[0025] Inflammation plays an important role in many diseases and tissue homeostasis. Macrophages, derived from myeloid precursors of bone marrow are often modulators of inflammation. Macrophages may be polarized into either of two distinct forms: a proinflammatory state referred to as M1, or an anti-inflammatory state referred to as M2. These states may be induced in vivo or in vitro. M2 macrophages may secrete anti-inflammatory factors such as cytokines, chemokines, or other metabolites, or may play a role in tissue repair, allergy, or regeneration. As such, it may be useful to polarize macrophages to the M2 state, and to avoid polarizing macrophages to M1.

[0026] Disclosed herein is a novel bacterial derived modality, termed MP003, with anti-inflammatory and anti-oxidative properties. MP003 may be considered as being within the thioredoxin (Trx) superfamily, which includes small ubiquitous redox proteins of approximately 12 kD. Some Trx superfamily proteins may play important roles in redox signaling, oxidative stress responses, or immunomodulation. Trx has thiol-disulfide reductase activity that may be influenced by a conserved active motif (-Cys32-Gly-ProCys35-) . Three human Trx homologues exist, Trx1, Trx2, and Trx3. Each may have its own sub-cellular location and inflammatory modulating effect. Trx1 promotes anti-inflammatory macrophages of the M2 phenotype.

[0027] An amino acid sequence for MP003 was identified, and found to have some degree of similarity to human Trx1 (hTrx1) . However, as disclosed herein, MP003 was found to have some surprisingly superior effects relative to Trx1 when used to polarize macrophages to M2, or used for treating subjects having an inflammatory disorder. For example, MP003 counteracted oxidative stress as demonstrated by one assay to a similar extent as Trx-1, which is an important characteristic of some Trx superfamily proteins. However, MP003 also displayed significantly stronger anti-inflammatory effects compared to hTrx1 at the same or even a lower protein concentrations. MP003 induced a stronger M2 polarization of macrophage cells (in vitro) , and ameliorated pathophysiology of animal subjects with inflammatory bowel disease (IBD) .
