[0002] Human skin forms a barrier between the external environment and the human body and is continually subjected to external aggressions. There are a large number of environmental factors and diseases affecting the skin and in particular the barrier function of the skin. This can lead to inflammation of the skin and thus reduce the barrier properties of the skin. Psoriasis is for example a skin disease involving an inflammatory skin reaction and altering the barrier function of the skin.

[0003] The ionization of water is known and ionized water is today mainly used as a descaler, particularly for household appliances.

[0004] It is known that the gradient of ions such as calcium or magnesium plays a crucial role in the homeostasis of the skin barrier.

[0005] Today there are many cosmetic compositions that can restore the skin's barrier function and moisturize the skin or even treat psoriasis.

[0006] These compositions comprise a large number of cosmetic ingredients, some of which may prove harmful to the body, in particular sulfated compounds.

[0007] To date, there are no compositions comprising hyperionized water that can improve the barrier function and hydration of the skin and treat psoriasis.

[0008] Thus, there is a need for cosmetic compositions with few or no ingredients that can treat psoriasis. Statement of the invention

[0011] According to a characteristic of the invention, the composition also comprises an anti-inflammatory agent.

[0012] According to another characteristic of the invention, the composition also comprises an antioxidant agent.

[0013] According to yet another characteristic of the invention, the composition also comprises a humectant.

[0014] According to yet another characteristic of the invention, the composition is in the form of a cream, a gel, a lotion, a serum, a balm or a mousse.

[0015] A very first advantage of the present invention lies in the restorative effect of the skin's barrier function.

[0016] Another advantage of the present invention lies in the improvement of the formation and strengthening of the stratum corneum.

[0017] Yet another advantage of the present invention is the improvement of skin hydration.

[0027] DETAILED DESCRIPTION OF EMBODIMENTS OF THE INVENTION

[0028] As described above, the invention relates to a composition comprising as active ingredient hyperionized water having a zeta potential of between -15 mV and -1 mV for use in treating psoriasis.

[0029] The inventors tested the effects of hyperionized water using a hyperionization device and discovered that treating water with a hyperionization device modified the electrical characteristics of the treated water. This type of device makes it possible to reduce the zeta potential of the treated water. This implies that positively charged ions such as calcium, potassium and magnesium, naturally present in water, combine with particles of opposite charges to form nanoparticles called colloids. Thus, the closer the zeta potential is to 0, the more the particles tend to aggregate.

[0030] Thus, treating water with a hyperionization device makes it possible to reduce the zeta potential of the treated water by 20% to 50% compared to untreated water. For example, tap water with a zeta potential of -18.3 mV sees its zeta potential drop to -13.8 mV after treatment with a hyperionization device.

[0031] This indicates the appearance of charged substances with a size of a few hundred nanometers and of a colloidal nature.

[0032] The inventors also discovered that treating water with a hyperionization device modified the RMS (Relative Magnitude Spectrum) conductivity of the treated water. This parameter was obtained by carrying out EIS impedance spectrometry measurements, which also allows ultrasensitive measurements of variations in the electrochemical parameters of electrolytic solutions.

[0033] Thus, the treatment of water by a hyperionization device makes it possible to increase the RMS conductivity of the treated water by 25% to 60% compared to untreated water. For example, tap water with RMS conductivity of 54 pS.cm 1 sees its RMS conductivity increase to 71 pS.cm 1 after treatment by a hyperionization device.

[0034] This indicates a modification of the arrangement of water molecules with the creation of coherence domains (ice-like structures in contact with colloids) which promote the transfer of protons through hydrogen bonds. This therefore makes it possible to create a proton current in the treated water which plays a role in many energy transduction processes along metabolic chains.

[0035] The inventors also carried out fluorescence spectroscopy measurements to evaluate the effect of magnetization of water treated by a hyperionization device. To do this, they diluted in the water a fluorophore having an excitation wavelength of 450 nm and an emission wavelength between 500 nm and 550 nm. The intensity of the light emission emitted in the range between 350 nm and 750 nm is then measured. The emission spectra of a Water treated by a hyperionization device and untreated water are then compared.

[0036] The inventors observed an increase in the intensity peak of 35% to 95% of the treated water compared to the untreated water.

[0037] The inventors therefore imagined a composition comprising hyperionized water having a zeta potential of between -15 mV and -1 mV to treat the skin and this is how they noted with surprise that this composition had a beneficial effect on the skin.

[0038] The composition according to the invention makes it possible to treat psoriatic skin, to hydrate the skin and to repair the skin barrier.

