Abstract
Description
Title of the invention: COSMETIC SUNSCREEN COMPOSITION CONTAINING AMINO ACIDS ANALOGUES OF MY-COSPORIN Scope of the invention
[0001] The present invention relates to a composition, in particular a cosmetic or dermatological composition, advantageously solar, comprising amino acids analogous to mycosporin and sun filters, as well as its use to protect the skin from ultraviolet radiation. Prior state of the art
[0002] Ultraviolet (UV) radiation consists of light with wavelengths between 100 nm and 400 nm, traditionally divided into three subgroups: UV-A (400-315 nm), UV-B (315-280 nm), and UV-C (280-100 nm). UV-C, with its shorter wavelength, is the most energetic and harmful form of UV radiation. It is filtered by the ozone layer in the atmosphere and does not reach the Earth's surface in significant quantities. Most skin damage is caused by UV-A and UV-B radiation.
[0003] UV-B rays induce the production of the skin's natural pigment called "melanin." This is the origin of the phenomenon commonly known as "tanning." UV-B rays also stimulate cells to produce a thicker epidermis. The reactions described above constitute a defense mechanism of the body against UV radiation.
[0004] The high energy of UV-B radiation generates molecular disorders (DNA alteration and protein damage) which, in the long term, saturate and disrupt the nuclear DNA repair system. This leads to permanent mutations in the genome of affected cells, which are the cause of skin cancers. This is a direct form of UV-B toxicity. As for UV-A radiation, it is known that it penetrates the deep layers of the skin, where it produces deleterious effects, particularly on connective tissue and blood vessels. It is notably responsible for the phenomenon known as "photoaging," that is, premature actinic aging of the skin.
[0005] Moreover, although UV-B rays are the primary cause of skin cancers, UV-A rays also make an indirect contribution to this type of damage. Indeed, both UV-A and UV-B rays are responsible for the production of free radicals, particularly ROS (Reactive Oxygen Species). These are highly unstable molecules with a very short half-life, on the order of from nanoseconds to milliseconds. ROS are capable of damaging intracellular structures (DNA, membranes, intracellular proteins, etc.) as well as extracellular structures (components of the extracellular matrix such as collagen fibers). ROS also exert an indirect harmful effect by causing the oxidation of membrane lipids, which induces the formation of reactive carbonyl species that contribute to amplifying tissue and cellular damage.
[0006] It is therefore necessary to combat the harmful effects of UV radiation, particularly UV-A and UV-B, taking into account inter-individual differences such as skin type (phototype) and sun exposure times. For this reason, sunscreen compositions have been developed to provide photoprotection for areas exposed to UV radiation. These compositions are available in the form of lotions, oils, oil-in-water or water-in-oil emulsions, foams, gels, sticks, or sprays. They contain a cosmetically acceptable base and one or more chemical filters or mineral sunscreens at various concentrations.
[0007] Mineral sunscreens, including zinc oxides and titanium dioxides, protect against UV radiation by reflecting sunlight. They therefore act like mirrors localized on the surface of the skin. Mineral sunscreens, at least in micrometric formulations, are safe for human use and pose no health risks.
[0008] However, they have other disadvantages: - Mineral sunscreens tend to dry out the skin and are difficult to spread, leaving white streaks on the skin which can be very unsightly; - The galenic assessment of compositions based on mineral screens is often poor, particularly in compositions free of silicones.
[0009] Chemical filters are organic compounds that absorb light, creating a filtering layer on the skin that neutralizes UV rays. Thus, when the sunscreen molecule receives UV radiation, it enters an excited state by absorbing the energy of the radiation and then returns to its stable state. Through this mechanism, the light radiation is converted into heat and dissipated. Chemical filters therefore act in the same way as melanin, the skin's natural photoprotective pigment. Most chemical or organic filters are lipophilic and are therefore suitable for use in highly water-resistant compositions. From a pharmaceutical standpoint, compositions based on organic filters are generally more popular than those based on mineral sunscreens, which improves user compliance.
[0010] In Europe, all organic filters or mineral screens authorized by regulations are listed (Annex VI of the European Directive on cosmetic products). This is a list currently comprising around thirty compounds, identified by their INCI designation, from which the cosmetics industry must choose ingredients to formulate sunscreens. However, this list is not fixed and is constantly evolving based on the results of scientific research. Indeed, several studies have shown that some of the organic filters on this list could pose risks to human health and the environment, particularly because they are suspected of acting as endocrine disruptors.
[0011] In particular, scientific results call into question the safety of benzophenone-4 or benzophenone-3 (Kinnberg et al. 2015; Zucchi et al. 2011), octyl methoxycinnamate (Szwarcfarb et al. 2008, Axelstad et al. 2011), 3-methylbenzylidene camphor and 4-methylbenzylidene camphor (Schlumpf et al. 2008; Petersen et al. 2007), octocrylene, isoamyl methoxycinnamate, or octyl dimethyl PABA (Schlumpf et al. 2001; Rehfeld et al. 2016; Ozäez et al. 2016).
[0012] In addition to the potential effects on humans, the impact of these molecules on natural environments cannot be ignored; indeed, the filters are continuously dispersed into bathing water or wastewater. This water pollution, combined with UV filters, inevitably affects the entire food chain, with unpredictable consequences for the balance of an ecosystem already compromised by human activities; among other examples of ecotoxic effects, sunscreens are believed to have a negative impact on marine reefs and are notably responsible for the phenomenon of coral bleaching (Danovaro et al. 2008).
