invention can achieve the purpose of quickly filling the fermentation mixture into all the reaction bottles 111 in the fermentation assembly 100, effectively improving the filling efficiency.

[0041] The operation process of the present invention is as follows: In the initial state, three layers of reaction bottles 111 are placed in the first frame 101 of each fermentation assembly 100. The reaction bottles 111 are hung on the hooks 104 at the lower ends of the two vertical rods 103 at the top of the corresponding layer through the upper hanging ears 116. The first main pipe 117 connected to the air supply pipe 415113 at the bottom of the reaction bottle 111 is in a closed state. Fig.15 As shown, the first valve assemblies 328 of all the first adding assemblies 300 in the adding assembly 200 are in an open state, the limiting block 312 on the first valve plug 309 is located at one end of the corresponding limiting groove 308, the first vortex spring 313 is in a compressed state, the first piston 317 and the second piston 318 in each first adding assembly 300 are located at the corresponding first feed port 306 and are respectively located on both sides of the first feed port 306, the first piston 317 is in a toggling state with the shifting block 314 on the first valve plug 309, the shifting block 314 is against the cylindrical surface of the first piston 317, and the seed liquid in the first feed pipe 320 fills the space between the first piston 317 and the second piston 318 through the opened first valve assembly 328. The first silo 401 in the second adding component 400 contains lignin particles, the second valve component 407 and the third valve component 408 are both in a closed state, the second vortex springs 412 in the second valve component 407 and the third valve component 408 are both in a compressed state, and the second baffles 409 in the second valve component 407 and the third valve component 408 are abutted against the corresponding limit strips 419 under the action of the corresponding second vortex springs 412 and close the second feed pipe 405 and the second discharge pipe 406 respectively. The space on the side of the fourth feed port 503 of the third piston 522 in the second material bin 501 of the third adding component 500 and the third discharge pipe 506 are filled with distilled water, the third piston 522 abuts against the second limiting ring 520 in the second material bin 501 near the fourth feed port 503, the fourth valve assembly 507 and the fifth valve assembly 508 are in a closed state, the second valve plug 510 in the fourth valve assembly 507 and the fifth valve assembly 508 are in a closed state to the third feed pipe 505 and the third discharge pipe 506 respectively, the first spring 511 in the fourth valve assembly 507 and the fifth valve assembly 508 is in a compressed state, and the third valve plug 517 in the sixth valve assembly 519 is in a closed state to the discharge port of the third discharge pipe 506 under the action of the compressed second spring 518.

[0042] When the present invention is needed to carry out large-capacity lignin fermentation and degradation, first adjust the length of the multi-section telescopic legs 106 in the fermentation component 100 to an appropriate length so that the three-layer reaction bottle 111 in the fermentation component 100 matches the height of the three-layer fifteen groups of the first adding component 300, the second adding component 400 and the third adding component 500 in the feeding component 200 respectively, loosen the locking screws 108 between the telescopic sections 107 on the multi-section telescopic legs 106, so that the length of the multi-section telescopic legs 106 is changed to a height that matches the height of the feeding component 200, and then tighten the locking screws 108 to lock the length of the multi-section telescopic legs 106.

[0043] After the length adjustment of the four multi-section telescopic legs 106 in the fermentation component 100 is completed, the fermentation component 100 is pushed to the side of the feeding component 200, so that the discharge ports of the three layers and fifteen groups of the first adding component 300, the second adding component 400 and the third adding component 500 in the feeding component 200 are first respectively opposite to the bottle openings of the three layers and fifteen reaction bottles 111 closest to the second frame 201 in the fermentation component 100.

[0044] Then, the motor 327 is started, and the motor 327 drives the first driving rods 316 in all the first adding components 300 through the gear 326, the rack 324 and the first connecting frame 319 to synchronously push the first piston 317 and the second piston 318 to move toward the first discharge port 303. The first piston 317 and the second piston 318 push the quantitative seed liquid between the two toward the first discharge port 303. When the second piston 318 reaches the end of the first feeding tube 301, the seed liquid between the first piston 317 and the second piston 318 reaches the first discharge tube 305 through the first discharge port 303 and the drainage groove 304 and is added into the reaction bottle 111.

[0045] During the movement of the first piston 317 and the second piston 318 toward the first discharge port 303, the first valve plug 309 in the first valve assembly 328 rotates and gradually closes the first feed pipe 320 under the action of the corresponding first vortex spring 313. When the first piston 317 passes over the first feed port 306, the restriction of the first piston 317 on the shift block 314 on the first valve plug 309 is released, and the first valve plug 309 completely closes the first feed pipe 320 under the action of the first vortex spring 313.

