Abstract
Description
[Technical field]
[0001] The present invention relates to an external skin preparation containing, as an active ingredient, an extract of one or more plants belonging to the family Rutaceae. [Background technology]
[0002] Skin aging or disorders is a phenomenon that occurs due to a complex intertwining of internal factors such as inactivation of cell proliferation and differentiation, decreased hormone secretion, and quantitative decrease in extracellular matrix components with aging, and external factors such as cell and tissue damage caused by active oxygen induced by sunlight (ultraviolet rays), or inflammation. Examples of skin aging or disorders include wrinkles, sagging, loss of elasticity, dryness, decreased barrier function, pigmentation, dullness, etc., and wrinkle improvement agents, moisturizers, age spot prevention agents, etc. have been proposed according to each skin condition.
[0003] Conventionally, antioxidants such as vitamins A and E and moisturizers (e.g., amino acids, organic acids, hyaluronic acid or its salts, chondroitin, heparinoids, etc.) have been proposed as ingredients for preventing or improving skin aging or disorders, but there are problems with stability, efficacy, and safety when blended into skin external preparations. In addition, the use of ingredients derived from natural products such as plants has also been proposed, but there are problems with efficacy. [Prior art documents] [Patent documents]
[0004] [Patent Document 1] Japanese Patent Application Publication No. 07-118132 [Patent Document 2] Japanese Patent Publication No. 08-295621 [Patent Document 3] Patent Publication No. 2000-096050 [Patent Document 4] JP2001-114634A DISCLOSURE OF THEINVENTION [Problem to be solved by the invention]
[0005] In view of the problems of the prior art, the present inventors have conducted extensive research to find new, highly effective plant-derived components. As a result, they have found that one or more extracts of Citrus tachibana and Citrus unshiu × Citrus noblis, which belong to the Rutaceae family, have the effect of continuously improving the moisturizing, translucency, and luster of the skin, and the effect of activating an enzyme that reduces "oxidized ascorbic acid" generated by oxidation in the body.
[0006] The use of Rutaceae plants in topical skin preparations has been known in the past, for example, from Patent Documents 1 to 4. However, it was not known that they have the effect of continuously improving the moisturizing, transparency, and luster of the skin, or of activating the enzyme that reduces oxidized ascorbic acid. [Means for solving the problem]
[0007] The present invention is an external skin preparation that contains an extract of either one or two of Citrus tachibana and Citrus unshiu × Citrus noblis as active ingredients, and that continuously improves the skin's moisturizing, clarity and luster. The present invention also relates to an activator of oxidized ascorbic acid reductase, which comprises an extract of Calamandarin (Citrus unshiu × Citrus noblis) as an active ingredient. Effect of the Invention
[0008] According to the present invention, by using an extract of any one or two of Citrus tachibana and Citrus unshiu × Citrus noblis as an active ingredient, it is possible to provide an external skin preparation having the effect of continuously improving moisturizing feeling, transparency and luster. Furthermore, according to the present invention, by using an extract of Calamandarin (Citrus unshiu × Citrus noblis) as an active ingredient, it is possible to activate the enzyme that reduces oxidized ascorbic acid, returning it to reduced ascorbic acid that exerts antioxidant properties, thereby suppressing the effects of active oxygen and the like on the skin and enhancing the effects of ascorbic acid (including derivatives) applied to the skin through external application. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0009] The Rutaceae plant used in the present invention is Tachibana (Citrus tachibana or Citrus aurantium var. tachibana) and Calamandarin (Citrus unshiu × Citrus noblis). Although the variety of Tachibana is not limited, Nippon Tachibana (also known as Yamato Tachibana) is more preferable.
[0010] Examples of parts of Tachibana or Calamansia that can be used include the fruit, the peel, and the flesh.
[0011] The preparation of the extract is carried out by first washing the part to be used for extraction with water if necessary to remove foreign matter, leaving it as it is or drying it, and then cutting or crushing it as necessary, and contacting it with an extraction solvent to carry out extraction. The extraction can be carried out by contacting it with an extraction solvent according to a conventional method such as the immersion method, but it is also possible to use a supercritical extraction method other than the immersion method.