[0013] The progressive reduction in the number of organic sunscreens, considered safe for human use and for the environment, is increasingly complicating the formulation of sun cosmetic products, in particular high protection factor sunscreens, i.e. with an SPF (Sun protection factor) greater than 20 or even 30. The formulation of high sun protection products is particularly difficult in the case of products intended for the Canadian and American markets, where only a portion of the latest generation filters used in Europe for the past two decades are authorized.
[0014] To compensate for the reduction in the number of usable sunscreens, sun protection agents are often formulated in cosmetic products. These sun protection agents are sometimes molecules similar to authorized sunscreens, or plastic derivatives, which can therefore suffer from the same drawbacks as organic sunscreens; in particular, their ecotoxicity could be significant.
[0015] An interesting family of protective agents is represented by mycosporine-like amino acids (MAAs). MAAs are small heterocyclic molecules (<400 Da) comprising cyclohexenone or cyclohexeminine chromophores.
[0016] MAAs are found naturally as secondary metabolites in marine organisms, such as corals, algae and cyanobacteria, so they are expected to be eco-compatible and respectful of aquatic ecosystems.
[0017] In particular, document EP2855441 discloses a family of MAA-type compounds having the following general IE formula:
[0018] [Chem.l]
[0019] This document specifies that these compounds, from the IE molecule family, absorb ultraviolet radiation and can be used as UV-blocking agents. As an example, compounds with formulas IAi and IE4 are given, having sun protection factors of 2.4 and 4.5 respectively.
[0020] Thus, although MAAs of the IE molecule family absorb UV radiation, the sun protection factor is insufficient for use in sun cosmetic compositions.
[0021] Although considered good candidates for the formulation of eco-friendly sun products, a solution must be found to improve the anti-UV potential of MAAs, in order to be able to use them in sun products.
[0022] Thus, the present invention aims to overcome the drawbacks of the prior art, by proposing a composition comprising MMAs and whose SPF is compatible with a topical solar application. Description of the invention
[0023] The Applicant found that, quite surprisingly, combining MMAs with organic filters resulted in an SPF for the combination that was higher than the sum of the respective SPFs of the MAAs and the organic filters.
[0024] This combination is therefore suitable for their implementation in high sun protection compositions with relatively low organic filter contents.

[0138] Example IV - Measurement of the sun protection factor (SPF) obtained with compositions according to the invention IV-1 Purpose of the study
[0139] The objective of the study was the development of compositions with a high sun protection factor, i.e. with an SPF>20, incorporating organic UV filters and MAAs according to the invention. IV-2 Materials and methods
[0140] An in vitro method for measuring the solar protection factor was used. A first UV absorption measurement is carried out on a plate (PMMA molded plate or SB6 sandblasted plate, Helioscreen) coated with 15 lp of glycerin (white), thus eliminating the absorption of the substrate.
[0141] For formulas fiel and fie 2, the measurements were carried out using molded HD6 plates, according to the following protocol.
[0142] Next, the product under study is sampled and deposited in 16 spots onto the Helioscreen HD6 PMMA molded plate (5 x 5 cm). The weighed quantity is 32.5 mg, corresponding to a concentration of 1.3 mg / cm². The sample is spread uniformly using a robotic spreader (HD-SPREADMASTER - Helioscreen). The plate is then left in the dark at room temperature for at least 15 minutes. The plate is placed in the measurement chamber of the Labsphère UV 2000 S spectrophotometer to perform UV absorption measurements between 290 and 400 nm. Note that one measurement actually corresponds to the average of 9 measurements (9 points on the PMMA plate are measured).
[0143] In the cases of formulas Fig. 3 to Fig. 7, the protocol is identical but SB6 sandblasted plates were used. For these plates, the weighed quantity is 30 mg, which corresponds to a concentration of 1.2 mg / cm².
[0144] The theoretical SPF values were calculated for individual filters, as in vitro SPF cannot be measured under these conditions. The BASF simulator was used for these measurements (Herzog et al. 2003).
[0145] The following formulas were developed and tested:

[0154] Table 7: SPF measurements from figures 3 to 7
[0155] In the case of tests relating to broad spectrum filters + UVA filter, a very low SPF was obtained for formula 1 (fig 1) containing an MAA according to the invention (INCI: Methoxyphenylimino dimethylcyclohexene glycine), a UVA filter and a broad spectrum filter.
[0156] The addition of a second broad spectrum filter made it possible to go from an SPF value of 5 to an SPF value of 21, whereas the theoretical expected contribution for the broad spectrum filter was 7.5.
[0157] For the tests relating to UVB +UVA filters, the addition of a UVB filter in formula 4, containing an MAA according to the invention (INCI: methoxyphenylimino dimethylcyclohexene glycine), makes it possible to increase the SPF value from 12.5 to an SPF value of 24.11. The addition of the same filter in the same concentration in formula 6, containing another MAA according to the invention (INCI: methoxyphenylimino dimethyl-hexahydro benzothiazine carboxylic acid) makes it possible to increase the SPF value from 11.73 to an SPF value of 25.93.
[0158] The expected theoretical contribution for the 9% homosalate UVB filter was 4.6.
[0159] The comparison between tests 3 and 5, which differ only in the presence of a MAA according to the invention, validates the solar protection amplifier function of MAA (SPF 18.62 ->24.11).
[0160] In conclusion, the combinations of filters and MAAs according to the invention make it possible to obtain products characterized by a good protection index (SPF > 20), while limiting the introduction of organic filters in the formulas thanks to the sun protection amplifier action of the MAAs.
[0161] Thus, the present invention makes it possible to reduce the quantity and concentration of organic filters required in a sunscreen product, while maintaining a high sun protection factor, i.e., greater than 20. The use of MAAs as a UV blocker, in combination with organic filters, is an eco-friendly, environmentally responsible, and safe solution for humans. References
