082907 on March 21, 2024, entitled with "COMPOUNDS FOR THE MODULATION OF IL-17" , which is incorporated herein by reference in its entirety.Technical FieldThe present invention pertains to compounds that are effective in modulating interleukin-17 (IL-17) activity. It further relates to pharmaceutical compositions comprising these compounds, as well as methods for utilizing such compositions in the modulation of IL-17 for therapeutic purposes.Background ArtIL-17 has become recognized as a crucial cytokine for defending against mucosal infections, while also being identified as a significant pathogenic cytokine and drug target in numerous autoimmune and inflammatory conditions. IL-17 also known as CTLA8 or IL17, was discovered in 1993 and subsequently renamed IL-17 (Rouvier et al., 1993; Yao et al., 1995b) . IL-17 family comprises six members (IL-17A to IL-17F) that mediate their biological functions through the IL-17 receptors (IL-17RA to IL-17RE) . IL-17A and its closest relative IL-17F, signal through the same receptor complex (IL-17R) composed of the subunits IL-17RA and IL-17RC (Mills et al., 2023; Monin et al., 2018) . IL-17A and IL-17F are produced as covalent homodimers and can also combine to form IL-17A-IL-17F heterodimers. Studies showed that lymphocytes including CD4 + T cells, CD8 + T cells, γδ T cells, innate lymphoid cells (ILCs) , natural killer (NK) cells were key producer of IL-17A. Furthermore, mucosal associated invariant T cells, mast cells, and Paneth cells can also serve as sources of this cytokine (Cua and Tato, 2010) .Interest in the study of IL-17A's role greatly increased following the discovery that IL-17A is the signature cytokine, characterized by its expression under the regulation of the "master" transcription factor RORγt and its activation by IL-23. The so-called "IL-23-IL-17A axis" has been identified as a critical driver of autoimmune diseases (McInnes et al., 2017) . In line with this, IL-17A was also found to regulate NF-κB and CCL20 via CARMA2, a locus linked to psoriasis and identified in genome-wide association studies (GWAS) (Tsoi et al., 2012) . There is substantial evidence indicating that cells producing IL-17A, including Th17 cells, play a role in various autoimmune and inflammatory conditions such as human psoriasis, rheumatoid arthritis, multiple sclerosis, inflammatory bowel diseases, and asthma (Korn et al., 2009; Genovese et al., 2014) .Secukinumab and Ixekizumab, which are anti-IL-17A neutralizing antibodies, as well as Brodalumab that targets the IL-17RA receptor, have been assessed for their efficacy in treating psoriasis, ankylosing spondylitis, and psoriatic arthritis, showing effectiveness. Although several antibodies targeting IL-17A or IL-17RA have been approved, there are currently no approved orally available modulators of IL-17A. However, the disadvantages of monoclonal antibodies (mAbs) , including the requirement for non-oral administration, limited tissue penetration, inability to cross the blood-brain barrier, and often prolonged half-life, restrict their application. Oral administration and a more flexible treatment regimen could significantly enhance patient convenience. Additionally, compounds that allow for oral intake might offer improved safety through the potential for quicker drug withdrawal in the event of adverse reactions. However, LY3509754, a potent small molecule inhibitor discovered by Eli Lilly and Company, was discontinued in Phase I clinical trial, because of Four participants experienced increased liver transaminases or acute hepatitis (Amita Datta-Mannan et al., 2012) .Therefore, intensive research is needed to identify potent small molecules with wider safety window that target the IL-17A / / AHEK-Blue assayThe IL-17A / AHEK-Blue assay to assess the potential inhibition effect of compounds on IL-17A / A.The Materials, Reagents, Plates and Instrumentation of the Cell Proliferation Assay as shown in the following Table 1:Table 1Each test compound was initially solubilized in DMSO to achieve a stock solution concentration of 10 mM. This solution was then subjected to a series of threefold serial dilutions, resulting in a sequence of test solutions of the concentrations at 3.33 mM, 1.11 mM, 0.37 mM, 0.12 mM, 0.04 mM, 0.01 mM, 0.004 mM, 0.002 mM, and 0.5 μM, respectively, for subsequent experimental evaluation. The cultivation of cells was conducted in accordance with the protocols stipulated by the technical data sheet for HEK-BlueTM IL-17, as recommended by the manufacturer. The assay of HEK-Blue IL-17 cells was performed during their exponential phase of growth, adhering to optimal cellular activity conditions. A test solution (25 nl) was dispensed into 384-well plates, followed by the seeding of 25 μl of HEK-Blue IL-17 cells with rhIL-17 protein (10 ng
