AI Technical Summary

Technical Problem

Current cancer diagnostic and therapeutic approaches targeting fibronectin, a key extracellular matrix protein, have not been translated into practical pharmaceuticals or diagnostic techniques despite decades of research, highlighting the need for novel methods to identify and analyze disease-specific epitopes, particularly in cancer.

Method used

Development of glycopeptides with specific sugar chains attached to threonine residues, which can serve as dynamic epitopes for cancer diagnosis and therapy, utilizing mass spectrometry for analysis and identification of these epitopes in serum or plasma samples.

Benefits of technology

The approach enables systematic identification of disease-specific dynamic epitopes, facilitating new diagnostic biomarkers and therapeutic antibodies, and provides a method for analyzing cancer progression by quantifying changes in glycopeptide concentrations, particularly in liver and pancreatic cancers.

Abstract

The present invention addresses the problem of providing a material for systematically searching for and identifying epitopes specific to various diseases including cancer. The present invention also addresses the problem of establishing a method for disease analysis using epitopes specific to various diseases including cancer. The present invention relates to a glycopeptide in which one sugar chain comprising not more than 6 sugars is bonded to each of one or two or more of the threonines in a peptide comprising the amino acid sequence : TTPPTTATPIR, wherein the sugar chain is an O-linked sugar chain. The present invention further relates to a method for analyzing glycopeptides, comprising analyzing whether an epitope glycopeptide is present in a sample containing glycopeptide obtained by protease digestion of serum or plasma, wherein the glycopeptide is glycopeptide in which one sugar chain comprising not more than 6 sugars is bonded to each of one or two or more of the threonines in a peptide comprising the amino acid sequence : TTPPTTATPIR, wherein the sugar chain is an O-linked sugar chain.
