[0045] The present invention will be described in more detail below with reference to examples, but the present invention is not limited to these examples.
[0046] <Test 1: Evaluation of epidermal keratinocyte activation> Epidermal keratinocyte activation was evaluated.
[0047] [Test substance] The test substances used were (A) procyanidin B1 reagent (manufactured by ChemFaces) as procyanidin B1, (B) procyanidin B3 reagent (manufactured by ChemFaces) as procyanidin B3, and (C) catechin reagent (manufactured by Nagara Science) as catechin.
[0048] [cell] Human epidermal keratinocytes (NHEK) were used as epidermal keratinocytes.
[0049] [culture] In a 37℃, 5% CO2 incubator, 75 cm 2 Human epidermal keratinocytes were cultured in a keratinocyte growth medium (Takara Bio) flask. The cells were then suspended by lactase treatment and placed in a 75 cm 2 From the flask, place 8 x 10 cells in each well of a 96-well plate. 3 The cells were seeded at a density of 100 cells / well and precultured for 24 hours in a 5% by volume CO2 incubator at 37°C.
[0050] [Epidermal keratinocyte proliferation test] After removing the medium from each well, 100 μL / well of medium containing a test substance prepared to a predetermined concentration was added, and the cells were cultured in a 37°C, 5% by volume CO2 incubator for 24 hours. As the test substance, the compositions of Examples 1 to 9 and Comparative Examples 1 to 5 shown in Table 1 below were used. The medium containing the test substance was prepared so that the total concentration of each component of the composition in the culture medium was 1 μg / mL. As a control, a culture medium containing no test substance was added.
[0051] [Table 1]
[0052] After 24 hours of culture, the medium was removed, and 150 μL of Cell Counting Kit-8 (Dojindo Chemical Industries, Ltd.) diluted 30-fold with serum-free DMEM was added to each well. The plate was placed in a 37°C, 5% by volume CO2 incubator to allow for adequate color development, and the absorbance at 450 nm was measured. Based on the data obtained, the % of control (relative number of live cells with the number of live cells in the control taken as 100%) was calculated. The results are shown in Figure 1. The absorbance ratio and the ratio of human epidermal keratinocyte counts are directly proportional to each other, so the absorbance ratio indicates the ratio of human epidermal keratinocyte counts. % of control=(Data sample-Data blank) / (Data control-Data blank)×100
[0053] [Test Results] 1, the group (Examples 1 to 9) in which procyanidin B1 (A) and procyanidin B3 (B) were added and the mass ratio of component (A) to component (B) [(A) / (B)] was 1 or more exhibited epidermal keratinocyte activation, compared to the group (Comparative Examples 1 to 4) in which procyanidin B1 (A) and procyanidin B3 (B) were added and the mass ratio of component (A) to component (B) [(A) / (B)] was less than 1, and the group (Comparative Example 5) in which catechin (C) was added alone. Among Examples 1 to 9, the group (Examples 4 to 9) in which catechin (C) was added exhibited superior epidermal keratinocyte activation, compared to the group (Examples 1 to 3) in which catechin (C) was not added. Furthermore, among them, the group (Examples 4 to 8) in which the mass ratio [(A) / (C)] of component (A) to component (C) is 0.5 or more was found to have particularly excellent epidermal keratinocyte activation, compared to the group (Example 9) in which the mass ratio [(A) / (C)] of component (A) to component (C) is less than 0.5. Therefore, by combining (A) procyanidin B1 and (B) procyanidin B3 and setting the mass ratio [(A) / (B)] of component (A) to component (B) to 1 or more, epidermal keratinocyte activation is exerted, so that the brightness of the skin is improved and a superior skin quality improvement effect can be obtained. In addition, by combining (C) catechin, a more excellent epidermal keratinocyte activation is exerted, so that a more excellent skin quality improvement effect can be obtained. Furthermore, by setting the mass ratio [(A) / (C)] of component (A) to component (C) to 0.5 or more, a particularly excellent epidermal keratinocyte activation is exerted, so that a particularly excellent skin quality improvement effect can be obtained.
