Description

Novel hemiasterline derivatives and uses thereof

[0001] The present invention provides a novel hemiasterlin derivative, a method for preparing the same, and its use. The novel hemiasterlin derivative synthesized in the present invention exhibits a significant survival inhibitory effect against cancer cell lines, and can be used as a safe form of anticancer agent.

[0002] Hemiasterlin can be isolated from the marine sponges Cymbastela sp., Hemiasterella minor, Siphonochalina sp., and Auletta sp. (Talpir et al., Tetrahedron Letters, vol. 35, no. 25, pp. 4453-4456, 1994).

[0003] Natural hemiasterlin is known as a drug that inhibits the formation of microtubules, which are essential for cancer cell division, and is being actively studied for use in the treatment of cancers such as ovarian cancer, but safety concerns still exist.

[0004] Accordingly, the inventors of the present invention completed the present invention by identifying the structure of a derivative of hemiasterlin with improved safety as a therapeutic drug and confirming an anticancer effect similar to that of natural hemiasterlin.

[0005] The present invention aims to solve the above-mentioned problems by identifying a novel hemiasterlin derivative structure with improved safety, and to provide it as a drug for preventing or treating cancer in the form of a drug-linker, drug-carrier conjugate (ADC), etc.

[0006] To achieve the above purpose, the present invention provides a novel hemiasterlin derivative, a compound of the following chemical formula 1, an isomer thereof, a pharmaceutically acceptable salt thereof, or a solvate thereof.

[0326] Cell viability analysis

[0327] 3000 FaDu or A549 cells were seeded per well in a 96-well plate and incubated (37℃, 5% CO2). After 24 hours, 100 μl of the drug at 9 concentrations (serial dilution of 1 / 5 each starting from 1000 nM) was treated to the cell lines. The drugs included the compound of chemical formula 2 prepared according to Example 1, SC209 (MedChemExpress), and MMAE. A control group (drug concentration 0) that was not treated with the drug was also prepared. After incubating for 3 days (37℃, 5% CO2), 100 μl of CellTiter-Glo reagent (CellTiter-Glo® Luminescent Cell Viability Assay kit (Promega, G7571)) was added to each well of the plate and pipetted. After incubation at room temperature (RT) for 10 minutes, luminescence was measured. When the luminescence value at 0 drug concentration is considered 100%, the concentration that shows 50% of the luminescence value is IC 50 It's worth it.