[0012] Examples of extraction solvents include water; lower alcohols such as methanol, ethanol, and propanol; polyhydric alcohols such as ethylene glycol, propylene glycol, 1,3-butylene glycol, and glycerin; esters such as ethyl acetate, butyl acetate, and methyl propionate; ketones such as acetone and methyl ethyl ketone; ethers such as ethyl ether and isopropyl ether; and hydrocarbon solvents such as n-hexane, toluene, and chloroform, and these may be used alone or in combination of two or more.
[0013] Among the above-mentioned extraction solvents, hydrophilic solvents such as water, lower alcohols, or polyhydric alcohols are preferred in the present invention from the viewpoint of the effectiveness of the obtained extract, furthermore, skin irritation, and also from the viewpoint of wide applicability to skin external preparations (cosmetics, quasi-drugs, external medicines, etc.).Preferred examples of the hydrophilic solvent include the use of water, lower alcohols (particularly ethanol), or polyhydric alcohols (particularly 1,3-butylene glycol, glycerin) alone, or a mixed solvent of water and lower alcohols (particularly ethanol), or a mixed solvent of water and polyhydric alcohols (particularly 1,3-butylene glycol, glycerin), etc.
[0014] When a mixed solvent is used, the mixing ratio, for example, of water and a lower alcohol or a polyhydric alcohol, is preferably 1:5 to 25:1 in terms of volume ratio (the same applies below).
[0015] In preparing the extract, the pH is not particularly limited, but is generally preferably in the range of 3 to 9. In this sense, if necessary, an alkalinity regulator such as sodium hydroxide, sodium carbonate, or potassium hydroxide, or an acidity regulator such as citric acid, hydrochloric acid, phosphoric acid, or sulfuric acid may be added to the extraction solvent to adjust the pH to a desired level.
[0016] The extraction conditions such as extraction temperature and extraction time vary depending on the type and pH of the solvent used. For example, when water alone or a mixture of water and a lower alcohol or a polyhydric alcohol is used as the solvent, the extraction temperature is preferably in the range of 0°C to 80°C, and the extraction time is preferably in the range of 1 to 168 hours (1 hour to 1 week), and more preferably in the range of 1 to 120 hours (1 hour to 5 days).
[0017] In order to remove impurities and improve stability, the extract of the present invention may be subjected to one or a combination of two or more of activated carbon treatment, ion exchange resin treatment, synthetic adsorbent treatment, silica gel, and recrystallization treatment to remove impurities other than the active ingredients.
[0018] The extract prepared as described above may be used as an ingredient in a skin external preparation as it is after adjusting the pH to 3 to 8, or may be used at a desired concentration by concentrating under reduced pressure, etc. The extract may also be dried by a conventional method such as spray drying.
[0019] Examples of topical skin preparations (cosmetics, quasi-drugs, topical medicines) containing the extract of the present invention include, but are not limited to, milky lotions, creams, lotions, essences, packs, lipsticks, foundations, sheet masks, liquid foundations, makeup press powders, blushers, face powders, facial cleansers, body shampoos, soaps and other cleansing cosmetics, hair growth products, and bath additives.
[0020] When the extract of the present invention is incorporated into an external skin preparation, the amount of the extract is generally in the range of 0.00001 to 5.0% by weight in terms of the solid content of each extract.

