Abstract
Description
Composition for improving skin swelling comprising nipa palm extract and fucoidan extract
[0001] The present invention relates to a composition for improving skin swelling comprising a nipa palm extract and a fucoidan extract, and more particularly, to a composition having an excellent effect in alleviating swelling by comprising a nipa palm extract and a fucoidan extract as active ingredients.
[0002] Skin swelling occurs when excessive water or salt accumulates in the skin. This reduces skin elasticity and hydration, contributing to wrinkles and aging. Severe swelling can lead to cracking, loss of elasticity and radiance, and increased wrinkles.
[0003] This skin swelling is known to be caused by various factors such as dehydration, excessive salt intake, stress, increased free radicals, lack of sleep, allergic reactions, and hormonal imbalance.
[0004] Various products, including functional foods and cosmetics, are being released to improve skin swelling, but most of them do not show a clear effect or cause skin irritation due to physical pressure or chemical components.
[0005] Meanwhile, as demand for products made with natural ingredients that are non-irritating to the human body and non-irritating to the skin grows, extensive research is being conducted to discover natural ingredients and enhance their efficacy. Furthermore, research is actively underway on mixtures combining different natural extracts, leveraging their functional properties. However, mixing extracts can lead to problems such as sedimentation, formulation instability, and reduced usability, preventing the desired effects from being fully realized.
[0006] Accordingly, the inventors of the present invention have continued their research to develop a composition effective in improving skin swelling by using a complex of natural extracts, and as a result, they have confirmed that a composition excellent in at least one of the effects of alleviating skin swelling, improving moisturizing, improving elasticity, and improving density can be prepared by including a nipa palm extract and a fucoidan extract as active ingredients, thereby completing the present invention.
[0007] The purpose of the present invention is to provide a composition having excellent effects of at least one of alleviating skin swelling, improving moisturizing, improving elasticity, and improving density by including a nipa palm extract and a fucoidan extract as active ingredients.

[0029] Unless otherwise defined, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which the present invention pertains. In general, the nomenclature used herein is well known and commonly used in the art. In addition, when describing embodiments of the present invention, if a detailed description of a related known structure or function is judged to hinder the understanding of the embodiments of the present invention, a detailed description thereof will be omitted. In addition, although embodiments of the present invention will be described below, the technical idea of the present invention is not limited or restricted thereto, and can be modified and implemented in various ways by those skilled in the art.
[0030] When a part in this specification is said to include a certain component, this does not exclude other components, unless otherwise specifically stated, but rather means that other components may be included. In this specification, the term "and / or" includes a combination of multiple related items or any one of multiple related items.
[0031]
[0032] According to an embodiment of the present invention, a composition for improving skin swelling comprising a nipa palm extract and a fucoidan extract is provided.
[0033] Nypa palm (Nypa fruticans) is the only species in the subfamily Nypa, a monotypic subfamily of the family Palmaceae. It is native to South Asia, Southeast Asia, and Oceania. Nypa palm, the young shoots of the palm tree, are extremely high in polyphenols, which have antioxidant, blood circulation-promoting, anti-inflammatory, and cancer-preventing properties. It is also rich in vitamin E and saponins, which help with cardiovascular disease, cataracts, and arthritis. It is also known to relieve constipation, detoxify, and help with hangovers.
[0034] Fucoidan is a type of sulfated polymer polysaccharide with a viscous structure that is contained in small amounts in brown algae such as wakame and kelp. It is a component that protects the algae from the flow of seawater and external stimuli, and acts as a protective film to defend against marine microorganisms.
[0035] In the present invention, "extract" refers to a substance obtained by extracting components from a natural product, and is a broad concept that includes, for example, a substance obtained by extracting a solvent-soluble component from a natural product using water or an organic solvent, a substance obtained by extracting only a specific component of a natural product, etc. The extract may include all extracts, fractions, dilutions, concentrates, or dried products thereof obtained at each stage such as extraction, fractionation, or purification.
[0036] In the present invention, the extraction method of the extract may utilize extraction methods commonly used in the art, such as hot water extraction, ethanol extraction, immersion extraction, cold extraction, reflux extraction, supercritical extraction, subcritical extraction, dissolution method, pressing method, high temperature extraction, high pressure extraction, or ultrasonic extraction. Specifically, hot water extraction or ethanol extraction may be used, but is not limited thereto.
