Abstract
Description
ARTThe present disclosure relates generally to methods and compositions for controlling diptera insects, such as mosquitos and house flies, particularly disease-bearing mosquitos and diptera insects. More particularly, the present disclosure relates, among other embodiments, to methods for controlling mosquito populations and / or killing mosquito larvae or pupae comprising contacting one or more mosquito larvae or pupae or a mosquito slide surface or an aqueous mosquito slide environment likely to be infested with mosquito larvae or pupae with a larvicidal composition comprising benzopyrimoxan.BACKGROUNDHematophageal insect species such as mosquitos, rubble mosquitos, sandflies, tsetse flies, ticks, lice, fleas, midges, predances, and triatoinae are the major arthropod vectors of diseases due to their mouth tools and chemical agents to penetrate and feed from the vasculature of the skin of host animals. Hematophage diptera species in particular are considered to be particularly important vectors in both the biological and mechanical transmission of a variety of diseases. Many flies function as mechanical disease vectors by taking infectious agents on the outside of their bodies and transporting the infectious agent to an animal host where they transfer the pathogen to the host by physical contact. In mechanical transmission, insects are contaminated with a virus or other pathogen during normal feeding, and the virus or pathogen is persistent on their mouth tools or body until the next feeding, by which the virus or pathogen is transferred to a new host animal. In contrast, other diptera species, such as mosquitos, function as biological vectors by not only transporting pathogens but also playing a role in the life cycle of the pathogens, allowing the pathogen to proliferate in the insect's body before delivery to a new animal host.Mosquitos can transmit diseases such as malaria transmitted from various mosquitos of the genus Anopheline (such as Anopheles gambiae or Anopheles funestus), or the Zika virus, yellow fever, Dengue fever and Chikungunya transmitted from mosquitos of the genus Aedes (e.g., Aedes aegypti or Aedes albopictus). Approximately 390 million people infect dengue virus each year, at risk of about half of the world's population. In 2017, around 97,400 people infected with yellow fever, although vaccination is available. The Zika virus is widely distributed in tropical and subtropical areas in 86 countries and 2.2 million cases have been reported in 2017. While targeted efforts have been made to combat Aedes aegypti, the major vector of these nonmalarial diseases, including chemical protection measures against adults and larvae, management of larval sources, exposure to sterile populations and the use of wolbachiain-infected mosquitos, the most important methods for combating A. aegypti vectors in Asia continue to rely heavily on the use of larvicides.Insecticide-treated bed nets (ITN) and indoor residual spray (IRS) have been developed with the aim of vector controlling adult disease-bearing mosquitos and have contributed to a decrease in malaria cases by almost about 80% from 2000 to 2015. ITNs and IRSs are both interventions which target mosquitos which sting in interiors and rest in interiors, such as, for example, An. gambiae and An. funestus. However, An. stephensi, an invasive species of the Asian continent, has recently become established in Subsahara Africa (SSA) and now propagates throughout the region. An. stephensi is considered to be an outdoor lancing and outdoor resting mosquito that cannot be efficiently controlled by internal space interventions such as ITN and IRS. Other mosquito deposits, such as Aedes species and Culex species, are also mosquitos which are free-piercing and resting in the open, requiring interventions other than those used in malaria vectors. In particular, the use of larvae is generally recommended to combat outdoor mosquito populations and diseases transmitted by these vectors. Larvicides control mosquito populations by killing mosquito larvae at mosquito breeds, thereby reducing or eradicating adult mosquito populations in the treatment zone.In the past decades, only a limited number of compounds have been available for mosquito control, including organophosphates, carbamates and pyrethroids. Recently, neonicotinoids, butenolides, benzoylureas and juvenile hormone mimetics have also become available. However, in most cases the latter are used together with a pyrethroid. Pyrethroids are the most commonly used insecticidal class of vector control products and in 2022, about 80% of the vector control products certified by the World Health Organization (WHO) contain a pyrethroid. Pyrethroids have three advantages: they have good contact efficiency, they are fast acting, which is important because the mosquito often comes into contact with the compound for only a few seconds, and they have a good toxicological profile over mammals. However, due to the frequent use of pyrethroids, mosquitos have developed resistance to pyrethroids and pyrethroids can no longer reliably provide good protection from disease-transferring mosquitos. In addition, there has also been reported a cross-resistance of pyrethroid-resistant mosquitos to pyriproxyfen, one of the most commonly used larvae. In 2000, there were around 1 million deaths worldwide due to malaria. By more financial means, new