Description

Anti-obesity composition containing powder of lactic acid bacteria derived from Phellodendron amurense

[0001] This invention was made possible by project number S3138172 (project unique number 1425175213) with the support of the Ministry of SMEs and Startups. The research management specialized organization of the project is the Small and Medium Business Technology Information Promotion Agency, the research project name is "New Product Development Project with Purchase Conditions (Purchase Linked Type)", the research project name is "Commercialization of Hypersensitive Immune Reaction Health Functional Food Raw Material of Killed Cell Powder Using Phellinus Linteus Lactic Acid Bacteria", the main organization is Bytech Co., Ltd., and the research period is from September 6, 2021 to September 5, 2023.

[0002] The present invention relates to an anti-obesity composition comprising a powder of killed lactic acid bacteria derived from Phellinus linteus, and more particularly, to an anti-obesity composition comprising a powder of killed lactic acid bacteria derived from Phellinus linteus, including killed Lactobacillus plantarum VIOAP03 KCTC14498BP and killed Lactobacillus fermentum VIMPP04 KCTC14499BP, as an active ingredient.

[0003]

[0004] Obesity is a condition in which excessive fat accumulation occurs due to metabolic disorders, which cause fat cells to proliferate and differentiate. When energy intake increases relative to energy expenditure, the number and volume of adipocytes increase, resulting in an increase in adipose tissue mass (Otto et al, Crit Rev Biochem Nol Biol., 40(4):229-242, 2005). At the cellular level, obesity is understood as an increase in the number and volume of adipocytes due to the promotion of adipocyte proliferation and differentiation (Hirsch et al, Clin Endocrinol Metab., 5(2):299-311, 1976).

[0005] Obesity is associated with serious complications and a very high morbidity rate. Furthermore, obesity is of increasing public health importance because it increases the risk of diabetes, hypertension, dyslipidemia, osteoarthritis, cardiovascular disease, and some cancers. However, preventing or treating obesity is challenging. While most obese patients can improve their obesity somewhat easily by maintaining a healthy weight through diet control and appropriate exercise, treating patients with genetic obesity or those diagnosed with obesity for a long time is challenging. Treatment for obesity begins with non-pharmacological therapies such as diet, exercise, and behavioral therapy. If sufficient weight loss is not achieved after three to six months of these non-pharmacological therapies or if obesity-related complications occur, pharmacological treatment is initiated.

[0006] Pharmacological treatments for obesity have long been attempted, but various side effects have hindered their effectiveness. Since thyroid hormones were introduced as an anti-obesity drug in the 1890s, numerous medications have been tried, but most have been discontinued due to ineffectiveness or adverse effects. Abuse of amphetamines and diuretics has been a problem in recent years, and in the 1990s, the combination drug fenfluramine and phentermine was banned due to heart valve problems. Furthermore, the appetite suppressant sibutramine, originally developed as an antidepressant, inhibits serotonin reuptake at the synapse, promoting a faster feeling of fullness. However, it has been reported to have various side effects, including cardiovascular effects, central nervous system effects, and kidney damage. Orlistat, a drug that inhibits lipase activity and promotes fat excretion, has been reported to be ineffective in the Korean diet, which is primarily based on carbohydrates.

[0007] Although various drugs have been used to treat obesity to date, their use is limited due to numerous side effects and minimal effects. Therefore, there is a need to develop new anti-obesity treatments that have excellent anti-obesity effects while minimizing side effects.

[0008] Recently, research is being conducted on probiotics as a natural alternative to anti-obesity drugs. In particular, various research results are being published on the correlation between obesity and the gut microbiota, as well as on probiotics with anti-obesity effects. Consequently, the need for screening for probiotics with anti-obesity effects is growing.

[0009] Accordingly, the inventors of the present invention have conducted extensive research efforts to overcome the problems of the above-mentioned prior arts, and as a result, have confirmed that an anti-obesity composition comprising a dead cell powder containing dead cells of Lactobacillus plantarum VIOAP03 KCTC14498BP derived from Phellinus linteus and dead cells of Lactobacillus fermentum VIMPP04 KCTC14499BP derived from Phellinus linteus as an active ingredient can prevent or improve obesity by suppressing fat accumulation, thereby completing the present invention.

