[0002] In recent years, many allergic diseases such as atopic dermatitis, bronchial asthma, and allergic rhinitis have become a problem. The main causes of these allergic diseases are mites that live in homes, particularly allergens (Dermatophagoides pteronyssinus, Dermatophagoides typhimurium) that are abundant in house dust, and cedar pollen allergens (Cryj1, Cryj2) that are airborne in large quantities mainly in spring.
[0003] Therefore, there is a need for a technology that can remove allergens from living spaces or inactivate them by denaturing them.
[0004] Patent Document 1 discloses a cellulosic fiber having allergen treatment ability, characterized in that it is loaded with an allergen treatment agent containing, as an active ingredient, a nitrogen-containing compound having a specific chemical structural formula or a solvate thereof.

[0068] Here, the allergen reducing agent refers to an agent having an allergen reducing effect. The allergen reducing effect can be determined by measuring the allergen reduction rate for cedar pollen allergen (Cryj1) or mite allergen (Derf1) as described below, and if the allergen reduction rate for at least either allergen is 60% or more, the agent can be determined to have an allergen reducing effect.
[0069] The allergen reduction rate of an allergen reducing agent can be measured as follows: 6 parts by mass of an allergen reducing agent is mixed with 94 parts by mass of an ultraviolet-curable acrylic paint to prepare an allergen-reducing paint.
[0070] Next, a polyester film is prepared as a substrate. One side of this substrate is coated with an allergen-reducing paint to a thickness of 18 μm, and then ultraviolet light with a wavelength of 365 nm is irradiated onto the coating layer at 25° C. with an integrated light intensity of 500 mJ / cm. 2 The ultraviolet-curable acrylic paint was cured by irradiating the paint so that an allergen-reduced substrate having a coating film with a thickness of 18 μm formed on one side was produced.
[0071] An allergen solution containing 10 μg / mL of allergen is prepared by dissolving the allergen in purified water, and then phosphate-buffered saline (pH 7.4, hereinafter referred to as "PBS-T") containing 0.05 w / v% polysorbate 20 is added to the allergen solution and mixed uniformly to prepare an allergen solution containing 15 ng / mL of allergen.
[0072] The cedar pollen allergen (Cryj1) may be, for example, a product commercially available from ITEA under the trade name "Cedar Pollen Extract, Code 10103." The mite allergen (Derf1) may be, for example, a product commercially available from ITEA under the trade name "Mite Allergen Extract, Code 10102."
[0073] The allergen-reduced substrate was cut into a flat square with sides of 5 cm, and 0.4 mL of the allergen solution was dropped onto the coating. The substrate was then covered with a flat square polyethylene film with sides of 4 cm and left at 25°C for 24 hours to prepare a test solution.
[0074] Next, the amount W1 (ng / mL) of cedar pollen allergen (Cryj1) present in the test solution is measured by ELISA using an allergen measurement kit.
[0075] A blank substrate with a coating film formed on one side was prepared in the same manner as above, except that a blank paint containing only an ultraviolet-curable acrylic paint was used instead of the allergen-reduced paint. The amount of allergen present in the test solution, W0 (ng / mL), was measured in the same manner as above, except that a blank substrate was used instead of the allergen-reduced substrate. The allergen reduction rate (%) was calculated using the following formula: Allergen reduction rate (%) = 100 - (W1 / W0) x 100
[0076] The allergen reduction rate of the allergen reducing agent is preferably 70% or more, more preferably 80% or more, more preferably 85% or more, more preferably 90% or more, and even more preferably 95% or more.
[0077] The antibody effect of the allergen reducing agent is preferably 12% or less, more preferably 10% or less, and even more preferably 9% or less.
[0078] The antibody effect of an allergen-reducing agent can be measured as follows. An allergen-reduced substrate is prepared in the same manner as when measuring the allergen reduction rate. 0.4 mL of PBS-T is dropped onto the coating film of the allergen-reduced substrate, and the substrate is then covered with a flat square polyethylene film measuring 4 cm on a side, and left to stand at 25°C for 24 hours to prepare a test solution.
[0079] 100 μL of the test solution is dropped into wells of an antibody-immobilized 96-well plate included in the allergen measurement kit, and left to stand for 1 hour at 25° C. Next, the wells are washed three times with PBS-T using a plate washer.
