[0004] In a so-called microscopic examination, in which formed elements in a biological specimen such as urine are measured under a microscope, a predetermined amount of the biological specimen is dispensed into a centrifuge tube and centrifuged to precipitate the formed elements.

[0005] However, if the amount of biological specimen dispensed into the centrifuge tube is not the specified amount, the proportion of sediment components in the sediment obtained by centrifugation will be different from that obtained when the amount of biological specimen is the specified amount, which may result in a decrease in the accuracy of the microscopic measurement.

[0006] An object of the present disclosure is to improve and enable measurement accuracy in microscopic examination even when the amount of biological sample dispensed into a centrifuge tube is not a predetermined amount. [Means for solving the problem]

[0007] In the technology disclosed herein, a portion of the biological specimen is removed from a centrifuge tube, leaving a specified amount of the biological specimen, and the amount of the removed biological specimen is obtained.Based on the actual amount of the biological specimen in the centrifuge tube obtained from the specified amount of the biological specimen remaining and the amount of the biological specimen removed, a correction factor for correcting the microscopic measurement value of the biological specimen is obtained, and the correction factor or a correction value obtained by multiplying the microscopic measurement value of the biological specimen by the correction factor is output. [Effects of the Invention]

[0008] The technology disclosed herein can improve the measurement accuracy of microscopic examinations even when the amount of biological sample dispensed into a centrifuge tube is not a predetermined amount. [Brief explanation of the drawings]

DETAILED DESCRIPTION OF THE INVENTION

[0010] Hereinafter, the first embodiment will be described with reference to the drawings.

[0011] Fig. 1 shows part of the configuration of a biological sample measurement support device 20 that performs the biological sample measurement support method according to the technique of the present disclosure. Fig. 2 shows an example of a centrifuge tube 12 used for measuring a biological sample.

[0012] The biological sample measurement support device 20 is a device that plays a part in preparing a specimen when measuring formed elements from a biological sample contained in a centrifuge tube 12, and also supports the measurement of the biological sample. In this embodiment, as an example, urine UR is used as the biological sample, as shown in FIG. 4A , and an examination (so-called "microscopic examination") is performed to observe formed elements such as cells, casts, and crystals in the urinary sediment UP generated by centrifugation under a microscope. In the following, for convenience of explanation, the urinary sediment examination method GP1-P4, which has been standardized as a standard procedure by the Japanese Committee for Clinical Laboratory Standards, may be used as an example. However, the technology of the present disclosure is not limited to this standardized urinary sediment examination method GP1-P4.

[0013] 2, the centrifuge tube 12 illustrated in this embodiment has a conical bottom and is sometimes called a Spitz-type centrifuge tube. The centrifuge tube 12 has a first graduation 14 and a second graduation 16.

[0014] The first graduation 14 indicates the amount of urine to be dispensed into the centrifuge tube 12. For example, the first graduation 14 is set at the 10 mL position in the urinary sediment testing method GP1-P4. The amount of urine indicated by the first graduation 14 is an example of the "standard amount" according to the technology of the present disclosure.

[0015] The second graduation 16 indicates the predetermined amount of sediment to be left in the centrifuge tube 12 after removing a portion of the urine UR (supernatant) after centrifugation. For example, in the urinary sediment testing method GP1-P4, the second graduation 16 is set at the 0.2 mL position. The sediment amount indicated by the second graduation 16 is an example of the "predetermined amount" according to the technology of the present disclosure. As will be described later, when urine UR is aspirated by the nozzle 28 with the suction port 28V of the nozzle 28 in this position, a predetermined amount of urinary sediment UP can be left in the centrifuge tube 12.

[0016] The biological sample measurement support device 20 comprises a base 22, a holding member 24, a support 26, a nozzle 28, a suction device 30, a flow meter 32, a reservoir 34, a display 36, and a control unit 38.