[0015]

[0016] Accordingly, the main purpose of the present invention is to provide an anti-obesity composition comprising a powder of lactic acid bacteria cells derived from Phellodendron amurense, which can prevent or improve obesity by suppressing fat accumulation.

[0017]

[0018] According to one aspect of the present invention, the present invention provides an anti-obesity composition comprising, as an active ingredient, a killed cell powder comprising killed cells of Lactobacillus plantarum VIOAP03 KCTC14498BP derived from Phellinus linteus and killed cells of Lactobacillus fermentum VIMPP04 KCTC14499BP derived from Phellinus linteus.

[0019] The Lactobacillus plantarum VIOAP03 and Lactobacillus fermentum VIMPP04 cells according to the present invention are derived from Phellinus linteus, specifically, from Phellinus linteus, a mulberry-derived Phellinus linteus mushroom. The specific separation and identification methods and the characteristics of the deposited strain are as disclosed in the applicant's published patent application No. 2022-0143231. The present inventors confirmed that the deposited strain isolated and identified as described above is significantly superior to commercially available strains in the effect of inhibiting fat accumulation, and intended to use it as a lactic acid bacterium for preventing or improving obesity.

[0020] In the anti-obesity composition of the present invention, the dead cell powder is prepared by mixing a culture of Lactobacillus plantarum VIOAP03 KCTC14498BP derived from Phellinus linteus in a medium containing a Phellinus linteus extract and a culture of Lactobacillus fermentum VIMPP04 KCTC14499BP derived from Phellinus linteus in a medium not containing a Phellinus linteus extract, and then heat-treating the mixture. In the case of the dead cell powder prepared using the deposited strain of the present invention, the number of bacteria increases and conjugated linolenic acid, which is an effective substance, is produced, so that the anti-obesity effect can be enhanced.

[0021] In the anti-obesity composition of the present invention, the dead cell powder contains conjugated linolenic acid, linolenic acid and oleic acid in a ratio of 1:0.5 to 0.7:0.3 to 0.6, and preferably contains conjugated linolenic acid, linolenic acid and oleic acid in a ratio of 1:0.7:0.6.

[0022] According to one experimental example of the present invention, as a result of confirming the active ingredients from the killed cell powder manufactured using the deposited strains of the present invention, Lactobacillus plantarum VIOAP03 KCTC14498BP and Lactobacillus plantarum VIMPP04 KCTC14499BP, it was confirmed that conjugated linolenic acid, linolenic acid, and oleic acid were produced as the active ingredients, and it was confirmed that these active ingredients were included in a ratio of 1:0.5~0.7:0.3~0.6. These results show that the killed cell powder according to the present invention contains active ingredients capable of reducing body fat and thus can be used as an anti-obesity composition (see Experimental Example 1).

[0023] The Phellinus linteus extract according to the present invention can be prepared using any solvent conventionally used to extract medicinal plants, and water, an organic solvent, or a mixed solvent thereof is suitably used as the solvent. The organic solvent may be selected from the group consisting of ethanol, methanol, propylene glycol, butylene glycol, glycerin, hydrous ethanol, hydrous butylene glycol, hydrous propylene glycol, hexane, and ethyl acetate, and a solvent in which these organic solvents and water are appropriately mixed may also be used. In addition, as the extraction method using the extraction solvent, a plant extraction method commonly used in the art, such as hot water extraction or organic solvent extraction, may be used. Specifically, in the case of Phellinus linteus, it is suitable to add the dried or pulverized Phellinus linteus mushroom to the extraction solvent and heat at 90 to 95°C for 3 to 5 hours while stirring as necessary to perform extraction, and the amount of the solvent used for extraction is preferably 2 to 5 times (weight ratio) the amount of the extraction raw material.

