[0002] This application claims the benefit of U.S. Provisional Application No. 63 / 354,516, filed on June 22, 2022, which is hereby incorporated by reference in its entirety. Background Art

[0003] Hair and scalp cleansing is a common grooming habit that is considered important for optimal cleanliness, health and maintenance of hair. Hair cleansers generally remove sebum, sweat, shed stratum corneum and environmental soils, which helps prevent the development of conditions such as folliculitis and seborrheic dermatitis. Current commercial formulations employ synthetic detergents, technically described as surfactant systems that are mixed in varying amounts and ratios to achieve the desired amount of cleansing power. The beauty industry offers consumers a myriad of options to assist in general hair cleansing, covering formulations with a wide variety of ingredients and compositions.

[0004] In addition to removing sebum, sweat, shed stratum corneum and environmental dirt, the secondary function of shampoo is to impart the desired sensory effects to the hair. Thoroughly cleaned hair (i.e., completely devoid of any sebum or product) tends to feel less smooth and look duller, and as a result is generally not considered cosmetically desirable. One strategy to address this problem is to incorporate large amounts of conditioners into the cleanser. These conditioners are deposited on the surface of the hair and generally only change the surface properties of the hair, i.e., improve the appearance and feel of the hair, but do not address any potential damage to the hair structure. Excessive deposition of these conditioners, especially when combined with inadequate cleaning, can result in conditioners accumulating in the hair and weighing the hair down or making it look duller. Commercial products generally focus on providing benefits to the hair or scalp. However, the health of the hair begins with the follicles and the treatments and products applied to the hair affect the scalp; the two are inseparable. Products targeting the overall health of the hair and scalp system will provide the greatest benefits. Therefore, there is a need for new hair cleansing compositions that cleanse both the hair and the scalp, provide desirable hair properties, without excessive deposition of conditioning agents. DETAILED DESCRIPTION

[0022] The fiber structure of human hair is composed of stratum corneum, cortex and medulla. The stratum corneum is composed of a thin layer of overlapping cells, like a scale system, and provides protection for the inner core (i.e., cortex) of the hair. Due to the presence of a large number of isopeptides and disulfide bonds in each lamellae of stratum corneum cells, stratum corneum cells are mechanically very stable and can resist many environmental factors, including the penetration of foreign molecules. The stratum corneum accounts for about 15% of the weight of a hair. The cortex is the middle region of the hair and accounts for about 80% of the weight of a hair. The cortex is composed of a hierarchical structure of elongated fibrous α-helical keratin, which extends along the fiber axis embedded in an amorphous protein matrix. It is the system of the cortex and its intertwined protein fibers and supporting matrix that gives hair its tensile properties (strength, elasticity). The cell membrane complex connects stratum corneum cells with other stratum corneum cells, cortex cells with other cortex cells, and stratum corneum cells with cortex cells. The medulla corresponds to the central cell bundle and is absent in some hair fibers.

[0023] Human hair contains about 65% to 95% protein. Proteins exhibit their active properties within a specific temperature and pH range and can unfold outside these limits. When treated with low pH solutions, thermal denaturation of fibrous hard α-keratin proteins (i.e., cortical proteins responsible for hair strength and elasticity) shows significantly improved stability. This is especially true for those keratins that have been subjected to oxidative damage (such as by bleaching), and this has been demonstrated by the significant increase in the enthalpy and peak temperature of thermal denaturation as quantified by DSC. Enthalpy is generally understood to reflect the amount and structural integrity of α-helical material in the hair intermediate filament (IF), while peak temperature reflects the cross-linking density of the matrix embedded with IF. Amino acid analysis, X-ray diffraction, Raman spectroscopy and tensile strength observations further support this phenomenon.

[0024] Alkaline pH creates a more favorable environment for cystine (disulfide) disruption, hydrolysis of amide bonds, and formation of lanthionine and lysalanine, all of which weaken the protein structure of hair. The duration of hair exposure to solutions of a particular pH does not significantly change thermal stability; combined with the fact that tap water pH varies widely from neutral to alkaline, this suggests that negative effects may be experienced for the duration of exposure that hair could reasonably be expected to experience under normal washing conditions.