[0080] Next, ELISA measurement is performed using the above wells to measure the amount of allergen present in the allergen solution containing 15 ng / mL of allergen, W2 (ng / mL). The effect on antibodies (%) is calculated using the following formula: Effect on antibodies (%) = 100 - (W2 / 15) x 100
[0081] [Allergen-reducing agent] The allergen-reducing agent contains magnesium oxide as an active ingredient. The method for producing the allergen-reducing agent is not particularly limited, and the allergen-reducing agent can be produced by mixing magnesium oxide, the active ingredient, with compounds to be added as needed in a commonly used manner.
[0082] Next, the usage of the allergen denaturing agent will be described. The allergen denaturing agent exerts an allergen denaturing effect on various allergens by the action of magnesium oxide. The allergen denaturing agent exerts an allergen denaturing effect on various allergens by coming into contact with the allergens.
[0083] Allergens that can be targeted by allergen reducing agents include animal allergens such as house dust mite allergens (Der1, Der2) and allergens caused by dogs and cats (Can f1, Fel d1), as well as airborne cedar pollen allergens (Cryj1, Cryj2) and plant allergens such as pollen. However, it is particularly effective against house dust mites, which are abundant in house dust, especially in bedding, and cause allergic diseases.
[0084] The allergen-reducing agent is used, for example, by being incorporated into a substrate to which an allergen-reducing effect is to be imparted, thereby constituting an allergen-reduced product. The substrate containing the allergen-reducing agent exhibits an allergen-reducing effect as an allergen-reduced product. The manner in which the allergen-reducing agent is incorporated into the substrate is not particularly limited, and examples include (1) adhering the allergen-reducing agent to the surface of the substrate, and (2) kneading the allergen-reducing agent into the substrate, thereby incorporating the allergen-reducing agent into the substrate.
[0085] For allergen-reduced products, the content of magnesium oxide constituting the allergen-reducing agent is preferably 0.1 parts by mass or more, more preferably 0.5 parts by mass or more, and more preferably 1 part by mass or more, per 100 parts by mass of the substrate. [0086] The allergen-reducing agent can be attached to the surface of a substrate by dissolving or dispersing the allergen-reducing agent in a solvent to form an allergen-reducing solution, and then applying the allergen-reducing solution to the substrate. The allergen-reducing solution may contain additives such as water-soluble solvents, oils, emulsions, and suspensions, as needed.
[0087] Examples of the solvent include water (preferably ion-exchanged water), alcohols (methyl alcohol, ethyl alcohol, propyl alcohol, etc.), hydrocarbons (toluene, xylene, methylnaphthalene, kerosene, cyclohexane, etc.), ethers (diethyl ether, tetrahydrofuran, dioxane, etc.), ketones (acetone, methyl ethyl ketone, etc.), and amides (N,N-dimethylformamide, etc.).

[0097] The paint contains a binder and a solvent. The binder and solvent may be the same as those described above. The paint may contain additives such as pigments, plasticizers, curing agents, extenders, fillers, antioxidants, thickeners, and surfactants, as long as the additives do not impair the paint's physical properties. Examples of methods for preparing an allergen-reducing paint by incorporating an allergen-reducing agent into the paint include a method in which the allergen-reducing agent and the paint are supplied to a dispersing device and mixed uniformly. Examples of dispersing devices include a high-speed mill, a ball mill, and a sand mill.

[0107] (Examples 1 to 12 and Comparative Examples 1 to 5) Magnesium oxide or magnesium hydroxide of the type shown in Table 1 was used as the allergen denaturing agent.
[0108] Examples 13 to 15 100 parts by mass of magnesium oxide of the type shown in Table 3 and 50 parts by mass of a sulfonate compound or 50 parts by mass of a sulfate ester salt compound of the type shown in Table 3 were prepared. 100 parts by mass of magnesium oxide and 50 parts by mass of the sulfonate compound or 50 parts by mass of the sulfate ester salt compound were supplied to 1,050 parts by mass of pure water and uniformly mixed to prepare a dispersion. The dispersion was then powdered using a spray dryer at an atomizer rotation speed of 20,000 rpm, to allow the sulfonate compound or sulfate ester salt compound to adhere (support) to the surface of the magnesium oxide. The magnesium oxide was then pulverized using a jet mill (manufactured by Nisshin Engineering Inc., trade name "SJ-500") under operating conditions of a raw material supply rate of 1 kg / h and a compressed air pressure of 0.75 MPa, yielding magnesium oxide as an allergen reducing agent having the entire amount of the sulfonate compound or sulfate ester salt compound supported (adhered) on its surface.
[0109] The allergen reduction rate and the effect on antibodies of the allergen reducing agents of the Examples and Comparative Examples were measured in the following manner, and the results are shown in Tables 1 and 3.
