Description

A preparation method of GLP1R agonist and its intermediate Technical Field

[0001] The present invention belongs to the technical field of drug synthesis, and particularly relates to a method for preparing 2-(2-fluoro-4-(oxetane-3-yl)benzyl)oxy)-6-(piperidin-4-yl)pyridine (Formula XI) and (S)-2-((4-(6-((2-fluoro-4-(oxetane-3-yl)methyl)oxy)pyridin-2-yl)piperidin-1-yl)methyl)-1-(oxetane-2-ylmethyl)-1H-benzimidazole-6-carboxylic acid (Formula I), intermediates of a GLP-1 receptor agonist. Background Art

[0002] (S)-2-((4-(6-((2-fluoro-4-(oxetan-3-yl)methyl)oxy)pyridin-2-yl)piperidin-1-yl)methyl)-1-(oxetan-2-ylmethyl)-1H-benzimidazole-6-carboxylic acid (compound of Formula I) is a non-peptide small molecule glucagon-like peptide-1 receptor (GLP-1R) agonist with a molecular weight of 586.26 and a chemical structure as follows:

[0003] Invention patent WO 2022 / 268152 Al discloses for the first time a compound of formula I and a feasible preparation method thereof, using 3-(4-(bromomethyl)-3-fluorophenyl)oxetane and tert-butyl 4-(6-hydroxypyridin-2-yl)piperidine-1-carboxylate as raw materials, undergoing nucleophilic substitution and removal of the Boc protecting group to obtain the intermediate XI of the present invention (specific steps are as follows), which is then substituted and hydrolyzed to obtain the compound of formula I. Among them, 3-(4-(bromomethyl)-3-fluorophenyl)oxetane is obtained by bromination of a benzyl alcohol derivative, and tert-butyl 4-(6-hydroxypyridin-2-yl)piperidine-1-carboxylate is obtained by hydroxylation of a 6-chloropyridine derivative catalyzed by a palladium catalyst.

[0004] In kilogram-scale synthesis scale-up experiments, the raw material 3-(4-(bromomethyl)-3-fluorophenyl)oxetane used in the above-mentioned route is unstable and highly allergenic. Even with proper protection, the experimenters still experienced skin rashes or ulcers, posing a safety hazard in industrial production. Furthermore, the 6-hydroxyl group in tert-butyl 4-(6-hydroxypyridin-2-yl)piperidine-1-carboxylate needs to be converted from a chloropyridine derivative in the presence of a precious metal palladium catalyst, which is costly. Therefore, it is highly necessary to seek a process route with stable raw materials, controllable costs, and ease of production.

[0005] Summary of the Invention

[0006] In order to overcome the problems of unstable and highly sensitized raw materials or excessively high costs described in the background art, the present invention provides a novel method for preparing 2-(2-fluoro-4-(oxetane-3-yl)benzyl)oxy)-6-(piperidin-4-yl)pyridine (Formula XI). This method overcomes the instability and safety hazards of 3-(4-(bromomethyl)-3-fluorophenyl)oxetane, effectively simplifies the steps, and reduces the cost of raw materials. The process is as follows:

[0007] Wherein, R is an amino protecting group, which is independently selected from one of benzyloxycarbonyl (Cbz), tert-butyloxycarbonyl (Boc), p-methoxybenzyl (PMB), benzyl (Bn), trityl (Trt), p-toluenesulfonyl (Tos), phthaloyl (Pht), and allyloxycarbonyl (Alloc); wherein, R x is selected from Cl, Br and I, such as Cl.

[0008] The process provided by the present invention uses compound 1 and a 6-halopyridine derivative as raw materials, generates compound X-1 through an affinity substitution reaction, removes the amino protecting agent to obtain a compound of formula XI, and then obtains a compound of formula I through substitution, hydrolysis and other steps of the compound of formula XI. [0087] Example 6: Preparation of Compound X-1 (When R is Boc, Preparation of Compound X2-1)

[0088] Under nitrogen, 100 mL of dry toluene, 18 g of compound X2, 10 g of compound 1, and 5.1 g of powdered potassium hydroxide were added sequentially to a reaction flask equipped with a water separator. The temperature was raised to 120°C and stirred for 1 hour. After cooling to room temperature, the reaction solution was diluted with 150 mL of ethyl acetate, followed by 200 mL of purified water. The mixture was stirred for 25 minutes, allowed to stand for stratification, and the organic phase was collected. The aqueous phase was back-extracted once with 150 mL of ethyl acetate. The combined organic phases were washed with purified water and saturated brine (2 × 200 mL each), and the organic phase was collected. The organic phase was dried over anhydrous sodium sulfate, filtered, and concentrated to dryness.

[0089] Post-processing

[0090] The concentrated crude product was dissolved in 50 mL of ethyl acetate, stirred for 1 h, and then filtered to remove insoluble matter. The filtrate was diluted with 100 mL of n-hexane. The organic phase was poured into a pre-packed silica gel column, and the eluate was collected. The eluate was collected and concentrated to dryness, and a mixed solvent of methyl tert-butyl ether and n-hexane (V / V = 1:20) was added and heated until the solution became clear. The mixture was then cooled to room temperature for crystallization, filtered, and the filter cake was rinsed with n-hexane and dried in vacuo to obtain compound X2-1 (13.5 g, molar yield 55%).

[0091] Another post-treatment method used in Example 6 is as follows: the concentrated crude product is dissolved in 150 mL of a mixed solvent of ethyl acetate and n-hexane (V / V = 1:2). The resulting organic phase is filtered through a suction funnel filled with silica gel (approximately 30 g), and the filter cake is rinsed with a mixed solvent of n-hexane and ethyl acetate (V / V = 1:2). The filtrate and the rinse are combined and concentrated to dryness. Then, a mixed solvent of methyl tert-butyl ether and n-hexane (V / V = 1:4) is added and heated until the solution is clear. The mixture is then cooled to room temperature for crystallization, filtered, and the filter cake is rinsed with n-hexane and dried under vacuum to obtain compound X2-1.

After stirring at room temperature for 60min, the temperature was raised to 50°C and the reaction was continued for 6h. After the reaction solution was cooled to room temperature, purified water was added, stirred for 2h and filtered. The filter cake was dissolved in ethyl acetate, and the organic phase was washed twice with saturated brine. The organic phase was dried over anhydrous sodium sulfate, filtered and concentrated to dryness. The concentrated crude product was slurried with methyl tert-butyl ether at 50°C for 2h, then cooled to room temperature and filtered. The filter cake was rinsed with ethyl acetate and dried under vacuum to obtain compound VI with a molar yield of 85%.

[0094] Compound VI and methanol were added to a reaction flask at room temperature. Lithium hydroxide monohydrate was dissolved in purified water and added to the reaction flask. The mixture was stirred at room temperature for 1 hour. The reaction temperature was raised to 50°C and stirred for 18 hours. After the reaction solution cooled to room temperature, purified water was added and acetic acid was added dropwise to adjust the pH of the system to 5-6. The precipitated solid was collected and then slurried with methyl tert-butyl ether at 50°C for 4 hours. After cooling to room temperature, the mixture was stirred for 4 hours and filtered. The filter cake was rinsed with methyl tert-butyl ether. The resulting solid was dried under vacuum to obtain Compound I in a molar yield of 90%.