[0024] In the present invention, the heat treatment can be performed through a tindile process, and preferably, it can be performed at 110°C for 15 minutes in the first step and at 90°C for 30 minutes in the second step. In addition, the dead cells (culture medium state) manufactured through the tindile process can be powdered through a drying process, and preferably, the tindile culture medium is rapidly frozen at -180°C to -195°C using a liquid nitrogen freezer (LNF), and then the rapidly frozen tindile culture medium is freeze-dried at -35°C to -45°C to powderize. If the step of rapidly freeze-drying using a liquid nitrogen freezer is performed before freeze-drying, the freeze-drying time can be shortened, the browning phenomenon of the raw material can be prevented, and the active ingredient can be maintained while minimizing damage to the strain.

[0025] In the anti-obesity composition of the present invention, the dead cell powder is 1Х10 of Lactobacillus plantarum VIOAP03 KCTC14498BP dead cells and Lactobacillus fermentum VIMPP04 KCTC14499BP dead cells. 6 cells / g or more, preferably 1Х10 8 1Х10 12 Contains cells / g concentration.

[0026] The anti-obesity composition of the present invention can prevent or improve obesity by inhibiting fat accumulation. Since adipocyte differentiation leads to fat accumulation and cell hypertrophy, inducing obesity, inhibiting fat accumulation through inhibition of adipocyte differentiation can prevent and improve obesity.

[0027] According to one experimental example of the present invention, the dead cell powder manufactured using the deposited strains of the present invention, Lactobacillus plantarum VIOAP03 KCTC14498BP and Lactobacillus plantarum VIMPP04 KCTC14499BP, was confirmed to have an inhibitory effect on adipocyte differentiation. As a result, the dead cell powder manufactured using the commercial strain did not exhibit an inhibitory effect on adipocyte differentiation, whereas the dead cell powder of the present invention exhibited an excellent inhibitory effect on adipocyte differentiation. These results show that the dead cell powder according to the present invention can prevent and improve obesity by inhibiting fat accumulation (see Experimental Example 4).

[0028] In the anti-obesity composition of the present invention, the anti-obesity composition can prevent or improve obesity by inhibiting the expression of one or more proteins selected from the group consisting of PPARγ, C / EBPα, SREB, and FABP4 in preadipocytes. Various transcription factors and coregulators are involved in the differentiation of adipocytes, and representative examples thereof include peroxisome proliferator-activated receptorγ (PPARγ), CCAAT / enhancer-binding protein α (C / EBPα), sterol regulatory element-binding protein, and fatty acid binding protein 4 (FABP4), which are known as coregulators of adipogenesis. Since the expression of coregulators of adipogenesis is increased by the interaction of these coregulators, thereby inducing the expression of fatty acid synthase (FAS), a specific gene of adipocytes, inhibition of the expression of coregulators can prevent and improve obesity.

[0029] According to one experimental example of the present invention, the dead cell powder manufactured using the deposited strains of the present invention, Lactobacillus plantarum VIOAP03 KCTC14498BP and Lactobacillus plantarum VIMPP04 KCTC14499BP, showed an inhibitory effect on the expression of adipocyte differentiation proteins. As a result, the dead cell powder manufactured using commercial strains did not show an inhibitory effect on the expression of adipocyte differentiation proteins, whereas the dead cell powder of the present invention showed an excellent inhibitory effect on the expression of adipocyte differentiation proteins. These results show that the dead cell powder according to the present invention can prevent and improve obesity by inhibiting the expression of fatty acid synthase (FAS), a specific gene of adipocytes, by inhibiting the expression of adipocyte differentiation proteins (see Experimental Example 5).

[0030] In the anti-obesity composition of the present invention, the composition may be a health functional food composition. If another composition of the present invention is a health functional food composition, it may have a dosage form convenient for intake and use, and preferably may be formulated into one or more dosage forms selected from the group consisting of capsules, tablets, powders, granules, liquids, pills, flakes, pastes, syrups, gels, beverages, jellies, and bars. In order to manufacture such dosage forms, conventional excipients, stabilizers, thickeners, etc. may be further included. In addition, food additives may be additionally included, and suitability as a "food additive" is determined by the specifications and standards for the relevant item in accordance with the general provisions and general test methods of the Food Additives Codex approved by the Ministry of Food and Drug Safety, unless otherwise specified.

