AI Technical Summary
The existing eutectic preparation methods are prone to form solvates when using solvents, and are difficult to be suitable for production of different scales, which limits its wide range of applications.
The anti-solvent suspension method is used to mix the poor solvent with the good solvent, and then the solid reactant and eutectic form are added, and eutectics are obtained by stirring, filtration and drying.
This method can effectively inhibit the generation of solvates, improve the purity and yield of the product, and is suitable for eutectic preparation from milligram to kilogram, with wide application prospects.

Abstract
Description
A method for preparing eutectic Technical Field
[0001] The present invention relates to the technical field of medicinal chemistry, and in particular to a method for preparing a cocrystal. Background Art
[0002] A cocrystal is a single crystalline material composed of two or more different solid-state compounds in a stoichiometric ratio. Typically, the components of a cocrystal are neutral molecules. When proton transfer occurs between the components, resulting in the inclusion of charged ions in the lattice, a salt is formed. When a cocrystal contains both neutral molecules and charged ions, an ionic cocrystal is formed. If one component of the cocrystal is an active pharmaceutical ingredient (API) and the other is a pharmaceutically acceptable cocrystal former (CCF), the cocrystal is called a pharmaceutical cocrystal. Cocrystals have attracted increasing attention in various solid materials fields, particularly in the pharmaceutical field, due to their ability to manipulate optoelectronic, mechanical, physicochemical, and biological properties and enrich their solid forms.
[0003] The formation of cocrystals involves the cocrystallization of two or more solid components. Compared to the crystallization of a single component, the synthesis of cocrystals faces greater challenges because the different components often have different polarities and properties. Many methods for preparing cocrystals have been reported, which can be roughly divided into methods using solvents (e.g., evaporation, cooling crystallization, antisolvent crystallization, and suspension methods) and methods using no or only a small amount of solvent (e.g., melt crystallization, hot melt extrusion, and grinding methods). In contrast, methods using solvents for preparing cocrystals have advantages in terms of online monitoring of the reaction process, product quality control, and industrial scale-up (Rodrigues et al, "Pharmaceutical cocrystallization techniques. Advances and challenges", Int. J. Pharm. 2018, 547, 404-420, DOI: 10.1016 / j.ijpharm.2018.06.024).However, for some components that easily form solvates, solvates or cocrystals mixed with solvates are often obtained when cocrystals are prepared using solvents (Johnston et al, "Carbamazepine N,N-dimethylformamide solvate", Acta Crystallogr. E 2005, 61, 01509-01511, DOI: 10.1107 / S1600536805012535; Burnett et al, "Investigating carbamazepine-acetone solvate formation via dynamic gravimetric vapor sorption", J. Therm. Anal. Calorim. 2007, 89, 693-698, DOI: 10.1007 / s10973-006-7957-8; Harris et al, "Structural Studies of the Polymorphs of Carbamazepine, Its Dihydrate, and Two Solvates", Org. Process Res.Dev.2005,9,902-910,DOI:10.1021 / op0500990; Fleischman et al, "Crystal engineering of the composition of pharmaceutical phases: Multiple-component crystalline solids involving carbamazepine", Cryst.Growth Des. 2003, 3, 909-919, DOI: 10.1021 / cg034035x).
[0004] The suspension method is a widely used solvent-based method for preparing cocrystals, in which solid reactants and cocrystal formers are suspended in a solvent in a specific ratio to form the cocrystal (Zhang et al, "Efficient Co-crystal Screening Using Solution-Mediated Phase Transformation", J. Pharm. Sci. 2007, 96, 990-995, DOI: 10.1002 / jps.20949). When the solubility of the cocrystal or certain components thereof in the solvent is high, excessive feeding is often required to obtain the cocrystal (Rodriguez-Hornedo et al, "Reaction crystallization of pharmaceutical molecular complexes", Mol. Pharmaceutics 2006, 3, 362-367, DOI: 10.1021 / mp050099m). This often reduces the utilization of the raw materials and also introduces a high level of impurities into the residual mother liquor.
[0005] Antisolvent crystallization is another method for preparing cocrystals using solvents. This involves dissolving the solid reactants and the cocrystal former in a certain good solvent. An antisolvent is then added to supersaturate the solution and precipitate the cocrystal from the solution (Zhang Jianjun et al., Theory and Application of Crystalline Drug Development, 2019, p191). The antisolvent crystallization method requires that the poor solvent and the good solvent are miscible (Douroumis et al., “Advanced methodologies for cocrystal synthesis”, Adv. Drug Deliver Rev. 2017, 117, 178-195, DOI: 10.1016 / j.addr.2017.07.008).
[0006] Another challenge in cocrystal preparation is the applicability of the preparation method. It is often difficult for a single preparation method to be suitable for milligram-scale cocrystal screening, gram-scale cocrystal sample preparation, kilogram-scale cocrystal pilot production, and even larger-scale industrial production.
[0007] Therefore, developing a new cocrystal preparation method can overcome the shortcomings of the above-mentioned suspension method and has wide applicability. It can be used for milligram-scale cocrystal screening, gram-scale cocrystal sample preparation, and kilogram-scale cocrystal pilot production, which has important practical significance.