[0037] According to an embodiment, the solvent used in the extract may be water, ethanol, or a mixture thereof, and specifically, purified water, ethanol, or alcohol. However, the present invention is not limited thereto, and various solvents applicable in the relevant technical field may be used alone or in combination. For example, as the extraction solvent, water (or distilled water); lower alcohols having 1 to 4 carbon atoms, such as methanol, ethanol, propyl alcohol, or butyl alcohol; polyhydric alcohols, such as propanediol, butanediol, hexanediol, methylpropanediol, glycerin, butylene glycol, propylene glycol, pentylene glycol, or dipropylene glycol; and hydrocarbon solvents, such as methyl acetate, ethyl acetate, acetone, benzene, hexane, diethyl ether, or dichloromethane; or mixtures thereof may be used.
[0038] In an embodiment, the nipa palm extract may be water extracted or alcohol extracted.
[0039] In an embodiment, the fucoidan extract may be hot water extracted.
[0040] Hot water extraction is a method of extracting effective substances using hot water at 60 to 140°C. Since pure water is used as the extraction medium, it has the advantage of not causing problems due to the toxicity of the organic solvent used during extraction.
[0041] Depending on the embodiment, the extract may be in liquid form or in dry powder form. In this case, various drying methods commonly used in the art, such as freeze-drying, vacuum drying, hot air drying, and spray drying, may be applied.
[0042] Depending on the embodiment, the extract may be manufactured or processed in various ways, such as by distillation under reduced pressure or concentration.
[0043]
[0044] The composition of the present invention comprises both a nipa palm extract and a fucoidan extract, and has at least one or more of the effects of improving skin swelling, specifically reducing swelling, improving moisturizing, improving elasticity, and improving density. These effects can be further enhanced when the nipa palm extract and fucoidan extract are combined, compared to when the nipa palm extract and fucoidan extract are used alone.
[0045] The composition of the present invention may include a nipa palm extract and a fucoidan extract in a weight ratio of 1:0.5 to 2. When the nipa palm extract and the fucoidan extract are included in the above-mentioned mixing ratio, the effects of reducing swelling, improving moisturizing, improving elasticity, or improving density may be excellent.
[0046] In an embodiment, the nipa palm extract and fucoidan extract may be mixed in a weight ratio of 1:1. In this case, the effects of reducing swelling, improving moisturizing, improving elasticity, or improving density may be maximized. However, this is not limited thereto.
[0047] In an embodiment, the nipa palm extract and fucoidan extract may be included in an amount of 40 to 70 wt% based on the total weight of the composition. When the nipa palm extract and fucoidan extract are included in the above amounts, an excellent effect of improving skin swelling may be exhibited. However, the present invention is not limited thereto.
[0048] In some embodiments, the composition may exhibit at least one of the following effects: alleviating swelling, improving hydration, improving elasticity, and improving density. Specifically, the composition of the present invention can alleviate skin swelling by improving hydration within the skin, increasing elasticity and density, and suppressing factors that cause skin swelling.
[0049] In an embodiment, a food composition comprising the composition is provided.
[0050] In this specification, "food" refers to a food manufactured and processed using raw materials or ingredients that have functionality useful to the human body, and means a food with high medical or healthcare effects that has been processed to efficiently exhibit a bioregulatory function in addition to providing nutrition, and may be used interchangeably with terms known in the art such as functional food. For example, it may be manufactured as a health functional food, health supplement, special nutritional supplement food, functional beverage, etc., or the composition according to the present invention may be added to a natural food, processed food, etc. The food may be manufactured in the form of a powder, granule, tablet, capsule, syrup, or beverage, and there is no limitation on the form the food may take, and may include all foods in the conventional sense.
[0051] For example, this includes beverages and various drinks, fruits and their processed foods (canned fruits, jams, etc.), fish, meat and their processed foods (ham, bacon, etc.), bread and noodles, cookies and snacks, dairy products (butter, cheese, etc.), and can include all functional foods in the conventional sense. Furthermore, it can also include foods used as animal feed.