[0031] In the anti-obesity composition of the present invention, the composition may be a food composition. If the composition according to the present invention is a food composition, it may be formulated as a food that can utilize the lactic acid bacteria cells as a main ingredient, and is preferably characterized by being one or more formulations selected from the group consisting of dairy products, beverages, sauces, jams, and confectionery.

[0032]

[0033] As described above, the anti-obesity composition comprising the killed cells powder including the killed cells of Lactobacillus plantarum VIOAP03 KCTC14498BP derived from Phellinus linteus according to the present invention and the killed cells of Lactobacillus fermentum VIMPP04 KCTC14499BP derived from Phellinus linteus as an active ingredient can prevent or improve obesity by inhibiting fat accumulation in adipocytes and inhibiting the expression of PPARγ, C / EBPα, SREB and FABP4 in preadipocytes.

[0040]

[0041] Hereinafter, the present invention will be described in more detail through examples. These examples are intended merely to illustrate the present invention, and therefore, the scope of the present invention is not to be construed as being limited by these examples.

[0042]

[0043] Example: Preparation of lactic acid bacteria cells derived from Phellodendron amurense

[0044] The production of lactic acid bacteria cells derived from Phellodendron amurense was carried out using a deposited strain isolated and identified by Bytech Co., Ltd.

[0045] After conducting flask seed cultures of the new strains L. Plantarum VIOAP03 KCTC14498BP and L. Fermentum VIMPP04 KCTC14499BP derived from Phellinus linteus, L. Plantarum VIOAP03 KCTC14498BP was cultured in a medium containing Phellinus linteus extract, and L. Fermentum VIMPP04 KCTC14499BP was cultured in a general glucose medium.

[0046] Specifically, in the first step, L. FermentumVIMPP04 KCTC14499BP was cultured in a flask for 18 hours, and then pre-cultured in a 50 L incubator containing general medium, followed by the addition of the pre-cultured culture in a 5,000 L incubator containing general medium to conduct the first main culture. After the first main culture was completed, a concentrate was prepared by centrifugation for 3 hours.

[0047] In the second step, L. PlantarumVIOAP03 KCTC14498BP was cultured in a flask separately from L. FermentumVIMPP04 KCTC14499BP for 18 hours, and then the second main culture was performed in a 700 L incubator containing a medium containing an extract derived from Phellinus linteus (origin) to prepare the culture.

[0048] In the third step, a mixed culture solution was prepared by mixing the concentrate prepared in the first step and the culture prepared in the second step in a 1:1 ratio.

[0049] Finally, in the fourth step, the mixed culture solution prepared in the third step was subjected to a tindile process, and after the tindile process was completed, a rapid freezing process was performed using a liquid nitrogen freezer (LNF), followed by freeze-drying to produce a powder of dead cells. The tindile process was performed three times, at a temperature of 80 to 110°C for 15 to 30 minutes.

[0050]

[0051] Comparative example: Preparation of lactic acid bacteria powder

[0052] According to the composition of Table 1 below, a lactic acid bacteria powder was manufactured using a commercially available strain rather than a strain isolated from a mushroom, and the manufacturing method was the same as in the examples.

[0053] Comparison of strains and media: commercialL. Plantarum+ medium containing Phellinus linteus extract and commercialL. Fermentum+ general medium

[0054]

[0055] Experimental Example 1: Confirmation of Effective Substances

[0056] The effective substances of the lactic acid bacteria killed powder manufactured in the example were confirmed.

