[0001] The present invention relates to a solid oral pharmaceutical dosage form comprising 6-acetyl-8-cyclopentyl-5-methyl-2- (5-piperazin-1-yl-pyridin-2-ylamino) -8H-pyrido [2, 3-d] pyrimidin-7-one (hereinafter palbociclib) and succinic acid.

[0003] Palbociclib is a potent and selective inhibitor of CDK4 and CDK6, having the following chemical structure:

[0004] Palbociclib is described in WHO Drug Information, Vol. 27, No. 2, page 172 (2013) .

[0005] Palbociclib and pharmaceutically acceptable salts thereof are disclosed for example in the international publication No. WO 2003 / 062236.

[0006] Palbociclib is approved in the European Union for the treatment of hormone receptor (HR) -positive, human epidermal growth factor receptor 2 (HER2) -negative locally advanced or metastatic breast cancer in combination with an aromatase inhibitor or in combination with fulvestrant in women who have received prior endocrine therapy, wherein in pre-or perimenopausal women, the endocrine therapy should be combined with a luteinizing hormone-releasing hormone (LHRH) agonist.

[0007] Palbociclib is marketed by Pfizer under the trade name in the form of an oral immediate release capsule and in the form of an immediate release tablet both containing palbociclib in the form of the free base.

[0008] Palbociclib is a dibasic compound and has two basic groups with pKa’s of approximately 7.3 (the secondary piperazine nitrogen) and 4.1 (the pyridine nitrogen) . The solubility of palbociclib free base is pH dependent. Palbociclib is water soluble at low pH (2.1-4.5) , while  the solubility dramatically decreases as pH rises above 4.5. Palbociclib has poor water solubility (9 mg / mL) at pH 7.9. Concomitant administration of agents which increase gastric pH can alter the solubility and absorption of palbociclib free base formulations.

[0009] The absorption and bioavailability of a therapeutic agent can be affected by numerous factors when dosed orally, including whether the subject is in a fed or fasted state, and the use of certain medications, such as proton pump inhibitors (PPIs) or H2 receptor antagonists, as well as certain medical conditions. Compounds having pH-dependent solubility, particularly basic compounds, may exhibit undesirable pharmacokinetic properties, such as poor absorption and / or reduced bioavailability, which may result in significant inter-patient and intra-patient variability.

[0013] It is an object of the present invention to provide dosage forms of palbociclib having a favorable dissolution profile and storage stability.Solution to problem

[0014] Said object has been solved by a dosage form comprising palbociclib free base as active pharmaceutical ingredient and succinic acid wherein the succinic acid has a surface area of more than 0.015 m2 / g.

[0015] Surprisingly it has been found that the dosage form according to the present invention demonstrates a favorable and stable dissolution profile and good storage stability. In particular, the dosage form according to the invention is characterized by a particularly low content of the palbociclib-succinyl adduct, which is regarded as an impurity. Moreover, it has been found that the dosage form according to the present invention demonstrates a favorable dissolution profile even if the dosage form contains succinic acid in relatively low amounts.

[0140] Dissolution tests of said tablets with five different succinic acid surface areas have been conducted. The respective formulations have been prepared as described above (Example 2) . The dissolution test refers to the dissolution of palbociclib. The dissolution test is conducted in a standard USP 2 rotating paddle apparatus as disclosed in United States Pharmacopoeia (USP) Dissolution Test Chapter 711, Apparatus 2. Paddles are rotated at 75 rpm and the dosage form is added to 900 mL of 10 mM pH 5.5 acetate buffer at 37° C. At appropriate times following test initiation (e.g., insertion of the dosage form into the apparatus) , filtered  aliquots (typically 1.5 mL) from the test medium are analyzed for palbociclib by high performance liquid chromatography (HPLC) . Dissolution results are reported as the percent of the total dose of palbociclib dissolved versus time in minutes.

[0145] Stress stability tests of said tablets with the five different succinic acid qualities have been performed.

