A LIVE BACTERIA STRAIN OF STAPHYLOCOCCUS SP.Technical Field

[0001] The invention relates to the field of biomedicine. In particular, the invention relates to a live bacteria strain of a species from Staphylococcus sp. such as Staphylococcus aureus. More particularly, the invention relates to a live bacteria strain of Staphylococcus sp., such as a live bacteria strain of Staphylococcus aureus, with reduced activity of saePQRS, reduced activity of adsA, and / or reduced production of capsules; to the uses of said live bacteria strain; to a vaccine against bacterial infection comprising said live bacteria strain; and to a method for preventing and / or treating bacterial infection in a subject by administering said live bacteria strain.Background

[0002] Staphylococcus aureus (S. aureus) is a highly-adaptable pathogen able to infect various tissues. Common manifestations of S. aureus infections range from mild skin and soft-tissue infections to invasive disease. Additionally, this pathogen has gained resistance against many anti-microbial drugs, leaving healthcare providers with few options for the treatment of infections. Therefore, the development of new methods for the treatment / prevention of Staphylococcus aureus infection is an urgent clinical need.

[0003] Attenuated vaccines are weakened forms of a pathogen that are used to stimulate an immune response without causing disease. Attenuation of virulence was achieved through natural selection by multiple passages of the pathogen on growth medium, random mutagenesis or genetic modification of target genes to inducing virulence attenuation. Genetic modification which can make the higher level of virulence attenuation has become one of the most suitable ways to develop attenuated vaccines. The selection of the target is the key to the successful development of attenuated vaccine.

[0004] S. aureus has evolved many mechanisms to circumvent killing by these potent innate immune cells. The production of secreted virulence factors during pathogenesis is primarily controlled by the combined influence of two-component systems (TCSs) that sense the host environment and respond accordingly. Of these, the Sae TCS has been shown to be essential for evasion of human neutrophil killing. The system is composed of a histidine kinase sensor, SaeS, a response regulator, SaeR, and two accessory proteins, SaeP and SaeQ. The sensor protein SaeS detects environmental cues, such as changes in pH or oxygen levels, and then phosphorylates the response regulator, SaeR. The phosphorylated SaeR then binds to the promoter regions of target genes, activating or repressing their transcription. The roles of the SaeR response regulator and its cognate sensor kinase SaeS have been demonstrated to be critical for causing infection. Public university of Navarre selected a mutant strain devoid of all TCS genes (including saeRS) as candidate to live attenuated strain for vaccination purposes (EP3342854A1) . So far, no one has reported the attenuated function of the saePQ gene, which is one of the TCSs. Instead, Collins, Madison M., et al. demonstrated that a saeP and saeQ double mutant (USA300ΔsaePQ) increased the virulence of USA300 during murine bacteremia [1] .

[0260] To measure the antibody-mediated immune responses, BALB / c mice were immunized with Newman, Sau0021, Sau0126 and PBS (s.c. 5x107, n=10) , and antibody titers were determined by enzyme-linked immunosorbent assay (ELISA) .