[0046] After the seed liquid between the first piston 317 and the second piston 318 is completely added into the reaction bottle 111, the motor 327 is started in reverse. The motor 327 drives the first driving rods 316 in all the first adding components 300 to reset through a series of transmissions. The first driving rods 316 drive the corresponding first pistons 317 and second pistons 318 to reset. When the first piston 317 reaches the first feed port 306, the side end face of the first limiting ring 315 of the first piston 317 first meets the upper end face of the first valve plug 309, and the first valve plug 309 rotates and resets under the action of the shifting block 314 during the resetting process of the first piston 317 and compresses the first vortex spring 313 again, and finally the first feed pipe 320 is closed as the first piston 317 is reset.

[0047] After the first piston 317 and the second piston 318 in the first adding component 300 are reset, the first liquid pump 322 is started, and the second main pipe 321 of the first liquid pump 322, the first feed pipe 320, the second feed port 310 on the first valve plug 309 of the first valve assembly 328, and the first feed port 306 refill the space between the first piston 317 and the second piston 318 with seed liquid.

[0048] At the same time, the air pump 417 is started, and the air pump 417 forms a negative pressure in the interlayer between the first silo 401 and the shell 414 in all the second adding components 400, and then forms a negative pressure in the first silo 401 through the second vent hole 402 on the wall of the first silo 401. The second baffle 409 of the second valve assembly 407 in the second feeding pipe 405 overcomes the corresponding second vortex spring 412 to open under the action of the negative pressure, and the suction nozzle at the port of the fourth main pipe 418 moves the lignin particles through the second vent hole 402 on the wall of the first silo 401. The second feed pipe 405 enters the first silo 401. The negative pressure in the first silo 401 will cause the second baffle 409 of the third valve assembly 408 in the second discharge pipe 406 to close the second discharge pipe 406 under the action of the corresponding second vortex spring 412 without leakage. The lignin particles in the sender's first silo 401 gradually fill the first silo 401. Since the aperture of the second vent 402 is smaller than the diameter of the lignin particles, the lignin will not leak into the outer shell 414 through the second vent 402. When the first silo 401 is full of lignin particles, the lignin particles block all the second air holes 402, and the air pump 417 runs in reverse to deliver air into the outer shell 414, thereby forming a positive pressure in the first silo 401. The second valve assembly 407 in the second feed pipe 405 closes the second feed pipe 405 under the combined action of the positive pressure and the corresponding second vortex spring 412 to prevent the material from leaking in the opposite direction. The second baffle 409 of the third valve assembly 408 in the second discharge pipe 406 overcomes the elastic force of the corresponding second vortex spring 412 under the action of the positive pressure and opens the second discharge pipe 406. The lignin particles in the first silo 401 are all quantitatively added into the reaction bottle 111 through the opened second discharge pipe 406.

[0049] During the process of the second adding component 400 adding lignin to the reaction bottle 111, the second liquid pump 526 and the electric push rod 524 are started at the same time. The electric push rod 524 drives all the second driving rods 521 in the third adding component 500 to move synchronously toward the outside of the second silo 501 through the second connecting frame 523. The second driving rods 521 drive the corresponding third pistons 522 to move synchronously. The second liquid pump 526 and the fifth main pipe 525 pump distilled water into the third feeding pipe 505. The second valve plug 510 in the fourth valve assembly 507 in the third feeding pipe 505 opens the third feeding pipe 505 under the combined action of the negative pressure in the second silo 501 and the distilled water in the third feeding pipe 505 and further compresses the corresponding first spring 511. The distilled water in the third feeding pipe 505 enters the second silo 501 and fills the second silo 501 when the third piston 522 reaches the second limiting ring 520 on the side of the second air hole 502.

[0050] After the second material bin 501 is full, the second liquid pump 526 is stopped and the electric push rod 524 is started in reverse. The electric push rod 524 drives the third piston 522 to perform a reset movement through a series of transmissions. The second valve plug 510 in the fourth valve assembly 507 in the third feed pipe 505 closes the third feed pipe 505 under the corresponding first spring 511 and the water pressure in the second material bin 501 to prevent the distilled water from leaking in the opposite direction. The second valve plug 510 of the fifth valve assembly 508 in the third discharge pipe 506 opens the third discharge pipe 506 under the action of the water pressure in the second material bin 501 and further compresses the corresponding first spring 511. The distilled water in the second material bin 501 reaches the sixth valve assembly 519 through the third discharge pipe 506 and overcomes the elastic force of the second spring 518 of the sixth valve assembly 519 to push open the third valve plug 517 to complete the quantitative filling of distilled water into the reaction bottle 111.