[0146] For the stress stability test the Al / Al-packaged tablets have been loaded in a climate chamber adjusted to 60℃ / 75%RH. After time periods as indicated in the following table the tablets were crushed and analyzed for impurities using a high-performance liquid chromatography (HLPC) method as follows: Column: Waters CSH C18, 2.1 x 100 mm, 1.7 mm column; mobile phase (gradient elution) A: 0.03%trifluoroacetic acid in water, and B: 0.03%trifluoroacetic acid in acetonitrile; column temperature of 45℃; flow rate of 0.5 mL / min; UV detection at 234 nm; injection volume of 2 mL; and run time of 10.72 minutes. The total amount of impurities was determined by summing up the peak areas of the HPLC- chromatogram. Peak areas were calculated as percentage of the total area of peaks in the HPLC chromatogram. The results are summarized in the following table.

Claims

1.A solid oral pharmaceutical dosage form comprising palbociclib as active pharmaceutical ingredient and succinic acid wherein the succinic acid has a surface area of more than 0.015 m2 / g.2.The dosage form according to claim 1, characterized in that the succinic acid has a surface area of more than 0.025 m2 / g, preferably a surface area of more than 0.03 m2 / g, more preferably a surface area of more than 0.035 m2 / g and most preferably a surface area of more than 0.038 m2 / g.3.The dosage form according to any one of the previous claims, characterized in that the succinic acid has a surface area of less than 0.13 m2 / g, preferably a surface area of less than 0.09 m2 / g and more preferably a surface area of less than 0.045 m2 / g.4.The dosage form according to claim 1, characterized in that the succinic acid has a surface area in a range from 0.026 m2 / g to 0.09 m2 / g, preferably a surface in a range from 0.03 m2 / g to 0.05 m2 / g and more preferably a surface area in a range from 0.035 m2 / g to 0.045 m2 / g.5.The dosage form according to any one of the previous claims, characterized in that the dosage form is a tablet, preferably a film-coated tablet.6.The dosage form according to claim 5, characterized in thata) the tablet comprises an intragranular phase and a continuous extragranular phase, wherein the palbociclib is contained in the intragranular phase and the succinic acid is contained in the continuous extragranular phase; orb) the tablet comprises a first intragranular phase, a second intragranular phase and optionally a first continuous extragranular phase, wherein the palbociclib is contained in the first intragranular phase and the succinic acid is contained in the second intragranular phase.7.The dosage form according to any one of the previous claims, characterized in that the dosage form contains palbociclib in an amount from 10 %by weight to 35 %by weight, preferably in an amount from 10 %by weight to 20 %by weight based on the total weight of the dosage form.8.The dosage form according to any one of the previous claims, characterized in that the dosage form contains palbociclib in an amount of 25 mg, 75 mg, 100 mg or 125 mg.9.The dosage form according to any one of the previous claims, characterized in that the dosage form contains the succinic acid in an amount from 5 %by weight to 1 %by weight, preferably in an amount from less than 5 %by weight to 1 %by weight, and more preferably in an amount from 4.5 %by weight to 3 %by weight based on the total weight of the dosage form.10.The dosage form according to any one of the previous claims, characterized in that the weight ratio of palbociclib to succinic acid is selected within a range from 1 : 0.1 to 1 : 0.5, preferably within a range from 1 : 0.2 to 1 : 0.4 and more preferably within a range from 1: 0.2 to 1 : 0.3.11.The dosage form according to any one of the previous claims, characterized in that the dosage form further comprises at least one diluent in an amount from 50 %by weight to 80 %by weight, preferably in an amount from 60 %by weight to 80 %by weight based on the total weight of the dosage form.12.The dosage form according to claim 11, characterized in that the diluent is selected from the group consisting of microcrystalline cellulose, lactose monohydrate, mannitol, sorbitol, xylitol, magnesium carbonate, dibasic calcium phosphate and tribasic calcium phosphate.13.The dosage form according to any one of the previous claims, characterized in that the dosage form further comprises at least one lubricant in an amount from 0.5 %by weight to 5 %by weight, preferably in an amount from 1 %by weight to 2 %by weight based on the total weight of the dosage form.14.The dosage form according to claim 13, characterized in that the lubricant is selected from the group consisting of magnesium stearate, calcium stearate, colloidal silicon dioxide, talc, zinc stearate and sodium