[0047] Test Example 1. Evaluation of the effect of promoting the expression of filaggrin Normal human epidermal cells (NHEK) were cultured in a 24-well plate at 3 × 10 4The cells were seeded at 1.000 cells / well and cultured at 37°C under 5% CO2 and saturated steam. Test sections were set up to add each of the sample solutions from Preparation Examples 1 and 4, and after 24 hours of culture, each sample solution was added, and normal human epidermal cells NHEK were further cultured for 48 hours. Here, the concentration of each sample solution was added to 1.0% as a solution. As a comparison control, a control section was set up in which a culture solution containing only 50% BG solution of the same concentration was added instead of the above sample solution, and cultured in the same manner. Next, the cells in each test section and control section were collected with 0.5 mL of ISOGEN II reagent (manufactured by Nippon Gene Co., Ltd.). 200 μL of RNase-free water was added to the collected cells, which were stirred and mixed, left at room temperature for 15 minutes, and then centrifuged for 15 minutes at 15,000 rpm and 4°C using a centrifuge (manufactured by TOMY Co., Ltd. / MX-160), and 500 μL of the supernatant was taken out. 500μL of isopropanol (Wako Pure Chemical Industries, Ltd.) was added to the collected supernatant, stirred and mixed, left at room temperature for 10 minutes, and then centrifuged at 15,000 rpm and 4℃ for 10 minutes to obtain a precipitate of total RNA. 0.5mL of 75% ethanol was added to the total RNA, and the precipitate was collected by centrifugation at 15,000 rpm and 4℃ for 3 minutes. The supernatant was discarded, and this process was repeated twice. After completely removing the supernatant, the mixture was air-dried and dissolved in RNase-free water. The collected total RNA was reverse-transcribed using a designated kit (PrimeScript RT reagent Kit with gDNA Eraser (Perfect Real Time) [Takara Bio]) to synthesize cDNA. Using the synthesized cDNA as a sample, the expression of various genes and the expression of the internal standard GAPDH gene were detected using Thermal Cycler Dice (registered trademark) Real Time System Single (manufactured by Takara Bio Inc.) and SYBR (registered trademark) Premix Ex TaqTM (Perfect Real Time) [manufactured by Takara Bio Inc.].Here, GAPDH (glyceraldehyde-3-phosphate dehydrogenase) is one of the housekeeping genes (a gene that is expressed at a constant level in many tissues and cells, is constantly expressed, and is essential for the maintenance and proliferation of cells), and since its expression level is always constant, it is used as an internal standard in PCR experiments. The test results were obtained by comparing the expression levels of each gene in each test group when the expression level of the GAPDH gene was kept constant. The expression level in the control group was set to 100% and a relative value was calculated.
[0048] The results of Test Example 1 are shown in Table 1. [Table 1] JPEG2025004740000001.jpg42126
[0049] As shown in Table 1, Test Example 1 confirmed that the extracts according to the present invention (extracts of Production Examples 1 and 4) significantly promote the expression of filaggrin. In the stratum corneum, filaggrin is deiminated by an enzyme to become deiminated filaggrin, and this deiminated filaggrin is decomposed by an enzyme to produce amino acids (NMFs) that are natural moisturizing factors. Therefore, applying the extracts of Production Examples 1 and 4 to the skin promotes the skin cells to produce natural moisturizing factors themselves, providing a sustained feeling of moisture and also enabling sustained improvement in transparency and luster.
[0050] Test Example 2: Evaluation test of the effect of promoting tight junction synthesis Normal epidermal cells (NHEK) were cultured in a 96-well plate at 1.5 x 10 4The cells were seeded at 1000 cells / well and cultured at 37°C under 5% CO2 and saturated water vapor. The cells were cultured for 48 hours in the well to make them subconfluent. Next, the extract according to the present invention (extracts of Production Examples 1 and 4) was added as an evaluation sample, and the cells were cultured for another 48 hours. The sample was adjusted to 1.0% as a solution. In addition, as a comparative control, a control group was set in which a culture solution containing only a 30% BG solution (1.0%) of the same concentration was added instead of the sample. After completion, the culture solution was removed, the cells were washed with D-PBS (containing no Ca or Mg, hereinafter referred to as PBS), and PBS containing 10% trichloroacetic acid was added as a fixative, and the cells were allowed to stand at 4°C for 30 minutes. After removing the fixative and washing with PBS, PBS containing 0.2% Triton-X (hereinafter referred to as wash solution) was added and incubated at room temperature for 1 hour. Then, the cells were incubated overnight at 4°C using Blocking One P (manufactured by Nacalai Tesque) as a blocking agent. After washing once with washing solution, anti-occludin (Life Span Bio Sciences) and anti-claudin-1 antibody (Bioward Technology) were added as primary antibodies at 500-fold dilution, and further incubated overnight at 4°C. After washing three times with washing solution, Alexa Fluor (registered trademark) 488 Goat Anti-mouse IgG and Alexa Fluor (registered trademark) 546 Goat Anti-rabbit IgG (both manufactured by Invitrogen) were added as secondary antibodies at 500-fold dilution. After incubation at room temperature for 2 hours, washing was performed five times with washing solution. Then, Hoechst 33342 (Dojindo Laboratories) was added at 1000-fold dilution with PBS, and incubated for 30 minutes. The fluorescence intensity of each antibody and Hoechst 33342 was measured using a Fluoroskan Ascent (ThermoLabsystems). The fluorescence intensity of the antibody was corrected using the Hoechst 33342 value as an internal standard, and then the detection value of each antibody was calculated as a relative value with the control group set as 100%.
