Description

LOW AVERAGE MOLECULAR WEIGHT COLLAGEN PEPTIDES COMPOSITIONField of the invention

[0001] The present invention relates to a collagen peptides composition characterized in that it has a high amount of di-and tripeptides and very low average molecular weight. The composition further has improved organoleptic properties and in particular has improved functional properties, such as dissolution good colour properties and low turbidity when put into solution. The present invention further relates to use of the present composition in food, pharmaceutical, nutraceutical and cosmetic applications.Background of the invention

[0002] Collagen peptides, also known as collagen hydrolysate, are recognized for their beneficial health effects. For some specific applications, there is a need in the very low molecular range of collagen peptides.

[0003] Existing low molecular peptides compositions have very poor organoleptic properties such as smell and taste, which renders their application into food quite difficult and the only way they can be consumed is in tablet or capsule form. Other formulations also show increased turbidity when put into solutions which limits their application in clear beverages. Existing collagen peptides compositions with better smell and taste profile typically have a higher average molecular weight distribution.

[0004] Low average molecular weight peptides compositions may be preferred as alternative to higher molecular weight peptide compositions as these allow faster absorption. In particular, collagen amino acids, di-and tri-peptides can pass directly into the intestine and enter the bloodstream, allowing for a fast absorption. Also for topical applications, low to ultra-low (500Da or lower) molecular weight collagen peptides are preferred as they can easily penetrate to the skin.

[0005] There is a need for an improved low average molecular weight collagen peptides composition.

[0006] The present invention aims to solve at least some of the problems of the prior art.Summary of the invention

[0007] The present invention is defined in the appended claims.Detailed description

[0008] It is an object of the present invention to provide a collagen peptides composition characterized in that:

[0009] a. it has an average molecular weight of from 200 to 800 Da; and

[0010] b. a 12.5%aqueous solution of said collagen peptides composition has a transmittance at 450nm of 80%or more.

[0011] Preferably, the composition of the present invention has an average molecular weight of from 200 to 800Da, more preferably from 300 to 700Da, even more preferably from 400 to 700Da, yet even more preferably from 500 to 700Da. In another preferred embodiment, the composition may have an average molecular weight of from 200 to 500Da. In another preferred range, the composition may have an average molecular weight of from 200 to 300Da; preferably from 200 to less than 300Da.

[0012] Transmittance at 450nm of a 12.5wt%aqueous solution of the collagen peptides composition is a measure for the colour of said solution. A lower transmittance reflects a more yellow product. Preferably, a 12.5%aqueous solution of said collagen peptides composition has a transmittance at 450nm of 80%or more, more preferably 85%or more, even more preferably 88%or more, yet even more preferably 90%or more. Transmittance may be measured as defined herein.

[0013] The composition of the present invention is further characterized in that it comprises high amounts of free amino acids, di-and tripeptides, preferably high amounts of di-and tripeptides. This is advantageous as collagen free amino acids, di-and tripeptides can pass through the intestine wall and enter the bloodstream directly, allowing for a fast absorption and fast bioavailability after ingestion. The present invention is preferably further characterized in that it comprises at least 30wt%, more preferably at least 35wt%, even more preferably at least 40wt%, yet even more preferably at least 45wt%, yet even more preferably at least 50wt%free amino acids, di-and tripeptides, preferably di-and tripeptides. Preferably the composition of the present invention comprises from 30 to 60wt%of free amino acids, di-and tripeptides, preferably di-and tripeptides.

[0014] It is a further object of the present invention to provide a composition with improved organoleptic properties, in particular improved taste and smell properties. This allows the composition of the present invention to have an increased flexibility for use in various applications and thus to be delivered to the consumer in other forms than tablets and capsules. Smell and taste can for example be measured by the trimethylamine content, total volatile basic nitrogen, or biogenic amines content.

[0015] The composition of the present invention further has advantageous properties when put into solution, in that it provides solutions which show low turbidity. Transmittance at 620nm can be used as a measure of turbidity. In that respect, a 12.5wt%aqueous solution of the composition of the present invention preferably has a transmittance of 95%or more at 620nm, preferably of 98%or more.