[0017] 3, the control unit 38 has a computer 40. The computer 40 has a processor 42, memory 44, storage 46, an input device 48, an output device 50, a storage medium reader 52, and an interface (I / F) 54, and each of these elements is connected to each other so that they can communicate with each other via a bus 56. The computer 40 controls the biological sample measurement support device 20.

[0018] Storage 46 stores a biological sample measurement support program (hereinafter referred to as "measurement support program 58") for executing the measurement support processing described below. Processor 42 is capable of executing various programs and controlling each element. Specifically, processor 42 reads the program from storage 46 and executes the program using memory 44 as a work area. In other words, processor 42 controls each element and performs various arithmetic processing in accordance with the program stored in storage 46.

[0019] The memory 44 can temporarily store programs and various data as a work area.

[0020] The storage 46 is, for example, a read-only memory (ROM), a hard disk drive (HDD), or a solid state drive (SDD), and stores various programs and data. These programs include not only application programs such as the measurement assistance program described above, but also an operating system.

[0021] The input device 48 is a device for performing various inputs to the computer 40. The input device 48 includes operation switches, operation buttons, etc., and may also include pointing devices such as a keyboard and a mouse used in a personal computer, etc.

[0022] The output device 50 is a device for outputting various types of information from the computer 40, and may include, for example, an indicator lamp, a speaker, etc. in addition to the display 36. A touch panel display may be used as the display 36, which is an example of the output device 50. In this case, the touch panel display also functions as the input device 48. In particular, in this embodiment, the control unit 38 displays a correction factor, which will be described later, on the display 36.

[0023] The storage medium reader 52 is a device that reads data stored in various storage media and writes data to the storage media, such as a CD (Compact Disc)-ROM, a DVD (Digital Versatile Disc)-ROM, a Blu-ray Disc, and a USB (Universal Serial Bus) memory.

[0024] The interface 54 is a device for communicating with other devices, and is compliant with standards such as Ethernet (registered trademark) and FDDI (Fiber Distributed Data Interface).

[0025] 1, in biological sample measurement support device 20, holding member 24 and support 26 are provided on the upper surface of base 22. Holding member 24 is a member that holds centrifuge tube 12 in an upright position on base 22.

[0026] The support 26 is fixed in a vertically standing position on the base 22. A nozzle 28 is held on the support 26 by a nozzle holder 60. The nozzle holder 60 moves up and down along the support 26 by being driven by a pulse motor 62. The driving of the pulse motor 62 is controlled by the computer 40.

[0027] The nozzle 28 extends downward from the nozzle holder 60. The lower end of the nozzle 28 is a suction port 28V. The nozzle 28 is concentric with the centrifuge tube 12 held by the holding member 24 and faces the centrifuge tube 12 from above. The nozzle holder 60 moves up and down to an initial position PP where the suction port 28V is separated from the upper end of the centrifuge tube 12, and a suction position VP where the suction port 28V is lowered to a predetermined position within the centrifuge tube 12. This suction position VP is the position of the second graduation 16, i.e., the position where 0.2 mL of sediment is left in the centrifuge tube 12 when urine is aspirated from the centrifuge tube 12 by suction. In other words, the insertion length of the nozzle 28 into the centrifuge tube 12 is determined so that the position of the suction port 28V is the position of the second graduation 16.

[0028] Furthermore, the support 26 is provided with a photointerrupter 66. The photointerrupter 66 detects the height position of the nozzle holder 60 and sends the obtained height position information to the computer 40. In other words, the photointerrupter 66 is an example of a detection device that detects the height position of the nozzle holder 60.

[0029] The suction device 30 has a suction pipe 64 and a pump 68. One end of the suction pipe 64 is connected to the upper end of the nozzle 28, and the lower end of the suction pipe 64 is open to the reservoir 34. This realizes a configuration in which the suction device 30 is equipped with the nozzle 28.

[0030] The suction tube 64 is provided with a pump 68. The pump 68 is controlled by the computer 40. When the pump 68 is driven, the biological sample in the centrifuge tube 12 is sucked from the centrifuge tube 12 via the nozzle 28 and the suction tube 64.

