Description

[Technical field]

[0001] The present invention relates to a composition for improving scalp conditions.

[0002] This application claims priority to Korean Patent Application No. 10-2022-0035446 filed on March 22, 2022 and Korean Patent Application No. 10-2022-0082719 filed on July 5, 2022, and the entire contents disclosed in the specification and drawings of said applications are incorporated herein by reference. [Background technology]

[0003] In modern society, many people use various products to cleanse and purify their scalp and hair. However, the main ingredients of these products contain surfactants, foam boosters, emulsifiers, thickeners, preservatives, etc., which may cause irritation and itching on the scalp.

[0004] In addition, many people use hair dyes or perms to change their hairstyle for beauty purposes. For some people with sensitive scalps, the physical and chemical irritation caused by hair dyes or perms can cause damage to the scalp.

[0005] In addition, changes in temperature and humidity in daily life, exposure to sunlight, and pollution such as fine dust can also irritate the scalp, and other factors such as stress and nutritional deficiencies can also worsen the condition of the scalp.

[0006] The scalp is composed of the epidermis, dermis, and subcutaneous tissue.

[0007] Normally, the stratum corneum of the scalp undergoes turnover and is replaced by new keratinocytes in a cycle of about one month. In the case of a healthy scalp, small keratinocytes fall off and are replaced by new keratinocytes during turnover. However, if the turnover is not steady and there is an excess of keratinocytes in certain areas, they may fall off in clumps that are visible to the naked eye, which is called dandruff. Although dandruff is a scalp problem, it is not a major problem for human health, and although there are differences in severity, it is a condition that occurs on most people's scalps.

[0008] The known causes of dandruff include increased sebum secretion on the scalp, increased proliferation and activity of microorganisms, abnormal keratinization due to abnormal scalp metabolism, and scalp irritation due to fatty acids generated by lipase.

[0009] When dandruff becomes excessive, the scalp may become scratched or rubbed due to itching, which may cause injury or inflammation. The itching that occurs at this time may be induced through the mediation of histamine and arachidonic acid metabolites, which causes a complex inflammatory reaction that stimulates nerve cells and induces the itching.

[0010] In addition, the secretion of sebum from the scalp, sweat, and the decomposition of keratin by microorganisms, which are related to the symptoms of dandruff, induce scalp odor. As these waste products oxidize, an unpleasant odor can be produced on the scalp. Scalp odor is related to the distribution of microorganisms, and this is because the scalp is more likely to form a habitat for microorganisms (hair, humidity, etc.) than other parts of the skin.

[0011] The active ingredient most commonly used in anti-dandruff shampoos currently on the market is zinc pyrithione. Zinc pyrithione has antimicrobial properties, and is water-insoluble with a high specific gravity, making it difficult to maintain a dispersed state within the formulation. In addition, zinc pyrithione is irritating, so products that can replace it with other ingredients are gradually being researched and developed in Korea.

[0012] There have been attempts to suppress dandruff formation or improve scalp itching by applying natural ingredients such as existing plant extracts to the scalp or taking them orally, but these have the disadvantage that they are not very effective in improving dandruff, excessive shedding of keratin, thickening of keratin, or itching, etc. Therefore, there is a need to develop a composition for improving scalp conditions that is effective in improving dandruff, excessive shedding of keratin, thickening of keratin, or itching, etc. Summary of the Invention [Problem to be solved by the invention]

[0013] An object of the present invention is to provide a composition for improving scalp conditions which is safe for the human body, has excellent formulation stability, and is capable of keeping the scalp healthy. [Means for solving the problem]

[0014] The present inventors have conducted extensive research to develop a composition capable of improving scalp conditions, and have found that the scalp condition is significantly improved when gluconolactone and climbazole are used in combination as active ingredients of the composition for improving scalp conditions. Specifically, the composition for improving scalp conditions of the present invention has a very excellent anti-inflammatory effect due to the synergistic effect of the combination of gluconolactone and climbazole, and therefore has remarkably excellent effects such as improvement of dandruff, improvement of scalp itching, improvement of keratin shedding, improvement of scalp keratin thickening, and improvement of seborrheic scalp inflammation. In addition, the composition for improving scalp conditions of the present invention has a very excellent effect of strengthening the scalp barrier by reducing the amount of transepidermal water loss. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS

[0033] Hereinafter, the present invention will be described in detail with reference to examples to aid in understanding the present invention. However, the embodiments according to the present invention may be modified in various other forms, and the scope of the present invention should not be construed as being limited to the following examples. The embodiments of the present invention are provided to more completely explain the present invention to those having average knowledge in the field to which the present invention pertains.