[0016] In a further embodiment, the composition of the present invention is characterized in that it comprises at least 10wt%, more preferably at least 15wt%, even more preferably at least 20wt%, yet even  more preferably at least 25wt%of tripeptides. The composition of the present invention preferably comprises from 10 to 30wt%, more preferably from 15 to 30wt%, yet even more preferably from 20 to 30wt%of tripeptides, yet even more preferably it comprises from 25 to 30wt%of tripeptides. This is advantageous because low molecular weight collagen containing high content of collagen-specific di-and tripeptides and free amino acids can be absorbed directly, bypassing long digestion steps, and enter the bloodstream, providing fast absorption. Additionally, bioactive collagen di-and tripeptides have anabolic stimuli potential to trigger collagen synthesis in the cells. A low molecular weight collagen product with high content of collagen di-and tripeptides and free amino acids can supply an abundant and accessible pool of these peptides for bioactivity in the body.

[0017] In a preferred embodiment, the composition of the present invention is characterized in that it is a free-flowing powder composition. In a further preferred embodiment, the composition is an agglomerated powder composition. Agglomeration is a well-known process in the art, which results in the cohesion of several powder particles together to form bigger agglomerates. Agglomeration is beneficial as it reduces dust formation, it improves the flowability of the product, and the like.

[0018] Collagen is the main component of connective tissues in animals and humans. Collagen consists of chains of amino acids bound together to form triple-helices which in turn form elongated fibrils. It is mostly found in fibrous tissues such as tendons, ligaments and skin. It is also found in bones, teeth, corneas, cartilage, intervertebral discs and blood vessels. Collagen for industrial processing is mostly derived from skin and / or bones and / or (dry) scales. Collagen may be processed to produce gelatin, which is obtained by irreversible, partial hydrolysis of collagen. According to the Food Chemical Codex, gelatin is defined as the product obtained from the acid, alkaline, or enzymatic hydrolysis of collagen, the chief protein component of the of the skin, bones, and connective tissue of animals, including fish and poultry. Gelatin is typically characterized by a gel strength and in warm solution by a certain viscosity. This gelatin can be further hydrolysed to shorter protein chains to produce collagen hydrolysates, i.e., collagen peptides, losing its ability to form a gel and becoming even soluble in water at ambient temperatures. Hydrolysis can be done by an enzymatic treatment for example. In the present invention, the collagen peptides can be derived from any animal such as pig, beef, poultry, or fish. Preferably the collagen peptides composition is derived from fish skins and / or bones and / or scales, more preferably it is derived from fish skins. In a further preferred embodiment, it is derived from farmed fish, and / or is preferably derived from one or more of the group of carp, catfish, salmon, and tilapia, more preferably is derived from tilapia fish.

[0019] Preferably, the composition of the present invention is produced by enzymatic hydrolysis of a gelatin raw material.

[0020] Due to its improved properties in taste and smell, the composition of the present invention can be used in various products, such as food and nutraceuticals without compromising on the smell and taste of these products. Also in view of its low molecular weight properties and high amount in free amino acids, di-and tripeptides, in particular high amount in di-and tripeptides, the dosage required to have an effective daily consumption of collagen peptides is lower with the composition of the present invention.

[0021] Thus, the invention relates to the use of the collagen peptides composition of the invention as an ingredient in food, nutraceutical compositions, cosmetic compositions, and the like.

[0022] Another object of the present invention is to provide a food or nutraceutical product comprising the collagen peptide composition as defined in the present invention and further food or nutraceutical ingredients. The food product includes, but is not limited to, bakery products, such as bread, cakes, sponge cakes, cereal bars and the like; dairy products, such as yogurts, dairy desserts and the like; beverages, such as smoothies, nutrition beverages, protein beverages, meal replacement beverages, waters, fruit juices, fruit concentrates and the likes. The food product includes supplements such as powder blends comprising the composition of the present invention and possibly other ingredients.

[0023] Preferably the food product is a beverage. More preferably, the beverage is a clear beverage, such as a protein water, or a flavoured water, and the like. The composition of the present invention may also be used in dry mixes to make instant beverages for examples, wherein the dry mix is mixed with water by the consumer before consumption. Preferably, the beverage has a transmittance at 620 nm of 50%, more preferably 60%, even more preferably 70%, yet even more preferably 80%, as measured against a deionised water reference.

