Abstract
Description
[Technical field]
[0001] The present invention relates to an external skin preparation that exhibits higher effectiveness by combining a plurality of ingredients. [Background technology]
[0002] Conventionally, the use of various compounds has been proposed as ingredients that can be incorporated into skin external preparations, and skin external preparations (external medicines, quasi-drugs, cosmetics, etc.) containing these compounds have been marketed. For example, antioxidants such as vitamin C and vitamin E; anti-inflammatory agents such as glycyrrhizic acid; various ultraviolet absorbers; cell activating ingredients such as α-hydroxycarboxylic acid, placental extract, and γ-amino-β-hydroxybutyric acid; extracellular matrix ingredients such as collagen, elastin, and hyaluronic acid; and moisturizing agents such as urea have been proposed. On the other hand, in consideration of safety, skin external preparations containing various plant-derived ingredients have been proposed. However, the effectiveness of these compounds and plant-derived ingredients alone is not sufficient, and a combination of ingredients that exhibit high effectiveness is required. [0003] [Patent Document 1] Patent No. 2022-138725 Summary of the Invention [Problem to be solved by the invention]
[0004] In view of the problems of the above-mentioned conventional techniques, the present inventors have conducted extensive research into combinations of highly effective compounds and plant-derived ingredients as active ingredients for skin external preparations. As a result, they have found that a combination of niacinamide (also known as nicotinamide), bamboo shoot extract, and perilla extract is highly effective as an ingredient in skin external preparations. Conventionally, a technique for blending niacinamide and bamboo shoot extract into skin external preparations is known from Patent Document 1, but it has not been known that a more effective skin external preparation can be provided by combining niacinamide, bamboo shoot extract, and perilla extract. [Means for solving the problem]
[0005] The present invention is a skin external preparation containing a composition containing niacinamide, bamboo shoot extract, and perilla extract as active ingredients. Furthermore, the skin topical preparation of the present invention may further contain one or more of glycyrrhizinic acid or a derivative thereof, tranexamic acid or a derivative thereof, ascorbic acid or a derivative thereof, pantothenyl alcohol, and tocopherol or a derivative thereof. Effect of the Invention
[0006] The present invention provides an external skin preparation that exhibits effects such as improved skin texture and transparency, moisturizing effect, and wrinkle reduction by combining niacinamide, bamboo shoot extract, and perilla extract as active ingredients. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0007] Next, the "bamboo shoots" used in the present invention will be described. "Bamboo shoots" are young shoots of bamboo belonging to the Poaceae family, Bambusoideae subfamily. The parts of bamboo shoots used in the present invention may be edible parts, the skin of the inedible parts, or bamboo water.
[0008] Examples of bamboos of the Poaceae subfamily Bambusoideae that can be used in the present invention include Phyllostachys pubescens, Phyllostachys bambusoides, Phyllostachys nigra, Phyllostachys aurea, Phyllostachys heterocycla, Bambusa multiplex, Semiarundinaria fastuosa, Sasa kurilensis, Sinobambusa tootsik, Chimonobambusa quadrangularis, Chimonobambusa marmorea, Pseudosasa japonica, Pleioblastus simonii), but the present invention is not limited thereto.
[0009] In the present invention, bamboo shoots refer to young shoots of bamboos of the Poaceae subfamily. The bamboo shoots preferred for use in the present invention vary depending on the bamboo species. For example, in the case of Moso bamboo, those harvested from early March to late June before the young shoots appear above ground are preferred. In the case of Madake, those harvested from June to September with an aboveground part of 10 to 500 cm are preferred, and those 30 to 200 cm above ground are more preferred. In addition, when the bamboo shoots grow beyond the above-mentioned range, the edible part and skin of the young shoots become hard, and the amount of tyrosine, which is undesirable as a cosmetic ingredient, increases, making them unsuitable for the present invention.