[0034] Experimental example 1: Confirmation of anti-inflammatory effect (1) Evaluation of inflammatory response inhibition rate To confirm the anti-inflammatory activity of gluconolactone and climbazole, their ability to inhibit the production of nitric oxide (NO), one of the substances that induce inflammation, was analyzed. The RAW 264.7 cells used in the experiment are murine macrophage cell lines, which are used as the main model for anti-inflammatory activity cell experiments and were purchased from ATCC (American Type Tissue Collection). The RAW 264.7 cells were cultured in RPMI1640 (Gibco) medium containing 10% FBS (fetal bovine serum, Gibco) and 1% penicillin / streptomycin (Gibco).

[0035] To confirm the anti-inflammatory activity of gluconolactone and climbazole, RAW 264.7 cells were cultured in a 96-well plate at 2.0 × 10 cells per well. 4 The cells were plated, allowed to attach, and cultured for 24 hours, after which they were treated with 1 μg / mL of lipopolysaccharide (LPS), gluconolactone, and climbazole for 24 hours simultaneously, and their ability to inhibit NO production was analyzed by the Griess reaction.

[0036] The Griess reaction is a method for quantitatively analyzing the amount of NO produced by cells treated with a sample and released into the culture medium. The Griess reaction solution is prepared by mixing a 1% sulfanilamide solution dissolved in a 5% phosphoric acid solution with a 0.1% naphthylenediamine dihydrochloride solution in a 1:1 ratio.

[0037] After 24 hours of treatment, 50 μL of cell culture medium was taken from each well and dispensed into a 96-well plate. The same amount of the prepared Griess reaction solution as the cell culture medium was then dispensed into wells at 50 μL each, and the color change due to NO production and inhibition was measured by absorbance at 540 nm using a multi-plate reader. The experimental results were shown as the average of three replicates for each gluconolactone and climbazole treatment combination.

[0038] To confirm whether gluconolactone and climbazole induce cytotoxicity, a CCK-8 assay (Cell Counting Kit-8, Dojindo) was performed using 50 μL of cell culture medium. The CCK-8 assay is a method for measuring cell viability. After adding 1 / 10 of the assay reaction solution to the cell culture medium and reacting at 37°C for one hour, the absorbance at 450 nm was measured using a multiplate reader, and no cytotoxicity was observed between the measured concentrations.

[0039] The NO inhibitory activity induced by LPS was expressed as a percentage of the control group (A) in which neither LPS nor the sample was treated, and the control group (B) in which only LPS was added and no sample was added. The formula for calculating the percentage of the inflammation inhibitory activity in each sample (C) is as follows: Inflammation inhibitory activity (%)={(BC) / (BA)}×100

[0040] A is the absorbance at 540 nm of the control group in which neither LPS nor the sample was treated, B is the absorbance at 540 nm of the control group in which only LPS was treated, and C is the absorbance at 540 nm of the experimental group in which both LPS and the sample were treated.

[0041] The calculated values for the inflammatory response suppression activity of gluconolactone and climbazole of the present invention are shown in Table 1 below. [Table 1]

[0042] From the experimental results shown in Table 1, it can be seen that gluconolactone or climbazole inhibits inflammatory responses, and in particular, when gluconolactone and climbazole are administered together, the inhibition rate of inflammatory responses is significantly increased.

[0043] (2) Examination of the synergistic anti-inflammatory effect of gluconolactone and climbazole When gluconolactone and climbazole are used together, the expected value (inhibitory rate of inflammatory reaction) was calculated using Colby's formula. The calculation process and results are shown below. "Synergy effect" is as defined in "Calculation of the synergistic and antagonistic responses of herbicide combinations", Weeds, (1967), 15, pp. 20-22. Colby's formula: Expected value = A + B - (A × B / 100) A: Efficacy observed from active ingredient A at the same concentration used in the mixture. B: Efficacy observed from active ingredient B at the same concentration used in the mixture.

