Description

Composition for improving skin blood circulation

[0001] This application claims priority to Republic of Korea Application No. 10-2024-0006084, filed January 15, 2024, the entire disclosure of which is incorporated herein by reference.

[0002] The present disclosure relates to a composition for improving skin blood circulation.

[0003] The skin is a vital organ responsible for various physiological functions, including protecting the body from the external environment, acting as a barrier to prevent the loss of internal moisture and beneficial substances, regulating body temperature, and excreting nutrients. However, skin cell activity can decline and skin condition worsen for the following reasons.

[0004] Collagen and elastin, found in the dermal layer of the skin, play a significant role in skin composition, providing mechanical strength, connective tissue resistance, maintaining tissue cohesion, and supporting cell adhesion. Collagen is depleted by stress, aging, and photoaging caused by UV exposure. Elastin, on the other hand, is known to lose its three-dimensional structure and activity after exposure to UV rays, due to elastase, a degrading enzyme whose activity increases. This loss of collagen and elastin activity can cause skin tissue to become loose and lose elasticity.

[0005] Furthermore, as epidermal cellular function declines, metabolism becomes impeded and dead skin cells may not shed properly. When exposed to UV rays while excessive dead skin cells accumulate, skin elasticity decreases, leading to wrinkles. Furthermore, if a sebum barrier is not formed on the epidermis, moisture and sebum secretion decrease, leading to dryness and the formation of wrinkles.

[0006] Blood circulation refers to the flow of blood through blood vessels to all parts of the body. Blood circulation is influenced not only by physical factors like blood cell count, viscosity, and shear stress, but also by changes in blood lipid composition and blood cells. Poor blood circulation can lead to stiff shoulders and neck, numbness in the extremities, cold hands and feet, and edema in the lower extremities, and can even contribute to cerebral or cardiovascular disease. Furthermore, improving blood circulation in the skin can transform dull, inelastic skin into vibrant, clear, and transparent skin, creating a more radiant, healthy complexion.

[0007] However, existing medications aimed at improving blood flow have side effects such as gastrointestinal upset and elevated blood pressure, making them difficult to take. Therefore, there is a need to develop products using natural ingredients that are safe for the human body with minimal side effects and can fundamentally improve blood flow.

[0008] The problem that the present disclosure seeks to solve is to provide a composition for improving skin blood circulation that is safe for the human body, has no side effects, and has an excellent effect of improving skin blood circulation.

[0009] To solve the above problems, the inventors of the present invention have diligently researched and made efforts to develop a composition that can improve problems such as side effects and precautions for use of conventional skin treatments, and can effectively improve skin blood circulation without the disadvantages such as weak effects of skin improvement agents, while being safe for the human body. As a result, the inventors of the present invention have discovered that although the skin blood circulation improvement effect of panthenol and niacinamide individually was minimal, when escin; and a combination of panthenol, niacinamide, or a mixture thereof was administered (applied) to the skin, skin blood circulation was significantly improved, and surprisingly, the combination of the above ingredients unexpectedly exhibited a remarkable synergistic effect in improving skin blood circulation, thereby completing the present invention.

[0010] The present disclosure provides a composition for improving skin blood circulation, comprising escin, panthenol, niacinamide, or a mixture thereof as active ingredients. This composition for improving skin blood circulation may be a composition for external application to the skin.

[0034]

[0035] Experimental Example 1: Effect of regulating angiogenesis-promoting signals (adenosine receptor pathway)

[0036] Adenosine receptor pathway signals, which are generally activated in vascular cells, play a key role in the formation of new blood vessels and the branching of capillaries. Therefore, this experiment examined how the combination of the present invention's escin and panthenol, niacinamide, or a mixture thereof contributes to the amplification of adenosine receptor pathway signals.

[0037] NECA was used as a positive control to amplify adenosine receptor pathway signals, and DMSO was used as a negative control. 3x10 CREB Reporter HEK293A cells were cultured in a 96-well plate. 4 After seeding each well, the dogs were treated with the escin, panthenol, niacinamide, or a mixture thereof according to the present invention at the treatment concentrations listed in Table 2 below. Reporter analysis was performed using the Luciferase assay kit (E1960) from Promega. The analysis was performed according to the manufacturer's experimental method, and adenosine receptor activity was measured using a luminometer Victor (PerkinElmer, Waltham, Massachusetts, USA).

[0039] As shown in the experimental results described in Table 2 above, when Escin and panthenol, niacinamide, or a mixture thereof were combined, the activity of the adenosine receptor signaling system, one of the main targets for improving blood flow, was significantly increased. Through this, it was confirmed that Escin and panthenol, niacinamide, or a mixture thereof had a remarkably excellent effect on improving blood flow.

[0040]

[0041] Experimental Example 2: Effect of a composition for improving skin blood circulation (skin lotion) on improving skin blood circulation

[0042] The skin blood circulation improvement effect of the composition for improving skin blood circulation of the present disclosure was tested on a total of 36 men and women. Using Comparative Examples 1 to 9 and Examples 1 to 12 described in Table 1 above, the skin blood circulation improvement efficacy test of each composition was conducted.

[0043] The test sequence for the composition for improving skin blood circulation was divided into 15-minute intervals: before and after skin application. Furthermore, during the human application test period, the use of any cosmetics containing active ingredients that could influence the test results (e.g., functional cosmetics for whitening or wrinkle improvement) other than the test composition was prohibited.

[0044] To measure the improvement in skin blood flow, the skin was washed and left for 10 to 15 minutes under constant temperature and humidity conditions (25°C, 40% relative humidity). Measurements were made around the hairline between the forehead and scalp using a Periflux System 5000 (Perimed, Jarfalla, Sweden). The compositions of the examples were applied to the measurement area and measurements were made 15 minutes later. This experiment was repeated three times, and the average value was calculated. The change in skin blood flow after use compared to before use was indicated. The measurement results are shown in Table 3 below.

[0046] In view of the above results, it was confirmed that the skin blood circulation improvement effect was remarkably excellent in Examples 1 to 12 containing escin; and panthenol, niacinamide, or a mixture thereof. Through this, it was confirmed that the skin blood circulation improvement composition of the present disclosure containing escin; and panthenol, niacinamide, or a mixture thereof as active ingredients can be very effectively used to promote skin blood circulation.

[0047] In the above experimental example, the composition for improving skin blood circulation according to the present disclosure was prepared in the form of a skin lotion, and the skin blood circulation-improving effect was confirmed. However, this formulation example is merely one example of the composition for improving skin blood circulation according to the present disclosure, and it will be apparent to those skilled in the art that the scope of the composition of the present disclosure is not limited to the formulation alone.