[0031] The aspirating tube 64 is provided with a flow meter 32. The flow meter 32 measures the amount (volume) of the aspirated biological sample, and sends information about the amount of the biological sample obtained to the computer 40.

[0032] The biological sample aspirated from the centrifuge tube 12 is temporarily stored in the storage tank 34. A discharge pipe 70 is connected to the bottom of the storage tank 34, and an on-off valve 72 is provided on the discharge pipe 70. By opening the on-off valve 72, the biological sample in the storage tank 34 can be discharged to the outside.

[0033] Next, the operation of this embodiment and the biological sample measurement support method will be described. In the following, the case where urine, which is a biological sample, is subjected to the urinary sediment test method GP1-P4 standardized by the Japanese Committee for Clinical Laboratory Standards will be described as an example.

[0034] First, before performing the biological sample measurement support method, the number of pulses of pulse motor 62 corresponding to moving nozzle 28 from initial position PP to suction position VP is obtained. Specifically, this is the number of pulses sent to pulse motor 62 when nozzle 28 is lowered from initial position PP to suction position VP.

[0035] When performing microscopic examination of formed elements in urine, a predetermined amount of urine UR is dispensed into a centrifuge tube 12. This predetermined amount is 10 mL according to the urinary sediment examination method GP1-P4, but in reality, the amount may differ from 10 mL.

[0036] In the initial state, as shown in Fig. 4A, nozzle 28 has suction port 28V at initial position PP. In addition, as shown in Fig. 4B, a predetermined amount of urine UR is centrifuged in centrifuge tube 12 to produce urinary sediment UP. This centrifuge tube 12 is held by holding member 24.

[0037] An example of the flow for executing measurement support processing in the biological sample measurement support method is shown in Figure 5. Measurement support program 58 that performs this measurement support processing is stored in advance in storage 46, for example, and after measurement support program 58 is called into memory 44, it is developed and executed in processor 42.

[0038] In step S102, the control unit 38 drives the pulse motor 62. Specifically, the processor 42 reads out the movement coefficient (number of pulses) stored in the storage 46, and transmits a drive instruction to the pulse motor 62 in accordance with this movement coefficient. As a result, the nozzle 28 is lowered from the initial position PP to the suction position VP, as shown in FIG. 6.

[0039] Next, in step S104, the control unit 38 drives the pump 68. As a result, the supernatant of urine in the centrifuge tube 12 is sucked through the nozzle 28 and the suction tube 64, as shown in FIG. 7A.

[0040] The suction port 28V of the nozzle 28 is located at the second marking 16. Therefore, when the pump 68 is driven to aspirate the supernatant urine, a predetermined sediment volume (prescribed volume) of 0.2 mL of urinary sediment UP remains in the centrifuge tube 12, as shown in Figure 7B. Here, the operator performs a microscopic examination using the 0.2 mL sediment volume of sediment components and obtains a measurement value.

[0041] When the supernatant of the urine UR in the centrifuge tube 12 is aspirated through the nozzle in this manner, the amount (volume) of aspirated urine is measured by the flow meter 32. In step S106, the control unit 38 receives information on the flow rate measured by the flow meter 32, i.e., the amount of urine aspirated by the pump 68, and acquires this value.

[0042] Next, in step S108, the control unit 38 calculates a correction factor using the acquired volume of aspirated urine (X mL). The calculation of the correction factor is performed using the following formula (1).

[0043] Correction multiple=10mL / (XmL+0.2mL) (1) Here, the acquired volume of aspirated urine (X mL) is an example of the "removal volume" in the technology of the present disclosure, and the predetermined sediment volume of 0.2 mL is an example of the "specified volume" in the technology of the present disclosure. Furthermore, (X mL + 0.2 mL) in formula (1) is the volume of urine (biological specimen) actually present in the centrifuge tube 12, i.e., the "actual volume" in the technology of the present disclosure.