[0039] To do this, the inventors tested the effect of water treated by a hyperionization device on delipidated skin explants.

[0040] In order to evaluate the effects of treated water on skin hydration, the inventors observed the expression of filaggrin, involucrin and aquaporin 3.

[0041] To do this, the inventors tested the effect of product P corresponding to hyperionized treated water having a zeta potential of -8 mV.

[0042] The inventors prepared explants of human skin from an abdominal plastic surgery and divided them into six batches: - lot T0 corresponding to the plastic surgery witness, - batch D0 corresponding to the delipidation witness, - batch T corresponding to the untreated control, - batch P corresponding to the batch treated with product P, - batch D corresponding to the defatted batch, and - the DP batch corresponding to the defatted batch treated with product P.

[0043] The delipidation of batches D0, D and DP is carried out by applying an ether / acetone mixture to the surface of the skin. All the explants are then cultured in a culture medium, in a humid atmosphere enriched with 5% CO2.

[0044] Immediately after delipidation (D=0), 1 day after delipidation (D=1), 4 days after delipidation (D=4), 6 days after delipidation (D=6) and 8 days after delipidation (D=8), product P was applied topically to batches P and DP at a rate of 2 μL per explant. The explants were then left at room temperature for 2 hours and then returned to culture.

[0045] Batch T received no treatment except for the renewal of culture medium.

[0046] AJ=0, the 3 explants of batches T0 and D0 were taken and cut in two. A part was fixed in buffered formalin solution and another part was frozen at -80°C.

[0047] 3 hours after delipidation (h=3), 3 explants from batches P, D and DP were taken and treated in the same way as the explants of batches T0 and D0 at D=0.

[0048] 24 hours after delipidation (h=24), 3 explants from batches T, P, D and DP were collected and treated in the same way as the explants of batches T0 and DO at D=0.

[0049] AJ=6, 3 explants from batches T and P were taken and treated in the same way as the explants from batches T0 and DO at D=0.

[0050] 11 days after survival (D=1 1), 3 explants from batches T and P were taken and treated in the same way as the explants of batches T0 and D0 at D=0.

[0051] After 24 hours in buffered formalin, the samples were dehydrated and impregnated in paraffin and then put into blocks. 5 μm sections were made and mounted on a histological glass slide.

[0052] The cell viability of the epidermal, dermal and stratum corneum structures of each explant was observed on paraffin sections after staining with Masson's trichrome. These observations showed that product P does not induce any modification of cell viability. However, the product improves the thickness and the flaky appearance of the stratum corneum, reflecting a better state of hydration of the stratum corneum.

[0053] Filaggrin expression was observed by immunostaining filaggrin from batches T0, D0, T at h=24, D at h=24, D at h=3 and h=24 and DP at h=3 and h=24. Filaggrin was labeled on paraffin sections with an anti-filaggrin monoclonal antibody, diluted 1 / 500 in PBS-BSA 0.3%-Tween 20 at 0.05% for 1 hour at room temperature, and revealed in AlexaFluor 488. The nuclei were counterstained with propidium iodide.

[0054] The expression of involucrin was observed by performing immunolabeling of involucrin from batches T0, T at D=6 and P at D=6. Involucrin was labeled on paraffin sections with an anti-involucrin monoclonal antibody, diluted 1 / 100 in PBS-BSA 0.3% overnight at room temperature with a biotin / streptavidin amplifier system and revealed in VIP.

[0055] The expression of FAQP3 was observed by performing immunolabeling of AQP3 from batches T0, T at D=6 and D=11, P at D=6 and D=11. AQP3 was labeled on paraffin sections with an anti-AQP3 polyclonal antibody, diluted to 1 / 1000 in PBS-BSA 0.3%-Tween 20 0.05% for 1 hour at room temperature with a biotin / streptavidin amplifier system and revealed in VIP.

[0056] In Figures 1 to 5, TF corresponds to very weak marking, F to weak marking, M to moderate marking, AN to fairly clear marking, N to clear marking, TF to very clear marking and Fo to strong marking.

[0057] [Fig. 1] illustrates the results of the labeling and therefore the expression of filaggrin at the base of the stratum corneum.