stearyl fumarate.15.The dosage form according to any one of the previous claims, characterized in that the dosage form further comprises at least one disintegrant in an amount from 1 %by weight to 10 %by weight, preferably in an amount from 2 %by weight to 5 %by weight based on the total weight of the dosage form.16.The dosage form according to claim 15, characterized in that the disintegrant is selected from the group consisting of crospovidone, croscarmellose sodium and sodium starch glycolate.17.The dosage form according to any one of the previous claims, characterized in that the palbociclib is present in the crystalline form.18.The dosage form according to any one of the previous claims, characterized in that the dosage form is an immediate release dosage form.19.The dosage form according to any one of the previous claims, characterized in that the dosage form when added to a test medium comprising 900 mL of 10 mM pH 5.5 acetate buffer at 37℃ in a standard USP 2 rotating paddle apparatus with the paddles spinning at  75 rpm dissolves: (a) 50 %to 90 %of the palbociclib in 15 minutes; (b) 60 %to 95 %of the palbociclib in 30 minutes; and (c) 75 %to 100 %of the palbociclib in 60 minutes.20.The dosage form according to any one of the previous claims, characterized in that the dosage form when added to a test medium comprising 900 mL of 10 mM pH 6.8 phosphate buffer (at 37℃ in a standard USP 2 rotating paddle apparatus with the paddles spinning at 75 rpm dissolves: (a) 10 %to 35 %of the palbociclib in 15 minutes; (b) 15 %to 40 %of the palbociclib in 30 minutes; and (c) 15 %to 50 %of the palbociclib in 60 minutes.21.The dosage form according to any one of the previous claims, characterized in that the succinic acid has a particle size distribution with a D90-value in the range from 100 μm to 1000 μm, a D50-value in the range from 50 μm to 700 μm and a D10-value in the range from 20 μm to 500 μm.22.The dosage form according to any one of the previous claims, characterized in that the succinic acid has a particle size distribution with a D90-value in the range from 500 μm to 700 μm, a D50-value in the range from 300 μm to 450 μm and a D10-value in the range from 150 μm to 260 μm.23.The dosage form according to any one of the previous claims, characterized in that the succinic acid has a particle size distribution with a D90-value in the range from 500 μm to 700 μm, a D50-value in the range from 300 μm to 450 μm and a D10-value in the range from 150 μm to 260 μm.24.The dosage form according to any one of the previous claims, characterized in that the palbociclib has a particle size distribution with a D90-value in the range from 300 μm to 650 μm, a D50-value in the range from 150 μm to 400 μm and a D10-value in the range from 40 μm to 250 μm.25.A process for the preparation of a dosage form according to any one of the previous claims, wherein the dosage form is a tablet, the tablet comprises palbociclib as active pharmaceutical ingredient and succinic acid, wherein the tablet comprises an intragranular phase and a continuous extragranular phase and wherein the palbociclib is contained in the intragranular phase and the succinic acid is contained in the continuous extragranular phase, wherein the process comprises the following steps:a) dry granulating palbociclib, optionally together with one or more pharmaceutically acceptable excipients, to obtain a dry granulate that forms the intragranular phase;b) mixing the dry granulate with succinic acid, optionally together with one or more pharmaceutically acceptable excipients, to obtain a mixture;c) compressing the mixture to obtain a tablet;d) optionally film coating the tablet to obtain a film coated tablet.26.A process for the preparation of a dosage form according to any one of the previous claims, wherein the dosage form is a tablet, the tablet comprises a first intragranular phase, a second intragranular phase and optionally a first continuous extragranular phase, wherein the palbociclib is contained in the first intragranular phase and the succinic acid is contained in the second intragranular phase, wherein the process comprises the following steps:a) dry granulating palbociclib, optionally together with one or more pharmaceutically acceptable excipients, to obtain a dry granulate that forms the first intragranular phase;b) wet granulating succinic acid, optionally together with one or more pharmaceutically acceptable excipients, to obtain a wet granulate that forms the second intragranular phase;c) mixing the dry granulate and the wet granulate, optionally together with one or more pharmaceutically acceptable excipients, to obtain a mixture;d) compressing the mixture to obtain a tablet;e) optionally film coating the tablet to obtain a film coated tablet.
