Description

[Technical field]

[0001] The present invention relates to personal care compositions that lighten the skin. More particularly, the present invention relates to topical compositions that are effective against hyperpigmentation, especially hyperpigmentation caused by exposure to the sun. [Background technology]

[0002] Most people consider the appearance of their skin, especially the appearance of the skin on their face, as one of the important indicators of their own beauty and health. Thus, having a light, blemish-free, even skin tone is desired by many people. The degree and evenness of skin pigmentation is affected by factors such as age, hormonal changes, acne development, and exposure to sunlight and environmental pollution. These can lead to age spots or freckles, hyperpigmentation in certain localized areas of the skin, and even dark circles under the eyes. Many people believe that certain lifestyle factors, such as hydration (amount of water consumed), type of diet, and amount and quality of sleep, also affect the appearance of the skin, including dark circles under the eyes. People are aware that changes such as living a healthier lifestyle can take a long time to even out the appearance of the skin, so they turn to cosmetics that provide a temporary solution to skin blemishes. They also want products to reduce the darkening of the skin caused by exposure to sunlight. To meet this need, many attempts have been made to develop products that reduce pigment production in melanocytes.

[0003] The inventors have been working in this field for a long time and have filed several patent applications and produced various cosmetic products on the market. In the present invention, they were specifically looking for a highly effective solution to this problem, which could also be used as a general cosmetic product to provide a uniform and light skin tone, especially on exposed skin surfaces.

[0004] They want to ensure that the product they develop is equal to or better than the most effective products on the market. After screening a large number of actives, they found that one or more of the skin care actives known for providing an even skin tone, such as alkylresorcinols or carboxylic acid functionalized heteroaromatic compounds, or retinoic acid compounds known for their anti-aging benefits, when combined with inulin, provide enhanced skin brightness and even skin tone. To the best of the inventors' knowledge, no combination of actives that provide such benefits has been known so far. Furthermore, it is generally desirable for conventional skin care actives known for this benefit, such as alkylresorcinols or carboxylic acid functionalized heteroaromatic compounds or retinoic acid compounds, to be included in lower amounts, which not only reduces costs but also ensures better compatibility with other ingredients in the formulation, thereby resulting in better formulation stability. The inventors were looking for a solution to identify additives that exhibit synergy with the above conventional skin care actives, allowing the additives to be included at very low concentrations, thereby providing all of the above benefits. Summary of the Invention [Problem to be solved by the invention]

[0005] It is therefore an object of the present invention to provide a solution to the problem of hyperpigmentation and uneven skin appearance, thereby resulting in a brighter skin appearance.

[0006] Another object of the present invention is to provide such a solution that allows the inclusion of lower amounts of skin care actives, thereby ensuring better formulation stability. [Means for solving the problem]

The inventors have found that including inulin in the topical compositions of the present invention aids in the production of beneficial metabolites that promote skin health. For example, S. epidermidis fermented inulin can provide depigmentation benefits.

[0037] The invention will now be illustrated with the following non-limiting examples. EXAMPLES

[0038] Examples A-F, Example 1, Example 2: % Cellular Melanin Content of Various Combinations of Actives vs. Control

[0039] Various samples containing actives were prepared and the % cellular melanin content was measured. The protocol for measuring cellular melanin is summarized below.

[0040] Cell culture:

[0041] Primary neonatal human epidermal melanocytes were obtained from Life Technologies. Melanocytes were maintained in Medium 254 supplemented with human melanocyte growth supplement (MGM). HaCaT keratinocytes were maintained in DMEM medium containing 10% heat-inactivated FBS. All media were also supplemented with 10 U / mL penicillin G and 0.1 mg / mL streptomycin sulfate. Both cell types were maintained at 37°C in a humidified incubator with a 5% CO2 atmosphere. Cell maintenance and subculture were performed according to the manufacturer's instructions.

[0042] To set up the co-cultures, 40,000 cells each of HaCaT keratinocytes and melanocytes were mixed in 1 mL of a 1:1 mixture of MGM:KGM (KGM: Epilife medium supplemented with human keratinocyte growth supplement) and directly seeded per well into a 12-well Nunc plate.

[0043] Addition of active substances:

[0044] The active substances were prepared in culture medium and the test concentrations were added to the cells along with the relevant controls 24 hours after seeding. After 72 hours of treatment with the active substances, measurements of cell viability (using the calcein method) and cellular melanin content were performed as described below.

[0045] Cell viability assay:

[0046] Briefly, spent medium was removed and cells were washed once with 0.4 mL of phosphate-buffered saline. Fresh 1 μM calcein-acetoxymethyl in phosphate-buffered saline was added to each well. Plates were incubated at 37°C in a 5% CO2 incubator for 30 min. Calcein fluorescence was measured using a TECAN M1000 plate reader (excitation wavelength 490 nm, emission wavelength 520 nm).

[0047] Melanin Content Assay:

[0048] After calcein fluorescence measurements were obtained, cells were drained and 0.15 mL of fresh 1N NaOH (in 10% DMSO) was added per well. Cells were lysed by resuspension and incubation (60°C, 1 h). 0.1 mL of this lysate was then transferred to a new 384-well plate and the OD at 405 nm was measured on a TECAN M1000 plate reader. Melanin content was calculated as % cellular melanin relative to control.

[0049] The following samples were prepared as listed in Table 1 and the % melanin content of each was measured as listed in Table 1.

[0050] [Table 1]

[0051] COM1 represents a combination of niacinamide (0.0122%) + 4-hexylresorcinol (0.00002%) + retinyl propionate (0.00003%)

[0052] * indicates p<0.05 vs. Example D # indicates p<0.05 vs. Example F $ indicates p<0.05 vs. Example C

[0053] The data in Table 1 above show that the compositions according to the present invention (Examples 1 and 2) provide significantly better reduction in cellular melanin content compared to each individual component. The results of Example 2 are comparable to the highly effective positive control (Example E).

[0054] It should be understood that the above experiments were performed in an in vitro assay to evaluate the % melanin at the cellular level. The concentrations actually used to prepare the compositions for topical use are expected to vary widely. The concentrations are generally orders of magnitude higher due to the following reasons that affect the difference in bulk concentration compared to the cellular concentration: The compositions may be formulated as emulsions or gels that contain numerous additional ingredients. The concentrations of the desired actives in the oil phase and the water phase are expected to be very different. They may also have very different physical and hydrodynamic properties such as partition coefficients, diffusion rates, convective transport rates, rheological properties, etc. Thus, the concentrations used when formulating the compositions are expected to be significantly different, often orders of magnitude higher, than the cellular concentration at which the experiments were performed.