[0058] Filaggrin is initially synthesized in the form of its precursor, pro-filaggrin, a highly phosphorylated and insoluble protein of approximately 400 kDa, which represents the main constituent of the keratohyalin granules of the granular layer. During the late phase of epidermal differentiation, pro-filaggrin is dephosphorylated and cleaved, by capsase-14, into several filaggrin monomers. Filaggrin thus binds to the keratin intermediate filaments, participating in their aggregation into macrofibrils and the formation of the horny envelope. At the intermediate zone of the stratum corneum, the filaggrin monomers are then proteolyzed. This proteolysis participates in the formation of a very hygroscopic complex, the natural moisturizing factor (NMF), which also helps maintain the hydration of the horny layer and the suppleness of the skin.

[0059] AJ=0, the expression of filaggrin is low on batch T0 and the delipidation carried out on batch D does not induce any modification of the expression of filaggrin compared to T0.

[0060] At h=3, the delipidation carried out on batch D induces a slight decrease in the expression of filaggrin compared to T0. The application of product P after delipidation on batch DP induces an increase in the expression of filaggrin compared to batch D. The treated water having a zeta potential equal to -8 mV therefore induces complete protection against the repression of the expression of filaggrin induced by delipidation.

[0061] At h=24, the filaggrin labeling is moderate on batch T and the delipidation carried out on batch D induces a decrease in the expression of filaggrin compared to T. The application of product P on batch P does not induce any modification of the expression of filaggrin compared to T. The application of product P after delipidation on batch DP induces an increase in the expression of filaggrin compared to batch D. The treated water having a zeta potential equal to -8 mV therefore induces complete protection against the repression of the expression of filaggrin induced by delipidation.

[0062] Thus, the delipidation of the skin surface leads to an alteration of the skin's barrier function. In response, the skin attempts to restore its barrier function, in particular by degrading filaggrin to increase NMFs, which are capable of binding a large quantity of water and ensuring good hydration of the stratum corneum.

[0063] When treated water with a zeta potential equal to -8 mV is applied after delipidation, filaggrin is not degraded. This implies that treated water with a zeta potential equal to -8 mV has rapidly compensated or restored the skin barrier function, making filaggrin degradation unnecessary.

[0064] Treated water with a zeta potential equal to -8 mV therefore has an excellent restorative effect on the skin's barrier function.

[0065] The inventors found that treated water with a zeta potential equal to -8 mV also improved the thickness and flaky appearance of the stratum corneum. This improvement results in a better hydration state of the stratum corneum.

[0066] Thus, treated water with a zeta potential equal to -8 mV improves skin hydration.

[0067] Thus, the composition according to the invention makes it possible to improve the hydration of the skin.

[0068] [Fig.2] illustrates the results of the labeling and therefore of the expression of involucrin in the granular layer of the epidermis.

[0069] Involucrin is a structural protein located in the upper epidermis. Involucrin is an essential component of the horny envelope, promoting its formation. Involucrin is commonly used in dermo-cosmetic research as a marker of terminal differentiation of keratinocytes.

[0070] AJ=0, the expression of involucrin is fairly clear to clear on batch T0.

[0071] AJ=6, the expression of involucrin is quite clear on batch T. The application of the product P on batch P induces a slight increase in the expression of involucrin compared to batch T.

[0072] Thus, treated water with a zeta potential equal to -8 mV induces an increase in the expression of involucrin. Treated water with a zeta potential equal to -8 mV thus improves the formation of the horny envelopes necessary during cell differentiation and the formation of the stratum corneum.

[0073] Treated water with a zeta potential equal to -8 mV therefore has a notable effect on the formation and strengthening of the stratum corneum.

[0074] Thus, the composition according to the invention makes it possible to improve the formation and strengthening of the stratum corneum and to participate in the repair of the skin barrier.

[0075] [Fig.3] illustrates the results of the labeling and therefore of the expression of AQP3 in the epidermis.

[0076] Aquaporins are a family of small, hydrophobic integral membrane proteins. They are involved in the transport of water across the plasma membrane and are notably expressed in keratinocytes. Aquaporin type 3 (AQP3) is a specific membrane transporter for water and glycerol and is expressed within the epidermis. Its expression can be affected by skin pathologies such as atopic eczema, by agents such as retinol or by abiotic agents such as UV rays.

[0077] AJ=0, the expression of AQP3 is clear to very clear on batch T0.

[0078] AJ=6, the expression of AQP3 is clear to very clear on batch T. The application of the product P on batch P induces a slight decrease in FAQP3 expression compared to batch T.

[0079] AJ=11, the expression of AQP3 is quite clear. The application of product P on batch P induces a fairly clear decrease in the expression of FAQP3 compared to batch T.