[0110] [Anti-allergen test (coating film)] [Measurement of allergen reduction rate of cedar pollen allergen (Cryj1) (coating film)] An allergen-reducing coating was prepared by mixing 6 parts by mass of the allergen-reducing agent obtained in the Examples and Comparative Examples with 94 parts by mass of an ultraviolet-curable acrylic coating (manufactured by Coattec Co., Ltd., product name "AI-N2"). Next, a polyester film was prepared as a substrate. One side of this substrate was coated with the allergen-reducing coating to a thickness of 18 μm using a wire bar coater #8, and then ultraviolet light with a wavelength of 365 nm was applied to the coating layer at 25°C using a UV conveyor device (manufactured by iGraphics Co., Ltd., "ECS301G1") at an integrated light dose of 500 mJ / cm. 2 The ultraviolet-curable acrylic paint was cured by irradiating the paint so that an allergen-reduced substrate having a coating film of 18 μm in thickness formed on one side thereof was produced.
[0111] A freeze-dried powder of cedar pollen allergen (Cryj1) (ITEA, product name "Cedar Pollen Extract, Code 10103") was dissolved in purified water to prepare an aqueous allergen solution containing 10 μg / mL of cedar pollen allergen (Cryj1). PBS-T (phosphate buffer containing 0.05% by mass of Tween 20 (Tokyo Chemical Industry Co., Ltd.), pH 7.4) was then added to the aqueous allergen solution and mixed uniformly to prepare an allergen solution containing 15 ng / mL of cedar pollen allergen (Cryj1).
[0112] The allergen-reduced substrate was cut into a flat square with sides of 5 cm, and 0.4 mL of the allergen solution was dropped onto the coating. The substrate was then covered with a flat square polyethylene film with sides of 4 cm and left at 25°C for 24 hours to prepare a test solution.
[0113] Next, the amount W1 (ng / mL) of cedar pollen allergen (Cryj1) present in the test solution was measured by ELISA using a cedar pollen allergen measurement kit (ITEA, product name "Cedar Pollen Allergen (Cryj1) ELISA Kit, code 10204").
[0114] A blank substrate with a coating film formed on one side was prepared in the same manner as above, except that a blank paint containing only an ultraviolet-curable acrylic paint was used instead of the allergen-reduced paint. The amount of cedar pollen allergen (Cryj1) present in the test solution, W0 (ng / mL), was measured in the same manner as above, except that a blank substrate was used instead of the allergen-reduced substrate. The allergen reduction rate (%) was calculated based on the following formula. This is shown in the "Reduction rate (%)" column for "Pollen (Cryj1) (coating film)" in Tables 1 and 3. Allergen reduction rate (%) = 100 - (W1 / W0) x 100
[0115] [Measurement of the effect of cedar pollen allergen (Cryj1) on antibodies (coating film)] An allergen-reduced substrate was prepared in the same manner as in the measurement of the allergen reduction rate of cedar pollen allergen (Cryj1). 0.4 mL of PBS-T (phosphate buffer containing 0.05% by mass of Tween 20 (Tokyo Chemical Industry Co., Ltd.), pH 7.4) was dropped onto the coating film of the allergen-reduced substrate and brought into contact with the substrate. The substrate was then covered with a flat square polyethylene film measuring 4 cm on a side and left to stand at 25°C for 24 hours to prepare a test solution.
[0116] 100 μL of the test solution was dropped into wells of an antibody-immobilized 96-well plate included in the cedar pollen allergen measurement kit (ITEA, trade name "Cedar Pollen Allergen (Cryj1) ELISA Kit, code 10204") and allowed to stand for 1 hour at 25° C. Next, the wells were washed three times with PBS-T using a plate washer (Tecan, trade name "HydroFlexPlus").