[0051] The results of Test Example 2 are shown in Table 2. [Table 2] JPEG2025004740000002.jpg70127
[0052] As shown in Table 2, Test Example 2 confirmed that the extract according to the present invention (extracts of Production Examples 1 and 4) exhibits the effect of promoting the synthesis of proteins (occludin, claudin-1) that constitute tight junctions, which are binding structures between cells. This suggests that the extract according to the present invention can improve the barrier function of the skin and prevent evaporation of moisture from the skin, thereby providing the skin with a sustained moist feeling. It also suggests that the extract exhibits the effect of preventing the invasion of bacteria and foreign substances (environmental pollutants, etc.) into the skin from the outside.
[0053] Test Example 3. Evaluation test of increased activity of oxidized ascorbate reductase NHEK epidermal cells were cultured in HuMedia KG2 medium (Kurabo) at 8.0 × 10 4The cells were seeded per well, and cultured for 24 hours under conditions of 37°C and 5% CO2, then replaced with HuMedia KB2 medium (Kurabo) containing each sample solution, and further cultured under the same conditions for 48 hours. Here, the extract of Production Example 4 was adjusted to a final concentration of 1.0% in the medium. After the culture was completed, the cells were detached using trypsin and collected in a microtube using PBS(-) containing 5% FBS. Then, the cells were washed twice with PBS(-), the supernatant was removed, and the cells were rapidly frozen using liquid nitrogen and stored at -80°C until the enzyme activity was measured. In this Test Example 4, the activity of an enzyme that reduces oxidized ascorbic acid (dehydroascorbic acid) (hereinafter referred to as "DHA reductase") was measured. The DHA reductase activity was measured as follows. 500 μL of Passive Lysis Buffer (Promaga) was added to the stored cells, and the cells were dissolved by pipetting. After centrifugation (12000g x 5 min, 4°C), the supernatant was collected and used as a crude enzyme solution. 50 μL of the obtained crude enzyme solution was added per well, and 50 μL of purified water (control) or 50 μL of 2 mM dehydroascorbic acid solution (test) was added. 50 μL of DTNB (5,5'-dithio-bis- (2-nitrobenzoic acid)) test solution (CAYMAN CHEMICAL COMPANY) was added, and the absorbance at 414 nm was measured. The difference obtained by subtracting the value of the test group from the value of the control group for each sample addition group was used as the DHA reductase activity. Meanwhile, the protein concentration of the crude enzyme solution was measured by the Bradford method using Protein Assay Dye (BIO-RAD), and the value obtained by dividing the absorbance difference by the protein concentration was used as the DHA reductase activity per protein for each sample addition group, and the relative value was calculated by taking the value of the solvent control addition group as 100.
[0054] The results of Test Example 3 are shown in Table 3. [Table 3] JPEG2025004740000003.jpg41127
[0055] As shown in Table 3, Test Example 3 confirmed that the extract according to the present invention (extract of Production Example 4) activates DHA reductase. As a result, the extract according to the present invention can reduce oxidized ascorbic acid (dehydroascorbic acid) that has been oxidized in the body and lost its functions such as antioxidant effect, returning it to the original ascorbic acid, thereby promoting the exertion of its functions.