[0080] Thus, treated water with a zeta potential equal to -8 mV induces a decrease in the expression of AQP3. However, it improves the repair of the skin barrier function, it improves the appearance of the stratum corneum and promotes the formation of horny envelopes. These characteristics show that treated water with a zeta potential equal to -8 mV improves the stratum corneum and its barrier function, thus reducing its water requirements. Since the barrier function is reinforced, water evaporation is reduced and does not require increasing the water flow from the dermis to the epidermis by overexpressing AQP3.

[0081] Which explains the decrease in the expression of AQP3.

[0082] Treated water with a zeta potential equal to -8 mV therefore has a good moisturizing effect on the skin.

[0083] The inventors arrived at the same results described previously with hyperionized waters having a zeta potential equal to -15 mV, -11 mV, -9 mV, -7 mV, -5 mV, -3 mV and -1 mV.

[0084] Similarly, the inventors arrived at the same results with hyperionized waters having an RMS conductivity equal to 65 pS.cm ', 70 pS.cm ', 75 pS.cm ', 80 pS.cm ', 85 pS.cm1, 90 pS.cm1 and 95 pS.cm *.

[0085] The inventors also analyzed the effect of hyperionized water having a zeta potential between -15 mV and -1 mV on psoriatic skin and realized that the composition according to the invention makes it possible to treat psoriasis.

[0086] The inventors then analyzed the biological activity of the composition according to the invention on ex vivo skin explants and realized that the composition according to the invention induced at the level of psoriatic human skin: - a reduction in abnormal thickening of the epidermis, - a reduction in the overexpression of cytokeratin 16 (CK16), - a reduction in the overexpression of psoriasin S100A7, and - a decrease in the release of TNF-alpha, and - a reduction in the inflammatory state.

[0087] To do this, the inventors tested the effect of the following products: • the PI product corresponding to untreated city water, and • the product P2 corresponding to the composition according to the invention.

[0088] The inventors prepared explants of human skin from an abdominal plastic surgery and divided them into seven batches: - batch T0 corresponding to the plastic surgery control, - batch T corresponding to the untreated control, - the PI batch corresponding to the batch treated with the PI product, - batch P2 corresponding to the batch treated with product P2, - The SI batch corresponding to the “psoriasis-induced” batch - batch SIP1 corresponding to the “psoriasis-induced” batch then treated with the PI product, and - batch SIP2 corresponding to the “psoriasis-induced” batch then treated with product P2.

[0089] At t = 0 days (DO), the explants of batches SI, SIP1 and SIP2 were treated to induce psoriasis. To do this, the explants were stripped 10 times with 3M tape and then Aldara 5% Imiquimol was applied topically at a rate of 2 mg / cm2.

[0090] 4h30 after the induction of psoriasis in batches SI, SIP1 and SIP2, at t = 1 day (D 1), t = 2 days (D2), t = 3 days (D3) and t = 4 days (D4), the PI product was applied to the explants of batches PI and SIP1 and the P2 product was applied to the explants of batches P2 and SIP2. The PI and P2 products were applied topically at a rate of 2 μL per explant.

[0091] The explants of batches T0 and T did not undergo any treatment except the renewal of the culture medium.

[0092] At D0, the explants from batch T0 were removed and cut into two parts. The first was fixed in a buffered formalin solution and the second part was frozen at -80°C for histological analysis.

[0093] On D4, the explants from batches T, PI, P2, SIP1 and SIP2 were collected and treated in the same way as the explants from batch T0. The culture media were also collected for biochemical assays.

[0094] After 24 hours in buffered formalin, the samples were dehydrated and impregnated in paraffin and then put into blocks. 5 μm sections were made and mounted on histological glass slides.

[0095] The general morphology of the dermal and epidermal structures was analyzed on paraffin sections after staining with Masson trichrome Goldner variant and evaluated by microscopic examination.

[0096] The expression of CK16 was observed by performing immunolabeling of CK16 from batches T0, T, PI, P2, SI, SIP1 and SIP2 at D4. CK16 was labeled on paraffin sections with an anti-CK16 monoclonal antibody diluted 1 / 400 in PBS-BSA 0.3%-Tween 20 at 0.05% for 1 h at room temperature with a biotin / streptavidin amplifier system and revealed in VIP, a peroxidase substrate giving a purple color once oxidized. The immunolabeling is then evaluated by microscopic observation.