[0057] Specifically, the lactic acid bacteria killed cell powder manufactured in the example was extracted at 60℃ for 4 hours with approximately 20 L of a 70% methanol aqueous solution. The extract was concentrated under reduced pressure to remove methanol, and the same amount of hexane was added to perform distribution extraction. The same amount of ethyl acetate was added to the aqueous layer again, distribution extraction was performed, and the activity assay was confirmed for each fraction.

[0058] In the case of the example, the hexane showing the activity was concentrated and subjected to silica gel column chromatography (elution solvent: chloroform-methanol = 100:1 → 1:1, v / v) to divide into five fractions (Sil-1, Sil-2, Sil-3, Sil-4, Sil-5) through normal phase TLC (elution solvent: chloroform-methanol = 20:1, v / v). Among these, the two fractions Sil-1 and Sil-4 showing activity were each concentrated under reduced pressure, and fractions were obtained by performing ODS-MPLC (elution solvent: 0→100% methanol aqueous solution), and then purified through preparative HPLC (ODS column Id 10 x 150 mm, 70% aq. acetonitrile, 3 mL / min) to obtain purified chemicals PL-1, PL-2, and PL-3.

[0059] The chemical structure of each chemical was determined by measuring and analyzing the NMR spectrum, and the chemical structures were confirmed to be linoleic acid for PL-1, trans-10-cis-12-octadecadienoic acid (10-trans,12-cis-CLA) for PL-2, and oleic acid for PL-3 (Figs. 1a to 1c).

[0060]

[0061] Experimental Example 2: Confirming the content of effective substances

[0062] The content ratio of the effective substance confirmed according to Experimental Example 1 was confirmed.

[0063] Specifically, a standard curve was drawn for each material standard and the HPLC peak area was input to confirm the results, and the results are shown in Table 2.

[0064] Type 10-trans, 12-cis-CLALinoleic acidOleic acidRatio 10.70.6

[0065] As a result, the content of 10-trans, 12-cis-CLA was the highest, followed by linoleic acid and oleic acid, and as shown in Table 2, when the content of 10-trans, 12-cis-CLA was set as 1, the ratios of linoleic acid and oleic acid were 0.7 and 0.6, respectively.

[0066]

[0067] Preparation example: Cell culture and test substance treatment

[0068] The body fat reduction effect was confirmed through in vitro testing. The cell line used in the test was the 3T3-L1 preadipocyte cell line, a cell line commonly used for evaluating body fat reduction functionality.

[0069] 1.5×10 3T3-L1 preadipocytes 5Cells were seeded per well in a 6-well plate and cultured until 90-100% confluent. Adipocyte differentiation was induced for 72 h with DMEM containing 10% Fetal Calf Serum (FCS) and 1% Penicillin / Streptomycin (P / S) with IDM (0.5 mM IBMX, 1 μM Dexamethasone, 1 μg / mL Insulin). Subsequently, the cells were cultured for 4-6 days with DMEM containing 10% FCS and 1% P / S, including 1 μg / mL insulin, as maturation medium. The maturation medium was changed once every 48 h, and the test substances were treated at each concentration and evaluated throughout the entire process of adipocyte differentiation.

[0070]

[0071] Experimental Example 3: Cytotoxicity Verification

[0072] 1.5×10 3T3-L1 preadipocytes 5 Cells / well were dispensed into 6-well plates and treated with test substances to determine cytotoxicity according to test substance treatment using a kit. When the control group's result was set to 100%, the results according to treatment with different test substance concentrations (1ug / mL, 5ug / mL, 10ug / mL, 15ug / mL, 30ug / mL, 60ug / mL) were determined, and the results are shown in Figure 2.

[0073] As a result, as can be confirmed in Fig. 2, there was no effect by the test substance up to a concentration of 10 ug / ml, but at a concentration of 15 ug / ml or higher, cytotoxicity was significantly reduced compared to the control group, indicating cytotoxicity. Based on these results, subsequent tests were conducted with the concentration of 10 ug / ml set as the highest concentration.