[0117] Next, ELISA was performed using the above wells to measure the amount W2 (ng / mL) of cedar pollen allergen (Cryj1) present in the allergen solution containing 15 ng / mL of cedar pollen allergen (Cryj1). The effect (%) on antibodies was calculated based on the following formula and is shown in the "Effect (%) on antibodies" column of "Pollen (Cryj1) (coating film)" in Tables 1 and 3. Effect (%) on antibodies = 100 - (W2 / 15) x 100
[0118] [Measurement of reduction rate of mite allergen (Derf1) (coating film) and effect on antibody (coating film)] Instead of freeze-dried powder of cedar pollen allergen (Cryj1), freeze-dried powder of mite allergen (Derf1) (ITEA, product name "mite (Df) culture medium extract, code 10102") was used, and instead of cedar pollen allergen measurement kit (ITEA, product name "cedar pollen allergen (Cryj1) ELISA kit, code 10204"), a mite allergen measurement kit (ITEA, product name "cedar pollen allergen (Cryj1) ELISA kit, code 10204") was used. The allergen reduction rate (%) of mite allergen (Derf1) and the effect on antibodies (%) were calculated in the same manner as for measuring the reduction rate (%) of cedar pollen allergen (Cryj1) and the effect on antibodies (%), except that a kit (trade name "mite allergen (Derf1) ELISA kit, code 10205") was used, and the results are shown in the "Reduction rate (%)" and "Effect on antibodies (%)" columns for "mite (Derf1) (coating film)" in Tables 1 and 3, respectively.
[0119] [Anti-allergen test (fiber)] [Measurement of allergen reduction rate of cedar pollen allergen (Cryj1) (fiber)] Purified water was added to 1 g of the allergen reducing agent of Examples 6 to 8, 10 to 12 and Comparative Example 4 and mixed uniformly to prepare an allergen reducing aqueous solution containing 1% by mass of the allergen reducing agent.
[0120] Next, as a substrate, polyester fiber (manufactured by Shikisen Co., Ltd., product name "Polyester Tropical Toray", basis weight 120 g / m 2 The fibers were immersed in 100 g of the allergen-reducing aqueous solution for 2 minutes. The immersed polyester fibers were squeezed with a manual mangle and dried at 120°C for 10 minutes to produce allergen-reducing fibers in which the allergen-reducing agent was fixed to the polyester fibers. In the allergen-reducing fibers, the allergen-reducing agent was fixed at 1 g / m 2 It was contained.
[0121] A freeze-dried powder of cedar pollen allergen (Cryj1) (ITEA, product name "Cedar Pollen Extract, Code 10103") was dissolved in purified water to prepare an aqueous allergen solution containing 10 μg / mL of cedar pollen allergen (Cryj1).
[0122] 0.4 g of the allergen-reduced fiber was placed in a zippered plastic bag, and 1 mL of the above allergen solution was dropped into it. The bag was then sealed and left to stand at 25° C. for 2 hours to prepare a test solution.
[0123] Next, the amount W1 (ng / mL) of cedar pollen allergen (Cryj1) present in the test solution was measured by ELISA using a cedar pollen allergen measurement kit (ITEA, product name "Cedar Pollen Allergen (Cryj1) ELISA Kit, code 10204").
[0124] The amount of cedar pollen allergen present in the test solution, W0 (ng / mL), was measured in the same manner as above, except that no allergen-reduced fiber was placed in the zippered plastic bag. The allergen reduction rate (%) was calculated using the following formula and is shown in the "Reduction rate (%)" column for "Pollen (Cryj1) (fiber)" in Table 2. Allergen reduction rate (%) = 100 - (W1 / W0) x 100
[0125] [Measurement of the effect of cedar pollen allergen (Cryj1) on antibodies (fiber)] Allergen-reduced fibers were prepared in the same manner as in the measurement of the allergen reduction rate for cedar pollen allergen (Cryj1). 0.4 g of the allergen-reduced fiber was placed in a zippered plastic bag, and 1 mL of PBS-T (phosphate buffer containing 0.05% by mass of Tween 20 (manufactured by Tokyo Chemical Industry Co., Ltd.), pH 7.4) was added dropwise to bring the fiber into contact with the bag. The bag was then sealed and left to stand at 25°C for 30 seconds to prepare a test solution.
[0126] 100 μL of the test solution was dropped into wells of an antibody-immobilized 96-well plate included in the cedar pollen allergen measurement kit (ITEA, trade name "Cedar Pollen Allergen (Cryj1) ELISA Kit, code 10204") and allowed to stand for 1 hour at 25° C. Next, the wells were washed three times with PBS-T using a plate washer (Tecan, trade name "HydroFlexPlus").
[0127] Next, ELISA was performed using the above wells to measure the amount W2 (ng / mL) of cedar pollen allergen (Cryj1) present in the allergen solution containing 15 ng / mL of cedar pollen allergen (Cryj1). The effect (%) on antibodies was calculated based on the following formula and recorded in the "Effect (%) on antibodies" column of "Pollen (Cryj1) (fiber)" in Table 2. It can be said that the smaller the effect (%) on antibodies, the less the allergen reducer's effect on antibodies. Effect (%) on antibodies = 100 - (W2 / 15) x 100
