[0018] According to the present invention, there are provided an efficient method for producing a halogeno-2,4-substituted benzoic acid derivative, and a method for producing a benzophenone derivative and a halogenobenzene derivative using the halogeno-2,4-substituted benzoic acid derivative obtained by this method. DETAILED DESCRIPTION OF THE INVENTION
[0019] The structure of SGLT-2 inhibitors is roughly divided into a sugar moiety consisting of gluconolactone or thiogluconolactone and various aglycone moieties linked to the sugar moiety. Halogeno-2,4-substituted benzoic acid derivatives can be used as intermediates for the aglycone moiety.
[0020] A method for producing a halogeno-2,4-substituted benzoic acid derivative represented by formula (VI) according to an embodiment includes contacting a 2,4-substituted benzoic acid derivative represented by formula (V) with a halogenating agent, the halogenating agent including at least one compound selected from the group consisting of N-bromosuccinimide, 1,3-dibromo-5,5-dimethylhydantoin, N-chlorosuccinimide, and 1,3-dichloro-5,5-dimethylhydantoin.
[0021] This production method uses the above-mentioned halogenating agent, allowing efficient production of a halogeno-2,4-substituted benzoic acid derivative. That is, simple substances such as bromine (Br2) and chlorine (Cl2) are sometimes used as halogenating agents. The present inventors have found that in the method for producing a halogeno-2,4-substituted benzoic acid derivative represented by formula (VI) using a 2,4-substituted benzoic acid derivative represented by formula (V), when simple substances such as halogen atoms are used, a large amount of by-products other than the target product is produced. This is thought to be due to the excessively high reactivity of these simple substances. Halogen compounds such as N-bromosuccinimide have lower reactivity than simple substances such as halogen atoms. Therefore, in the method according to the embodiment, the 2,4-substituted benzoic acid derivative represented by formula (V) and the halogenating agent can react appropriately, resulting in a small amount of by-products. Furthermore, simple substances such as halogen atoms are volatile liquids at room temperature and pressure, and are highly toxic. In contrast, halogen compounds such as N-bromosuccinimide are solid at room temperature and pressure, and have low toxicity, making them easy to handle.
[0022] Hereinafter, the invention according to the embodiment will be described in detail.