campaigns, enhanced vector control, new medical treatments and antimalarial institutions, malaria cases and malaria deaths decreased to 400,000 in 2015. However, malaria deaths increased again to 600,000 in 2021, insecticide resistance and the spread of invasive an. stephensi being essential factors contributing to this trend. A problem with current insecticides used for mosquito vector control is the limited number of chemical classes available for mosquito control, which results in a limited number of pesticidal activities, leading to the rapid development of insecticide resistance in mosquito populations. Thus, Applicant has recognized that novel compositions and methods for killing or controlling mosquitos, particularly insecticide resistant mosquitos, are desirable for controlling malaria and other diseases transmitted by mosquitos.SUMMARYTo address these circumstances, Applicant has developed novel methods and compositions for killing mosquito larvae and for controlling populations of adult mosquitos. In particular, the methods disclosed herein involve the use of compositions containing a pesticidally effective amount of benzpyrimoxane or a salt, solute, hydrate or solvate thereof. The Applicant has surprisingly found that benzopyrimoxan exhibits good larvicidal activity against various mosquito species, including mosquito populations resistant to pesticides such as pyrethroid insecticides (e.g., mosquito populations having a deltamethrin resistance ratio of greater than 50). Surprisingly, mosquito larvae, when exposed to the presently disclosed benzpyrimoxan compositions, often develop through all four larval stages and into the pupae stage, but do not come out of the pupae stage or do not unpunculate to become stinging, disease-bearing adult vectors. Applicant has found that mosquito larvae contacted with compositions comprising higher benzpyrimoxan concentrations are killed without traversing all stages of larvae or are killed while still in the stage of development of a pupa. The present disclosure provides methods for killing mosquito larvae and methods for controlling populations of adult mosquitos by killing mosquito larvae and / or preventing further development of mosquito larvae beyond the stage of development of a pupa or depoping to form an adult mosquito. Specifically, benzpyrimoxan test solutions were prepared using a benzpyrimoxan stock solution formed by combining benzpyrimoxan and acetone in a mixing ratio of 1 mg (mg) of benzpyrimoxan per 1 ml of acetone. Benzpyrimoxane test solutions containing 10.0 ppm, 1.0 ppm, 0.1 ppm, 0.01 ppm, 0.001 ppm and 0.0001 ppm of benzpyrimoxane were prepared by diluting the benzpyrimoxane stock solution with water. Each test solution was placed in a round polypropylene test container 47 millimeters (mm) high and 100 mm in diameter. Ten (10) larvae stage 2 mosquito larvae were placed in each container, together with tetramine as feed source. Larvae were scored after one (1) day and then every two (2) to three (3) days until hatching. When the larvae pupate, the containers were capped. The rate of hatching of mosquito larvae from pupae to adult was determined for both Aedes aegypti and Anopheles stephensi mosquitos fourteen (14) days after treatment with the benzpyrimoxan test solution. The slip rate values versus the concentration of benzpyrimoxan in the test solutions are shown in Figure 1 for both pesticidally resistant and pesticidally acceptable Aedes aegypti larvae and in Figure 2 for Anopheles stephensi larvae. Error bars indicate the standard deviation (n=270 for FIG. 1 ; n=180 for FIG. 2 ). As shown in Figure 1, the slip rate of susceptible A. aegypti mosquito larvae decreased by about 25% when exposed to a 1.0 ppm benzpyrimoxane solution. When exposed to a 10.0 ppm benzpyrimoxan solution, 100% of the A. aegypti susceptable mosquito larvae were prevented from hatching and died, while over 90% of the A. aegypti pesticide resistant larvae did not hatching. As shown in Figure 2, the slip rate of An. stephensi mosquito larvae was decreased by over 90% when contacted with the 1.0 ppm of benzpyrimoxane solution.Mortality of A. aegypti and An. stephensi mosquito larvae was also determined for 1 ppm and 10 ppm of benzpyrimoxan test solutions during the fourteen (14) day test period (Figs. 3-4). Specifically, Figure 3 illustrates mortality of A. aegypti mosquito larvae after contact with a 10 ppm benzpyrimoxane test solution. Error bars indicate the standard deviation (n=270). As shown in Figure 3, significant mortality occurred 2 days after contact with the 10 ppm test solution, with maximum mortality observed 7 days after contact. After 7 days, over 90% of the susceptible and over 60% of the pesticidally resistant A. aegypti mosquito larvae were killed upon contact with the 10 ppm benzpyrimoxane test solution. Figure 4 illustrates mortality data for An. stephensiwhen exposed to either the 1 ppm or 10 ppm benzpyrimoxane test solution. Error bars indicate the standard deviation (n=180). As shown in Figure 4, 100% mortality was observed for An. stephensi larvae 2 days after contact with the 10 ppm benzpyrimoxan test solution. Upon contact with the 1 ppm test solution, greater than 50% mortality was observed after 2 days and 70% mortality was observed after 14 days.Other objects, features, and advantages of the disclosure will be apparent from the above-mentioned figures, detailed description, and examples.