[0044] Next, in step S110, the control unit 38 causes the display 36 to display the volume of aspirated urine, as shown in FIG. 8, and also to display a message indicating that the measured value of the number of formed elements contained in the microscopically examined sample will be corrected by multiplying it by a correction factor. The operator corrects the value of the number of formed elements contained in the microscopically examined sample by multiplying the measured value of the number of formed elements contained in the microscopically examined sample by this correction factor. The display 36 may display the volume of aspirated urine and the correction factor on separate screens, or may display them on two or more different displays. Alternatively, the display 36 may display a correction value calculated by multiplying the measured value of the number of formed elements contained in the microscopically examined sample by the correction factor. Either the correction factor or the correction value may be displayed, or both may be displayed.

[0045] As described above, in the urinary sediment examination method GP1-P4 standardized by the Japanese Committee for Clinical Laboratory Standards, the amount of urine dispensed into the centrifuge tube 12 is 10 mL. However, in practice, it is difficult for an operator to accurately dispense 10 mL of urine into a centrifuge tube.

[0046] Furthermore, prior to the urinary sediment test according to the technology of the present disclosure, other tests using the urine, such as a so-called urinary qualitative test, may be performed. In this case, if a portion of the 10 mL of urine dispensed into centrifuge tube 12 is used for the urinary qualitative test, no 10 mL of urine will remain in centrifuge tube 12 when the urinary sediment test is performed.

[0047] In addition, there are cases where it is not possible to obtain 10 mL or more of urine from a test subject. In this case, it is not possible to dispense 10 mL of urine into the centrifuge tube 12.

[0048] Due to these various factors, a state may occur in which there is no 10 mL of urine in the centrifuge tube 12, as shown in Figure 9. If the urine UR in the centrifuge tube 12 is centrifuged in this state and then the supernatant is aspirated to obtain a urinary sediment UP with a sediment volume of 0.2 mL, the proportion of formed elements in this urinary sediment UP will be different from the proportion when 10 mL of urine is present in the centrifuge tube 12.

[0049] For example, as shown in Figure 4B, consider the case where 10 mL of urine UR is dispensed into the centrifuge tube 12. After centrifuging this urine UR, the sediment volume UP is set to 0.2 mL, and the formed element value obtained by microscopic examination is 6. This value is correct.

[0050] In contrast, consider the case where the amount of urine UR dispensed into the centrifuge tube 12 is, for example, 5 mL, as shown in Figure 9. In this case, the amount of urine UR dispensed into the centrifuge tube 12 is half that shown in Figure 4B, and the measured value of formed elements obtained by microscopic examination is also half that shown in Figure 4B, i.e., 3 mL.

[0051] In the technology disclosed herein, a correction factor is calculated using the volume of urine aspirated (X mL) received from flow meter 32. This correction factor is a ratio indicating how many times the value (measured value) of formed elements measured by microscopic examination must be multiplied to obtain the correct value when the specified volume (standard volume) of 10 mL of urine UR is not present in centrifuge tube 12. Therefore, according to the technology disclosed herein, even when the specified volume (standard volume) of 10 mL of urine UR is not present in centrifuge tube 12, the accuracy of the value of formed elements can be improved by multiplying the measured value of formed elements obtained by microscopic examination by this correction factor to obtain a corrected value.

[0052] In particular, in this embodiment, the correction factor is obtained by the above formula (1). Therefore, when using urine as a sample and performing a microscopic examination according to the urinary sediment examination method GP1-P4 standardized by the Japanese Committee for Clinical Laboratory Standards, it is possible to improve the accuracy of the formed element values.

[0053] If a microscopic examination is performed using a method other than the urinary sediment examination method GP1-P4 standardized by the Japanese Committee for Clinical Laboratory Standards, the values in formula (1) can be replaced with the values corresponding to the microscopic examination method in question. This makes it possible to improve the accuracy of the formed element values according to the respective microscopic examination method.