[0025] The observed differences in thermal stability of hair proteins exposed to different pH are due to changes in the environment of the intermediate filaments, i.e., the interface between the IF and the surrounding matrix proteins. This scaffold structure at the surface of the IF is formed by side chain interactions that anchor the filament to the matrix. Strong adsorption of protons at the interface of the IF may help to shield this interface; disulfide bonds as well as electrostatic forces further stabilize this region.

[0026] During the cleansing process, hair is in a structurally fragile state, as water molecules disrupt the hydrogen bonds that stabilize the protein structure. This causes swelling of certain parts of the cuticle and makes the hair fiber susceptible to damage. Furthermore, it has been determined that the swelling behavior of the hair cuticle is highly dependent on pH: the more alkaline the environment, the greater the swelling. In addition to the effect on thermal stability, this modification of cuticle swelling by pH also has an impact on the properties and damage propensity of hair, which is directly relevant to the consumer. When the cuticle swells, the interfiber friction of the hair fiber increases, resulting in increased hair tangling and a rough, uncomfortable feel. Hair is also in a more fragile state when wet; the need for greater combing forces combined with the lower breaking stress of hair in the wet state can easily lead to cuticle flaking, protein loss, and even severe breakage during hair washing and shampooing. The minimal cuticle swelling exhibited at acidic pH can help to counteract this effect.

[0027] The proteins in hair are at their best (most structurally resilient) at a pH of around 5. This more acidic pH environment can also minimize excessive swelling of the stratum corneum, thereby reducing protein loss, preserving color, and minimizing electrostatic repulsion. This is also the ideal pH range for skin with an acidic mantle. Skin with a disturbed pH can experience barrier disturbances, inflammation, and irritation. The pH of tap water varies widely, from 6.5 to 9.53, and can contain metal ions such as divalent calcium and magnesium (hard water minerals) that can affect the texture of the hair, as well as trace levels of transition metals such as copper, iron, nickel, lead, etc. that can cause oxidative degradation of the hair fiber.

[0028] Additionally, peptides can be used to treat hair that has been damaged during the cleansing process. These peptides can interact with the hair primarily via covalent (disulfide) bonds, polar bonds, and hydrophobic interactions. However, penetration of peptides into the cortex of the hair strand, which is necessary for successful treatment, can be difficult. Molecular size and polarity play an important role in determining the ability of a molecule to enter the interior of the fiber structure. Additionally, the presence of sebum and products on the hair strands can hinder the ability of the peptides to successfully migrate to the cortex. While a properly acidic pH helps stabilize the protein structure of the hair (as detailed above), these peptides can work in a complementary and synergistic manner to add additional strengthening effects. Overall, this system can both protect and repair the hair protein structure, which can be fragile during the cleansing process.

[0029] To ensure that no residue is left on the hair fiber that could interfere with the ability of the peptides to penetrate the cortex, optimize protein structural reinforcement and enhance the benefits, a clarifying shampoo should be used before applying a repair treatment containing peptides. Mineral buildup can also interfere with the efficacy of peptides, so the formula must be strong enough to remove 1) product, 2) sebum, 3) hard water minerals and any other type of buildup from the hair. Salicylic acid can be challenging to formulate in a surfactant system, so choosing a strong acid-compatible surfactant that is both ion-tolerant and stable in hard water helps achieve the removal of the above three types of buildup. Detoxifiers such as charcoal, for example, isolate metal ions at low levels without the need to complicate formulation with viscosity-changing chelating agents. Charcoal contains many small pores that create a large surface area that can adsorb heavy metals, including iron (Fe), chromium (Cr), cadmium (Cd) and copper (Cu).

[0030] Thus, detoxifying hair and scalp cleansing compositions with optimized pH that reduce swelling and protein loss are provided herein. Formulating shampoos with a more acidic pH can help counteract this alkaline pH environment and minimize the net negative charge on hair proteins, which minimizes cuticle swelling and reduces binding sites for dissolved metal cations. While the use of protectants such as softeners, sealants, cations, and polymers on hair fibers minimizes friction and associated styling damage and provides a barrier to heat, UV, and pollution, excessive product accumulation can weigh hair down and prevent other products from performing. Hair compositions can include peptides that strengthen the protein structure within the hair cortex to restore the tensile strength of the hair fiber. These peptides can be incompatible with traditional conditioners that form films or coatings and can prevent the peptides from interacting with hair proteins, most particularly preventing the peptides from entering the cortex.