[0044] In order to calculate the expected inflammatory response inhibition rate (expected value) when gluconolactone and climbazole are used together, the inflammatory response inhibition rate of the experimental group treated with only LPS and gluconolactone was substituted into A of the Colby formula, and the inflammatory response inhibition rate of the experimental group treated with only LPS and climbazole was substituted into B to calculate the expected value according to Colby's formula, and the values are shown in Table 2 below.

[0045] In addition, the degree of synergy effect was calculated and shown in Table 2 below. The values in Table 2 were rounded off to one decimal place and expressed to one decimal place. [Table 2]

[0046] From the results shown in Table 2, it can be seen that the actual inflammatory response inhibition rate when gluconolactone and climbazole are actually used together is about 14.4% higher than the expected value predicted when gluconolactone and climbazole are used together, when both substances are used at 25 μM each. In other words, it can be seen that the combination of gluconolactone and climbazole shows a remarkable synergistic effect in inhibiting inflammatory responses.

[0047] Experimental Example 2: Evaluation of scalp condition improvement In order to confirm the efficacy of the composition for improving scalp conditions prepared according to the present invention, a composition (shampoo) for improving scalp conditions containing gluconolactone and climbazole was prepared in Example 1, and a composition not containing gluconolactone and climbazole was prepared in Comparative Example 1 (see Table 8).

[0048] The experimental group consisted of 12 adults aged 26-45 who suffered from scalp itching, divided into two groups of six people each. The product evaluated was a wash-off type shampoo, and after using 3ml of the product once a day for two weeks, the degree of improvement in scalp condition was evaluated in the following items.

[0049] (1) Evaluation of transepidermal water loss (TEWL) of the scalp Transepidermal water loss indicates the degree to which water evaporates from within the skin, and tends to increase when the skin's barrier function is damaged.

[0050] To measure the transepidermal water loss of the subjects' scalps more than 8 hours after washing the hair, the scalps were left to stand for 20 minutes under constant temperature and humidity conditions (23°C, relative humidity 40%) for application. After 20 minutes of application, the transepidermal water loss at the top of the head, parietal area, forehead, back of the head, and the intersection with the midline of the head was measured using a transepidermal water loss meter (Tewameternano probe, MPA6, C+K Electronic Co., Germany).

[0051] Thereafter, the subjects used the scalp condition improving composition (Comparative Example 1, Example 1) for two weeks each, and the transepidermal water loss was measured at the same site one week and two weeks after using the composition. One of the subjects who used Comparative Example 1 discontinued the test midway.

[0052] The measurement results, which are the average of the five subjects in each Comparative Example 1 group and the average of the six subjects in each Example 1 group, are shown in Table 3 and FIG. [Table 3]

[0053] As can be seen from Table 3 and FIG. 1, when Example 1, which is a composition for improving scalp conditions containing gluconolactone and climbazole, was used, the amount of transepidermal water loss was significantly reduced compared to when Comparative Example 1, which is a control group not containing gluconolactone and climbazole, was used.

[0054] (2) Evaluation of improvement in scalp keratin hyperplasia In order to evaluate the efficacy of the composition for improving scalp condition according to the present invention in improving keratinocyte thickening, keratin was collected from the scalp of the subject and the thickness of the keratin was analyzed.

[0055] The thickened keratin of the scalp was analyzed using Visioscan VC98 (C+K Electronic Co., Germany). To collect and measure keratin, a special tape, Black D-Squame tape, was prepared with a size of 0.5cm x 2.5cm and attached to the scalp surface of the subjects at the four intersections of the top of the head, the parietal region, the frontal region, the back of the head, and the scalp center line for 10 seconds, and then the tape was peeled off and attached to a Visioscan (CCD-camera) to take a picture, and the mass of the collected keratin was measured. Since it is difficult to eliminate the interference of hair between the desquamation index provided after analysis by the software and the total amount of collected keratin, the ratio of the most thickened keratin (G4) to the total area of the collected and analyzed keratin (G0+G1+G2+G3+G4) was defined as the "proportion of thickened keratin" and the analysis was performed. Percentage of thickened keratin = G4 / (G0+G1+G2+G3+G4)

[0056] The parameters such as G0, G1, G2, G3, and G4 are as defined in "Relationship between lip skin biophysical and biochemical characteristics with corneocyte unevenness ratio as a new parameter to assess the severity of lip scaling", International Journal of Cosmetic Science (2021), Vol. 43, Issue 3, pages 275-282.

