AI Technical Summary

Technical Problem

Current 2D culture methods for dental pulp stem cells are physiologically deficient due to the lack of an extracellular matrix, limiting their differentiation and regenerative potential for dentin and pulp tissue repair.

Method used

A 3D graft culture platform using a scaffold made of matrigel, with varying concentrations for the lower and upper parts, allows for the culture of pulp tissue, promoting odontoblast growth and avascular dentin formation, and is maintained with a specific culture medium that includes L-Glutamine, Phenol Red, and HEPES.

Benefits of technology

The platform supports the growth of odontoblasts and formation of avascular dentin, with positive expression of stem cell and odontoblast markers, enabling the regeneration of dental tissue and potential tooth formation.

Abstract

The present invention provides a platform for culturing a 3D graft of a human dental pulp tissue and a method for culturing a dental pulp tissue using same. As a result of culturing an undamaged dental pulp tissue obtained from teeth separated from a human body in a support made of Matrigel for 4 weeks, odontoblast growth, reticular growth, and orthodentin formation were found on the surface of the dental pulp tissue; the stem cell marker NESTIN and the odontoblast markers PHEX and DMP1 were positive in a derivative of the dental pulp tissue through culturing; and the mineralization marker OSTERIX was detected at the edge of the derivative. Thus, the present invention provides a method for culturing odontoblasts and forming teeth derived from dental pulp tissues through three-dimensional graft culture of dental pulp tissues.