[0051] After the quantitative filling of lignin, distilled water and seed liquid in the fifteen reaction bottles 111 on the first side and three layers in the fermentation component 100 is completed, the fermentation component 100 is moved to the second frame 201 again, so that the bottle mouths of the fifteen reaction bottles 111 on the second side and three layers in the fermentation component 100 correspond one by one to the three layers and fifteen groups of the first adding component 300, the second adding component 400 and the third adding component 500 in the adding component 200. Then, the above process is installed to complete the material filling of the fifteen reaction bottles 111 on the second side and three layers again. In this way, the quantitative filling of lignin, distilled water and seed liquid in all the reaction bottles 111 in the fermentation component 100 is completed in sequence.

[0052] like Figure 1 , Figure 2 , Figure 3As shown, after all the reaction bottles 111 in all the fermentation components 100 have completed the quantitative filling of lignin, distilled water and seed liquid, all the fermentation components 100 are grouped into three groups, and the lengths of the three multi-section telescopic legs 106 in each group are adjusted to the longest, the middle and the shortest, respectively, so that the top of the first frame 101 corresponding to the middle-length multi-section telescopic leg 106 is slightly lower than the bottom of the first frame 101 corresponding to the longest multi-section telescopic leg 106, and the shortest multi-section telescopic leg 106 is slightly lower than the bottom of the first frame 101 corresponding to the longest multi-section telescopic leg 106. 06 The top of the corresponding first frame 101 is slightly lower than the bottom of the middle and long multi-section telescopic legs 106. At the same time, the spacing between the two pairs of multi-section telescopic legs 106 in each fermentation component 100 in each group is adjusted so that the overall width of the two pairs of middle and long multi-section telescopic legs 106 in each group is slightly smaller than the spacing between the two pairs of the longest multi-section telescopic legs 106 in the corresponding group, and the overall width of the two pairs of the shortest multi-section telescopic legs 106 in each group is slightly smaller than the spacing between the two pairs of middle and long telescopic legs in the corresponding group.

[0053] Then, if Figure 2 As shown, the fermentation assembly 100 of the medium-length multi-section telescopic legs 106 in each group is pushed laterally under the first frame 101 of the longest multi-section telescopic legs 106 in the corresponding group, and the fermentation assembly 100 corresponding to the shortest multi-section telescopic legs 106 is pushed under the first frame 101 of the medium-length multi-section telescopic legs 106 in the corresponding group, thereby completing the upper and lower combination and merging of the three fermentation assemblies 100 in each group, thereby saving the space they occupy in the warehouse.

[0054] Next, a membrane sleeve 118 is provided on the outside of each group of three fermentation assemblies 100 that have been merged, so that a gap is formed between the lower edge of the membrane sleeve 118 and the first baffle 105 at the bottom of the first frame 101 in the fermentation assembly 100 corresponding to the shortest multi-section telescopic legs 106 in the corresponding group of fermentation assemblies 100, so as to avoid the subsequent rapid leakage of oxygen-rich clean air in the membrane sleeve 118 and ensure that the reaction bottle 111 in the membrane sleeve 118 is in an oxygen-rich and clean environment.

[0055] After each group completes the installation of the membrane sleeve 118, oxygen-rich clean air is transported to the interlayer of each reaction bottle 111 through the first main pipe 117 of the three fermentation components 100 in each group via the air supply pipe 415113 at the bottom of each reaction bottle 111. The air entering the interlayer of the reaction bottle 111 enters the reaction liquid through the first air vent 115 on the inner wall 114. The oxygen-rich clean air squeezed into the reaction liquid stirs the material while transporting oxygen inward, thereby improving the degradation reaction efficiency of the material in the reaction bottle 111.

[0056] The air entering the reaction bottle 111 enters the film sleeve 118 through the bottle mouth, so that a clean environment surrounding the reaction bottle 111 is formed in the film sleeve 118, which is conducive to the reaction bottle 111 to perform a fully and effectively degradation reaction in a sterile environment.