[0057] The rate of thickened keratin is expressed as an improvement rate relative to the measured value before use of the product (0 weeks), and the formula for calculating the improvement rate is as follows. Improvement rate of keratinocyte hyperplasia = (ab) / a x 100 a: Percentage of thickened keratin before product use b: Percentage of thickened keratin after product use

[0058] The measurement results are shown in Table 4 as the average of the five people in each Comparative Example 1 group and the average of the six people in each Example 1 group, and representative images of the Example 1 group before and after use of the product are shown in Figure 2. [Table 4]

[0059] As can be seen from Table 4 and FIG. 2, when Example 1, which is a composition for improving scalp conditions containing gluconolactone and climbazole, was used, keratinocyte hyperplasia was improved more than when Comparative Example 1, which is a control group not containing gluconolactone and climbazole, was used.

[0060] (3) Analysis of interleukin-8 in the collected stratum corneum In order to confirm the efficacy of the composition for improving scalp condition of the present invention in improving the secretion of inflammatory factors, the content of interleukin-8 in the stratum corneum was analyzed using stratum corneum collected from the scalp of the subject.

[0061] Interleukin-8 (IL-8) is a representative inflammatory factor known to be increased in keratin samples from lesions such as dandruff and seborrheic scalp dermatitis in literature such as "Dandruff / seborrhoeic dermatitis is characterized by an inflammatory genomic signature and possible immune dysfunction: transcriptional analysis of the condition and treatment effects of zinc pyrithione", British Journal of Dermatology (2012), Vol. 166, Issue S2, pp. 33-40.

[0062] To collect keratin, a special tape, Black D-Squame tape, measuring 0.5cm x 2.5cm, was applied to the scalp surface of the subjects at the four intersections of the top, parietal, frontal, back of the head, and the head center line for 10 seconds, and then the tape was removed. The collected keratin was dissolved using 1xPBS containing 0.05% Tween 20 + 0.5mM EDTA. The keratin was sonicated with a buffer solution for 60 minutes, and the dissolved keratin solution was quantitatively analyzed using a Human IL-8 ELISA kit (R&D system).

[0063] In addition, the amount of total protein in the eluted keratin solution was determined using a BCA Protein assay (ThermoFisher Scientific), and the amount of IL-8 in the keratin was analyzed after normalization with the amount of total protein.

[0064] The results of IL-8 analysis are shown in Table 5 as the average of the five subjects in each Comparative Example 1 group and the average of the six subjects in each Example 1 group. [Table 5]

[0065] As can be seen from Table 5, when Example 1, which is a composition for improving scalp conditions containing gluconolactone and climbazole, was used, IL-8 secretion was improved more than when Comparative Example 1, which is a control group not containing gluconolactone and climbazole, was used.

[0066] (4) Sensory evaluation In order to confirm the efficacy of the composition for improving scalp condition prepared according to the present invention, the experimenters who participated in the evaluation were surveyed on the following items and subjected to a sensory evaluation. The products evaluated were wash-off type shampoos of Comparative Example 1 and Example 1. After using each product once a day for one week, the subjects were surveyed to confirm the subjective improvement of their scalp condition, and the evaluation scale was 0 to 5 points. The self-diagnosis criteria for each item are shown in Table 6 below. [Table 6]

[0067] The results of the sensory evaluation were the average of the group of five people in Comparative Example 1 and the group of six people in Example 1, and are shown in Table 7 below and FIG. [Table 7]

[0068] As can be seen from Table 7 and FIG. 3, when Example 1, which is a composition for improving scalp conditions containing gluconolactone and climbazole, was used, scalp itching, keratin loss, and scalp odor were significantly improved and scalp irritation was also reduced compared to when Comparative Example 1, which is a control group not containing gluconolactone and climbazole, was used.

[0069] From the above experimental results, it can be seen that the composition for improving scalp condition of the present invention has remarkably excellent effects of improving transepidermal water loss, keratinocyte thickening, IL-8 secretion, scalp itching, keratin shedding, and scalp odor due to the combination of gluconolactone and climbazole.