[0074]

[0075] Experimental Example 4: Confirmation of inhibition of adipocyte differentiation

[0076] The effectiveness of the dead cell powder manufactured through examples and comparative examples on adipocyte differentiation was evaluated. The effectiveness on fat accumulation was confirmed through an Oil-Red O staining test. Specifically, the 3T3-L1 preadipocytes prepared in the preparation example were fixed with 4% formaldehyde at room temperature for 1 hour and washed with 60% isopropanol. Then, the cells were stained with Oil-Red O reagent for 20 minutes, washed three times with distilled water, and the absorbance of the stained cells was measured.

[0077] The test group was a negative control group that did not treat anything to 3T3-L1 cells, which are preadipocytes, a control group that induced adipocyte differentiation, and a comparative example and an example that induced adipocyte differentiation were simultaneously treated to confirm the test results (Table 3). The treatment concentration was 10ug / ml, which is a concentration that does not show toxicity to preadipocytes.

[0078] Lipid accumulation Negative control Control group Preliminary comparative example Abs (OD at 450 nm) 0.12 ± 0.01 0.47 ± 0.01 0.25 ± 0.01 0.38 ± 0.04

[0079] As a result, as can be confirmed in Table 3, in the case of the example, the negative control group was found to inhibit adipocyte differentiation to a similar level as the fat stained with Oil Red O due to adipocyte differentiation, and the comparative example did not show a significant effect in inhibiting adipocyte differentiation.

[0080] From the results in Table 3 above, it was confirmed that the dead cell powder according to the present invention inhibits fat cell differentiation, and the effect of inhibiting fat cell differentiation according to the concentration of the dead cell powder was additionally confirmed, and the results are shown in Figures 3 and 4.

[0081] As a result, as can be confirmed from the microscopic observation results of Fig. 3, as the treatment concentration of the example increased, the staining of fat cells was clearly reduced, which shows that the lactic acid bacteria killed cell powder according to the present invention can inhibit fat cell induction. In addition, as can be confirmed from the fat accumulation results through absorbance measurement of Fig. 4, it was confirmed that the absorbance of the control group and the killed cell powder (Example) treatment group according to the present invention differed by 2 times, which shows that the killed cell powder according to the present invention has a remarkably excellent effect in inhibiting fat cell accumulation.

[0082]

[0083] Experimental Example 5: Identification of Biomarkers Related to Adipocyte Differentiation

[0084] The protein expression levels of PPARγ, C / EBPα, SREBP-1, and FABP4, which are biomarkers related to adipocyte differentiation, were evaluated using the Western-blot test. Briefly, 3T3-L1 cells, which are preadipocyte cells, were treated with a negative control group that was not treated with anything, a control group that induced adipocyte differentiation, and a comparative example and an example group while inducing adipocyte differentiation simultaneously. The cells were harvested, and proteins were extracted using protein lysis buffer, quantified, and Western-blot was performed. The quantitative values were derived from the intensities of the protein bands and are shown in Table 4.

[0085] Relative Protein Level Negative Control Control Preliminary Comparative Example PPARγ / β-actin 1.0 3.2 0.8 2.9 C / EBPα / β-actin 1.0 2.7 0.8 2.4 SREBP-1 / β-actin 1.0 2.3 0.9 1.9 FABP4 / β-actin 1.0 3.4 1.0 2.8

[0086] As a result, as can be confirmed in Table 4 above, the protein expression level in the signal transduction system related to adipocyte differentiation was increased by differentiation stimulation, whereas in the case of the example, the protein expression level was found to be decreased compared to the control group, and in the case of the comparative example, it was found to not be significantly involved in adipocyte differentiation.

[0087] From the results in Table 4 above, it was confirmed that the dead cell powder according to the present invention reduces the expression of the adipocyte differentiation biomarker, and the degree of expression of the adipocyte differentiation biomarker according to the concentration of the dead cell powder was additionally confirmed, and the results are shown in Fig. 5.

[0088] As a result, as can be confirmed in Fig. 5, it was confirmed that the expression of PPARγ, C / EBPα, SREBP-1, and FABP4 proteins was inhibited at all concentrations of the dead cell powder according to the present invention.