[0052] The food composition according to the present invention may further comprise food additives and other suitable auxiliary ingredients commonly used in the art. For example, the food composition may further comprise flavoring agents, natural carbohydrates, sweeteners, vitamins, electrolytes, colorants, pectic acid, alginic acid, organic acids, protective colloid thickeners, pH regulators, stabilizers, preservatives, glycerin, alcohol, carbonating agents, and the like.
[0053] For example, as a flavoring agent, natural flavoring agents including thaumatin, stevia extract, etc., or synthetic flavoring agents including saccharin, aspartame, etc. can be used; natural carbohydrates can be monosaccharides such as glucose and fructose, disaccharides including maltose and sucrose, polysaccharides including dextrin and cyclodextrin, and sugar alcohols including xylitol, sorbitol, erythritol, etc.; and as a sweetening agent, natural sweeteners such as thaumatin, stevia extract, or synthetic sweeteners such as saccharin, aspartame, etc. can be used. It is preferable that the food additives and other auxiliary components, etc. are added and used in a trace amount as long as the purpose for which they are added can be achieved. The trace amount, when expressed numerically, may be in the range of 0.0005 wt% to about 0.5 wt% based on the total weight of the food composition. Those skilled in the art can easily select the blending amount of the above components within a range that does not impair the purpose and effect of the present invention.
[0054]
[0055] In an embodiment, a cosmetic composition comprising the composition is provided. The mixing amount of the above ingredients can be easily selected by those skilled in the art within a range that does not impair the purpose and effects of the present invention.
[0060]
[0061] Hereinafter, examples and experimental examples are presented to explain the present invention more specifically, but the present invention is not limited thereto.
[0062]
[0063] Preparation of Nipa Palm Water Extract
[0064] Nipa palm was extracted by boiling water at 95±2℃ for 6 hours by adding 10 times the amount of distilled water per gram of nipa palm. The solid content of the extract obtained by boiling water extraction was removed and concentrated under reduced pressure. After concentration, it was mixed with dextrin in a 7:3 ratio and spray dried to obtain a nipa palm hot water extract.
[0065]
[0066] Preparation of nipa palm alcohol extract
[0067] 50% alcohol 10 times the volume per g of nipa palm was used, and the extraction temperature was set to 70℃. The first extraction was performed for 4 hours with a solvent volume 10 times (v / w) of the sample, and the second extraction was performed for 2 hours with a solvent volume 10 times (v / w) of the sample. After the extraction was completed, the extract from which the solids were removed was concentrated using a vacuum evaporator, and then freeze-dried to obtain a nipa palm alcohol extract.
[0068]
[0069] Preparation of fucoidan hot water extract
[0070] Dried seaweed was extracted with purified water, and the extract was concentrated after solid-liquid separation to prepare a first concentrate. After removing alginate from the concentrate, a second concentration was performed. Then, in the detailed process of polysaccharide separation, alcohol was added and immersed, and the sediment was purified. After the purification was completed, it was dissolved in water and freeze-dried to prepare a fucoidan hot water extract.
[0071]
[0072] Experimental Example 1. Measurement of antioxidant activity (DPPH radical scavenging assay)
[0073] Compositions containing only the above-prepared nipa palm hot water extract, nipa palm alcohol extract or fucoidan hot water extract were used in Manufacturing Examples 1 to 3, and compositions mixed in the ratio shown in [Table 1] below were used in Manufacturing Examples 4 to 9. Meanwhile, vitamin C was used in Comparative Example 1.
[0074]
[0075]
[0076] To measure the antioxidant capacity of the manufacturing examples and comparative examples, a DPPH radical scavenging assay was performed. DPPH powder was dissolved in ethanol to make a 0.5 mM DPPH solution, and 100 μL of each of the manufacturing examples and comparative examples were added to a 96-well plate. 100 μL of 99% ethanol was added to the DPPH non-reaction group, and 100 μL of 0.5 mM DPPH was added to the DPPH reaction group, and the absorbance was measured at 540 nm.
[0077] Figure 1 is a diagram showing the antioxidant effect (radical scavenging ability) of manufacturing examples and comparative examples. As shown in Figure 1, the antioxidant effect was confirmed in all manufacturing examples, and in particular, it can be confirmed that the antioxidant effect was superior to that of manufacturing examples 1 to 3, which contained only nipa palm extract or fucoidan extract, in manufacturing examples 4 to 9, which mixed the extracts. Furthermore, manufacturing examples 5 and 8, which mixed nipa palm extract and fucoidan extract in a 1:1 ratio, showed superior antioxidant effects compared to the other manufacturing examples.