[0024] In another embodiment, the present invention relates to a cosmetic product comprising the composition of the present invention. The cosmetic product includes, but is not limited to creams, lotions for skin care, shampoos, and lotions for hair care, and the like. Preferably the cosmetic composition is a cream or lotion for skin care.

[0025] The invention further relates to a process to produce the collagen peptides composition of the invention, comprising the following steps:

[0026] a. providing raw material comprising collagen,

[0027] b. preferably washing the raw material in water,

[0028] c. treating the raw material with alkaline and / or acid,

[0029] d. performing a heat extraction of gelatin from the raw material,

[0030] e. hydrolysing the gelatin extracted in step e. to collagen peptides,

[0031] f. purifying the collagen peptides, and

[0032] g. drying.

[0033] In step a., raw material comprising collagen is provided. The raw material comprises collagen and can be derived from any animal such as pig, beef, poultry, or fish. Preferably, the raw material comprises fish skins and / or bones and / or scales, more preferably it comprises fish skins and / or scales. More preferably, it is derived from farmed fish, and / or is preferably derived from one or more of the group of carp, catfish, salmon, and tilapia, more preferably is derived from tilapia fish. The present inventors have found that, in particular for materials such as hides and skins, in particular fish skins, by limiting the time between slaughter / capture and further processing, organoleptic properties, colour and turbidity of the final composition can be improved. The raw material hides or skins are thus preferably coming from animals which have been slaughtered / captured at most 12h, preferably at most 10h, more preferably at most 8h, even more preferably at most 5h, yet even more preferably at most 3h prior to the following treatment step. The raw material may comprise hide or skins, which may alternatively be frozen at most 12h, preferably at most 10h, more preferably at most 8h, even more preferably at most 5h, yet even more preferably at most 3h after slaughter / capture and defrosted before the following treatment step. The raw material may also comprise fish scales and / or bones. In case of scales and / or bones, they preferably are coming from fish which have been captured at most 12h, preferably at most 10h, more preferably at most 8h, even more preferably at most 5h, yet even more preferably at most 3h prior to the following treatment step, or alternatively frozen at most 12h, preferably at most 10h, more preferably at most 8h, even more preferably at most 5h, yet even more preferably at most 3h after slaughter / capture and defrosted before the following treatment step.

[0034] Preferably, step b., washing the raw material in water, is performed. In step b., the raw material is washed in water, and in particular for hides or skins, preferably such as to obtain a conductivity of 700 micro siemens or less, more preferably 600 micro siemens or less, even more preferably 500 micro siemens or less. This step allows to wash away any surface dirt, which can comprise compounds that have bad organoleptic properties and colour. This step also allows to wash away fat and meat / flesh which is not suitable for gelatin extraction. This washing step may comprise one or more washing steps.

[0035] In step c., the optionally washed material is treated with an alkaline and / or acid solution. It is thought that such a step allows to halt enzymatic processes which degrade the material in undesirable manners. Furthermore, this step increases the yield of gelatin. For raw material comprising hides or skins, step c. preferably comprises treatment with an alkaline solution, preferably at a pH between 8 and 13.5, more preferably between 9 and 13.5, even more preferably 10 and 13.5, yet even more preferably 11 and 13.5, yet even more preferably 12 and 13.5. For raw material comprising scales and / or bones, step c.  preferably comprises treatment with acid in order to demineralize the scales and / or bones before extraction of gelatin. In step c., alkaline treatment may be done with NaOH for example and acid treatment may be done with H2SO4 or HCl.

[0036] In step d., gelatin is extracted from the raw material using heat. This step may be performed by adding 2-7 parts water by weight of the raw material, and applying heat. This step may be performed at a temperature of about 60 to about 90 ℃. This step may be performed for about 1 to 5 hours. This step may be repeated several times to increase the yield.

[0037] In step e., the extracted gelatin is hydrolysed to collagen peptides. The gelatin may be hydrolysed using any suitable hydrolysis process, such as enzymatic hydrolysis, steam hydrolysis, acid hydrolysis or alkali hydrolysis. Preferably, the hydrolysis is enzymatic hydrolysis, which allows a preferential hydrolysis to free amino acids, di-or tripeptides.