[0010] The extract can be prepared by first washing the skin or edible parts of the bamboo shoots with water if necessary to remove foreign matter, then either as is or dried, and then shredded or crushed as necessary, and contacting them with an extraction solvent according to a conventional method such as the soaking method.
[0011] In the present invention, "perilla" refers to perilla of the genus Perilla in the family Lamiaceae, and generally includes those represented by the scientific name Perilla frutescens var. crispa or Perilla frutescens var. Britton, such as Perilla aorticulatus, Perilla chirimen shiso, Perilla akasan, Perilla madara, Perilla katamen shiso, Perilla chirimen aorticulatus, or their variants, subspecies, or hybrids, but the present invention is not limited thereto. The part to be used may be the whole plant, leaves, inflorescences, stems, seeds, fruits, roots, etc., but it is preferable to use leaves or the whole plant including leaves. In addition, the harvest time and size of the part of perilla to be used are not particularly limited, and any size or harvest time may be used.
[0012] Examples of extraction solvents used in preparing bamboo shoot and perilla extracts include water; lower alcohols such as methanol, ethanol, and propanol; polyhydric alcohols such as ethylene glycol, propylene glycol, 1,3-butylene glycol, and glycerin; esters such as ethyl acetate, butyl acetate, and methyl propionate; ketones such as acetone and methyl ethyl ketone; ethers such as ethyl ether and isopropyl ether; and hydrocarbon solvents such as n-hexane, toluene, and chloroform, and these may be used alone or in combination of two or more.
[0013] Among these extraction solvents, the use of water or a mixed solvent of water and a lower alcohol or water and a polyhydric alcohol is preferred in the present invention, since it can be widely applied to external preparations for skin.
[0014] In preparing the extract, the pH of the extract is not particularly limited, but is preferably in the range of pH 2 to 8. In preparing the bamboo shoot extract, the pH is more preferably in the range of pH 2 to 4 in order to suppress the elution of "tyrosine" contained in the bamboo shoot. Therefore, in preparing the bamboo shoot extract, examples of the pH adjustment agents include acidity adjusters such as lactic acid, citric acid, hydrochloric acid, phosphoric acid, and sulfuric acid.
[0015] The extraction conditions such as extraction temperature and extraction time vary depending on the type and pH of the solvent used. For example, in the case of a immersion method using water as the extraction solvent, the extraction temperature is generally 1 to 90°C, preferably in the range of 5°C to 80°C, and the extraction time is generally 0.5 to 7 days, preferably in the range of 1 to 24 hours.
[0016] The extract obtained under the above conditions may generally be adjusted to a pH of 4 to 8 and used as is as an ingredient of an external skin preparation, or may be concentrated under reduced pressure or the like to a desired concentration.
[0017] When the composition containing niacinamide, bamboo shoot extract and perilla extract according to the present invention is incorporated into an external preparation for skin, in addition to the composition which is the essential component, components usually used in external preparations for skin (cosmetics, quasi-drugs or external medicines) can be appropriately incorporated as necessary, such as oily components, surfactants (synthetic or natural), moisturizers, thickeners, emulsifiers or emulsifier assistants, preservatives / bactericides, powder components, ultraviolet absorbers, antioxidants, sequestering agents, pigments, fragrances, anti-wrinkle agents, pigmentation prevention agents and other physiologically active components.

The concentration of the sample solution was adjusted so that the final concentration as a solution in the medium was 0.5%, 1.0%. After the culture was completed, the supernatant was discarded and the cells were washed once with PBS(-), and then 100 μL / well of Hoechst 33342 reagent diluted 500 times with PBS(-) was added and incubated at 37° C. for 30 minutes to fluorescently stain the DNA. Thereafter, the fluorescence intensity [excitation wavelength: 355 nm, fluorescence wavelength: 460 nm: fluorescence microplate reader (Fluoroscan Ascent, Thermo Fisher Scientific)] was measured to determine the amount of DNA. The same procedure was performed on a control group to which 30% 1,3-butylene glycol was added at 1.0% instead of the above sample solution, and the relative fluorescence intensity (DNA amount) of each sample-added group was calculated to obtain the cell proliferation rate (%).
