Abstract
Description
[0001] IMINOAMIDE-TYPE COMPOUNDS AS LIGANDS FOR COPPER-CATALYZED COUPLING REACTIONS OF (HETERO)ARYL HALIDES
[0002] The present invention relates to novel iminoamide-type compounds of the general formula (I) and use thereof as ligands in copper-catalyzed coupling reactions of (hetero)aryl halides forming C-C, C-N, C-S, C-O, C-P-bonds and other bonds, in particular the use of the compounds of formula (I) as ligand in copper- catalyzed coupling reactions of aryl halides forming C-O-bonds.
[0003] Copper-catalyzed hydroxylation reactions of (hetero)aryl halides to form phenols are known in the literature and can be achieved by reacting (hetero)aryl halides (X = I, Br, Cl) with overstochiometric amount of alkali hydroxide (LiOH, NaOH, KOH, CsOH) in the presence of a copper salt and various different ligand classes (Ma D. et al. Org. Proc. Res. Dev. 2022, 26, 1690-1750). Known ligands for this type of transformation can be categorized into first-generation bidentate ligands (Ma et al., Org. Process Res. Dev. 2022, 26, 1690- 1750) such as N.O-bidcntatc ligands (e.g. 8 -hydroxy quinoline (Ma et al., Synlett 2010 (6), 976-978), Li-Proline (Zhou et al., RSC Adv. Comm. 2013, 3, 22837-22840), DMEDA (Leadbeater et al., Cat. Comm. Vol 12(1) 2010, 64-66)), O,O-bidentate ligands (e.g. TMHD or 2, 2,6,6- tetramethyl-3,5-heptanedione (Taillefer et al., Angew. Chem. 2009, 121, 8881-8884), D-glucose (Sekar et al., Chem Comm. 2011, 47, 6692-6694), sucrose (Takita et al., Org. Biomol. Chem. 2020, 18, 7827- 7831), glycolic acid (Chae, J. Org. Chem. 2013, 78, 5804-5809), 8 -hydroxy quinolin-N-oxide (Jiang et al, Org. Lett. 2011, 13, 16, 4340-4343)), W-bidcntatc ligands (e.g. phenanthroline (You et al., Angew. Chem. Int. Ed. 2009, 48, 8729-8732), pyridine-2-aldoxime (Fu et al., Chem. Eur. J. 2010, 16, 2366-2370), 5-bromo-2-(lH-imidazol-2-yl)pyridine (Ma et al., Tet. Lett. 52 (2011) 5593-5595)). The second- generation of bidentate ligands such as oxalic diamides and related amides (e.g. N,N'-bis(4-hydroxyl-2,6- dimethylphenyl)oxalamide (Ma et al., J. Am. Chem. Soc. 2016. 138, 13493-13496), N,N’-bis(thiophene- 2-ylmethyl)oxalamide (Nett et al., ChemCatChem 2019, 11, 5748-5753)) also represent ligand systems for the copper-catalyzed hydroxylation to form phenols. WO2017 / 012379 discloses oxalic acid monoamides and WO2017 / 177979 amides as ligands for copper-catalyzed coupling reactions.
[0004] CN112142567 describes the preparation of 3-chloro-2-fluorophenol starting from l,3-dichloro-2-fluoro- 4-nitrobenzene through a multistep procedure. The preparation of 3-chloro-2-fluorophenol through Sandmeyer hydroxylation under harsh conditions is described in Finger et al., J. Am. Chem. Soc. 1959, 81, 1, 94-101.
[0005] Overall, the prior art syntheses of 3-chloro-2-fluorophenol are not suitable for production on large scale. The number of steps, harsh conditions and low product yield make the processes inefficient for large scale production. Previously identified ligands for this specific substrate led to unsatisfactory reaction outcomes under desirable reaction conditions. Literature known copper-coupling reaction conditions, require high copper salt and ligand loadings, extremely high temperatures for aryl-bromides and aryl-chlorides (130- 180°C) and predominantly rely on using dimethyl sulfoxide as (co-)solvent to ensure high solubility of the substrate and ensure efficient coupling of challenging substrates. Due to cost, sustainability and safety concerns these conditions are not suitable for large scale production scale.
[0006] The present invention provides novel iminoamide ligands of formula (I) enabling copper-catalyzed coupling reactions of (hetero)aryl halides forming C-C, C-N, C-O, C-S and C-P-bonds and other bonds, in a technically feasible, more economical process which is suitable for industrial scale. In particular, the present invention relates to the use of the novel iminoamide compounds of formula (I) as ligand in copper- catalyzed coupling reactions of aryl halides providing an efficient method for the preparing of halogenated phenols, more specifically 3-chloro-2-fluorophenol.
[0007] Surprisingly, it has been found that copper-catalyzed coupling reactions of (hetero)aryl halides forming C-C-, C-N-, C-S-, C-O- and C-P-bonds are easily accessible applying iminoamide-type ligands of formula (I).
[0008] The object of the present invention is to provide a catalytic system that is useful for copper-catalyzed coupling reaction of (hetero)aryl halides forming C-C-, C-N-, C-S-, C-O- and C-P-bonds applying compounds of formula (I) as ligands.

The inner temperature remained under 2°C during addition. After dosing was finished, the clear yellow solution was stirred for 15 min at 0 °C. In a 2L double jacketed vessel equipped with a mechanical stirrer under nitrogen at 22 °C were placed aqueous sodium iodide solution (50%, 224 g, 0.75 mol, 1.11 eq.), 100 g water and 160 ml acetonitrile and the resulting solution cooled to 0 - 1 °C with a jacket temperature of -1°C. Within 1 h the diazonium salt solution in the IL vessel was dosed into the mixture in the 2L vessel. The temperature rose up to 9 °C. The mixture turned brown and the evolution of gas (N2) is observed. After completed addition, stirring was continued for 15 min at 5 °C. 200 ml water and 200 ml dichloromethane were added, the mixture was stirred for 20 min while adding a total of 9.5 g sodium metabisulfite in several portions. Then the phases were separated. The lower organic phase was removed and the upper aqueous phase was extracted again with 140 ml dichloromethane. To achieve a complete phase separation 100 ml water was added. The lower organic phase was removed and combined with the organic phase of the first extraction. The aqueous phase (1435 g, 1staq. phase) was discarded. The combined organic phases were washed with 100 ml water and additional 2 g sodium metabisulfite and then washed with 100 ml water. Finally, 700 g organic phase and 213 g aqueous phase (2ndaq. phase) were obtained. The solvent of the organic phase was distilled off at 45 °C and a pressure down to 5 mbar to afford 159 g of crude product as a red-orange oil.