[0054] In the technology of the present disclosure, a flow meter 32 is provided in the suction device 30 that removes urine UR from the centrifuge tube 12. When the urine UR is aspirated from the centrifuge tube 12, the amount of urine removed from the centrifuge tube 12 (removed amount) is obtained. Compared to when the amount of urine removed from the centrifuge tube 12 is obtained in a process separate from the suction (for example, after storing in the storage tank 34), the removed amount can be obtained easily at the same time as the suction.

[0055] In the technology of the present disclosure, a portion of the urine UR is removed from the centrifuge tube 12 by inserting the nozzle 28 provided on the suction device 30 into the centrifuge tube 12 and aspirating the urine UR. Since there is no need to tilt the centrifuge tube 12, it is easy to remove a portion of the urine from the centrifuge tube 12.

[0056] In particular, in the above embodiment, the pump 68 is driven to aspirate a portion of the urine UR in the centrifuge tube 12 with the suction port 28V at the bottom end of the centrifuge tube 12 positioned at the second scale 16. This allows the urine UR to be aspirated while leaving a predetermined amount of sediment without adjusting the driving time or suction power of the pump 68.

[0057] Moreover, in this embodiment, by sending a predetermined number of pulses to the pulse motor 62, the suction port 28V of the nozzle 28 is lowered to the position of the second scale 16. In other words, the suction port 28V can be automatically lowered to a position corresponding to the amount of urine sediment in the centrifuge tube 12.

[0058] It should be noted that, in the technology disclosed herein, the method for controlling the descent of nozzle 28, i.e., the method for aligning the position of suction port 28V with the position of second scale 16, is not limited to the method described above. For example, the relationship between the drive time of the motor and the amount of descent of nozzle 28 may be acquired in advance, and the drive time of the motor may be adjusted to lower nozzle 28 so that the position of suction port 28V of nozzle 28 coincides with the position of second scale 16. Furthermore, the position of suction port 28V may be detected by a sensor, and the motor may be controlled to stop the descent of nozzle 28 when the position of suction port 28V reaches the position of second scale 16. In this case, the motor for moving nozzle 28 up and down is not limited to pulse motor 62.

[0059] In the technology disclosed herein, it is also possible to adjust the lowering position of the suction port 28V of the nozzle 28 in accordance with multiple types of centrifuge tubes 12. For example, if the positions of the second scale 16 for multiple types of centrifuge tubes 12 are known in advance, the number of pulses corresponding to the position of the second scale 16 for each centrifuge tube 12 may be stored in the storage 46 or the like. Alternatively, the centrifuge tube 12 to be used may be held in the holding member 24 in advance, and the nozzle 28 may be moved from the initial position PP to the suction position VP to obtain the number of pulses required to align the lowering position of the suction port 28V with the position of the second scale 16, and the result may be recorded in the storage 46. Furthermore, if the relationship between the lowering distance of the nozzle 28 and the number of pulses is known in advance, the operator may set the number of pulses so that the position of the suction port 28V coincides with the position of the second scale 16 when the nozzle 28 is lowered.

[0060] Furthermore, in the technology of the present disclosure, a portion of the urine may be sucked in by positioning suction port 28V of nozzle 28 below second graduation 16. In this case, the urine level in centrifuge tube 12 is detected by a sensor, and when the urine level reaches second graduation 16, the pump 68 is controlled to stop driving.

[0061] In the technology of the present disclosure, the control unit 38 may be part of the biological sample measurement support device 20, like the above-mentioned computer 40. Alternatively, for example, a control device provided as a separate component from the biological sample measurement support device 20 may function as the control unit 38 and remotely control the biological sample measurement support device 20.

[0062] Furthermore, the following notes are disclosed: (Appendix 1) removing a portion of the biological specimen from the centrifuge tube to leave a predetermined amount of the biological specimen and obtaining the amount of the removed biological specimen; A biological sample measurement support method that obtains a correction factor for correcting the microscopic measurement value of the biological sample based on the actual amount of the biological sample in the centrifuge tube, which is obtained from the specified amount of the biological sample remaining and the amount of the biological sample removed, and outputs the correction factor or a correction value obtained by multiplying the microscopic measurement value of the biological sample by the correction factor.