[0031] In some embodiments, the detoxifying hair and scalp cleaning compositions described herein comprise low molecular weight peptides that can penetrate into the cortex of the hair and improve the mechanical properties of the hair. In some embodiments, the detoxifying hair and scalp cleaning compositions described herein provide a durable improved texture and improved manageability for the hair when dry. After long-term use, the detoxifying hair and scalp cleaning compositions described herein provide a soft, shiny, undamaged texture and other desired features for the hair. The detoxifying hair and scalp cleaning compositions described herein do not strip any essential oils from the hair, reduce protein loss and repair potential hair damage, thereby avoiding over-reliance on conditioners that cover the hair fibers to achieve the desired properties. In addition, due to its limited ingredients, the detoxifying hair and scalp cleaning compositions described herein do not result in the accumulation of excess conditioners or other products that can eventually weigh the hair down.

[0032] In some embodiments, the detox hair and scalp cleaning compositions provided herein comprise one or more solvents, one or more peptides, one or more hair conditioners, one or more skin conditioners, one or more cleaning agents, and one or more cosmetically or dermatologically acceptable excipients. In some embodiments, one or more peptides have a peptide sequence of 6 to 15 amino acids. In some embodiments, 2 to 5 amino acids of the peptide sequence are cysteine. The composition of the peptide provides improved resistance, strength, toughness and elasticity for hair.

[0033] When striving for a “less is more” minimalist approach, choosing multifunctional ingredients is key. Salicylic acid has proven efficacy in skin care, providing amyloid and anti-comedogenic activity to promote healthy cell turnover while balancing sebum production, imparting photoprotection, and providing antimicrobial and anti-inflammatory effects. While these properties are expected to transfer to the scalp, the compound exhibits further scalp-specific benefits: when applied topically it can be used to treat conditions such as dandruff, ichthyosis, and psoriasis. 31 In addition to the acidic pH benefits discussed above, its pKa of approximately 2.8 and associated low pH make salicylic acid an ideal ingredient for application to hair fibers.

[0034] The detox hair and scalp cleaning compositions described herein maintain an optimal pH environment for the hair while cleaning the hair to minimize fiber swelling and maintain structural integrity. In addition, the hair cleaning compositions maintain an optimal pH environment while cleaning the scalp to avoid over-drying the scalp and will not damage the natural barrier function of the scalp. The detox hair and scalp cleaning compositions described herein have a pH optimized to clean the hair and scalp. In some embodiments, the pH of the detox hair and scalp cleaning compositions is optimized to promote the efficacy of certain ingredients in the composition. In some embodiments, the pH of the detox hair and scalp cleaning compositions is optimized to promote the efficacy of salicylic acid. In some embodiments, the pH of the composition is optimized to promote the chelation of certain minerals and metals. In some embodiments, the pH of the composition is optimized to minimize the swelling of the stratum corneum. In some embodiments, reducing stratum corneum swelling reduces the tangling of hair. In some embodiments, the pH of the composition is optimized to protect the structural integrity of the hair and mitigate the loss of proteins and lipids.

[0035] In some embodiments, the pH of the detoxifying hair and scalp cleansing composition is between about 1 and about 7. In some embodiments, the pH of the detoxifying hair and scalp cleansing composition is about 1, about 1.25, about 1.5, about 1.75, about 2, about 2.25, about 2.5, about 2.75, about 3, about 3.25, about 3.5, about 3.75, about 4, about 4.25, about 4.5, about 4.75, about 5, about 5.25, about 5.5, about 5.75, about 6, about 6.25, about 6.5, about 6.75, about 7, about, and any value therebetween. In some embodiments, the pH of the detoxifying hair and scalp cleansing composition is between about 1 and about 7, between about 2 and about 7, between about 3 and about 7, between about 3 and about 6, between about 4 and about 6, and any value therebetween.