[0038] In step f., the collagen peptides are purified. Purification may be done using any one or more suitable technique known in the art such as use of resins, evaporation, membrane filtration, use of active carbon, or use of other specific techniques known in the art. Preferably purification is done with active carbon. It is believed that this step improves the colour and organoleptic properties of the final collagen peptides composition.

[0039] In step g., the collagen peptides are dried. Drying may be done using any suitable drying technique such as spray drying for example. Before drying, the collagen peptides may be concentrated and / or sterilized. Concentration may be performed by heating to evaporate water. Sterilization is performed by heating to a temperature of 130-140℃ for about 3-4 seconds, in a closed vessel. Finally, a free-flowing collagen hydrolysate composition, i.e., collagen peptides composition, can be obtained by drying the concentrated hydrolysate solution.

[0040] Methods of measurement

[0041] Average molecular weight

[0042] The average molecular weight is measured by size exclusion chromatography according to the Gelatin Manufacturers Association of Asia-Pacific (GMAP) method.

[0043] Di-and tripeptides content

[0044] The di-and tripeptide content is estimated first determining the molecular weight distribution by size exclusion chromatography, followed by matching this to known collagen dipeptide and tripeptides, or free amino acids.

[0045] Calculation of di-and tripeptide content

[0046] The amino acid sequence of tilapia collagen is as follows (where every letter represents a distinct amino acid) :

[0047] A python code was written to randomly cut this sequence in chains of 2 to 6 / 10 amino acids (repeated analysis) to see distribution, amino acid sequence and average molecular weight.

[0048] The Tilapia collagen amino acid sequence can be generalized to collagen molecules from other raw materials, such as other fish collagen, but also collagen from pig, beef, chicken and the like.

[0049] From this analysis it comes out that collagen tripeptides typically have a molecular weight between 217Da and 428Da, with an average of 291Da, while collagen dipeptides typically have a molecular weight between 145Da and 301Da, with an average of 192Da.

[0050] Based on the model, an estimation was made about the fractions of dipeptides, tripeptides, tetrapeptides and pentapeptides present per molecular weight fraction. These are given in the table below for free amino acids and dipeptides, tripeptides and tetrapeptides.

[0051] From the molecular weight distribution of a given collagen peptide composition, the content of di-peptide / free amino acids, tripeptides, tetrapeptides can be deduced using the above table.

[0052] Transmittance

[0053] Transmittance is measured as follows: a 12.5 wt%aqueous solution is prepared by adding 5 g of the collagen peptide to 35 g deionised water, and heated at 40 ℃ to dissolve. Transmittance is measured at the wavelength of choice (e.g., 450 nm or 620 nm) and expressed as %vs a deionised water reference.

[0054] The invention will be illustrated in the following non-limiting examples:

[0055] Examples

[0063] f. hydrolysing the gelatin extracted in step e.,

[0064] g. purifying the hydrolysate with active carbon, and

[0065] h. concentrating, sterilizing, and spray-drying.

[0066] Several commercially available samples were analysed and benchmarked against the composition of the present invention. 7 commercially available products are marketed as low molecular weight collagen peptides (A-G) .

[0067] The taste and smell ratings were evaluated as follows:

[0068] 10g of product is added under continuous stirring to 40ml of demineralised water. As reference product, commercially available  Optima (PB Leiner) was used, which is a higher molecular weight collagen hydrolysate (2000-3000Da) . The samples are rated as follows: rating should be made based on intensity and not based on preference. Participants start with tasting / smelling the reference. The reference gets a score of 3. A blind reference sample was used as a validation, which the testers also scored as 3. All products are ranked in comparison to this reference using the following scoring system:

[0069] The results are provided in the table below:

[0070] It can be observed that the average molecular weight and free amino acid, di-and tripeptide content varied across samples, and that higher average molecular weight in the commercially available samples is associated with improved taste and smell. On the contrary, a higher free amino acid, di-and tripeptide content in commercially available samples is associated with poor taste and smell properties.

[0071] The composition of the invention showed a good combination of organoleptic properties (taste and smell ratings) , good colour (450 nm transmittance) , low turbidity (high 620 nm transmittance) , low average molecular weight and high free amino acids, di-and tripeptide content.