[0054] <Test 2: Evaluation of skin fibroblast activation> Skin fibroblast activation was evaluated.
[0055] [Test substance] As in Test 1, the test substances used were (A) procyanidin B1 reagent (manufactured by ChemFaces) as procyanidin B1, (B) procyanidin B3 reagent (manufactured by ChemFaces) as procyanidin B3, and (C) catechin reagent (manufactured by Nagara Science) as catechin.
[0056] [cell] Human dermal fibroblasts (NHDFs) were used as skin fibroblasts.
[0057] [culture] In a 37℃, 5% CO2 incubator, 75 cm 2 Human dermal fibroblasts were cultured in 10% FBS-DMEM medium using a flask. The cells were then suspended by trypsinization and placed in a 75 cm 2 From the flask to each well of a 96-well plate, 5 x 10 3 The cells were seeded at a density of 100 cells / well and precultured for 24 hours in a 5% by volume CO2 incubator at 37°C.
[0058] [Skin fibroblast proliferation test] After removing the medium from each well, 100 μL / well of a medium containing a test substance prepared to a predetermined concentration was added, and the cells were cultured in a 37°C, 5% by volume CO2 incubator for 24 hours. After removing the medium from each well, 100 μL / well of a medium containing a test substance prepared to a predetermined concentration was added, and the cells were cultured in a 37°C, 5% by volume CO2 incubator for 24 hours. As test substances, the compositions of Examples 1 to 9 and Comparative Examples 1 to 5 shown in Table 1 were used. The test substance-containing medium was prepared so that the total concentration of each component of the composition in the culture medium was 1 μg / mL. In addition, a culture medium not containing a test substance was added as a control.
[0059] After 24 hours of culture, the medium was removed, and 150 μL of Cell Counting Kit-8 (Dojindo Chemical Industries) diluted 30-fold with serum-free DMEM was added to each well. The plate was left to stand in a 37°C, 5% by volume CO2 incubator to allow for adequate color development, and the absorbance at 450 nm was measured. Based on the data obtained, the % of control (relative number of live cells with the number of live cells in the control taken as 100%) was calculated. The results are shown in Figure 2. The absorbance ratio and the ratio of human dermal fibroblast numbers are directly proportional to each other, so the absorbance ratio indicates the ratio of human dermal fibroblast numbers. % of control=(Data sample-Data blank) / (Data control-Data blank)×100
[0060] [Test Results] 2, the group (Examples 1 to 9) to which procyanidin B1 (A) and procyanidin B3 (B) were added and the mass ratio of component (A) to component (B) [(A) / (B)] was 1 or more exhibited skin fibroblast activation, compared to the group (Comparative Examples 1 to 4) to which procyanidin B1 (A) and procyanidin B3 (B) were added and the mass ratio of component (A) to component (B) [(A) / (B)] was less than 1, and the group (Comparative Example 5) to which catechin (C) was added alone. Among Examples 1 to 9, the group (Examples 4 to 9) to which catechin (C) was added exhibited superior skin fibroblast activation, compared to the group (Examples 1 to 3) to which catechin (C) was not added. Furthermore, among them, the group (Examples 4 to 8) in which the mass ratio [(A) / (C)] of component (A) to component (C) is 0.5 or more was found to have particularly excellent skin fibroblast activation, compared to the group (Example 9) in which the mass ratio [(A) / (C)] of component (A) to component (C) is less than 0.5. Therefore, by combining (A) procyanidin B1 and (B) procyanidin B3 and setting the mass ratio [(A) / (B)] of component (A) to component (B) to 1 or more, skin fibroblast activation is exerted, thereby improving skin moisture and skin elasticity, and obtaining an excellent skin quality improvement effect. In addition, by combining (C) catechin, more excellent skin fibroblast activation is exerted, thereby obtaining an even more excellent skin quality improvement effect. Furthermore, by setting the mass ratio [(A) / (C)] of component (A) to component (C) to 0.5 or more, particularly excellent skin fibroblast activation is exerted, thereby obtaining an especially excellent skin quality improvement effect.