[0078]
[0079] Experimental Example 2. Measurement of Skin Swelling
[0080] A clinical trial was conducted using Manufacturing Examples 5 and 8, which showed the best antioxidant effect.
[0081] Adult men and women aged 20 to 60 (final effective evaluation number: 61, mean age: 43.541 years, 6 men, 55 women) were randomly divided into a placebo control group and an experimental group, and a skin swelling measurement test was performed. The experimental group and the placebo control group took 1000 mg tablets containing the ingredients in Table 2 below twice a day (morning and evening) for 2 weeks.
[0082] The E-001 administration group refers to an experimental group that took a composition in which a nipa palm hot water extract and a fucoidan hot water extract were mixed in a 1:1 ratio (Manufacturing Example 5), and the E-002 administration group refers to an experimental group that took a composition in which a nipa palm ethanol extract and a fucoidan hot water extract were mixed in a 1:1 ratio (Manufacturing Example 8). The placebo control group took placebo tablets of the same shape as the experimental group. Unlike the nipa palm ethanol extract, dextrin is added to the nipa palm hot water extract during the spray drying process. Therefore, by using the nipa palm extract of Manufacturing Example 5 in an amount of 35.7143 wt% relative to the total weight of the composition, the mixing ratio of the active ingredients was made 1:1 as in Manufacturing Example 8.
[0083]
[0084]
[0085] The degree of improvement in the subjects' swelling was measured before and after starting to take the drug.
[0086] Specifically, skin swelling was measured by using a 3D skin imaging device, Antera 3D CS (Miravex Ltd., Ireland), to take pictures of the upper eyelids, cheeks, and backs of both hands in the placebo control group, E-001 group, and E-002 group before and after administration. The saved images (Elevation - Custom mode) were analyzed to determine the skin volume (mm³) within a specific range. A decrease in the value indicates a greater effect in improving swelling.
[0087] Skin swelling of the calves, ankles, and dorsum of the feet was measured by analyzing the volume (cc) values from the saved images after taking pictures of both calves, both ankles, and both dorsum of the feet in the placebo control group, E-001 group, and E-002 group before and after taking the drug using Vectra XT / Vectra H2 (Canfield Imaging Systems, USA), which can measure skin volume with 3D images. A decrease in the value indicates a greater effect in reducing swelling.
[0088] The measurement results and specific data are shown in [Table 3] below.
[0089]
[0090]
[0091] Based on the above data, the results of improvement in skin swelling of both cheeks, both calves, and both ankles before and 2 weeks after taking the placebo control group, E-001, and E-002 groups are shown in graph form in Fig. 2. In addition, in order to visually confirm the swelling improvement effect, images taken before and after taking the medication are shown in Fig. 3. Specifically, in Fig. 3(a), (a) is an image of the left eyelid before and after taking the medication, (b) is an image of the left cheek before and after taking the medication, (c) is an image of the left back of the hand before and after taking the medication, and in Fig. 3(b), (d) is an image of the calf before and after taking the medication, and (e) is an image of the ankle and back of the foot before and after taking the medication.
[0092] Referring to Table 3, Figures 2 and 3, the placebo control group showed no statistically significant difference after 2 weeks of administration compared to before administration, but the E-001 and E-002 groups showed a statistically significant decrease after 2 weeks of administration compared to before administration. In other words, it was confirmed that the E-001 and E-002 groups showed a statistically significant difference compared to the placebo control group. In addition, in the captured images, it was confirmed that the placebo control group showed no change, whereas the E-001 and E-002 groups showed a significant decrease in swelling before and after administration.
[0093] Therefore, it can be seen that the composition of the present invention is effective in improving swelling throughout the skin, including the eyelids, cheeks, back of the hand, calves, ankles, and back of the foot.
[0094]
[0095] Experimental Example 3. Measuring Skin Moisture
[0096] The moisture content in the skin of the subjects of Experimental Example 2 above was measured.