[0175] Example 2. Evaluation of the efficacy of detoxifying hair and scalp cleansers during and after cleansing

[0176] A detoxifying hair and scalp cleanser composition was prepared according to Example 1. The detoxifying hair and scalp cleanser composition was evaluated using a survey to determine the sensory effects of the detoxifying hair and scalp cleanser on shine, texture (i.e., smoothness), bounce, and firmness. The sensory effects were evaluated during the cleansing process and after drying.

[0177] A human hair tress having a length of 25 cm, a width of 5.5 cm, and a weight of 10 g was gently rinsed with warm water at 40°C. Excess moisture was removed, and the tress was fully foamed for about 1 minute, using 1.5 g of the detoxifying hair and scalp cleanser composition per 10 g of the tress. The foamed hair was subjected to a sensory evaluation to determine its smoothness. 20 people performed the evaluation, and the total evaluation value is shown. The evaluation criteria were: 4: good sliding, 3: slight sliding, 2: not very good sliding, and 1: no sliding.

[0178] A human hair tress of 25 cm in length, 5.5 cm in width, and 10 g in weight was gently rinsed with warm water at 40°C. Excess moisture was removed, and the tress was fully foamed for about 1 minute, using 1.5 g of the detoxifying hair and scalp cleanser composition per 10 g of the tress. The foamed hair was rinsed with tap water (2 L / min) at 40°C for 1 minute. Excess moisture was removed with a towel and the tress was air-dried. After drying, the dry hair was subjected to a sensory evaluation to determine its glossiness and uniformity. 20 people performed the evaluation, and the total evaluation value is shown. The evaluation criteria are: 4: good, 3: slightly good, 2: not very good, and 1: not good.

[0179] A tress of human hair having a length of 25 cm, a width of 5.5 cm, and a weight of 10 g was thoroughly rinsed with warm water at 40°C. The tress was thoroughly foamed for about 1 minute using 5 g or 10 g of the detoxifying hair and scalp cleanser composition. The foamed hair was thoroughly rinsed with tap water at 40°C (2 L / min). Excess moisture was removed using a towel and the tress was dried with warm air from a hair dryer. After drying, the dry hair was subjected to a sensory evaluation to determine its elasticity. 25 people performed the evaluation, and the total evaluation value is shown. The evaluation criteria were: 4: significant improvement in elasticity, 3: visible improvement in elasticity and hardness, 2: slight improvement in elasticity, 1: no improvement in elasticity, and 0: loss of elasticity.

[0180] Example 3. Evaluation of cleaning efficacy using FTIR

[0181] Bleached hair samples can be obtained. Some of the bleached hair samples can be treated with the detoxifying hair and scalp cleanser composition described herein. For the treated and untreated samples, cross sections will be prepared using a cryostat, and the cross sections will be examined using attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR). Using this technology, a hyperspectral image will be generated, which can be used to visualize the lipid content within the hair fiber, assess hair damage (i.e., chemical treatment or UV exposure) within the hair fiber, identify modifications in the hair structure (i.e., protein conformation or lipid organization), evaluate the hydration of the hair (i.e., water content), visualize active penetration within the hair fiber, and determine the efficacy of the cleanser in removing sebum and deposits on the hair strands.

[0182] ATR-FTIR can be used to scan individual fibers of treated and untreated samples longitudinally, which can provide infrared images of approximately the outer 1-2 microns of the fiber surface. These hyperspectral images can be used to determine lipid content in the cuticle, study hair damage along the fiber (i.e., chemical treatment or UV exposure), analyze the deposition of active ingredients along the hair fiber, identify changes in hair structure (i.e., protein conformation or lipid organization), evaluate hair hydration (i.e., water content), and determine the efficacy of cleansers and products in removing sebum from the hair strands.

[0183] Example 4: Evaluation of Metal Reduction / Chelation

[0184] Bleached hair samples can be obtained. Some of the bleached hair samples can be treated with the detoxifying hair and scalp cleanser compositions described herein. Treated and untreated samples can be rinsed with hard water and examined using time-of-flight secondary ion mass spectrometry (TOF-SIMS). Spectra of calcium, magnesium, copper, aluminum, iron, chromium, zinc, nickel, cadmium, lead, and mercury can be obtained.