[0044] [Table 1] JPEG2025037673000001.jpg50104
[0045] As shown in Table 1, the extracts of Production Examples 4 and 5 were confirmed to have the effect of promoting the proliferation of epidermal cells. The epidermal cells that proliferate when cultured are the basal cells in the lowest layer of the epidermis, and it is suggested that the proliferation of these basal cells pushes the basal cells up to the upper layers of spinous cells, granular cells, and keratinocytes, normalizing or improving the turnover of the epidermis until the keratin peels off. It is also suggested that the normalization or improvement of this turnover has the effect of improving the texture of the epidermis and improving its transparency, as well as the effect of promoting the discharge of melanin from skin cells and promoting the prevention or improvement of pigmentation such as age spots, freckles, and melasma.
[0046] Test example 2: Monitor test It was confirmed that the extracts of Production Examples 4 and 5 above have the effect of proliferating epidermal cells, and furthermore, the following test confirmed that the combination of niacinamide with the extract of Production Example 1 or Production Example 3 and the extract of Production Example 4 or Production Example 5 exerts the effect of improving skin texture, transparency and moisturizing feeling.
[0047] [Sample preparation] First, inventive samples 1 and 2 and comparative samples 1 to 4 were prepared as follows. Note that % indicates weight % in the sample solution. 1. Sample 1 of the present invention (1) Extract of Production Example 1 (1.0%) (2) Extract of Production Example 4 (1.0%) (3) Niacinamide (5.0% (4) 1,3-butylene glycol (5.0%) (5) Purified water (88.0%) 2. [0048] [Test Method] Ten randomly selected male and female subjects aged 25 to 60 years were divided into two groups. In group A, two test areas were applied to the forearm of each subject, and two comparison areas were applied to comparative samples 1 and 2. In addition, one control area (5% 1,3-butylene glycol aqueous solution) was set. Each test area was applied twice a day (morning and evening) for 14 days, and the subjects were asked to evaluate (a) texture, (b) transparency, and (c) moisturizing sensation on a four-point scale: A: very good, B: good, C: no change, D: slightly bad. In group B, two test areas were applied to the forearm of each subject, and two comparison areas were applied to comparative samples 3 and 4. In addition, one control area (5% 1,3-butylene glycol aqueous solution) was set. Each test area was applied twice a day (morning and evening) for 21 days, and the same evaluation was performed.
[0049] The results of Test Example 2 are shown in Table 2. [Table 2] JPEG2025037673000002.jpg8696
[0050] [Table 3] JPEG2025037673000003.jpg8796
[0051] As shown in Tables 2 and 3, it was confirmed that the samples of the present invention containing niacinamide, the extract of Production Example 1 or Production Example 2, and the extract of Production Example 4 or Production Example 5 exhibit the effect of improving skin texture, transparency, and moisturizing feeling. From this result, it was confirmed that wrinkles caused by dryness of the epidermis can also be improved. Furthermore, since niacinamide is known to have the effect of improving wrinkles and sagging of the dermis, it is suggested that the composition of the present invention also has the effect of preventing and improving aging of both the epidermis and dermis.
[0052] The following are examples of formulations of skin external preparations (cosmetics, quasi-drugs, etc.) containing the composition according to the present invention, but the present invention is not limited thereto. In addition to the composition according to the present invention, as shown in the following formulation examples, it is more preferable to use in combination with one or more of "glycyrrhizinic acid or its derivatives," "tranexamic acid or its derivatives," "ascorbic acid or its derivatives," "pantothenyl alcohol," and "tocopherol or its derivatives," which are known to have effects of preventing or improving rough skin and preventing or improving pigmentation.