[0097] The amount of moisture in the skin was measured using MoistureMeter D Compact (Delfin Technologies Ltd, Finland) in the placebo control group, E-001 group, and E-002 group before and after administration of the drug. The parameter is the tissue dielectric constant (TDC) value, the unit is %, and the measured value and the amount of moisture are proportional.
[0098] As a result of the measurement, specific data are shown in [Table 4] below.
[0099]
[0100]
[0101] Based on the above data, Fig. 4 shows the results of measuring the moisture content in the skin of both cheeks and the backs of both hands before and 2 weeks after taking the placebo control group, E-001, and E-002 groups in a graph format.
[0102] Referring to Table 4 and Figure 4, the placebo control group showed no statistically significant difference after 2 weeks of administration compared to before administration, but the E-001 and E-002 groups showed a statistically significant decrease after 2 weeks of administration compared to before administration. In other words, it can be confirmed that the E-001 and E-002 groups showed a statistically significant difference compared to the placebo control group.
[0103] Therefore, it can be seen that the composition of the present invention is effective in improving swelling by reducing the amount of moisture in the skin that causes swelling.
[0104]
[0105] Experimental Example 4. Measurement of Skin Elasticity
[0106] Skin elasticity was measured for the subjects of Experimental Example 2 above.
[0107] The skin elasticity of the eyelids, cheeks, and backs of the hands was measured using Cutometer MPA580 (Courage+ Khazaka electronic GmbH, Germany) in the placebo control group, E-001 group, and E-002 group before and after administration. The measurements were performed three times, and the average value was used as the skin elasticity evaluation data. The R2 value, which is the skin's re-deformation force, is the overall elasticity. The closer the value is to 1, the more elastic it is, and the measured value and skin elasticity are proportional.
[0108] Skin elasticity of the calves, ankles, and dorsum of the feet was measured using a Ballistometer BLS780 (DIA STRON, UK) in the placebo control group, E-001 group, and E-002 group before and after administration. Measurements were performed three times, and the average value was used as the skin elasticity evaluation data. The measurement parameter is CoR, and the measured value is proportional to skin elasticity.
[0109] The measurement results and specific data are shown in [Table 5] below. Figure 5 is a graph showing the results of improvement in skin elasticity on both cheeks, the backs of both hands, and both calves in the placebo control group, E-001, and E-002 groups before and two weeks after taking the drug.
[0110]
[0111]
[0112] Referring to Table 5 and Figure 5, the placebo control group showed no statistically significant difference after 2 weeks of administration compared to before administration, but the E-001 and E-002 groups showed a statistically significant increase after 2 weeks of administration compared to before administration. In other words, it can be confirmed that the E-001 and E-002 groups showed a statistically significant difference compared to the placebo control group.
[0113] Therefore, it can be seen that the composition of the present invention is effective in improving skin elasticity.
[0114]
[0115]
[0116] Experimental Example 5. Measurement of Skin Density
[0117] Skin density was measured for the subjects of Experimental Example 2 above.
[0118] Skin density was measured using a Skin Scanner DUB-USB (TPM taberna pro medicum, Germany). Scans were taken of the subjects' cheeks, the backs of both hands, the calves, and the tops of both feet before and after administration. The parameter is Density, expressed in %, and the measured value is proportional to skin density.
[0119] The measurement results and specific data are shown in [Table 6] below. Fig. 6 is a graph showing the results of improvement in skin density on both cheeks, the backs of both hands, and both calves in the placebo control group, E-001, and E-002 administration groups before and 2 weeks after administration, and Fig. 7 is a photographic image showing the measurement results, in which (a) is an image of the left cheek before and after administration, and (b) is an image of the right calf before and after administration.
[0120]
[0121]
[0122] Referring to Table 6, Figures 6 and 7, the placebo control group showed no statistically significant difference 2 weeks after administration compared to before administration, but the E-001 and E-002 groups showed a statistically significant increase 2 weeks after administration compared to before administration. In other words, it can be confirmed that the E-001 and E-002 groups showed a statistically significant difference 2 weeks after administration compared to before administration compared to the placebo control group. In addition, in the photographed images, it was confirmed that the skin density of the E-001 and E-002 groups before and after administration was significantly improved, while the placebo control group showed no change.
[0123] Therefore, it can be seen that the composition of the present invention is effective in improving skin density.
[0124]