[0097] The expression of psoriasin S100A7 was observed by performing immunolabeling of psoriasin from batches T0, T, PI, P2, SI, SIP1 and SIP2 at D4. Psoriasin was labeled on paraffin sections with an anti-S100A7 monoclonal antibody diluted 1 / 200 in PBS-BSA 0.3%-Tween 20 at 0.05% for 1 h at room temperature, with a biotin / streptavidin amplifier system revealed in VIP, a peroxidase substrate giving a purple color once oxidized. The immunolabeling is then evaluated by microscopic observation.

[0098] The release of TNF-alpha was observed by performing a TNF-alpha assay in the culture medium of batches T0, T, PI, P2, SI, SIP1 and SIP2 at D4. TNF-alpha was assayed using a suitable ELIS A kit.

[0099] The thickness of the epidermis was measured on the explants of all batches at T = 4 days (F4).

[0100] [Fig.4] illustrates the results of the labeling and therefore of the expression of CK16 in the epidermis.

[0101] Cytokeratins (CK) are structural proteins that form intermediate filaments within all epithelial tissues. In healthy tissues, CK16 is constitutively expressed at low levels in the palmar and plantar epidermis, tongue, oral mucosa, and hair follicles. CK16 plays a role in modulating cell proliferation. In psoriasis, for example, it is common to correlate the amount of CK16 with disease severity.

[0102] Indeed, CK16 is particularly overexpressed in psoriatic skin. It induces a proliferation of keratinocytes which leads to a strong increase in the thickness of the epidermis.

[0103] It can be seen in [Fig.4] that the marking of CK16 at the level of the epidermis is weak on batch T0, weak to moderate on batches PI and P2, moderate on batch T, moderate to fairly clear on batch SIP2, fairly clear to clear on batch SIP1 and clear on batch SL

[0104] Thus, the P2 product induces a decrease in the expression of CK16 compared to the SI and SIP2 batches.

[0105] The composition according to the invention makes it possible to reduce the expression of CK16 in the epidermis of psoriatic skin. It therefore makes it possible to reduce one of the characteristic parameters of psoriatic skin and to treat psoriasis.

[0106] [Fig.5] illustrates the results of the labeling and therefore of the expression of psoriasin in the epidermis.

[0107] S100 proteins are part of a multigene family that encodes proteins involved in the terminal differentiation complex. Psoriasin S100A7 is further expressed in human epidermis and keratinocytes and has been identified as an antimicrobial peptide overexpressed in psoriatic lesions.

[0108] It can be seen in [Fig.5] that the psoriasin marking at the epidermis level is weak on batch T0, moderate on batches T, PI and P2, fairly clear to clear on SIP2, clear to very clear on batch SIP1 and very clear on batch SI.

[0109] Thus, the P2 product induces a decrease in the expression of psoriasin compared to batches SI and SIP1.

[0110] The composition according to the invention makes it possible to reduce the expression of psoriasin in the epidermis of psoriatic skin. It therefore makes it possible to reduce the psoriatic condition of the skin and to treat psoriasis.

[0111] The table below illustrates the results of the dosage and therefore of the release of TNF-alpha in the culture medium of batches T, PI, P2, SI, SIP1 and SIP2 at D4.

[0112] [Tables 1] TNF-alpha concentration (pg / mL) T PI P2 SI SIP1 SIP2 Mean 12.3 12.7 17.8 15.2 13.7 13.3

[0113] Thus, it is observed that the product P2 induces a reduction in the release of TNF-alpha in the culture medium compared to batches SI and SIP1.

[0114] TNF-alpha is a biological marker of inflammation which is one of the characteristics of psoriatic skin.

[0115] The composition according to the invention makes it possible to reduce the release of TNF-alpha into the extracellular medium by the keratinocytes of psoriatic skin. It therefore makes it possible to reduce the inflammatory reaction induced by psoriasis and therefore to treat psoriasis.

[0116] The table below illustrates the effect of the composition according to the invention on the thickness of the epidermis of psoriatic skin.

[0117] [Tables2] Epidermal thickness (pm) T0 T PI P2 SI SIP1 SIP2 Average 35.8 46.3 45.2 44.3 79.2 79.1 69.1

[0118] Thus, it is observed that product P2 induces a reduction in the thickness of the epidermis compared to batches SI and SIP1.

[0119] The composition according to the invention makes it possible to reduce the thickness of the epidermis of psoriatic skin.

[0120] The composition according to the invention makes it possible to reduce the characteristic parameters of psoriatic skin, which are increased keratinocyte proliferation resulting in an abnormally thick epidermis, an inflammatory state of the skin and overexpression of psoriasin.
